미역의 섭취가 화학적으로 유도된 쥐의 대장암화 표지자인 Aberrant Crypt 형성 및 상피세포 분열에 미치는 영향에 관한 연구 (Effect of Fiber-Rich Sea Mustard Feeding on AOM-Induced Colon Aberrant Crypt Formation and Colonic Cell Proliferation in Sprague Dawley Rats)

  • The modulating effect of feeding sea mustard (Undarina pinnatifida), a fiber-rich seaweed, during initiation and post-initiation phases of colon carcinogenesis was investigated in Sprague Dawley rats. Four groups of animals were exposed to the two weekly injections of a chemical carcinogen, azoxymethane (AOM). Animals were placed on diet containing 15% sea mustard during initiation. post-initiation or initiation+post-initiation phase of carcinogenesis for 10 weeks, and colonic aberrant crypt formation and cell proliferation were compared to those of rats fed semi-synthetic control diet. Results showed that sea mustard feeding significantly reduced the numbers of both aberrant crypts and aberrant crypt foci. Also, labeling indices and proliferation zones were significantly reduced in the colon of the rats fed sea mustard diets. These results indicate that the diet containing sea mustard may decrease the risk of colon cancer development, and a part of such effect may be mediated through both the blocking of initiation and the suppression of cell proliferation in the colonic crypts, although precise mechanisms should be further examined.

Protective Effects of Bifidobacterium spp. on Experimental Colon Carcinogenesis with 1,2-Dimethylhydrazine

  • The protective role of Bifidobacterium spp. (B. breve K-110, B. breve K-111, and B. infantis K-525) isolated from the fecal samples of healthy Koreans was investigated on 1,2-dimethylhydrazine (DMH)-induced aberrant crypt foci(ACF) formation in mouse colon. In mice fed normal diet with DMH treatment, an average of 68.5 ACF/colon was formed, whereas in mice administered with B. breve K-110, B. breve K-111, and B. infantis K-525, the numbers of DMH-induced ACF decreased to 7.2, 10.9, and 6.6 ACF/ colon, respectively. The mean number of crypts/focus was not significantly altered. Fecal harmful enzymes, such as β-glucuronidase, tryptophanase, and urease, were effectively inhibited during the administration of these bifidobacteria to mice. These results suggest that bifidobacteria could prevent colon cancer.

Protective Roles of Mushrooms in Experimental Colon Carcinogenesis

  • There is epidemilogical evidence that the population with high fecal ${\beta}-glucuronidase$activity has greater risk of colon cancer than the population with low facal ${\beta}-glucuronidase$. This relationship was investigated by using the mouse-dimethylhydrazine colon carcinogenesis model and the fraction of Glucidum which is a .${\beta}-glucuronidase$inhibitor. Mice with low facal ${\beta}-glucuronidase$activity induced by consumption of the ether fraction of G lucidum had significantly fewer aberrant crypts(AC) after injections of 1, 2-dimethylhydeazine (DMH) than mice treated with DMH alone. The result supports the hypothesis that the inhibitor such as the ether fraction of G lucidum can protect an animal against the induction of colon cancer.

  • 본 연구는 로페라마이드로 처리된 흰쥐의 결장점막에 1,2-dimethylhydrazine (DMH)로 유도시 aberrant crypt foci(ACF)에 대한 길항 작용 효과를 갖는 초임계 열처리된 무 복합추출물의 화학적 암 예방 효과를 알아보고자 하였다. 초임계 열처리된 무 복합물추출물을 DMH유발 발암의 개시(12주 동안) 또는 개시 후 단계(9주 동안)에 식수(3.2 mg/ml)에 투여 하였다. 12주 동안 DMH와 함께 초임계 열처리된 무 복합추출물 투여군은 비정상적인 선와(전신경병변)의 총 수와 ACF의 수를 각각 35 %와 30 %로 크게 줄였다. 12주 동안 초임계 열처리된 무 복합추출물을 투여한 흰쥐의 결장에서 4가지 이상의 이상선와소(ACF)가 있는 ACF의 수가 현저히 감소하였다. 9주 동안 개시후 초임계 열처리된 무 복합추출물을 투여한 그룹에서, 4 이상의 이상선와소(ACF)의 수만이 현저히 감소하였다. 12주 동안 단독으로 투여한 초임계 열처리된 무 복합추출물은 ACF의 발생을 유발하지 않았다. 이러한 결과는 초임계 열처리된 무 복합추출물이 화학적으로 유도된 흰쥐의 결장 발암에 화학적 암예방 효과를 가지고 있음을 시사한다.

