호랑나비 유충 혈림프 Transferrin의 특성과 분포 (Characterization and Distribution of Transferrin from the Last Larval Haemolymph of Papilio xuthus)

  • Transferrin은 저장된 철분자를 운반하고 살아있는 유기체에서 철의 항상성을 유지한다. 호랑나비의 종령 유충 혈림프내 철 운반 단백질인 transferrin을 KBr 밀도구배 초원심분리와 Superose 6 HR을 이용한 fast protein liquid chromatography법으로 분리 정제하였으며, transferrin의 조직에 따른 분포는 면역화학적 방법에 따라 확인하였다. 정제된 transferrin의 아미노산 조성은 아스파르트산(Asp), 발린(Val), 루이신(Leu), 글루타민(GIu)이 많이 존재하였으며, subunit의 분자량은 78, 80 kDa으로 확인되었다. Immuno-diffusion을 통해 각 조직에서 분포를 확인한 결과 혈림프와 지방체에서 transferrin이 전시기에 걸쳐 뚜렷한 동질성을 나타냈다. 또한 rocket immuno-electrophresis법에 의해 transferrin의 양적 분포를 살펴보면 혈림프에서 용화 3일과 용화 5일에 증가하였으며, 지방체에서는 전용기와 용화직후에 양적인 증가를 나타냈다.

  • Expression of transferrin gene in various organs of rat was studied using rat transferrin cDNA. The hybridization method of $[^{35}S]-labeled$ transferrin cDNA with transferrin mRNA in cytoplasmic preparations was used to measure the level of transferrin mRNA. The rat from 15-day old fetus to 21-day old postnatal were employed as an animal model. In the liver, the level of transferrin mRNA increased with increasing age. However, the level of transferrin mRNA in brain was significantly lower than that in liver and the level did not increase with age.

감염병소 영상을 위한 $^{99m}Tc$-Transferrin 개발 (Development of $^{99m}Tc$-Transferrin as an Imaging Agent of Infectious Foci)

  • 목적 : 본 연구의 목적은 $^{99m}Tc$-방사성표지 트렌스페린 ($^{99m}Tc$-transferrin)을 개발하여 감염/염증병소의 진단에 이용할 수 있는지 알아보고, 이를 $^{67}Ga$-Citrate 영상과 비교하고자 하였다. 대상 및 방법 : Succinimidyl 6-hydrazino-nicotinate hydrochloride -chitosan -transferrin (Transferrin)을 합성하고, 여기에 $^{99m}Tc$ 방사성 표지를 시행하였다. $^{99m}Tc$-transferrin의 방사성표지효율은 표지 후 10분, 30분, 1시간, 2시간, 4시간, 8시간에 측정하였다. 포도상구균(ATCC 25923, $2{\times}10^8$ colony forming unite, 0.2 ml)을 접종한 쥐농양모델에서 $^{99m}Tc$-transferrin과 $^{67}Ga$-citrate의 생체내 분포를 평가하고 영상 검사를 실시하였다. 결과: 질량분석계를 이용하여 Transferrin이 성공적으로 제조되었음을 알 수 있었다. $^{99m}Tc$-transferrin의 방사성표지효율은 10분, 30분, 1시간, 2시간, 4시간, 8시간에 각각 $96.2{\pm}0.7%,\;96.4{\pm}0.5%,\;96.6{\pm}1.0%,\;96.9{\pm}0.5%,\;97.0{\pm}0.7%\;and\;95.5{\pm}0.7%$ 이었다. $^{99m}Tc$-transferrin의 단위섭취량은 감염병소에서 $0.18{\pm}0.01\;and\;0.18{\pm}0.01$, 정상근육에서 $0.05{\pm}0.01\;and\;0.04{\pm}0.01$이었고, 감염병소 대 정상근육 섭취비는 30분과 3시간에 각각 $3.7{\pm}0.6\;and\;4.7{\pm}0.4$이었다. $^{99m}Tc$-transferrin 영상에서 10분, 30분, 1시간, 2시간, 4시간 그리고 10시간에서의 병소/배후방사능비는 각각 $2.18{\pm}0.03,\;2.56{\pm}0.11,\;3.08{\pm}0.18,\;3.77{\pm}0.17,\;4.70{\pm}0.45$ 그리고 $5.59{\pm}0.40$이었고, $^{67}Ga$-citrate의 경우 2시간, 24시간, 48시간에 $3.06{\pm}0.84,\;4.12{\pm}0.54\;4.55{\pm}0.74 $이었다. 결론 : Transferrin에 $^{99m}Tc$을 이용한 방사성표지가 성공적으로 이루어졌고, $^{99m}Tc$-transferrin의 표지효율은 8시간까지 95% 이상의 안정된 방사성표지효율을 보였다. $^{99m}Tc$-transferrin을 이용한 감염영상을 성공적으로 얻을 수 있었으며, $^{67}Ga$-citrate 영상과 비교하여 더 빠른 시간 안에 우수한 영상을 얻을 수 있었다. 그러므로 $^{99m}Tc$-transierrin이 감염 병소의 영상진단에 사용될 수 있을 것으로 기대된다.

