Controlling Effect of Some Plant Extracts on Pathogenic Fungi and Pest of Rice (몇 가지 식물추출물의 벼 병해충 방제 효과)

  • This study was performed to test the insecticidal and antimicrobial activity of plant extracts from clove, Sophora flavescens Aiton and neem. As the result of antimicrobial activity test, clove extract showed the strongest activity against Botrytis cinerea. In insecticidal activity test, the extract of Sophora flavescens Aiton was the highest against Nilaparvata lugens. The pyroligneous liquor (10%) and emulsified spreader (10%) were added to the extracts of clove and Sophora, respectively, to apply the environment-friendly rice field. In the field treated clove extract, disease damage occurred 49.1% and insect damage occurred 29.5% compared to control plot (100%). In the field treated Sophora extract, disease damage was 56.7% and insect damage was 21.0% compared to control plot (100%). In conclusion, plant extracts from Sophora and clove could control about 50% of disease and about 70% of insect damage that they could be used as environment-friendly resources to control disease and insect in rice farming.

A Cytotoxic Constituent from Sophora flavescens

  • A cytotoxic constituent was isolated by bioassay-guided procedure from the roots of Sophora flavescens Aiton (Leguminosae). The constituent was identified as sophoraflavanone G (I) by means of chemical methods and in comparsion with spectral data of standard compound. The $ED_50$ values of constituent I were 0.78, 1.57, 2.14 and $8.59{\mu}g/ml$ against A549, HeLa, K562 and L1210 cell lines, respectively. Constituent I exhibited highly cytotoxic activities against A549, K562 and HeLa cells, but showed a mild activity $$(ED_50 value, 5{\mu}g/ml)$$ against L1210 cells. Among the tested cell lines, A549 cells were the most sensitive to constituent I.

The Effect of Allergic Inflamation by Sophora Flavescens Aiton Extract Ion Through Inhibition of the $NF{\kappa}B$, JNK and p38 Pathway (고삼(苦蔘)에탄올 추출물이 $NF{\kappa}B$ 및 JNK, p38 조절을 통한 알레르기성 염증에 미치는 영향)

  • 1. Objectives The roots of Sophora flavescens Aiton (SFA) are widely used as a herbal remedy for allergic inflammation. In this study, we invested the effect of SFA on passive cutaneous anaphylaxis reaction and histamin releas and we demonstrated that SFA suppressed the production of pro-inflammatory cytokines, such as tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin- 6 (IL-6), and interleukin -8 (IL-8), through inhibition of the $NF{\kappa}B$, JNK and p38 pathway in the human mast cell line, HMC-1. 2. Methods To accomplish this, we invested passive cutaneous anaphylaxis reaction and histamin release at an animal experiment. In addition, we investigated the effect of SFA on the production of inflammation-related cytokines in HMC-1 cells that were co-treated with PMA and A23187, which can induce production of pro-inflammatory cytokines. 3. Results and Conclusions SFA induced passive cutaneous anaphylaxis reaction and histamin releas and supressed the expression of TNF-${\alpha}$, IL-6, and IL-8. In addition, the protein levels of TNF-${\alpha}$ were also decreased by SFA treatment. Furthermore, SFA inhibited the nuclear translocation of nuclear factor $NF{\kappa}B$ through inhibition of the phosphorylation and degradation of $I{\kappa}B-{\alpha}$, which is an inhibitor of $NF{\kappa}B$. Moreover, SFA also inhibited induction of MAPKs (JNK, p38) and $NF{\kappa}B$ promoter-mediated luciferase activity. Taken together, these results suggest that SFA could be used as a treatment for mast cell-derived allergic inflammatory diseases.

  • Ethanol 75% extracts of Morus alba Linne(Ma) and Sophora flavescens AITON(Sf) were fractionated by chloroform($CHCl_{3}$), ethyl acetate(EtOAc), butanol(BuOH) and water. Each fractionate was added to tryptic soy broth varying in extract levels and 5 strains of Listeria monocytogenes(Lm) were inoculated in them for comparing growth inhibition at $32^{\circ}C$. The $CHCl_{3}$ and EtOAc fraction of Ma were superior in growth inhibition of 5 strains but only $CHCl_{3}$ fraction of Sf showed comparatively higher inhibition to Lm ATCC 19113. Lm ATCC 19111 and 19113 were totally inhibited by 500 ppm of EtOAc fraction and Lm ATCC 19112, 19114 and 15313 were also absolutely inhibited by 1000 ppm of $CHCl_{3}$ fraction of Ma 75% ethanol extract.

Susceptibility of Oral Bacterial to Sophoraflavanone G isolated from the Root of Sophora flavescens

  • Objective: The aim of this work is to investigate the antibacterial activity of the Sohporaflavanone G isolated from Sophora flavescens (S. flavescens), as the development of microbial resistance to antibiotics make it essential to constantly look for new and active compounds effective against pathogenic bacteria. Method : Sophoraflavanone G was isolated from the dried roots of Sophora flavescens Aiton (Leguminosae) by bioassay?guided fractionation. We investigated the effect of sophoraflavanone G on oral bacterial at various concentrations after incubation of 24 h in strains in the dose?dependent manner. Results: The structure of active compound, Sophoraflavanone G having a lavandulyl group at C?8, was elucidated on the basis of spectral data especially 1H?NMR and I3C?NMR. The antimicrobial activity showed that Sophoraflavanone G exhibited antimicrobial activilies against all the bacteria tested (MICs, 0.39 - 6.25 ㎍/ml). Sophoraflavanone G showed the strong antimicrobial activity against all the facultative bacteria and microaerophilic bacteria (MICs, 0.78 - 1.56 ㎍/ml) and also Sophoraflavanone G showed the strong antimicrobial activity against obligate anaerobic bacteria (MICs, 0.39 - 6.25 ㎍/ml).

