Camelina (Camelina sativa L.) is a potential bio-energy crop that has short life cycle about 90 days and contains high amount of unsaturated fatty acid which is adequate to bio-diesel production. Enhancing environmental stress tolerance is a main issue to increase not only crop productivity but also big mass production. CsRCI2s (Rare Cold Inducible 2) are cold and salt stress related protein that localized at plasma membrane (PM) and assume to be membrane potential regulation factor. These proteins can be divide into C-terminal tail (CsRCI2D/E/F/G) or no-tail group (CsRCI2A/B/C/H). However, function of CsRCI2s are less understood. In this study, physiological responses and functional characterization of CsRCI2s of Camelina under salt stress were analyzed. Full-length CsRCI2s (A/B/E/F) and CsPIP2;1 sequences were confirmed from Camelina genome browser. Physiological investigations were carried out using one- or four-week-old Camelina under NaCl stress with dose and time dependent manner. Transcriptional changes of CsRCI2A/B/E/F and CsPIP2;1 were determined using qRT-PCR in one-week-old Camelina seedlings treated with NaCl. Translational changes of CsRCI2E and CsPIP2;1 were confirmed with western-blot using the antibodies. Water transport activity and membrane potential measurement were observed by cRNA injected Xenopus laevis oocyte. As results, root growth rate and physiological parameters such as stomatal conductance, chlorophyll fluorescence, and electrolyte leakage showed significant inhibition in 100 and 150 mM NaCl. Transcriptional level of CsPIP2;1 did not changed but CsRCI2s were significantly increased by NaCl concentration, however, no-tail type CsRCI2A and CsRCI2B increased earlier than tail type CsRCI2E and CsRCI2F. Translational changes of CsPIP2;1 was constitutively maintained under NaCl stress. But, accumulation of CsRCI2E significantly increased by NaCl stress. CsPIP2;1 and CsRCI2A/B/E/F co-expressed Xenopus laevis oocyte showed decreased water transport activity as 61.84, 60.30, 62.91 and 76.51 % at CsRCI2A, CsRCI2B, CsRCI2E and CsRCI2F co-expression when compare with single expression of CsPIP2;1, respectively. Moreover, oocyte membrane potential was significantly hyperpolarized by co-expression of CsRCI2s. However, higher hyperpolarized level was observed in tail-type CsRCI2E and CsRCI2F than others, especially, CsRCI2E showed highest level. It means transport of $Na^+$ ion into cell is negatively regulated by expression of CsRCI2s, and, function of C-terminal tail is might be related with $Na^+$ ion influx. In conclusion, accumulation of NaCl-induced CsRCI2 proteins are related with $Na^+$ ion exclusion and prevent water loss by CsPIP2;1 under NaCl stress.

This study is designed to investigate cytoprotective effects of Platycodon grandiflorus (Jacq.) A.DC on C6 glioma cell apoptosis by oxidative stress. Experimental C6 glioma cells were classified into four groups as follows: normal group, PGE group, chemical groups, PGE+chemical groups. Oxidative stress that caused by chemicals in the C6 glioma cell, check the impact to Chemical group was administered normal group. Apoptotic effect protecting in order to observe the chemical group was administered PGE. We to observe effects of PGE on SOD inhibition, total glutathione production in C6 glioma cells were administered PGE. In case of administration PGE, apoptosis induced by Paraquat was significantly decreased. In case of administration PGE, apoptosis induced by SNP was significantly decreased. In case of administration PGE, apoptosis induced by $H_2O_2$ was significantly decreased. In case of administration PGE, apoptosis induced by Rotenone was decreased, but the statistical significance was not. In case of administration PGE, SOD inhibition activities significantly decreased. In case of administration PGE, Total glutathione did not affect the content. These results suggest that PGE is able to treat a disease caused by oxidative stress and prevent a aging. These results suggest that PGE is a disease caused by oxidative stress and aging, the prevention and treatment of food shall be able to be applied.
