This study was conducted to determine the effect of pupal development and adult longevity of Spodoptera litura larvae reared on leaves of different leguminos plants of 11 varieties or cultivars. Pupal duration varied from 8.7 to 9.5 days depending upon different diets and duration of female pupa (8.3~9.1 days) was about 1 day shorter than that of male's (9.4~10.1 days). Prepupal an postpupal duration were 1.7 to 2.0 days and 6.7 to 7.7 days, respectively. Pupal weight ranged from 0.23g to 0.43g for female and 0.24g to 0.35 g for male. Percent change of pupal weight was lowest (11%) on geomjeongkong-1 and highest (16%) on bukwangkong. Adult longevity was in ranging from 7.8 days on dongbu to 11.2 days on bukwangkong and tended to become slightly longer in female than tin male. Significant relationships were recognized between moisture content of feeding leaf, the pupal duration and weight and adult longevity.
This experiment was conducted to investigate pathogenicity of Spodoptera litura nucleopolyhedrovirus (SINPV) with different temperatures to mass production. In laboratory, LC$_{50}$ values of SINPV were 5.534$\times$10$^3$ PIBs/ml and 4.l0$\times$10$^2$PIBs/ml in 1st instar larvae at 2$0^{\circ}C$ and 28$^{\circ}C$, respectively, but those were increased at 32$^{\circ}C$. LC$_{50}$ at 3rd and 5th instar larvae were showed the lowest value at 28$^{\circ}C$. LT$_{50}$ values of SINPV in 1.0$\times$10$^{5.7}$ PIBs/ml were determined at various temperature conditions (20-32$^{\circ}C$). The result showed that treatments at 28$^{\circ}C$ and 32$^{\circ}C$ shortened LT$_{50}$ values, but treatments at 2$0^{\circ}C$ and 24$^{\circ}C$ was relatively longer. Also LT$_{50}$ values was shortened in high concentration and young larva. LT$_{50}$ values are dependent on the temperature, viral concentration and larval instar.

Insect lysozymes are basic, cationic proteins synthesized in fat body and hemocytes in response to bacterial infections and depolymerize the bacterial cell wall. The c-type lysozyme of the insect Spodoptera litura (SLLyz) is a single polypeptide chain of 121 residues with four disulfide bridges and 17 rare codons and is approximately 15 kDa. The full-length SLLyz cDNA is 1039 bp long with a poly(A) tail, and contains an open reading frame of 426 bp long (including the termination codon), flanked by a 54 bp long 5' UTR and a 559 bp long 3' UTR. As a host for the production of high-level recombinant proteins, E. coli is used most commonly because of its low cost and short generation time. However, the soluble expression of heterologous proteins in E. coli is not trivial, especially for disulfide-bonded proteins. In order to prevent inclusion body formation, GST was selected as a fusion partner to enhance the solubility of recombinant protein, and fused to the amplified products encoding mature SLLyz. The expression vector pGEX-4T-1/rSLLyz was then transformed into E. coli BL21(DE3)pLysS for soluble expression of rSLLyz, and the soluble fusion protein was purified successfully. Inhibition zone assay demonstrated that rSLLyz showed antibacterial activity against B. megaterium. These results demonstrate that the GST fusion expression system in E. coli described in this study is efficient and inexpensive in producing a disulfide-bonded rSLLyz in soluble, active form, and suggest that the insect lysozyme is an interesting system for future structural and functional studies.
This experiment was conducted to improve activity of Spodoptera litura Nucleopoly-hedrovirus (SINPV) combined with organic and functional matters. In combination of SINPV mixed with organic matters, LT$_{50}$ values of SINPV 1.0${\times}$10$^{5}$ PIBs/$m\ell$ combined with boric acid of 2,000 ppm were 4.5 days. It was 1.5 days shorter than SINPV 1.0${\times}$10$^{5}$ PIBs/$m\ell$ alone. The body weight of larva infected with SINPV 1.0${\times}$10$^{5}$ PIBs/$m\ell$ combined with boric acid of 2,000ppm was not increased, and S. litura was completely dead in 7 days after treatment. It suggested that addition of boric acid in SINPV application enhanced the pathogenicity against S. litura larvae. In laboratory experiment of combination of SINPV with functional matters, LT$_{50}$values of SINPV 1.0${\times}$10$^4$ PIBs/$m\ell$ alone were 7.3 days, but those of SINPV 1.0${\times}$10$^4$ PIBs/$m\ell$ with electrolyzed oxidizing water, pyroligneous liquor or kitosan were 10.4, 9.3 and 11.2 days, respectively. In the laboratory study, the toxicities of chlorpyrifos-methyl, chlorpyrifos, etofenprox, etofenprox+PAF' and deltamethrin for different larval instars of S. litura decreased significantly as larvae aged. LCso values for chlorpyrifos- methyl and chlorpyrifos increased significantly from 3rd instar larvae, while those for etofenprox+PAP and deltamethrin-increased from 2nd instar larvae. In pot study, no significant differences in control efficacy were observed among each treated plots and over 90% control efficacy at 5 d after treatment was obtained in all of the treated plots, except treatment with deltamethrin. Therefore four insecticides excluding deltamethrin will be effective for controlling S. litura. However, it will be probably important to select appropriate insecticides and decide a proper time of treatment because the developmental stage is a significant factor in deciding insecticide efficacy because various developmental stages of the tobacco cutworm inhibit in fields.

Bacillus thuringiensis with selected high toxicities against tobacco cutworm, Spodoptera litura were isolated from domestic soils. When being observed under a phase-contrast microscope, the insecticidal crystal proteins were showed a bipyramidal crystal types. New CAB 109 isolate was identified to B. thuringiensis subsp. aizawai in the H serotype. As a results of insecticidal activities between CAB 109 isolate and 3 existing ready-made products against 3rd larva of S. litura, CAB 109 isolate showed 100% mortality with spore concentration $(1.3{\times}10^7cfu/ml)$. It was a very high insecticidal activity compared with a existing ready-made B. t. products. $LD_{50}$ values of CAB 109 isolate was $9.78{\times}10^5,\;6.87{\times}10^6\;and\;1.83{\times}10^7cfu/ml$ spore concentration against 2nd, 3rd and 4th larva of S. litura, respectively. Unlike Plutella xylostella, S. litura was slowly died after application up to 7 days. The weight of S. litura larva applied with CAB 109 isolate were 6-7 times less than controlled group. Even though it didn't die, it did not grow into next larva. The result observed with scanning electron microscope was that CAB 109 isolate of B. t. aizawai formed a typical bipyramidal crystal protein type. Otherwise, when CAB 109 isolate was examined with SDS-PAGE and with trypsin, there was no difference between CAB 109 strain and ready-made products of B. thuringiensis.
Midgut tissues from 4 insect specIes were exammed for the activity of cytochrome P-450 monooxygenases, a major enzyme involved in chemical detoxification. When Helicoverpa assulta larvae were reared on an artificial d;et, the specific activity of the midgut cytochrome P-450 monooxygenases (MFO) was :3 times higher than that of the fat body, The specific activity of the midgut cytochrome P-450 monooxygenases was higher in H. assul/a larvae when reared on Nicotiana tabacum leaves than when on CapsIcum annuum fruits or an artificial diet. In the case of Hyphantria cunea larvae, Tilia megaphyllo leaves were the best in inducing midgut cytochrome P-450 monooxygenases activity. When larvae of H. assulta, Spodoptera exigua, H. cunea and Spodoptera litura were reared on their own artificial diet, the highest activity was seen in S. exigua larvae which is a polyphagous and insecticide-resistant strain.