The Activity of Apo-transferrin on the Growth of Staphylococcus pseudintermedius

  • Apo-transferrin is an iron-binding protein that has been reported to have an antimicrobial effect. It is considered a major component of the host defense mechanism as it limits microbial access to iron. This study was performed to investigate whether bovine apo-transferrin would have an inhibitory effect on the growth of S. pseudintermedius, which is one of the most isolated bacteria from dogs, and to compare the antimicrobial efficacy with bovine holo-transferrin. S. pseudintermedius were grown at $37^{\circ}C$ in 96-well culture plates using Muller Hinton broth containing bovine apo-transferrin or bovine holo-transferrin at concentrations ranging from 0.5 or 2.5 to 5.0 mg/ml. The optical densities of the wells were then measured at 570 nm. In this study, the apo-transferrin showed dose-dependent antimicrobial effect against S. pseudintermedius while holo-transferrin did not inhibit the growth of S. pseudintermedius effectively. The results suggest that iron deprivation is an important pathway for inhibiting bacterial growth and bovine apo-transferrin has great antimicrobial effects against S. pseudintermedius.

배양 Caco-2 세포 단층막 실험계에서 트란스페린과 옥시다아제효소 포합체의 세포막투과 (Transport of Transferrin-Horseradish Peroxidase Conjugate Through Cultured Caco-2 Cell Monolayer)

  • Transport study of horseradish peroxidase and transferrin-horseradish peroxidase conjugate was performed using an in vitro Caco-2 cell cultured monolayer grown on a polycarbonate membrane of $Transwell^{\circledR}$, Horseradish peroxidase was not transported across Caco-2 cell monolayer. Transferrin-horseradish peroxidase conjugate was transported through Caco-2 cell monolayer. The apparent membrane permeability coefficient $(P_{app})$ of transferrin horseradish peroxidase conjugate was $6.54{\times}10^{-7}\;cm/sec$. The $P_{app}$ value of transferrin-horseradish peroxidase conjugate across Caco-2 cell monolayer was increased to $11.9{\times}10^{-7}\;cm/sec$ in the presence of $50\;{mu}g/ml$ brefeldin-A. These results suggest the transferrin receptor mediated transcytosis of transferrin-horseradish peroxidase conjugate across Caco-2 cell monolayer.

Staphylococcus aureus Siderophore-Mediated Iron-Acquisition System Plays a Dominant and Essential Role in the Utilization of Transferrin-Bound Iron

  • Staphylococcus aureus is known to be capable of utilizing transferrin-bound iron, via both siderophore­and transferrin-binding protein (named IsdA)-mediated iron-acquisition systems. This study was designed in order to determine which iron-acquisition system plays the essential or dominant role with respect to the acquisition of iron from human transfenin, in the growth of S. aureus. Holotransferrin (HT) and partially iron-saturated transferrin (PT), but not apotransferrin (AT), were found to stimulate the growth of S. aureus. S. aureus consumed most of the transferrin-bound iron during the exponential growth phase. Extracellular proteases were not, however, involved in the liberation of iron from transferrin. Transferrin-binding to the washed whole cells via IsdA was not observed during the culture. The expression of IsdA was observed only in the deferrated media with AT, but not in the media supplemented with PT or HT. In contrast, siderophores were definitely produced in the deferrated media with PT and HT, as well as in the media supplemented with AT. The siderophores proved to have the ability to remove iron directly from transferrin, but the washed whole cells expressing IsdA did not. In the bioassay, the growth of S. aureus on transferrin-bound iron was stimulated by the siderophores alone. These results demonstrate that the siderophore-mediated iron-acquisition system plays a dominant and essential role in the uptake of iron from transferrin, whereas the IsdA-mediated iron-acquisition system may play only an ancillary role in the uptake of iron from transferrin.