Caffeine has both positive and negative effects on physiological functions in a dose-dependent manner. C. elegans has been used as an animal model to investigate the effects of caffeine on development. Caffeine treatment at a high dose (30 mM) showed detrimental effects and caused early larval arrest. We performed a comparative proteomic analysis to investigate the mode of action of high-dose caffeine treatment in C. elegans and found that the stress response proteins, heat shock protein (HSP)-4 (endoplasmic reticulum [ER] chaperone), HSP-6 (mitochondrial chaperone), and HSP-16 (cytosolic chaperone), were induced and their expression was regulated at the transcriptional level. These findings suggest that high-dose caffeine intake causes a strong stress response and activates all three stress-response pathways in the worms, including the ER-, mitochondrial-, and cytosolic pathways. RNA interference of each hsp gene or in triple combination retarded growth. In addition, caffeine treatment stimulated a food-avoidance behavior (aversion phenotype), which was enhanced by RNAi depletion of the hsp-4 gene. Therefore, up-regulation of hsp genes after caffeine treatment appeared to be the major responses to alleviate stress and protect against developmental arrest.
The aim of this study was to understand the effects of skin pigmentation on physiological parameters of thermoregulation and grazing behaviour of dairy goats in a hot and humid climate. The study used 26 crossbred Saanen yearling goats (95% Saanen+5% Local Hair Breed). The animals were raised at semi-intensive private farms in Adana (36 59'N, 35 18'E). Groups were selected 2 d before the start of observations. Goats were categorized as predominantly pigmented (P) skin and unpigmented (UP) skin. All observations and measurements were collected on grassland during the grazing period of June and July 2007 (60 d). Air temperature and relative humidity were recorded at 10 min intervals by a portable data logger. The physiological data (rectal temperature, respiration and pulse rate, and skin temperatures from head and udder) were recorded twice weekly in the morning (07:00-08:00); at midday (13:00-14:00); and in the evening (18:00-19:00). Additionally, the activity of the animals was observed and classified (eating, ruminating, drinking, standing, walking, lying) for 12 h during the day twice weekly, using a portable camera system linked directly to a computer. Panting behaviour was also observed. According to the THI values, the experimental goats were subjected to stressful conditions. The pigmented goats had significantly lower rectal temperatures (39.68 vs. $29.89^{\circ}C$), pulse rate (74.08 vs. 84.10 beat/min) and respiration rate (65.65 vs. 88.23 breath/min.) compared with unpigmented goats at midday when the THI exceeded 92. The rectal, head and udder temperatures, pulse and respiration rates of the non-pigmented group exceeded $40^{\circ}C$, $37^{\circ}C$, $37.5^{\circ}C$, 84 beats/min and 78 breaths/min, respectively. Higher activity was observed among pigmented compared with unpigmented goats. Unpigmented goats grazed (4.3 vs. 5.6 h), ruminated (2.0 vs. 2.4 h), and stood (0.8 vs. 1.2 h) less, but lay down (2.2 vs. 1.8 h) more than pigmented goats. The data obtained in this experiment support the hypothesis that unpigmented goats are more adversely affected by climatic stress, likely due to their decreased activity and increased water consumption, as demonstrated by previous studies.

To evaluate the thermal environments and the workload of farmers in the rice field in summer, this study investigated rice farmers' physiological, psychological responses, work postures, work clothes, air temperature and air humidity during the spraying pesticide in the rice field. Five career farmers (3 males, 2 females) volunteered as the subjects. During the spraying pesticide in the rice field, physiological responses were monitored continuously. The results were as follows. l. Farmers wore only raincoats not pesticide-proof clothing. 2. The value of WBGT, rectal temperature($T_{re}$), mean skin temperature(${\={T}}_{sk}$) were $24.9∼28.9^{\circ}C,\;37.8({\pm}0.3)^{\circ}C\;and\;33.6({\pm}0.6)^{\circ}C$, respectively. Clothing microclimate temperature($T_{cl}$) on the chest and back were $32.5({\pm}2.6)^{\circ}C\;and\;33.6({\pm}2.6)^{\circ}C$, respectively(p<0.00l). Humidity inside of the clothing ($H_{cl}$) was over 80%RH and heart rate(HR) was 112(${\pm}27$)bpm. We evaluated that the spraying pesticide was 'heavy work' by the Tre and HR. To four subjective questionnaires, all farmers expressed 'hard, hot, humid and uncomfortable' without individual difference at the end of works. We suggested that 1) the spraying pesticide in the rice field was a heavy work, 2) because the workload of farmers in the raincoat/pesticide-proof clothing can't be evaluated by only WBGT, assessors should measure physiological, psychological responses as well as thermal environments, 3) to alleviate farmers' heat strain, clothing manufacturers must consider not only the improvement of textile materials and clothing weight but also the designing of personal cooling equipment.
