A spsB Gene Putatively Encoding Glucosyl-Isopreny Phosphate-Transferase in Sphingomonas chungbukensis DJ77 (Sphingomonas chungbukensis DJ77의 Glucosyl-Isoprenyl Phosphate-Transferase를 암호화할 것으로 추정되는 spsB 유런자)

  • Some genes, which are involved in the biosynthesis of polysaccharides, could be found by the genome project of Sphingomonas chungbukensis DJ77. In this study, we identified the complete nucleotide sequence of a gene, encoding the glucosyl-isoprenyl phosphate-transferase, which catalyzes the first step in the biochemical pathway for the synthesis of the sphingan type polysaccharide. This gene, named spsB, is initiated by the ATG codon and terminated by the TGA, and its open reading frame consists of 1392 bp, encoding 463 amino acids. The predicted amino acid sequence of this enzyme indicates $50\%$ similarity to SpsB of Sphingomonas spp S88, also produces sphingan, and $48\%$ to GelB of Sphingomonas paucimobilis ATCC 31461.

Nucleotide Sequence and Secondary Structure of 16S rRNA from Sphingomonas chungbukensis DJ77 (Sphingomonas chungbukensis DJ77의 16S rRNA 염기서열과 이차구조)

  • A 16S ribosomal RNA gene from S. chungbukensis DJ77 has been sequenced. This sequence had a length of 1,502 bp and was extended for 29 bp at 5' and for 37 bp at 3' from the partial sequence (1,435 bp) registered in 2000 year. Besides, 1 bp was newly added near to the 3' end. We made the secondary structure of the 16S rRNA based on E. coli model and found four specific regions. We found constant and variable regions in genus Sphingomonas as the result of multiple alignment of 16S rRNA gene sequences from Sphingomonas spp. and S. chungbukensis DJ77. We found a stem loop structure in S. chungbukensis DJ77, which was only discovered in C. jejuni to date. It showed the structural agreement despite the difference of the sequences from the both organisms. Finally, S. chungbukensis DJ77 belonged to cluster II (Sphingobium) group, after the classification using phylogenetic analysis and nucleotide signature analysis.

Production of Algal Biomass and High-Value Compounds Mediated by Interaction of Microalgal Oocystis sp. KNUA044 and Bacterium Sphingomonas KNU100

  • There is growing interest in the production of microalgae-based, high-value by-products as an emerging green biotechnology. However, a cultivation platform for Oocystis sp. has yet to be established. We therefore examined the effects of bacterial culture additions on the growth and production of valuable compounds of the microalgal strain Oocystis sp. KNUA044, isolated from a locally adapted region in Korea. The strain grew only in the presence of a clear supernatant of Sphingomonas sp. KNU100 culture solution and generated 28.57 mg/l/d of biomass productivity. Protein content (43.9 wt%) was approximately two-fold higher than carbohydrate content (29.4 wt%) and lipid content (13.9 wt%). Oocystis sp. KNUA044 produced the monosaccharide fucose (33 ㎍/mg and 0.94 mg/l/d), reported here for the first time. Fatty acid profiling showed high accumulation (over 60%) of polyunsaturated fatty acids (PUFAs) compared to saturated (29.4%) and monounsaturated fatty acids (9.9%) under the same culture conditions. Of these PUFAs, the algal strain produced the highest concentration of linolenic acid (C18:3 ω3; 40.2%) in the omega-3 family and generated eicosapentaenoic acid (C20:5 ω3; 6.0%), also known as EPA. Based on these results, we suggest that the application of Sphingomonas sp. KNU100 for strain-dependent cultivation of Oocystis sp. KNUA044 holds future promise as a bioprocess capable of increasing algal biomass and high-value bioactive by-products, including fucose and PUFAs such as linolenic acid and EPA.

Antimicrobial Activities of Medicinal Herbs and Seaweeds Extracts Against Microorganisms Isolated from the Rice Warehouses (미곡창고 오염균주에 대한 천연추출물의 항균활성)

  • In order to prevent the quality deterioration of rice from microbial infection, we investigated antimicrobial activities of Morun albalinne, Glycyorrhiza uralensis, Sargassum siliquastrum, and Ecklonin cava against isolated microorganisms from the paddy and warehouses. The major types of the bacteria grown in the paddy and rice warehouses were Sphingomonas paucimobilis, Arthrobacter atrocyaneus, and Bacillus spp. such as Bacillus cereus Additionally Deuteromycetes, Aspergillus spp. and Penicillum spp. were considered as major contaminant microorganisms in the paddy and rice warehouses. As results of the paper disc assay against the isolated microorganisms, the ethanol extracts with Morun albalinne, Glycyorrhiza uralensis, Sargassum siliquastrum, and Ecklonia cava showed effective antimicrobial activities. Especially, Morun albalinne showed the strongest growth inhibition on the isolated bacteria at $0.0025{\sim}0.0075%$.

