Drect Interaction of Streptozotocin with TBA (thiobarbituric acid) in Lipid Peroxidation Analysis (Streptozotocin에 의한 lipid peroxidation 측정시 TBA법의 적합성에 관한 연구)

  • This study was conducted to evaluate the usage of TBA method for the analysis of lipid peroxidation induced by streptozotocin. 5 mM streptozotocin and 1 % TBA alone showed the maximum peak at 309 nm and 358 nm respectively, although no peak was observed at 532 nm which was the wavelength to determine the absorbance for TBA method. When 5 mM streptozotocin was mixed together with 1 % TBA in vitro, new peaks at 439 nm and 532 nm had been detected, suggesting TBA did interact directly with streptozotocin forming new colored products. These results suggest that TBA method is not adequate for determination of lipid lperoxidation induced by streptozotocin.

High Performance Liquid Chromatography (HPLC) Detection of Malonaldehydethiobarbituric Acid (MA-TBA) Complex in Ground Pork

  • For monitoring lipid oxidation development in cooked ground pork during refrigerationm, malonaldehydethiobarbituric acid(MA-TBA) contents were measured using high performance liquid chromatography(HPLC). As the oxidation proceeded during refergeration, TBA-reaction substances(TBARS) absorbances increased and the corresponding HPLC peak areas also increased proportationately. The correlation coefficient between the HPLC peak areas and MA-TBA absorbance were 0.9979. The treatemtn of cetrimide, an ion pairing agent, gave a complete resolution of the MA-TBA complex and the butanol extraction of the complex increased its recovery by 37.8%. Both cetrimide treatment and butanol extraction are essential steps for analyzing MA-TBA complex in ground pork wiht HPLC. A reliable and specific measurement of NA-TBA in ground pork was successfully performed using HPLC.

  • A new microassay was proposed for the determination of serum thiobarbituric acid(TBA) value greatly enhanced by ferric ion under anaerobic condition. One ${\mu}mole$ of $FeCl_{3}$ per $10{\mu}l$ of serum was added to the TBA reaction mixture containing serum protein precipitate. The reaction mixture was heated on boiling water-bath for 50min under $N_{2}$ flushing. The sensitivity of this assay was greatly enhanced by 40 times comparing with that of Yagi's method (1976). In favour of the enhancement, this test could be measured by colorimetry or spectrophotometry with the sample size of $10-20{\mu}l$ serum. The sensitivity and reproducibility were also improved by means of partial dehydration of the butanol extract with $(NH_{4})_{2}SO_{4}$ salting-out. Serum TBA values of healthy human at different age groups were determined by this proposed method.

  • 동결기간과 해동후 재냉장이 우육의 지방산 조성. TBA가 생육향 및 가열육향에 미치는 영향을 파악하기 위하여 홀스타인의 등심부위를 -2$0^{\circ}C$에서 6, 12 및 18개월 동경, 해동한 것과 이것을 1$^{\circ}C$에서 3일간 재냉장한 것에 대하여 실험하였다. 신선육의 총지질 중성지질 및 인지질의 단일 불포화지방산에 대한 포화지방산의 비율(MUFA/SFA)은 각각 1.35, 1.10 및 1.34이었으며 동결기간이 길어지면서 점점 감소하였다. 동결 6개월째 우육은 TBA가는 0.37 mg/MA/kg으로 신선육의 0.15 mg/MA.kg 보다 높아졌으나 산패취는 느끼지 못했다. 동결 12개월과 18개월째 우육의 TBA가는 각각 0.53과 0.64mg/MA/kg이었으며 산패취를 느낄수 있었다. 한편 재냉장한 우육은 동결기간에 관계없이 TBA가가 높아지고 생육향 평가 점수가 낮아졌으나 MUFA/SFA와 가열육향은 동결 6개월째에 향상되었고 12개월 및 18개월 째에는 저하되어 동결기간에 따라 재냉장 효과의 차이를 보였다.

  • Various concentrations of malonaldehyde (MA) produced upon hydrolysis of 1, 1, 3,3-tetraethoxypropane (TEP) were reacted with 2-thiobarbituric acid (TBA)and th e contents of MA-TBA complex were measured both with spectrophotometer and high performance liquid chromatography (HPLC). As the concentrations of MA-TBA increased, their absorbances and the corresponding HPLC peak areas increased. The correlation coefficient between absorbances and HPLC peak areas of MA-TBA peaks from the other compounds and butanol extraction of the complex increased its recovery its recovery by 29.4% . Measurement of the content of MA-TBA complex for monitoring the development of lipid oxidation was proven to be successful with the use of high performance liquid chromatography.

  • Acrylonitrile(AN) was solution-polymerized in dimethyl sulfoxide(DMSO) and tertiary butyl alcohol(TBA) at 30, 40, $50^\circ{C}$ using a low temperature initiator, 2,2'-azobis(2,4-dimethylvaleronitrile) (ADMVN). The low temperature polymerization using ADMVN, DMSO, and TBA is to be successful in obtaining high molecular weight polyacrylonitrile(PAN) with less branches by solution polymerization. Throug a polymerization of AN in DMSO at $30^\circ{C}$, PAN having viscosity-average molecular weight$(M_v)$ of 931,000 was obtained. And then, during AN solution polymerization in DMSO and TBA using a cosolvent system the in-situ formation of microfibrillar structure has been discovered at the cosolvent composition of 24/1$(V_{DMSO}/V_{TBA})$. The simultaneous process of gelation and phase separation of long chain molecules may explain the in-situ formation of PAN fibers during polymerization.

