Isolation and Characterization of A Thiodiglycol-Degrading Cupravidus sp. (Thiodiglycol를 분해하는 Cupriavidus sp.의 분리와 특성)

  • A Gram-negative bacterium capable of degrading thiodiglycol (TDG), main hydrolysis product of sulfur mustard, was isolated from ginseng field in enrichment medium supplemented with TDG as carbon source. The isolate, WS-32, grew optimally at $30-37^{\circ}C$ and pH 6.0-8.0. It was found to be similar to the genus Cupriavi연 on the basis of 165 rRNA sequence, while its biochemical properties were highly homologous to Alcaligenes faecalis. The cell growth of WS-32 strain was slightly inhibited on LB broth by TDG, but the maximum level of its growth was maintained stably in the presence of TDG. After incubation of inoculated LB medium or uninoculated LB medium containing TDG for 2 days, TDG amount of the culture filtrate was analyzed to decrease noticeably by HPLC. TDG and alcohols were also oxidized by cell-free extract of the isolate with maximum activities at pH 8.0 and $45^{\circ}C$.

Effect of Protein and Threonine Level in the Diet on Liver Threonine Dehydrogenase Activity in Growing Rat (성장하는 랫트에 있어서 단백질과 Threonine의 급여 수준이 간 Threonine Dehydrogenase 활성에 미치는 영향)

  • The aim of this investigation was to determine the influence of different protein and threonine (Thr) levels on the liver threonine dehydroaenase (TDG) activity b\ulcorner rats. In rats fed on CP (crude Protein) - diets, TDG activity was increased during an CP rise to 12.0% CP, decreased slightly down to 18.0% CP and showed .a trend to increase from 18.0 to 24.0% CP. In rats the feeding with graded Protein supply gavee no indication for additional stimulation of threonine-oxidation by TDG over a wide range of CP-content in the diets. The increase in threonine content from 0.28 to 0.72% in the presence of 12.0% CP caused a gradual increase in TDG activity in rat liver. This similarly applied to the feed admixed with 18.0% CP, but at a higher level.

  • Today's Internet is one of the necessaries of our life. Anomalies of the Internet provoke social problems. For that reason, Internet Measurement which studies characteristics on Internet traffic attracts pubic attention. Recently, Traffic Dispersion Graph (TDG), a novel traffic analysis method, was proposed. The TDG is not a statistical analysis method but a graphical visualization method on interactions among network components. In this paper, we propose a new anomaly detection paradigm and its technique using TDG. The existing studies have focused on detecting anomalous packets of flows. On the other hand, we focus on detecting the sources of anomalous traffic. To realize our paradigm, we designed the TDG Clustering method. Through this method, we could classify anomalous hosts infected by various worm viruses. We obtained normal traffic through dropping traffic of the anomalous hosts. Especially, we expect that the TDG clustering method can be applied to real-time anomaly detection because calculations of the method are fast.

Effects of Dietary Levels of Glycine, Threonine and Protein on Threonine Efficiency and Threonine Dehydrogenase Activity in Hepatic Mitochondria of Chicks