Biocidal Effects of Chlorine Dioxide on Isolated and Identified Pathogens from Nosocomial Environment - Biochemical and Technical Covergence (병원내 환경으로부터 분리 및 확인된 병원균에 대한 이산화염소의 살균 효과 - 생화학 및 기술 융합)

  • In this study, microorganisms were isolated from nosocomial environment and are identified by biochemical analysis as the part of biochemical and technical convergence. Microorganisms were collected at intense care unit of general hospital located in Pyeongtak (2014.11.28. - 2014. 11. 30). Using a VITEK2 equipment of biochemical approaches, eleven microorganisms e.g., Micrococcus luteus (or M. lylae), Granulicatella adiacens (M. luteus or M. lylae), Staphylococcus caprae, Sphingomonas paucimobilis, Kocuria kristinae, G elegans, Aerococcus viridans (or Staphylococcus arlettae), Methylobacterium spp., Dermacoccus nishinomiyaensis (or Kytococcus sedentarius), Kocuria kristinae (or M. luteus, M. lylae), Pseudomonas oryzihabitans were identified. And then identified bacteria plates were applied with a plastic stick, so called with "FarmeTok (medistick/Puristic) to produce ClO2. ClO2-releasing plastic stick showed the very strong inhibition of bacterial growth with about 99.9%. There were no bacterial colonies on the ClO2-incubated plate. Taken together, it is suggested that chlorine dioxide should be very strong inhibitor to microorganisms of nosocomial infections.

Isolation and Characterization of Marine Bacterium Producing Arylsulfatase

  • A bacterial strain capable of hydrolyzing sulfate ester bonds in p-nitrophenyl sulfate and agar was isolated from the Southeast coast of Korea. The isolated strain (AS6330) is aerobic, Gram-negative, rod-shaped, and motile. Octadecanoic acid was the major cellular fatty acid in the isolate. An almost complete 16S rDNA sequence of the isolate was determined and the sequence similarity of the 16S rDNA with those of known Sphingomonas spp. was found to be at most $96.4\%$, implying that the isolate was a new Sphingomonas species. The organism was grown optimally at NaCl concentration of $1.5-3.5\%$. Optimum culture conditions were determined to be $30^{\circ}C$ and pH 7.0 for 48 h fermentation using a laboratory fermentor under constant culture conditions. Partially purified arylsulfatase through Q-Sepharose and phenyl­Sepharose chromatographies catalyzed hydrolysis of sulfate ester bonds in agar, and $97\%$ of sulfates in agar were removed after 4 h reaction at $45^{\circ}C$ and pH 7.0. The arylsulfatase from the isolated bacterium might be useful for the removal of sulfate groups in agar.

  • 몇몇의 박테리아들은 바이오필름을 형성하여 그들 스스로를 보호한다. 하지만 바이오필름으로 인해 악취와 질병 등의 문제가 많이 발생되고 있기 때문에 바이오필름을 형성하는 박테리아의 성장을 효율적으로 억제하기 위해 은 나노, 구리 나노입자들이 포함된 다양한 나노스케일의 재료들에 대한 연구가 활발히 진행되어오고 있다. 이들 연구의 주된 목표는 체내에서 독성은 나타내지 않으면서 항균성을 증가시키는 것에 있다. 특히, 구형으로 이루어진 나노입자와 높은 종횡비를 가지는 탄소나노튜브와 같이 차원이 다른 나노물질들의 복합체들은 세포독성을 최소화하면서 특정 박테리아에 대한 항균성을 향상시킬지도 모른다. 이번 연구에서는, 산 처리된 탄소나노튜브에 화학적인 방법을 이용하여 구리 이온을 각각 환원시켜 구리 나노-탄소나노튜브 복합체를 합성하였다. 이들 복합체는 transmission electron microscopy, X-ray diffractometry, energy-dispersive X-ray spectroscopy 를 이용하여 특성이 분석되었고 Methylobacterium spp., Sphingomonas spp. 와 E. coli 에 대하여 항균성이 평가되었다. 추가적으로 구리 나노-탄소나노튜브 복합체는 human fibroblast cells 에 대하여 세포독성이 평가되었고 제작된 마이크로칩 안에 형성된 바이오필름의 성장억제효과가 평가되었다. 결과적으로, 구리 나노-탄소나노튜브 복합체에서 바이오필름을 형성하는 Methylobacterium spp. 에 대하여 특이적으로 항균성을 나타냈으며 바이오필름이 형성된 마이크로칩에서 바이오필름을 제거 하는 것이 확인되었다.

