Development of Specific Primer for Tricholoma matsutake

  • In this study, in an effort to develop a method for the molecular detection of Tricholoma matsutake in Korea from other closely related Tricholomataceae, a species-specific PCR primer pair, TmF and TmR, was designed using nuclear ribosomal intertranscribed spacer (ITS) sequences. The DTmF and DTmR sequences were 5'-CCTGACGCCAATCTTTTCA-3' and 5'- GGAGAGCAGACTTGTGAGCA-3', respectively. The PCR primers reliably amplified only the ITS sequences of T. matsutake, and not those of other species used in this study.

The Mycodiversity and Resources of Fungi in Mt. Nam (남산의 균류 다양성과 균류자원)

  • Many fungal fungi were collected at Mt. Nam areas during 3 days in August 1997 and 1 day in September 1998. They were identified. According to the resulting, they were composed of 2 divisions, 3 subdivisions, 5 classes, 4 subclasses, 15 oders, 30 families, 58 genera and 95 species. Dominant species was Fomiltella fraxinea and dominant family was Tricholomataceae. Among them genera of henningsomyces, Roseoformes and Physarum were newly to Korea. Henningsomyces candidus, Roseoformes subflexibilis, Lentaria micheneri and Physarum mutans were newly to Korea. Resources of fungi were 25 species in edibility, 4 species in clulture, 11 species in toxine, 13 species in medicne, 18 species in anticancer, 15 species in ectomycorrizahe and 50 species in rotten wood.

Distribution of Higher Fungi in NaeJangSan National Park (내장산국립공원의 고등균류 분포)

  • This study was conducted to investigate the diversity of higher fungi in NaeJangSan National Park from April 2004 to November 2006. The obtained results from investigation were as follows. The total of 5 classes, 19 orders, 60 families, 168 genera and 418 species (including 10 families, 13 genera and 15 species unrecorded) including saprophytic and ectomycorrhizal fungi was investigated. The higher fungi were classified into 47 families, 143 genera and 384 species in Basidiomycotina, 9 families, 19 genera and 28 species in Ascomycotina and 4 families, 6 genera and 6 species in Myxomycota. It was turned out that most of the higher fungi belong to Hymenomycetidae in Basidiomycotina, for which 34 families 122 genera, and 353 species were observed. Dorminant species belonged to Tricholomataceae(64 species) Russulaceae(39 species), Polyporaceae(36 species) and Boletaceae(36 species). The mushroom occurrence of higher fungi was closely related to climatic conditions such as high air temperature and lots of rainfall from July to September. The environment which has a favorable influence of mushroom occurrence was air temperature, relative humidity and rainfall of climatic environment.

Cultural Characteristics of Ectomycorrhizal Mushrooms

  • Ectomycorrhizal (ECM) mushrooms play a major role in plant growth promotion through symbiotic association with roots of forest trees. They also provide an economically important food resource to us and therefore they have been studied for their artificial cultivation for decades in Korea. We have secured bio-resources of ECM mushrooms from Korean forests and performed their physiological studies. To investigate the cultural characteristics, the fungi were cultured under different conditions (medium, temperature, pH of the medium, inorganic nitrogen source). More than 90% of total 160 strains grew on three solid media (potato dextrose agar, PDA; sabouraud dextrose agar, SDA; modified Melin-Norkrans medium, MMN). The rate of mycelial growth on malt extract agar (MEA) was lower than those of three media (PDA, SDA, MMN). None of the Tricholomataceae strains grew on MEA. Many strains of ECM mushrooms were able to grow at the temperature range of $15{\sim}25^{\circ}C$ on PDA, while they showed poor growth at $10^{\circ}C$ or $30^{\circ}C$. In particular, the growth rates of both Gomphaceae and Tricholomataceae were significantly lower at $10^{\circ}C$ than at $30^{\circ}C$. The optimal pH of many strains was pH 5.0 when they cultured in potato dextrose broth (PDB). Fifty-seven percent of tested strains grew well on medium containing ammonium source than nitrate source. Many strains of Tricholomataceae showed a notable growth on ammonium medium than nitrate medium. Twenty-three percent of strains preferred nitrate source than ammonium source for their mycelial growth. The production and activity of two enzymes (cellulase and laccase) by ECM fungi were also assayed on the enzyme screening media containing CMC or ABTS. Each strains exhibited different levels of enzymatic activities as well as enzyme production. The number of laccase-producing strains was less than that of cellulase-producing strains. We found that 77% of tested strains produced both cellulase and laccase, whereas 2% of strains did not produce any enzymes. The morphological characteristics of mycelial colony were also examined on four different solid media. Yellow was a dominant color in mycelial colony and followed by white and brown on all culture media. ECM mushrooms formed mycelial colonies with a single or multiple colors within a culture medium depending on the strains and culture media. The most common shape of mycelial colony was a circular form on all media tested. Other families except for Amanitaceae formed an irregular colony on MMN than PDA. All strains of Tricholomataceae did not form a filamentous colony on all media. The pigmentation of culture media by mycelial colonies was observed in more than 50% of strains tested on both PDA and SDA. The degree of pigmentation on PDA or SDA was higher than MMN and brown color was dominant than yellow color. The production of exudates from mycelial colony was higher on PDA than MMN. Brown exudates were mainly produced by many strains on PDA or SDA, whereas transparent exudates were mainly produced by strains on MMN. We observed the mycelial colonies with a single or multiple textures in just one culture plate. Wrinkled or uneven colony surfaces were remarkably observed in many strains on PDA or SDA, while an even colony surface was observed in many strains on MMN. Sixty percent of Tricholomaceae strains formed wrinkled surface on PDA. However, they did not form any wrinkle on MMN plate. Cottony texture was observed in mycelia colonies of many strains. Velvety texture was often observed in the mycelial colonies on SDA than PDA and accounted for 60% of Suillaceae strains on SDA.