Effects of Transferrin on Enhancing Biological Availability of Iron

  • In this study, transferrin which is an iron-carrying glycoprotein in plasma was evaluted for its iron binding capacities(TIBC), iron solubilizing abilities, and enhancing effect of biological availbability of iron. Results of TIBC showed that 1 mg of transferrin could blind 1.28$\mu\textrm{g}$ of iron indicating that one molecule of transferrin can bind about 2 molecules of iron. Also, solubility of iorn (7.5$\mu\textrm{g}$ Fe/ml) was significantly incresed to 96.0% with addition of transferrin (5mg/ml) .When FeCl3(80$\mu\textrm{g}$ Fe/ml) was injected to iron-deficient rats by intestinal segment in situ technique, 18.4% of injected iron was absorbed wherease 48.49 and 48.76% of injected iron was absorbed with addition of 10 and 20 mg transferrin/ml , respectively.

Tilapia(Oreochromis niloticus)의 혈청과 단백질의 transferrin의 다형현상에 관한 연구 (Studies on Polymorphism of Transferrin of Serum Proteins in Tilapia (Oreochromis niloticus))

  • 본 실험은 rivanol 침전법, iron-staining method, SDS-Polyacrylamide gel electrophoresis 그리고 $^{59}FeCl_3$ sutoradiography를 이용하여 tilapia(Oreochromis niloticus) 혈청으로부터 transferrin을 분리 추출하며, Hardy-Weinberg법칙을 이용하며 유전자 빈도와 유전자형 빈도 추정하기 위하여 시도하였다. 본 실험에서 얻어진 결과는 다음과 같다. 1. Tilapia의 transferrin은 albumin보다 상대적으로 느린 전기영동도와 높은 분자양의 크기를 가졌고, globulin 보다는 빠른 전기영동도와 낮은 분자양을 나타내었다. 2. 염색방법을 달리하여 transferrin의 band를 비교하였을 때, 별다른 차리를 발견하지 못했다. 3. Rivanol 대 혈청의 비율은 2 : 1이 가장 적합했고, 모든 분획에 이 비율을 이용하였다. 4. Transferrin의 분자양은 각 70,000$\pm$2,000을 나타내었다. 5. Transferrin의 종내 다형현상을 나타내었다. 6. 3개의 transferrin 변이체(A,B 그리고 C)가 발견되었고, Tf형은 3개의 공동 우성 대립유전자(Tf A, Tf B 그리고 Tf C)에 의해 조절되며, 6개의 가능한 표현형(Tf AA, Tf AB, Tf AC, Tf BC 그리고 Tf CC)중에서 5개의 표현형만이 발견되었고, Tf CC형은 발견되지 않았다. 7. 3개의 대립유전자 Tf A, Tf B 그리고 Tf C의 빈도는 각각 0.795, 0.15 그리고 0.055 이었다.

  • Lactococcus lactis subsp. cremoris FC는 카스피해 요구르트의 명칭으로 강한 점성을 가지는 것을 특징으로 하는 코카커스 지방 유래의 발효유로부터 분리된 균주로 있다. 본 연구는 Lc. lactis subsp. cremoris FC의 생장에 대한 lactoferrin group(native-lactoferrin, apo-lactoferrin, 그리고 holo-lactoferrin)과 transferrin (apo-transferrin과 holo-transferrin)의 첨가가 생장에 미치는 영향을 알아보기 위하여 수행하였다. Lc. lactis subsp. cremoris FC의 생장에 lactoferrin과 transferrin의 첨가효과는 0.5 또는 1 mg/mL의 농도로 첨가하는 것이 생장 촉진에 좋은 효과를 나타내는 것으로 조사되었다.

Gene Expression and Regulation of Wax Moth Transferrin by PAMPs and Heavy Metals

  • A complete mRNA sequence of transferrin from the wax moth, Galleria mellonella, was obtained, and compared with those of other species. We previously reported that the sequence was most similar to those of Manduca sexta and Bombyx mori. As in other moths, G. mellonella transferrin had only one iron-binding site at its N-terminal region. Semi-qRT PCR was conducted to investigate tissue-specific distribution and transcriptional regulation of the wax moth transferrin mRNA. Larval muscle and fat body contained larger quantity of mRNA than other tested tissues. In this study, it was observed that iron and cadmium regulated transferrin transcription, and this regulation pattern was tissue specific. Iron up-regulated transferrin mRNA level in fat body, while suppressed it in the Malpighian tubules and silk glands. Cadmium decreased the mRNA level in fat body, muscle, and Malpighian tubules, but significantly increased the mRNA level in silk glands. In addition, the mRNA expression was induced by all tested pathogen-associated molecular patterns (PAMPs) including LPS, lipoteichoic acid (LTA), glucan, and even chitin.