  • This study was carried out to evaluate the relationship between threonine (Thr) efficiency and Thr dehydrogenase (TDG) activity as an indicator of Thr oxidation on chicks fed with levels of diets (CP [17.5% and 21.5%] and Thr [3.8 and 4.7 g/100 g CP]; glycine [Gly][0.64% and 0.98%] and true digestible Thr [dThr] [0.45% and 0.60%]). Calculation of the Thr efficiency was based on N-balance data and an exponential N-utilization model, and TDG activity was determined as accumulation of aminoacetone and Gly during incubation of hepatic mitochondria. This study found that in the liver of chicks who received a diet containing up to 0.79% Thr (4.7 g Thr/100 g of CP) in the 17.5% CP diet, no significant (p>0.05) effect on TDG activity was observed. However, significantly (p = 0.014) increased TDG activity was observed with a diet containing 21.5% CP (4.7 g Thr/100 g of CP) and the efficiency of Thr utilization showed a significant (p = 0.001) decrease, indicating the end of the Thr limiting range. No significant (p>0.05) effect on the total TDG activity and accumulation of Gly was observed with addition of Gly to a diet containing 0.45% dThr. In addition, addition of Gly to a diet containing 0.60% dThr also did not result in a change in accumulation of Gly. Due to an increase in accumulation of aminoacetone, an elevated effect on total TDG activity was also observed. No significant (p>0.05) reduction in the efficiency of Thr utilization was observed after addition of Gly at the level of 0.45% dThr. However, significantly (p<0.001) reduced efficiency of Thr utilization was observed after addition of Gly at the level of 0.60% dThr. Collectively, we found that TDG was stimulated not only by addition of Thr and protein to the diet, but also by addition of Gly, and efficiency of Thr utilization was favorably affected by addition of Gly at the level near to the optimal Thr concentration. In addition, no metabolic requirement of Gly through the TDG pathway was observed with almost the same accumulation of Gly and a slight increase in TDG activity by addition of Gly. Thus, our findings suggest that determination of TDG activity and parameter of efficiency of Thr utilization may be useful for evaluation of dietary Thr level.

Effects of Dietary Protein and Threonine Supply on In vitro Liver Threonine Dehydrogenase Activity and Threonine Efficiency in Rat and Chicken

  • This study was conducted to assess the relation between threonine (Thr) oxidation rate and threonine efficiency on rat and chicken fed with graded levels of protein and threonine. The increase in threonine content from 0.28 to 0.72% in a diet containing 12.0% crude protein (CP) caused a gradual increase in threonine dehydrogenase (TDG) activity in rat liver. Similar, but more pronounced results were observed after 18.0% CP in the diet. Both protein levels in combination with the highest level of threonine supplementation increased liver TDG activity significantly, indicating enhanced threonine catabolism. Parameters of efficiency of threonine utilization calculated from parallel nitrogen balance studies decreased significantly and indicated threonine oversupply after a maximum of threonine supplementation. At the lower levels of threonine addition the efficiency of threonine utilization was not significantly changed. In the chicken liver up to 0.60% true digestible threonine (dThr) in the 18.5% CP diet produced no effect on the TDG activity. However, TDG activity in the liver was elevated by the diet containing 22.5% CP (0.60% dThr) and the efficiency of threonine utilization decreased, indicating the end of threonine limiting range. In conclusion, the in vitro TDG activity in the liver of rat and growing chicken has an indicator function for the dietary supply of threonine.

Isolation and Characterization of High Viscosity Polysaccharide Producing Endophytic Bacteria from Pueraria Root (고점도 다당류를 생산하는 갈근 내생균의 분리 및 특성)

  • Fifty endophytic bacteria, which produced slime around the colonies, were isolated from Pueraria roots. In particular, HDN-14, TDG-3, and TNB-3 strains, which appeared to be high viscosity producers, were selected. These strains produced high levels of polysaccharides in Puerara root medium extract. The purified polysaccharide was digested with 1N HCI and analyzed by HPLC, with glucose ($45.6{\sim}63.1%$), maltose ($14.6{\sim}23.7%$), and fructose ($17.4{\sim}23.7%$) detected as constitutive sugars. When determined by the homology relationship of the 16S rDNA sequence with the relative taxa, the HDN-14 and TNB-3 strains were closely ($99.06{\sim}99.32%$) related to the Pseudomonas $koreensis^T$ and Pseudomonas $jessenii^T$, while TDG-3 were closely ($99.48{\sim}99.74%$) related to Pseudomonas $plecoglossicida^T$, Pseudomonas $mosselii^T$, and Pseudomonas $monteilii^T$. The major cellular Pseudomonas acids are $3OH-C_{10:0}$, $2OH-C_{12:0}$, $3OH-C_{12:0}$, and $3OH-C_{12:1}$, with these strains being further differentiated in species belonging to the genus Pseudomonas.

