Microbacterium esteraromaticum CS3-1의 toluene 분해능에 미치는 benzene, ethylbenzene, xylene의 영향

  • Toluene-degrading bacterium, Microbacterium esteraromaticum CS3-1 was isolated from the biofilter for the removal of BTEX. Microbacterium esteraromaticum CS3-1 was shown to utilize toluene as a primary carbon and energy source. Effect of mixed BTEX gases on toluene degradation rate by M. esteraromaticum CS3-1 was investigated in this study. Toluene degradation rate was 2.26(only toluene), 2.06(toluene+benzene), 2.57(toluene+ethylbenzene), and 4.74(toluene+xylene) mmole $toluene\;{\cdot}\;g-DCW^{-1}\;{\cdot}\;h^{-1}$. Toluene degradation rate was 2.26(only toluene), 1.23(toluene+benzene+ethylbenzene), 1.52 (toluene+ethylbenzene+xylene), and 1.76(toluene+benzene+ethylbenzene+xylene) mmole $toluene\;{\cdot}\;g-DCW^{-1}\;{\cdot}\;h^{-1}$. The presence of BTEX compounds over three mixtures had a negative effect on toluene degradation rate. Toluene degradation rates were enhanced by the presence of ethylbenzene or xylene, whereas the presence of benzene had a negative effect on toluene degradation rate in comparison with toluene degradation rate when only toluene is existent.

Effect of the Combination of Ethanol with Toluene Treatment for a Short Time Period on the Toluene Metabolizing Enzyme Activity (흰쥐에 Toluene과 Alcohol의 병행투여가 Toluene 대사 효소활성에 미치는 영향)

  • To elucidate the effect of acute ethanol pretreatment on some toluene metabolizing enzyme activities, rats were divided into 4groups: control, alcohol-treated, toluene-treated, rat's and toluene-treated rats pretreated with ethanol. The alcohol or toluene-treated rats showed the significantly increased activities of hepatic aniline hydroxylase(AH) and aminopyrine demethylase(AD) compared to the control group. And the toluene-treated rats pretreated with ethanol showed somewhat decreased tendency of these enzyme activities compared to only toluene-treated rats. Liver benzylalcohol or aldehyde dehydrogenase activities were higher in alcohol or toluene-treated rats than those of the control group. The toluene-treated rats showed the decreased tendency of benzylalcohol dehydrogenase activities by the pretreatment of alcohol. Furthermore, toluene treated-rats showed the markedly decreased activity of benzaldehyde dehydrogenase by the ethanol pretreat-ment. On the other hand, hepatic xanthine oxidase activity in toluene-treated animals pretreated with ethanol was significantly higher than those of the toluene alone-treated rats. These results indicate that the combination of ethanol with toluene treatment for a short period of time possibly results in decreased activity of some toluene metabolizing enzymes in rats.

  • Blood toluene concentrations of thirty nine Korean toluene-exposed workers in shoes making factory were checked by headspace-gas chromatographic analysis. Air toluene concentrations in each worker's working region also checked by personal sampler during workshift and analyzed by gas chromatography. The range of blood toluene concentration was 0.15-0.84mg/L. The range of toluene concentration of each worker's working area was 8.46-189.9ppm. The correlation between blood and air concentration of toluene was 0.824.

Effect of Benzene, Acetone, and Methyl Mercaptan Vapor on Photocatalytic Decomposition of Toluene Vapor (톨루엔의 광촉매 분해시 벤젠, 아세톤 및 메틸메르캅탄 증기가 미치는 영향)

  • In this study, the photocatalytic decomposition characteristics of single toluene, toluene mixed with benzene, toluene mixed with acetone, and toluene mixed methyl mercaptan (MM) by UV reactor installed with $TiO_2$-coated perforated plate were studied. The photocatalytic decomposition rate of single toluene, toluene mixed with benzene, toluene mixed with acetone, and toluene mixed with MM fitted well on Langmuir-Hinshelwood (L-H) kinetics equation. The maximum elimination capacity was obtained to be $628g/m^3{\cdot}d$ for single toluene, $499g/m^3{\cdot}d$ for toluene mixed with benzene, $318g/m^3{\cdot}d$ for toluene mixed with acetone, and $513g/m^3{\cdot}d$ for toluene mixed with MM, respectively. The negative effect in photocatalytic decomposition of toluene are found to be in the order of acetone>benzene>MM.

  • Burkholderia cepacia strain G4 (pHG-2) containing toluene 2-monooxygenase and toluene dioxygenase, was able to grow on toluene and accumulate cis-3-methyl-3,5-cyclohexadien-1,2-diol (cis-toluene dihydrodiol) in the liquid culture. The cis-toluene dihydrodiol produced was identical to the authentic compound, as judged through mass spectrometry and nuclear magnetic resonance analysis. Our results indicate that pHG-2 provides an economical means to produce chemically-important chiral synthons while growing on toluene.

