The Proposed Validation Guidelines and Their Application for Performance Verification of UV-VIS Spectrophotometer (UV-VIS 분광기기의 성능검증을 위한 밸리데이션 가이드라인의 제정 및 응용)

  • For the increasing demand for the validation of analytical results, one of the important concerns of the analyst is to be assured that the instrumentation is working to qualified conditions. In this study, we have investigated and compared several validation guidelines of the performance verification of UV-VIS spectrophotometer used in pharmacopoeias of major countries and UV-VIS instrument analytical companies, and we proposed the practically useful validation guidelines of UV-VIS spectrophotometer. Based on our proposed validation guidelines of UV-VIS spectrophotometer, several UV-VIS spectrophotometers used in drug manufacturing laboratories were validated for their performances.

The Analysis of the UV cut-off effect of tinted lens and the Characteristic of UV tester (착색렌즈의 UV 차단효과와 UV TESTER 평가 분석)

  • We obtained the UV cut-off effect of tinted CR-39 Ophthalmic lens with the respect of concentration of green, brown, blue color. First, we analyzed the UV spectrum of tinted CR-39 lens measured by Spectrophotometer and then compared the UV data by Spectrophotometer with UV data by UV tester used in an optician shop. The result obtained show that the UV cut-off effect is increased with color concentration of tinted CR-39 lens, and the transmittance of 0%~5% at 400nm by Spectrophotometer is correspondence with transmittance of 0% by UV tester.

  • This study was conducted to find out analytical method for diesel fuel using uv-spectrophotometer in water samples. The optimum wavelength, detection limit and recoveries for desel fuel extracted with n-pentane were 257nm, $100{\sim}800mg/l$ and $102{\sim}121%$, respectively. This method were stable for measuring concentrations of desel fuel for 1 hour at $20{\pm}1^{\circ}C$ condition.

  • Through sol-gel process, inorganic/organic composite as a dye host was made. It was observed that this composite has about 90% of trnasmittance by UV/VIS spectrophotometer. The transmittance is the most important requirement to decide whether the composite could apply or not as the dye host. The transmittance was increased with decreasing HCl as a catalyst, aging time, and amount of PDMS. Also, fluorescent and transmittant charateristics of Rhodamine 6G Perchlorate that was doped into ORMOSIL were measured. The fluorescent intensity was increased and the trnasmittance was decreased with rising gelling temperature.

  • 5,15-Bis(mesityl)-10,20-bis(4-amino)porphyrin was synthesized with meso-(mesityl)dipyrromethane and 4-nitrobenzaldehyde. It was characterized with $^1H$-NMR spectroscopy, UV spectrophotometer, and GC-MASS. In UV spectrum, Soret-band(438 nm) and Q-bands(526, 572, 611 nm) were confirmed. In NMR spectrum, the characteristic peak of porphyrin was detected at -2.65 ppm. Copolyimide containing porphyrin structure was synthesized with mesityl-$TPP(NH_2)$, 3,3',4,4'-benzophenonetetracarboxylic dianhydride(BTDA) and 2,2-bis(4-(4-aminophenoxy)phenyl) hexafluoropropane(BAPHF) and characterized with UV and FT-IR spectrophotometer. The $T_g$ and $T_m$ of the synthesized copolyimide were 220.7 and $369.2^{\circ}C$, respectively.

  • In this study, CdSe-$TiO_2$ photocatalyst were synthesized by a facile solvothermal method and characterized by X-ray diffraction (XRD), Scanning electron microscopy (SEM) with energy dispersive X-ray (EDX) analysis and UV-vis diffuse reflectance spectrophotometer. The photocatalytic activity was investigated by degrading methylene blue (MB) in aqueous solution under irradiation of UV light as well as visible light. The absorbance of degraded MB solution was determined by UV-vis spectrophotometer. The results revealed that the CdSe-$TiO_2$ photocatalyst exhibited much higher photocatalytic activity than $TiO_2$ both under irradiation of UV light as well as visible light.

  • In this study, Ultraviolet (UV) curable urethane acrylates for ink binder was prepared by reaction of IPDI, polyether polyol and HEA. The UV curing behavior of compositions with HEA/PETA ratio on UV irradiation time was investigated by FT-IR spectrophotometer, probe tack tester and Photo-DSC. Conversion % and gel content were increased with increasing UV irradiation time, but probe tack was decreased. Storage modulus, tensile strength and decomposition temperature were increased as PETA content increased. In case of HEA/PETA ratio was 30/70, adhesion property of UV-cured composition on PMMA sheet was excellent.

Effects of Gagam-Jawoonaek about Erythema by UV Exposure (가감자운액(加減紫雲液) 도포가 자외선으로 유발된 피부 홍반에 미치는 영향)

  • We studied to investigate the erythema reduction effects generated by Gagam-Jawoonaek(GJ) application(appl.) after UV exposure. Twenty women in their twenties to fifties with no skin diseases were recruited. We exposed UV as a 6 subsites on the left upper arm of subjects using multi-port solar simulator. After setting Gagam-Jawoonaek(GJ) application(appl.) subsites and non-appl. subsites, we measured erythema degrees($a^*$ values) of the subsites using spectrophotometer. We measured $a^*$ values four times(before UV exposure, before application of GJ, twenty-four and forty-eight hours after first application of GJ). We analyzed data using student's t-test. After UV exposure, $a^*$ values on the left upper arm increased. Twenty-four hours after first GJ treat., the changes of $a^*$ value on GJ treat. subsites($1.22{\pm}0.13AU$) were bigger than GJ non-treat. subsites($1.04{\pm}0.12AU$), but there was no statistically significance. Forty-eight hours after first GJ treat., the changes of $a^*$ value on GJ treat. subsites($1.95{\pm}0.11AU$) were bigger than GJ non-treat. subsites($1.58{\pm}0.13AU$), a statistically significance. Gagam-Jawoonaek could decrease erythema by UV exposure.

Some considerations for the analytical approaches to measure atmospheric ammonia: tests on a relative performance of a portable colorimeter against UV/VIS spectrophotometer (대기 중 암모니아 성분의 분석법 연구: UV/VIS 분광계를 이용한 간이비색계의 상대 성능평가)

  • In this study, we investigated the performance of a portable colorimeter used for the measurements of atmospheric ammonia against the more reliable method like UV/VIS spectrophotometer. For the purpose of this study, we tested a portable colorimeter and UV/VIS spectrophotometer for such basic analytical parameters as detectibility, reproducibility, linearity, and accuracy. According to our study, the performance of the colorimeter was fairly good to show a good agreement with the UV/VIS system. However, the performance of colorimeter suffered from both low and high concentration ranges: it was found to have very poor reproducibility at lower concentration (below $10\mu}g$), while its linear dynamic range was limited at the upper end (e.g., near $100{\mu}g$). If we assume that air samples are collected up to 30 liter, it implies that the colorimeter can be used to measure samples containing NH3 in the range of 500 ppb to 5 ppm (without the aid of diluting technique). Consequently, we recommend the use of such instrument with the awareness of its basic properties pertaining to the fundamental performance (such as its detectable range).

  • Petroleum visible markers (dye) have been used to distinguish different fuel classes and to prevent illegal mixing. It is difficult to recognize the real color of visible marker when the small amount of petroleum product was mixed in another fuel oil. In this study, we determined the two wavelengths (370 nm, 645 nm) which detect all Korean petroleum visible marker using UV/Vis spectrophotometer. Then we analyzed the visible marker using high performance liquid chromatography (HPLC) in two wavelength detectors. Also, we optimized the analytic method for petroleum visible marker in illegal mixed fuel oil.