Grapefruit Juice Suppresses Azoxymethane-induced Colon Aberrant Crypt Formation and Induces Antioxidant Capacity in Mice

  • In the present report we determined the protective capacity of grapefruit juice (GJ) against molecular and cellular damage in azoxymethane (AOM) treated mice. Animals were daily administered GJ orally (0.8, 4.1, and 8.2 ${\mu}l/g$) for seven weeks, as well as intraperitoneally (ip) injected with AOM twice (weeks 2 and 3 of the assay). Control groups administered with water, with the high dose of GJ, and with AOM injected in weeks 2 and 3 were also included. The results showed a significant, dose-dependent protection of GJ on the number of colon aberrant crypts (AC) induced by AOM. The highest inhibitory effect was reached with the highest tested dose of GJ, decreasing ACF by 51% and 43% at weeks 4 and 7 of the assay. Regarding protein and lipid oxidation we also found a dose-dependent decrease caused with GJ in comparison with the increased levels produced by AOM. Therefore, our results established chemopreventive potential for GJ, and suggested effects related to its antioxidant capacity. Finally, we found that the tested agents induced neither micronuclei increase nor alteration in bone marrow cytotoxicity.

Soy Isoflavones Do Not Alter the Effects of Fructooligosaccharide on the Intestinal Ecosystem of Colon-Cancer Model Rats

  • This study sought to investigate any additive or interactive effects that soy isoflavones may have on the ecosystem of the gut, which is influenced by fructooligosaccharide (FOS) in colon-cancer model rats. Male Sprague-Dawley rats treated with 1,2-dimethylhydrazine were given experimental diets containing 0, 3, 6, or 9% FOS with or without 0.1% soy isoflavone for 12 weeks. In addition to the effects of FOS dosage on the gut ecosystem, dietary supplementation with soy isoflavone reduced the number of colonic aberrant crypts (ACs). The fecal weight, fecal pH, and gut transit time significantly decreased in a dose-dependent manner in rats fed FOS and the fecal concentration of bifidobacteria was higher in rats fed FOS than in control rats. The fecal output of total short-chain fatty acids, acetate, and propionate was significantly increased by the presence of FOS and was negatively correlated with the number of ACs, whereas the fecal output of butyrate showed no significant correlation with FOS dosage. The addition of soy isoflavone to the diet did not result in any significant differences in gut ecosystem parameters. Therefore, we conclude that the suppressive effect of soy isoflavone on ACs was not associated with the intestinal ecosystem, which was significantly altered by the dosage of FOS.

Protective Effect of Antler in Experimental Colon Carcinogenesis

The result supports the hypothesis that the inhibitor of ${\beta}-glucuronidase$ such as antler extract can protect an animal against the induction of colon cancer.

The Preventive Effects of Colon Cancer and Imflammatory Bowel Disease of Supercritical Heat-Treated Radish Extracts