  • 클럽 피팅(Club Fitting)이란 클럽을 구성하는 요소와 골퍼 자신의 신체적 특성 및 스윙 스타일에 맞춰 과학적으로 분석하고 내 몸과 내 스윙에 맞는 클럽으로 조정하는 과정이다. 본 논문에서는 피팅 관련 자료 수집 및 분석을 통해 클럽 피팅을 위한 클럽 피팅 마스터 프로그램 개념 설계 및 관련 DB 자료 수집 분석을 통하여 클럽 피팅 기술을 분석 및 연구한다. 본 프로그램 개발을 위해 Visual Basic(VB) 프로그램을 사용 하였으며, 피팅 마스터 프로그램 관련 자료의 데이터 관리를 위해 MDB를 이용하였다. 그리하여 측정한 클럽 정보를 골퍼에게 스펙정보 및 신체적 조건에 맞는 골프 클럽을 제시하는 연구를 한다.

  • 본 연구에서는 골프 피팅 장비인 스윙 피팅 장비, 바디 피팅 장비, 클럽 피팅 장비 같은 다양한 골프 피팅 장비들의 데이터를 컨트롤러로 연동한다. 이 과정에서 RS-232C통신을 통해 클럽 마스터 프로그램 서버와 통신하는 시스템을 설계한다. 클럽마스터 서버는 자동으로 데이터를 수집, 저장, 분석하여 알맞은 결과를 표출하기 위한 시스템이다. 측정된 데이터를 클럽 마스터 서버에 제공하기 위해 피팅 장비 복합기 연동시스템의 새로운 클럽 피팅 마스터 시스템 모델 설계를 제안한다.

Time-discontinuous Galerkin quadrature element methods for structural dynamics

  • Three time-discontinuous Galerkin quadrature element methods (TDGQEMs) are developed for structural dynamic problems. The weak-form time-discontinuous Galerkin (TDG) statements, which are capable of capturing possible displacement and/or velocity discontinuities, are employed to formulate the three types of quadrature elements, i.e., single-field, single-field/least-squares and two-field. Gauss-Lobatto quadrature rule and the differential quadrature analog are used to turn the weak-form TDG statements into a system of algebraic equations. The stability, accuracy and numerical dissipation and dispersion properties of the formulated elements are examined. It is found that all the elements are unconditionally stable, the order of accuracy is equal to two times the element order minus one or two times the element order, and the high-order elements possess desired high numerical dissipation in the high-frequency domain and low numerical dissipation and dispersion in the low-frequency domain. Three fundamental numerical examples are investigated to demonstrate the effectiveness and high accuracy of the elements, as compared with the commonly used time integration schemes.

  • 디지털 컴퓨터와 측정시스템의 발달로 대전력시험 데이터들은 시험자에 의한 분석이 아닌, 분석프로그램을 이용한 자동 분석이 가능해 졌다. 그러나 분석 프로그램을 이용하여 시험데이터를 분석해야 할 경우, 분석프로그램의 객관적인 신뢰성이 먼저 확보되어야 한다. 최근 들어 이러한 문제점을 보완하기 위하여, STL(Short-circuit Testing Liason)에서는 분석프로그램의 신뢰성평가를 위해 TDG(Test Data Generator)를 개발하였다. 이를 이용하면 대전력시험기관에서 사용하는 분석프로그램간의 객관적인 비교 및 평가가 가능하다. 따라서 본 논문에서는 STL에서 배포한 TDG를 이용하여, Crest, RMS, %DC 분석프로그램 알고리즘의 신뢰성을 평가해 보았다. 또한, STL에서 제시한 reference curve는 50[Hz]를 기본주파수로 하고 있기 때문에, 평가의 신뢰성을 높이기 위해 본 논문에서는 우리나라의 실정에 맞는 60[Hz]의 reference curve도 추가로 고려하였다. 신뢰성 평가 결과 모든 reference curve에 대한 분석알고리즘의 측정불화도가 STL에서 제시하는 범위에 존재함을 확인하였다.