Temporal Changes of Fungal and Bacterial Populations in Rice under Indoor Storage Conditions

  • This research was conducted to evaluate fungal and bacterial populations in unhulled and brown rice under indoor storage conditions, and to examine the relationship between microbial populations and environmental conditions such as temperature and relative humidity. The temperature and relative humidity of the storage room ranged from $22.6^{\circ}C\;to\;27.0^{\circ}C$ and 23.3% to 44.2%, respectively. Total fungal and bacterial populations remained relatively stable over the storage period. Predominant fungi included Aspergillus candidus, A. flavus, A. fumigatus, and Penicillium spp.; the predominant bacteria were Bacillus, Microbacterium, Sphingomonas, and Methylobacterium spp. Total fungi and bacteria were not significantly correlated with either unhulled (r=0.448, P=0.372) or brown (r=0.466, P=0.351) rice. In unhulled rice, total fungi showed positive correlations with total Aspergillus (r=0.994, P<0.001) and total Penicillium (r=0.906, P<0.05); A. flavus was positively correlated with total Aspergillus (r=0.913, P<0.05) and total fungi (r=0.868, P<0.05). In brown rice, Bacillus spp. was also positively correlated with total bacteria (r=0.998, P<0.001). Mean temperature was negatively correlated with A. candidus (r=-0.852, P<0.05) and total fungi (r=-0.961, P<0.01), and mean relative humidity was positively correlated with total Penicillium spp.(r=0.884, P<0.05) in brown rice. Hence these results could provide basic information on the fungal and bacterial populations in unhulled and brown rice stored under room conditions, and on the effect of environmental conditions on the populations of fungi and bacteria, especially Aspergillus and Penicillium spp.

Bacterial contaminants in extended boar semen and selection of effective antimicrobials (돼지 희석정액의 세균오염도 및 유효 항생제 선발)

  • Bacterial contamination is an unavoidable finding of the semen collection process in boar and can lead in deleterious effects on semen quality and longevity if left uncontrolled. The purpose of this study is to identify the bacteria in extended boar semen and to select the effective antimicrobials to control of the contaminants. Of 116 extended boar semen samples submitted from eight AI centers in Korea, 39 (33.6%) samples were positive for bacterial contamination. Among 39 contaminated semen, most of them (84.6%) were contaminated with one or two bacterial species and there was no significant difference between two age groups $(\leq\;24\;and\;>\;24\;month\;old).$ Stenotrophomonas maltophilia (n = 18) was the most predominant bacterium followed by Elizabethkingia meningoseptica (n = 12), Sphingomonas paucimobilis (n = 12), Myroides spp. (n = 5), Ochrobactrum anthropi (n = 3), and so on. Enrofloxacin (72.9%), florfenicol (72.9%), bacitracin (49.2%) and tylosin (49.2%) showed higher sensitivity compared with penicillin (13.6%) or aminoglycosides (6.8%-18.6%). Brucella spp., Leptospira spp., Mycoplasma hyopneumoniae, Mycoplasma hyorhinis, Mycobacterium tuberculosis complex were not detected in semen by PCR.

Distribution and Characteristics of Heterotrophic Plate Count Bacteria in Water Samples from Drinking Water Dispensers (냉온수기에서 일반세균의 분포 및 분리한 세균의 특성)

  • To evaluate bacteriological water quality, samples were taken from drinking water dispensers placed at S company (S-C) and U highschool (U-H) in Ulsan. The medians of heterotrophic plate counts (HPCs) were 53 CFU/ml for the 74 water samples of S-C and 80 CFU/ml for the 36 cold water samples of U-H, and 38% of the S-C and 42% of the U-H samples showed HPC bacterial concentrations higher than 100 CFU/ml. Coliform bacteria were detected from one sample of S-C. To determine the major source of bacterial contamination, water samples were taken daily for $6\sim8$ days from the bottled water containers as well as the faucets of an experimental water dispenser. While the average HPCs in the bottled water containers were 33 CFU/ml for the first and 132 CFU/ml for the 2nd analysis, the HPC concentration in the cold water samples was 1,022 CFU/ml for the 2nd analysis. These results suggest that the majority of bacteria detected in the cold water samples were originated from the biofilms on the surface of water passages within the water dispensers. There was no significant increase in HPC bacterial concentrations within the bottled water container after installation on the water dispenser. We could isolate and tentatively identify 3 genera 6 species of Gram-positive and 7 genera 7 species of Gram-negative bacteria from the plate count agar plates of U-H samples. Among the isolates, 72% were observed as Gram-positive, and Micrococcus spp. was the most abundant with 54% of the total, followed by Sphingomonas paucimobilis with 16%. It appears that most of the HPC bacteria detected in water dispensers originate from indoor airborne bacteria, which may play important roles in the formation of biofilms on the surface of water passages within the water dispensers.