  • A study was performed to evaluate an effect of toluene or ethanol pretreatment on the toluene metabolism. A slight liver damage in rats was induced by administration of 0.2 ml of toluene or 0.2 ml of 50% ethanol per 100 g of body weight intraperitoneally every other day for four weeks except the last day before sacrifice. One day before sacrifice, toluene was administered to rats pretreated ethanol or toluene and the control. Rats were sacrificed at the 1st, the 2nd, the 3rd and the 4th week after the first administration of xenobiotics mentioned above. Based on the histopathological findings, liver weight per body weight, serum alanine aminotransferase and hepatic glutathione content, toluene- or ethanol-pretreated groups showed the reversible liver injury. By the treatment of one dose of toluene, the contents of hippuric acid in urine was higher in the group pretreated with toluene or ethanol than control. The contents of cytochrome P-450, benzylalcohol dehydrogenase and benzaldehyde dehydrogenase activities were generally more increased in toluene- or ethanol-pretreated rats than control. $K_m$ values of the benzylalcohol dehydrogenase in pooled liver samples from toluene- and ethanol-pretreated groups were similar each other. $V_{max}$ values of toluene- or ethanol-pretreated group was higher than control. In conclusion, the toluene metabolism is accelerated in rats pretreated with toluene or ethanol.

  • An experimental investigation of the gas-phase photooxidation of toluene-NO$_{x}$-air mixtures at sub-ppm concentrations has been carried out in a 6.9 m3, indoor smog chamber irradiated by blacklights. Measured parameters in the toluene-NO$_{x}$ experiments included $O_3$, NO, NO$_2$, NO$_{x}$, CO, SO$_2$ toluene, and air temperature. The initial toluene concentration ranged from 225 ppb to 991 ppb and the initial concentration ratio of toluene/NO$_{x}$ in ppbC/ppb was in the range of 5~20. It was found that the variation of gaseous species with irradiation time caused by the photooxidation of toluene-NO$_{x}$-air mixtures depended on the initial toluene concentration for similar concentration ratio of toluene/NO$_{x}$. The dependency of initial toluene concentration on the photooxidation of toluene-NO$_{x}$-air mixtures for toluene/NO$_{x}$=5~6 seemed to be opposite to that for toluene/NO$_{x}$=10~11. The arriving time at maximum ozone concentration depended on both initial toluene concentration and initial concentration ratio of toluene/NO$_{x}$. However, the maximum concentration of ozone formed by photooxidation depended only on the initial toluene concentration.luene concentration.

The Beneficial Effect of Melatonin for Toluene Hepatotoxicity in Rats

  • Toluene is mainly metabolized in liver by oxidative pathway. Oxigen free radicals occur through the process of toluene metabolism Therefore it causes tissue and cell min by the oxygen free radicals from the metabolism of toluene. Melatonin acts as a highly efficient free radical scavenger that protects cells from damage by oxygen free radicals. To test this hypothesis, toluene hepatotoxicity was induced by an abdominal injection of toluene. To see if the melatonin protects the rat's liver, melatonin was administrated orally, at the time of each toluene injection. Aspartate aminotransferase(AST), alanin aminotransferase(ALT), latic dehydrogenase(LDH) and alkaline phosphatase(ALP) levels in serum were measured to estimate hepatic function. Malondialdehyde(MDA), which gives an indirect index of oxidative injury was also measured. Hippuric acid is the last metabolic Production of toluene was measured by HPLC. There were significantly higher in AST, ALT, LDH, MDA and hippuric acid in toluene group, but there were no significant difference in melatonin group except ALT and hippuric acid. There was significantly lower in ALP level in toluene group, but there was no significant difference melatonin group, suggesting a significant hepatotoxicity due to oxygen free radicals through the process of toluene metabolism Melatonin treatment significantly protected hepatic function and free radical-mediated injury in the liver against toluene-induced changes. Accordingly, this study shows that melatonin is helpful in protecting liver injury by acute toluene intoxication.

Effect of Ethanol Pretreatment on the Toluene Metabolism in Toluene-treated Rats (흰쥐에 있어서 주정중독이 Toluene 대사에 미치는 영향)

  • To evaluate an effect of ethanol pretreatment on the toluene metabolism, toluene (50% in olive oil) was given three times at 0.2 ml/100g body weight at the interval of one day to the rats fed with 5% ethanol during two months. The ethanol pretreated rats were not identified particular liver injury by the histopathologic findings. In case of toluene treatment, the ethanol pretreatment to the rats led to more increased concentration of urinary hippuric acid than those treated with only toluene. The ethanol pretreatment to the rats led to the increased activities of hepatic aniline hydroxylase and these enzyme activities were higher both in toluene treated and those pretreated with ethanol, but no differences were found in two groups. Ethanol pretreated rats showed the more increased activities of benzylalcohol dehydrogenase than control group. Moreover, the ethanol pretreatment to the toluene treated rats led to significantly more increased activities of benzylalcohol dehydrogenase compared with those treated with toluene only. Furthermore, the alcohol pretreatment to the toluene treated rats also led to somewhat higher activities of benzaldehyde dehydrogenase than those treated with toluene. In conclusion, these results indicate that the chronic pretreatment of ethanol at not so much liver damage as normal may rather activate the toluene metabolism.
