Effect of Seaweeds added in preparation of Tofu (두부제조시 해조류 첨가 효과)

  • This study was performed to investigate some quality characteristics of 새려 prepared from soybean milk and various seaweed(Undaria pinnatifida, Laminaria japonica, Porpyra tenera, Enteromorpha sp., Codium sp.) pulps in the ration of 9:1(v:v) with 20% MgCl2. The yields of tofu containing Undaria pinnatifida, Laminaria japonica, or Enteromorpha sp. increased but porphyra tenera, Codium sp. decreased in comparison with tofu prepared from whole soybean milk. The protein content of tofu containing Undaria pinnatifida, Laminaria japonica, Porphyra tenera, or Codium sp. increased but Enteromprpha sp. decreased in comparison with the tofu prepared from whole soybean milk. The content of Ca in Undaria pinnatifida, Porphyra tenera added tofu was higher than that of the tofu prepared from whole soybean milk or other seaweeds added tofu. In sensory evaluation the texture, color, taste of tofu were favored with the addition of sea mustard(Undaria pinnatifida) pulp than that of the tofu prepared from whole soybean milk or tofu prepared other seaweed. Tofu prepared was possible with adding 0.5~1.5% sea mustard to soybean milk but the feasible added amount level was 1% of sea mustard. The yields, protein Ca, and K content of tofu were increased by the more adding amount of sea mustard tan tat of the tofu prepared from whole soybean milk. The hardness values of 1% sea mustard added tofu were decreased than that of the tofu prepared from whole soybean milk ; on the other hand, elasticity, cohensiveness, gumminess and brittleness of tofu with sea mustard increased. The L and a values of tofu were lower and b values were higher with the addition of 1% sea mustard. The content of histidine, tyrosine, leusine, and phenylalanine were decreased but the other amino acid were increased in tofu prepared from 1% sea mustard pulp added to soybean milk. The saturated fatty acid and monoene fatty acid content of tofu were increased and C18:2, C18:3(${\gamma}$), C18:3, C20:5 of polyene fatty acid were slightly decreased in tofu prepared from 1% sea mustard pulp added to soybean milk.

Isolation of Marine Bacterium Decomposing Sea tangle (Laminaria japonica) to Single Cell Detritus (다시마(Laminaria japonica)를 single cell detritus로 분해하는 해양세균의 분리)

  • Seventy-one marine bacteria decomposing sea tangle (Laminaria japonica) into single cell detritus (SCD) were isolated from sea water, sea tangle, sea mustard (Undaria pinnatifida), sea urchin (Anthocidaris crassispina), star fish (Acanthaster planci), and turban cell (Batillus cornutus), among which 14 strains decreased cutting strength of sea tangle and had alginate-degrading activity. Marine bacterium No. 34 isolated from turban cell showed lowest cutting strength of sea tangle, strongest alginate-degrading activity, and produced high content of $5-10\;{\mu}m$ SCD from sea tangle. This strain was identified as Vibrio sp. based on morphological, physiological, and biochemical characteristics and named as Vibrio sp. YKW-34.

  • The characteristics of heavy metal biosorption on the seaweeds were investigated to develop a biological treatment technology for wastewater polluted with heavy metals. The heavy metal biosorption on seaweeds ranked in the tallowing order: U. pinnatifida$\geq$E. stolonifera$\geq$Laminaria sp.>G. amansii. The Pb was biosorbed in the range of $93{\sim}99%$, and the Cu and Cd were biosorbed in the range of $70{\sim}80%$ at the concentration of the heavy metal of $100mg/{\ell}$ respectively. The seaweed which was pretreated with $CaCl_2$ solution improved the biosorption of the heavy metals. The temperature and pH didn't affect the biosorption of heavy metals. The Langmuir isotherm reasonably fit the data of heavy metal biosorption compared to the Freundlich isotherm. The affinity of metals on the biosorption ranked in the following order: Pb>Zn>Cu>Cd. The biosorption efficiency of the heavy metals on the U. pinnatifida decreased in the multi-component rather than the single component. The heavy metals adsorbed on the U. pinnatifida were recovered using 0.3%-NTA. U. pinnatifida among the seaweed used in this work showed the best performance for the biosorption of the heavy metals.