  • Taxonomic relations among three species of some edible fleshy fungi(Lentinus edodes, Pleurotus ostreatus, Flammulina velutipes) in the family Tricholomataceae were examined by using polyacrylamied gel disc electrophoresis. The soluble, crude extract of mycelium grown on potato sucrose broth was subjected to electrophoresis. Similarities in the protein bands for each isolated of one species were compared with those for others. In the banding patterns there was a closer relationship between isolates within one species than among isolates of different species. However, the isozyme patterns obtained from each isolate of Peurotus ostreatus (esterase, peroxidase, tyrosinase) were appeared to represent the degree of geographical variability within one species.

Biodiversities and Fungal Resources of Mt.Daedun Provincial Park (대둔산 도립공원의 균류다양성과 균류자원)

  • Many fungi were collected at Mt. Daedun Provincial Park between the periods from March 2001 to October 2002 and they were identified. According to the results, the fungi diversity included 2 division, 3 subdivisions, 5 classes, 4 subclasses, 12 orders, 35 families, 75 genera and 153 species. Among them Ileodictyon is unrecorded genus and Ileodictyon gracile, Mycena flosum-nivium and Tricholoma lascivum are newly to Korea. Dominant family are Tricholomataceae, Russulaceae and Polyporaceae;genus Amanita and species Collybia confluens and Russula emetica In useful fungi and ecological resources, edible fungi included 51 species; culture fungi 10; poisonous fungi 18: medicinal fungi 17: anticancer fungi 22: ectomycorrhizal fungi 26 and wood rotting fungi 51, In geographical distributions of Chonbuk areas, same species are 50 spcies of Jeongup ; 56 of Mt.Manduck : 84 of Mt.Jiri : 28 of Pyonsan national park : 59 of Cheonju arboretum and 51 of Mt,Sunun.

  • To investigate a possibility of producing the antitumor component by shake culture method, the mycelia of Flammulina velutipes were cultured in flasks on a shaker at $26{\sim}28^{\circ}C$ at 180 rpm for seven days. The extract of the mycelia was concentrated under vacuum. The precipitate obtained by adding a three-fold volume of ethanol was centrifugated and freeze-dried after dialysis. The fraction was tested against sarcoma 180 in the mice. The inhibition ratio of the fraction against the tumor was 68.0% at the dose of 20 mg/kg/day for the period of ten days and the tumors in three of the mice were completely regressed. The results showed, therefore, that the antitumor component was produced by the shake culture method.

A Checklist of Mushrooms of Phousabous National Protected Area (PNPA) of Lao PDR

  • Mushroom survey and collection were conducted in the Phousabous National Protected Area (PNPA), which is located at the Northeastern area 230km from the capital city of Lao-PDR, Vientiane, from April 2017 to September 2018. During this periods, mushroom specimens were collected from 8 different locations, and then they were identified and classified into 284 species, 142 genera, 56 families, 20 orders and 7 classes by morphological and molecular analyses. The mushrooms belongs to Ascomycota were classified into 10 species, 5 genera, 5 families, 4 orders and 3 classes, while those belongs to Basidiomycota were classified into 274 species, 137 genera, 51 families, 16 orders, and 4 classes, respectively. Among these mushrooms, the most species-rich families are Boletaceae (16.06%), Polyporaceae (10.12%), Russulaceae (9.94%), Amanitaceae (7.68%), Agaricaceae (4.54%), Marasmiaceae (4.36%), Cortinariaceae (3.66%), Entolomataceae (3.66%), Mycenaceae (2.79%), Xylariaceae (2.44%), Physalacriaceae (2.09%), Omphalotaceae (2.09%), Hydnangitaceae (2.09%), Lyophyllaceae (1.92%), Tricholomataceae (1.75%) and comprised 75.19% of the total specimens identified.

The Higher Fungal Flora in the Areas of Mt. Daesung and Mt. Daeduck (대성산(大成山)과 대덕산(大德山) 지역(地域)의 고등균류(高等菌類))

  • More than 300 specimens of higher fungi were collected from both Mt. Daeduck and Mt. Daesung areas in Kangwon Province during the period from 13 to 19 August, 1990. These fungi were identified and classified into 4 orders, 15 families, 35 genera, 52 species and 1 variety. Dominant species were considered to be Collybia confluens in Tricholomataceae, and Russula emetica in Russulaceae of Agaricales, and Phlogiotis hevelloides in Auriculariales throughout our survey areas. Phlogiotis hevelloides (Fr.) Martin. (장미주걱목이) and Pseudohydnum gelatinosum (Scop. ex Fr.) P. Karst. (혓바늘목이) were newly described in Korea.