Antigastritic and Anti Helicobacter pylori of Trifolirhizin from Sophora Radix (고삼 Trifolirhizin의 항위염 및 항 H. pylori 균 효과)

  • Sophorae Radix, the dried roots of Sophora flavescens Aiton (Leguminosae), has been used in oriental traditional medicine for treatment of skin and mucosal ulcers, sores, gastrointestinal hemorrhage, diarrhea, inflammation and arrhythmia. Present study was carried out for the gastroprotective effect of trifolirhizin from Sophora flavescens. This reports were evaluated antibacterial activity against Helicobacter pylori and HCl ethanol-induced gastric lesion in rats and showed the significant effectiveness. In pylorus ligated rats, the treatments of trifolirhizin showed decrease in the volume of gastric secretion and acid output. Also we evaluated the antibacterial activity against H. pylori treated with methanol extract and trifolirhizin from Sophora flavescens had a equivalent antibacterial activity with ampicillin against H. pylori at the dose of $100{\mu}g/mL$. It may be regarded that the antigastritic effects and antibacterial activity of trifolirhizin from Sophora flavescens are originated from reduction of total acid output identified by gastric secretion reduce, free radical scavenging effects and the antibacterial activity against H. pylori.

Induction of Apoptosis in Human Oral Epidermoid Carcinoma Cells by Sophoraflavanone G from Sophora flavescens

  • Sophora flavescens AITON (Leguminosae) is a typical traditional Korean medical herb considered to exhibit antibacterial, anti-inflammatory, and antipyretic effects, and is also used for the treatment of skin and mucosal ulcers, sores, diarrhea, gastrointestinal hemorrhage, arrhythmia, and eczema. In this study, the compound sophoraflavanone G was isolated from the dried roots of S. flavescens by bioassay-guided fractionation. We then investigated the effects of various concentrations of sophoraflavanone G on cell viability and the induction of apoptosis in KB cells after an incubation of 24 hr. The results were determined by the following methods: 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-terazolium bromide (MTT) assay, Hoechst-33258 dye staining, flow cytometry (cell cycle), and Western blotting for caspase-3 and poly (ADP-ribose) polymerase (PARP). We found sophoraflavanone G induced the apoptosis of KB cells in a dose-dependent manner that was verified by DNA fragmentation, apoptotic bodies, the sub-G1 ratio, caspase-3 activity, and cleavage of PARP. These results suggest that sophoraflavanone G has potent anti-proliferative effects on human oral epidermoid carcinoma cells, with the induction of apoptosis.

Effects of Sophorae Radix on skin condition in mice with contact dermatitis induced by Dinitrofluorobenzene (고삼(苦蔘) 추출물이 접촉피부염이 유발된 생쥐의 피부에 미치는 영향)

  • Objectives : The dried root of Sophora flavescens Aiton have been used to treat patient with skin diseases such as eczema in Eastern countries. S. flavescens can clear away heat and dry dampness and purge sthenic-fire from the liver and gallbladder. Recently, anti-allergic effect of S. flavescens has been reported. However, the effect on skin condition of contact dermatitis still remains unknown. Thus, the purpose of this study is to evaluate the efficacy of S. flavescens on contact dermatitis. Methods : In order to investigate the effects of S. flavescens (methanol extract of S. flavescens, MESR) on skin condition, the contact dermatitis was induced in mice skin by using 1-fluoro-2,4-dinitrofluorobenzene (DNFB). As the effective endpoints, skin thickness, skin weight, histopathological change were checked. In addition, effects on skin lesion, body weight and spleen/body weight ratio were also investigated. Results : Topical application of MESR ($500{\mu}g/day$) lowered skin thickness (P < 0.05) and skin weight (P < 0.05), respectively. MESR-treated group showed diminished spongiosis and immune cell infiltration in skin tissues compared to those of non-treated control group. The inhibition of skin lesions was also observed in MESR-treated group. In addition, MESR did not affect body weight gain and spleen/body weight ratio in contrast with those in dexamethasone-treated group. Conclusion : These data suggest that Sophorae Radix could improve skin lesion of contact dermatitis. This indicates the possibility for Sophorae Radix to be used to patients with skin diseases such as contact dermatitis.

  • A new lavandulylated flavonoid, 8-lavandulylkaempferol (1), was isolated from the roots of Sophora f1avescens AITON (Leguminosae). The structure of this compound was determined via spectroscopic analysis. Compound 1 was determined to be a scavenger on both 1, 1-diphenyl­2-picrylhydrazyl radicals and $ONOO^{-}$.

  • We investigated the inhibitory effect of an oral administration of a Sophora flavescens Aiton ethanol extract (SFE) on the development of atopic dermatitis (AD) by using NC/Nga model mice. The induction of atopic dermatitis-like lesion was conducted by the removal of the back hairs and topical application of a mite antigen (Dermatophagoides farinae, Df) on to the back skin twice a week for 8 weeks. SFE was orally administered at a different doses (100-400 mg/kg). Atopic dermatitis-like skin lesions were evaluated by dermatitis scores, skin histology and immunological parameters (serum levels of IgE, TARC/CCL17, MDC/CCL22, and CTACK/CCL27). Oral administration of SFE significantly inhibited the clinical sign of Df-induced atopic dermatitis, including dermatitis score and leukocyte infiltration. Moreover, SFE suppressed significantly the serum IgE and Th2 chemokine (TARC/CCL17, MDC/CCL22, and CTACK/CCL27) levels in a concentration dependent manner. These results suggest that oral administration of SFE could reduce significantly the clinical signs and Th2 chemokines in Df-induced atopic dermatitis model mice. Therefore, SFE may be effective substances for the management of AD in human.