  • There is a strong connection between the diet rich in antioxidants and the decreased incidence of inflammatory bowel disease and cancerous diseases. Diets that are rich in anti-oxidants particularly include fruits and vegetables containing the high amounts of vitamin A-E, carotenoids, and minerals. The supercritical heat-treated radish extracts of the research result had an inhibitory effect on the development of aberrant crypt foci (ACF), namely, preneoplastic lesions having a potential to become cancer cells and reduced the number of the aberrant crypt foci (ACF) consisting of four or more aberrant crypts (AC) having high risk to become tumors by about half. The supercritical heat-treated radish extracts can reduce the incidence of preneoplastic lesions having a high risk of developing cancer by about 28 %. DSS-treated mice developed symptoms similar to those of human UC, such as severe bloody diarrhea and weight loss. Supercritical heat-treated radish extracts, as well as sulfasalazine, suppressed colonic length and mucosal inflammatory infiltration. In addition, supercritical heat-treated radish extracts treatment significantly reduced the expression of pro-inflammatory signaling molecules through suppression both mitogen-activated protein kinases (MAPK) and nuclear factor-kappa B (NF-kB) signaling pathways, and prevented the apoptosis of colon. Moreover, supercritical heat-treated radish extracts administration significantly led to the up-regulation of anti-oxidant enzyme including SOD and Catalase.

The Effect of Pine Needle Powder on AOM-induced Colon Aberrant Crypt Formation and Antioxidant System in Fisher 344 Male Rats

  • Pine needles are known as a traditional medicine and their ingestion has been shown to be beneficial to human beings. Following induction of the neoplastic process in rats by azoxymethane (AOM), we determined the effects of pine needle supplementation on colon carcinogenesis and on antioxidant systems in the blood and liver. Five week old male Fisher 344 rats were injected with AOM (15 mg/kg) once a week for two weeks. After the second injection, 18 rats were randomly assigned into two groups and were fed a casein-based high-fat diet (120 g fat and 1 g cholesterol/kg diet) with or without pine needle powder (10%w/w). After 6 weeks, rats receiving pine needle powder showed a 40% lower incidence of the number of colonic preneoplastic lesions (aberrant crypts) and a 52% lower incidence of aberrant crypt foci (p<0.01). A significantly elevated level of erythrocyte catalase activity was observed in the pine needle supplemented group (17.4$\pm$1.1 vs. 24.5$\pm$1.5, p<0.01). Pine needle supplementation also increased liver glutathione peroxidase activity (7.5$\pm$0.6 vs. 14.6$\pm$0.6, p<0.01). Other antioxidant parameters such as erythrocyte glutathione peroxidase, liver catalase activity, and plasma total antioxidant potential (TRAP), showed no statistical differences between the two groups. Our data demonstrate that pine needle supplementation improves the antioxidant system and suppresses the formation of colonic preneoplastic lesions in AOM-treated rats. This result provides additional insights into the chemo-preventative properties of pine needles.

Modifying Effect of Indole-3-carbinol on Azoxymethane-induced Colon Carcinogenesis

  • Indole-3-carbinol (I3C), one of naturally occurring main components in cauliflower vegetables, is supposed to have a chemopreventive potential in experimental animals and humans. This study was investigated to examine chemopreventive effect of I3C on colon carcinogenesis induced by azoxymethane (AOM) using C57BL/6J mice. Mice were divided into three groups (10 or 9 mice/group). All mice were subcutaneously injected with AOM (5 mg/kg body weight, four times at weekly interval). After AOM treatment, animals of group 1 were fed by AIN-76A pellets as a basal diet. Animals of groups 2 and 3 were given I3C containing diets (100 and 300 ppm in diets, respectively) for 6 weeks until sacrifice. All mice were sacrificed at week 10 and the aberrant crypt foci (ACF) of the colonic mucosa were assessed after staining with methylene blue. Total numbers of ACF/colon in group 2 ($10.1{\pm}5.1$) or group 3 ($10.6{\pm}5.3$) were decreased compared to the values of group 1 ($14.4{\pm}10.2$). Among numbers of ACF formation, 5, 7, 8 and 10 ACF in group 2 and 3 were greatly different those of group 1. Total numbers of aberrant crypts (AC)/colon of group 2 ($20.1{\pm}10.1$) or group 3 ($22.0{\pm}10.9$) were decreased compared to the value of group 1 ($33.7{\pm}24.7$). Taken together, it suggests that I3C treatment may retard mouse colon carcinogenesis even after administration of AOM.