Synthesis of High Molecular Weight Poly(vinyl alcohol) by Low Temperature Polymerization of Vinyl Acetate in Tertiary Butyl Alcohol and the Following Saponification (아세트산비닐의 삼차부틸알코올계 저온 중합 및 비누화에 의한 고분자량 폴리비닐알코올의 합성)

  • Vinyl acetate (VAc) was polymerized at 30, 40, and 5$0^{\circ}C$ using 2,2'-azobis (2,4-dimethylvaleronitrile) (ADMVN) and tertiary butyl alcohol (TBA) as the initiator and the solvent, respectively. High molecular weight (HMW) atactic poly(vinyl alcohol) (PVA) was prepared by saponifying the poly(vinyl acetate) (PVAc) synthesized. The effect of polymerization conditions were investigated in terms of conversion, degree of branching for acetyl group of PVAc, and molecular weight of both PVAc and PVA. The polymerization rate of VAc in TBA was proportional to the 0.49th power of ADMVN concentration in good accordance with the theoretical value of 0.5. HMW-PVA with high yield could be obtained successfully, probably due to lower polymerization temperature and decreased chain transfer reaction rate which was achieved by adopting ADMVN and TBA. PYAc having average degree of polymerization (P$_{n}$) of 10000~13000 was obtained at the conversion of 35~70%. Saponification of so prepared PVAc yielded PVA having P$_{n}$ of 2400~6100. The syndiotactic diad content increased with decreasing polymerization temperature and increasing VAc concentration due to a steric hindrance effect of TBA during polymerization.

Effect of Exposure Concentration and Time of Fuel Additives on the Indigenous Microbial Community in Forests (산림 토착 미생물 군집에 미치는 유류 첨가제 노출 농도 및 시간의 영향)

  • The toxicity of methyl tert-butyl ether (MTBE), tert-butyl alcohol (TBA) and formaldehyde (FA) on the indigenous microbial community in forest soil was studied. MTBE, TBA and FA with different concentrations were added into microcosms containing forest soil samples. After 10 and 30 days, total viable cell number and dehydrogenase activity in the microcosms were evaluated. Bacterial communities in the microcosms were also analyzed using a denaturing gradient gel electrophoresis (DGGE). Dehydrogenase activity and total viable cell number were decreased according to the increase of MTBE, TBA and FA concentrations (P<0.05). FA toxicity was the highest, but TBA toxicity was the lowest. The results of principal component analysis using DGGE fingerprints showed that the microbial communities contaminated MTBE, TBA and FA were grouped by exposure time not exposure concentration. Dominant species in the microcosms were as follows: Photobacterium damselae sub sp. and Bacillus sp. KAR28 for MTBE; Mycobacterium sp. and Uncultured Clostridium sp. for TBA; and Uncultured Paenibacillaceae bacterium and Anxynobacillus, Flavithermus for FA.

Tissue Preparation with t-Butyl Alcohol Freeze-drying Method for Scanning Electron Microscopy: Application for Rat Liver (t-Butyl Alcohol 동결건조법을 이용한 흰쥐 간장의 주사전자현미경적 관찰)

  • T-butyl alcohol (TBA) freeze-drying method originally designed by Inoue and Osadake (1989) was adopted to dry specimens for scanning electron microscopy and the results were compared with those dried using critical point dryer (CPD). Small pieces $(1\times1\times3mm)$ of liver of Sprague-Dawley rats were cut and fixed in 2% glutaraldehyde in 0.1 M sodium cacodylate buffer after anesthesia, and processed for scanning electron microscopy by several modifications of TBA freeze-drying methods and by the standard preparation method using CPD. The bile canaliculi and sinusoidal endothelial surface were observed. Tissue dehydrated with TBA before TBA freeze-drying preserved the structures best comparable to those prepared with CPD. This result suggests that combination of dehydration with TBA and TBA freeze-drying is a superior method to the original TBA freeze-drying method dehydrated with ethanol.

Improvement of Efficiency in Dye-Sensitized Solar Cells with Addition of TBA to the TiO2 Paste and Its Electrochemical Analysis (이산화티타튬 페이스트에 TBA 첨가에 따른 염료감응 태양전지의 효율향상 및 전기화학적 분석)

  • Dye-sensitized solar cells (DSSCs) are considered as promising alternatives to conventional photovoltaic device. However, commercialization of the DSSCs is restricted due to the low efficiency. In this paper, highly efficiency DSSCs were fabricated by the adding the TBA to the $TiO_2$ paste. $TiO_2$ photoanode added 0.2 M TBA in DSSCs are shown the best efficiency of 9.14 %. This result ascribed to improvement of the connection between the $TiO_2$ nanoparticles by the addition of the optimized amount TBA. The morphology of the photoanode was observed by FE-SEM. Further investigation about the kinetics of the electrochemical processes are performed by the EIS analysis. Longest diffusion length was obtained in case adding 0.2 M of TBA to $TiO_2$ paste, which was matched well with the improved efficiency.