Marine harpacticoids of genus parathalestris (Copepoda, Harpacticoida, Thalestridae) from Korea (한국산 Parathalestris속의 요각류 (요각아강, 갈고리노벌레목, Thalestridae과)의 기재)

  • Marine harpacticoid copepods of the genus parathalestris, which are suspected as a chief cause of pinholes or galls of cultivated seaweeds like Undaria pinnatifida, are studied taxonomically. As a result of examining the specimens from 23 localities in Korea since 1989, the authors confirmed sic species of the genus Parathalestris including a new species, P. parviseta n. sp., and four species new to korean fauna: P. bulbiseta Lang, 1965; P. verrucosa Ito, 1970; P. pacificus Tschislenko, 1971 and P. arelata Ito, 1972. A key to these six species of parathalestris was made out, with the illustrations for them.

  • Dietary value of eight species of benthic diatoms, considered as essential live food for larvae and spat of abalone, was assessed on the retarded spat (ca 4.4 m shell length). Survival of the spat, fed on Caloneis schroderi, Phaeodactylum tricornutum and Raphoneis sp. was over 93 % and significantly higher than that of others; spat fed on C かwがwi gained the highest daily growth of 112.6 mm, which was two times faster than that fed on Undaria pinnatifida Spat fed on natural or cultured diatoms of C. schroderi grew at rates, that were not significantly different. C. schroderi is recommended to restore the retarded growth of abalone spat.

  • The conversion of marine biomass to renewable energy has been considered an alternative to fossil fuels. Butanol, in particular, can be used directly as a fuel. In this experiment, the brown alga Undaria pinnatifida was selected as a biomass for biobutanol production. Hyper thermal (HT) acid hydrolysis was used as an acid hydrolysis method to produce monosaccharides. The optimal pretreatment conditions for U. pinnatifida were determined as slurry with 10% (w/v) U. pinnatifida content and 270 mM $H_2SO_4$, and heating at $160^{\circ}C$ for 7.5 min. Enzymatic saccharification was carried out with Celluclast 1.5 L, Viscozyme L, and Ultraflo Max. The optimal saccharification condition was 12 U/ml Viscozyme L. Fermentations were carried out for the production of acetone, butanol, and ethanol by Clostridium acetobutylicum KCTC 1724, Clostridium beijerinckii KCTC 1785, and Clostridium tyrobutyricum KCTC 5387. The fermentations were carried out using a pH-control. The optimal ABE fermentation condition determined using C. acetobutylicum KCTC 1724 adapted to 160 g/l mannitol. An ABE concentration of 9.05 g/l (0.99 g/l acetone, 5.62 g/l butanol, 2.44 g/l ethanol) was obtained by the consumption of 24.14 g/l monosaccharide with $Y_{ABE}$ of 0.37 in pH 5.0.

Use of Exo-polygalacturonase to Improve Extraction Yields of Alginic Acid from Sea Mustard (Undaria pinnatifida)

  • Exo-polygalacturonase (EPG) from Rhizopus sp. was applied to the extraction of alginic acid from sea mustard to increase extraction yield. EPG digestion was examined under distinct conditions within temperatures from $25^{\circ}C$ to 5$0^{\circ}C$, pH 5 to 9, and treatment times from 0 to 36 hr. The optimal conditions fur alginic acid extraction with EPG were: pH 7.0 at 3$0^{\circ}C$ for 24 hrs. The EPG hot water extraction yield was 3.4 times higher yield than hot water extraction alone. Using EPG to extract alginic acid from sea mustard should be considered a viable alternative to conventional extraction, with the advantage of reducing hazardous wastes such as strong acid and alkali solutions.