This study was conducted to determine the extent of drought resistance based on physiological responses of Calystegia soldanella under water deficit. In order to investigate the changes of plant growth, stomatal density, photosynthesis, chlorophyll fluorescence, the contents of chlorophyll and carotenoid, osmolality, total ion contents, the contents of carbohydrate and proline, C. soldanella was grown under well watered and drought stressed conditions for 12 days. In this study, water-deficit resulted in remarkable growth inhibition of C. soldanella. The effect of water-deficit on plant growth was associated with low osmotic potential of soil. On day 12 after drought treatment, dry weight, relative water contents, number and area of leaves and stem length were lower than those of control. The stomatal conductance and net photosynthetic rate were significantly reduced in water stressed plant to regulate inner water contents and $CO_2$ exchange through the stomatal pore. Chlorophyll fluorescence and chlorophyll contents were not different in comparison with the control, indicating that the efficiency of photosystem II was not affected by drought stress. This results could be explained that water-deficit in C. soldanella limits the photosynthetic rate and reduces the plant's ability to convert energy to biomass. A significant increase in total ion contents and osmolality was observed on day 7 and day 12. Accumulation of proline in leaves is associated with the osmotic adjustment in C. soldanella to soil water-deficit. Consequently, this increase in osmolality in water stressed plant can be a result in the increase of ion contents and proline.
Coenzyme $Q_{10}$ ($CoQ_{10}$) production from Rhizobium radiobacter T6102 was monitored under various oxygen supply conditions by controlling the agitation speeds, aeration rates, and dissolved oxygen levels. As the results, the $CoQ_{10}$ production was enhanced by limited oxygen supply. To investigate whether the $CoQ_{10}$ production is associated with its physiological functions of electron carrier and antioxidant, the effects of sodium azide and hydrogen peroxide on the $CoQ_{10}$ production were studied, showing that the $CoQ_{10}$ contents were slightly enhanced with increasing sodium azide (up to 0.4 mM) and hydrogen peroxide (up to $10\;{\mu}M$) concentrations. These results suggest the plausible mechanism where the limited electron transfer stimulating the environments of limited oxygen supply and oxidative stress could accumulate the $CoQ_{10}$, providing the relationship between the $CoQ_{10}$ physiological functions and its regulation system.