A Harpacticoid Copepod Parasitic in the Cultivated Brown Alga Undaria pinnatifida in Korea

  • We have examined harpacticoid copepods inhabiting the phaeophyte Miyok, Undaria pinnatifida (Harvey), on the southern coast of Korea and cultured in the laboratory pieces of Miyok frond heavily infested by frond-mining nauplii to identify the harpacticoid copepod to which those nauplii belong. Of the harpacticoids found in Miyok-washings, only Amenophia orientalis Ho and Hong and an unidentified species of the genus Scutellidium occurred consistently in all Miyok samples examined. Many females of both species carried egg sacs. In the cultures the frond-mining nauplii developed, in 15 days at$15^{\circ}C$, into cope-podid stages or adults that were all identified with Amenephia orientalis. It is therefore concluded that this species is the causative agent for the so-called pinhole disease of the cultivated Miyok on the southern coast of Korea, which is diagnosed by the appearance on the thallus of numerous pinholes occupied by developing nauplii.

Marine Algal Flora and Community Structure in Dokdo, East Sea, Korea (독도의 해조상 및 군집구조)

  • We investigated species composition and variation of benthic marine algae between 2003 and 2006 at Dokdo on the East Sea. The total number of benthic algae species identified was 96, which included 15 Chlorophyta, 30 Phaeophyta, 50 Rhodophyta and 1 Spermatophyta species. The representative species in this study were Ulva pertusa, Caulerpa okamurae, Codium adhaerens, C. frgile, Undaria pinnatifida, Ecklonia cava, Eisenia bicyclis, Dictyopteris undulata, Padina arborescens, Sargassum horneri, Hildenbrandtia rubra, Lithophyllum okamurae, Amphiroa dilatata, Corallina officinalis, C. pilulifera, Prionitis cornea, Grateloupia elliptica, Plocamium telgairiae, Chondrus ocellatus, Chondracanthus intermedia, Chondria crassicaulis, Polysiphonia morrowii, Melobesioidean algae and Phyllospadix sp. Wet weight biomass according to various depths ranged between 1,094.8 to $2,595.3\;g\;m^{-2}$ during the study period. Mean biomass at the investigated sites was greater in the 15m depth range than in the 5 and 10m depths. Vertical distribution was characterized by Ulva intestinalis, U. linza, Endarachne binghamiae, Bangia atropurpurea, Gloiopeltis furcata and Chondria crassicaulis at intertidal zone, Amphiroa dilatata, Corallina pilulifera, Gelidium amansii, Eisenia bicyclis and Ecklonia cava at 2m depth, Ecklonia cava, Eisenia bicyclis, Padina arborescens, Undaria pinnatifida and Sargasum horneri at 5m depth, Ecklonia cava, Sargassum horneri and Padina arborescens at 10m depth, Ecklonia cava, Myagropsis myagroides, Padina arborescens and Sargassum horneri at 15m depth. The R/P, C/P and (R+C)/P value were 1.67, 0.50 and 2.17, respectively.

Biosorption and Desorption of Heavy Metals using Undaria sp. (미역 폐기물의 중금속 흡탈착 특성)

  • The adsorption and desorption of Pb, Cd, Co, Zn, Cr, Co, Ni, and Mo on the waste Undaria sp. were studied. Except for Pb. the mono adsorption rate for all heavy metals were lower than that of the heavy metals mixed. However, the adsorption capacity of the heavy metals by 1g of biosorption, in mixed heavy metals increased According to FT-IR analysis of the biosorbent after heavy metal biosorption, the replacement of the functional group by the heavy metals ions could be confirmed and the inverted peaks became larger after heavy metals adsorption. The adsorption equilibrium of heavy metals was reached in about 1 hour. The equilibrium parameters were determined based on Langmuir and Freundlich isotherms. The affinity of metals on the biosorbent decreased in the following order: Pb>Cu>Cr>Cd>Co. The desorption rate decreased in the following sequence: NTA>$H_2SO_4$>HCl>EDTA. The desorption rate of heavy metals by NTA increased with increase in the concentration from 0.1 to 0.3% but the desorption rate became constant beyond 0.3%. Therefore, it represented that desorption rate of heavy metals was suitable under optimized condition ($30^{\circ}C$, pH 2 and 0.3% NTA solution) and was fast with 80% or more the uptake occurring within 10 min of contact time.