Cold stress influence plant growth through a wide range of growth characters. Adverse effects of low temperature to plant growth come from results of colligative and complex physiological responses to cold stress. To evaluate more exactly cold tolerance of crop plant, it is needed to observe physiological changes induced by cold stress and to analyze relationships between intraspecific variations in physiological factors related to cold tolerance and the extent of cold tolerance in the field. Therefore, the composition and unsaturation ratio of fatty acids in phospholipid, a constituent of membrane, the transition-temperature in respiratory activity of mitochodria, the chlorophyll fluorescence as a factor related to photosynthesis were investigated in rice plant and data on these factors were compared with the degree of cold tolerance obtained in the field experiment. Also, effects of hardening and Mn++ treatment were evaluated as a method to reduce chilling injuries. The unsaturation ratio of fatty acids, whether rice plants were grown in a natural condition or under the chilling stress, was higher in the cold- tolerant varieties and was significantly correlated with the degree of cold tolerance (1-9) observed in the field experiment. And it was also increased by chilling treatment or hardening treatment, due to a reduction in palmitic acid content and an increase in linolenic acid content. The transition-temperature of respiratory activity of mitochodria isolated from etiolated rice seedlings ($25^{\circ}C$, two week-grown in the dark), was correlated with the degree of cold tolerance in the field, cold -tolerant varieties showing a lower transition-temperature. It was not influenced by growth stages. The intensity of chlorophyll fluorescence was highly correlated with the degree of cold tolerance, cold-tolerant varieties having a higher fluorescence intensity. By foliar application of Mn, the transition-temperature of respiratory activity was lowered as much as 0-2$^{\circ}C$ in all tested varieties. Soil application of Mn induced more significant effect in cold-susceptible varieties with a possibility of reducing chilling injuries. On the whole, there were high correlationships among the degree of cold tolerance, the unsaturation ratio of fatty acids in phospholipid, the transition- temperature of respiratory activity and chlorophyll fluorescence except for a few varieties. The transition- temperature of respiratory activity appeared to be negatively correlated with the unsaturation ratio of fatty acids. and the chlorophyll fluorescence to be positively correlated with the unsaturation ratio. This implies that these physical and physiological factors were very closely related to cold tolerance and can be used as an effective index of the evaluation of cold tolerance of crop plant. But other factors as well as three factors discussed above are needed to be considered colligatively and altogether with a systematic analysis for the more exact evaluation of cold tolerance. in rice cultivars. in rice cultivars.
To find the preliminary environmental conditions for a short-time transport of living olive flounder, Paralichthys olivaceus, the stress response and physiological metabolic activity of the cultured fish to feed deprivation and slow temperature descending ($15.8^{\circ}C{\rightarrow}13.3^{\circ}C$) were monitored for 8 days. The monitored variables were the plasma alanine aminotransferase (ALT), aspartate aminotransferase (AST), glucose (GLU), total protein (TP), electrolytes ($Na^+$, $K^+$, $Cl^-$) and thyroid hormones ($TT_4$, $TT_3$, $FT_4$ and $FT_3$). In food deprivation experiment for 8 days, we did not find any statistical change of level in AST, ALT and electrolytes ($Na^+$, $K^+$, $Cl^-$), but found a significant decrease in TP and GLU. In thyroid hormones, the levels of four hormones in plasma were all showing a tendency to decrease. Especially, $FT_4$ and $TT_3$ were significantly decreased, indicating a withering of physiologic activity. In the temperature test, although no any significant change in AST, TP and electrolytes ($Na^+$, $K^+$, $Cl^-$), we observed a significant decrease of ALT and GLU following to temperature descending from $15.8^{\circ}C$ to $13.3^{\circ}C$ (P<0.05). In the levels of thyroid hormones, any significant change was not observed for experimental period. We conclude that the stress response and physiological activity of olive flounder were more influenced by feed deprivation than slow temperature descending at a transport of living fish, and plasma GLU appears to be sensitive factor to physiological metabolic activity, indicating that it could be used as a monitering mark or index for a health inspection of the fish.

In this study, we investigated the effects of light spectra on physiology stress and DNA damage in juvenile rock bream (Oplegnathus fasciatus) using light-emitting diodes (LEDs; green, 520 nm; red, 630 nm) at two intensities (0.25 and $0.5W/m^2$ ) with application of thermal stress (25 and $30^{\circ}C$). We measured the mRNA expression of heat shock protein 70 (HSP70) and the levels of plasma cortisol, glucose, aspartate aminotransferase (AspAT), and alanine aminotransferase (AlaAT). Additionally, DNA damage was measured using comet assays. Our findings showed that HSP70 mRNA expression and plasma cortisol, glucose, AspAT, and AlaAT levels were significantly higher after exposure to high temperatures and were significantly lower after exposure to green LED light. Thus, although high water temperatures induced stress in juvenile rock bream, green LED light inhibited stress. In particular, green LED light reduced stress and DNA damage to a greater degree than other light sources.
