Cytotoxic Constituents of Bombycis corpus (백강잠의 세포독성성분)

  • The acivity-guided fractionation on the MeOH extract of Bombycis corpus inoculated by Beauberia bassiana 101A led to the isolation of two steroids, 24-ethycholest-4-ene-3,6-dione (1) ergosterol peroxide (2), as active principles. Compounds 1 and 2 exhibited cytotoxicity against cultured human tumor cell lines, A-549, SK-OV-3, SK-MEL-2, XF-498 and HCT-15 with ED50 values ranging from 3.42 to $11.37{\;}\mu\textrm{g}/m$.

Effect of Several Sericultural Products on Blood Glucose Lowering for Alloxan-induced Hyperglycemic Mice (잠상산물의 장기간 투여에 따른 혈당강하효과)

  • This study designed to investigate the blood glusose lowering effect of sericultural products using allozan-induced byperglycemic mice. The administration of sericultural products did not affect body weight between normal mice and alloxan induced hyperglycemic mice. High blood glucose level in alloxan-induced mice group was maintained, during all the experimental period. In the 2-3 weeks, a significant decrease in the blood glucose level was observed in the group treated with sericultural products. Up to 5 weeks, blood glucose level of those group was maintained. Sericultural products treatment group have an activity for glucose tolerance test. Sericultural products showed the blood glucose lowering acivity for loading maltose and sucrose.

  • Roots of Rumex crispus (Rc) (Polygonaceae) has been used as therapeutic agents of acute and chronic cutaneous diseases, cathartics, fever and jaundice in folk medicines. Recently, Seeds of Rc has known as a digestive, an anticancer agent and a remedy of acute hepatitis, among many traditional folk medicines. So far it isn't reported about pharmacological effects of Rumecis Semen. The present study describes the preliminary evaluations of biological activities, anti-inflammatory activity (AA, Carrageenan) analgesic activity (writhing test), and hepatoprotective activities ($CCl_4$), of its methanol extract, ethyl acetate fraction and butanol fraction. Among them butanol fraction showed the highest activity in analgesic acivity.

Mutagenicity of the Essential Oils in Ames Test (Ames Test에 의한 정유의 돌연변이원성)

  • Mutagenic acivity of essential oils was tested using Salmonella typhimurium TA100 in the presence or absence of 59 fraction prepared from the mouse liver. Growth inhibitory effect of the oils on the bacteria was measured to warrant the mutagenic effect. Most oils were (round to be very strongly toxic against the bacteria at a high dose $(2,000{\mu}g/plate)$. At lower doses than this concentration, the Curcuma longa oil was found to be the most mutagenic with S9 fraction whereas it was not mutagenic without the fraction suggesting that this oil could undergo activation for the mutagenicity by cytochrome P45O. However, the mutagenicity of the Eugenia caryohpylata oil was disappeared under S9 fraction. Other oils obtained from Cinnamomum cassia, Chrysanthemum sibiricum, Paeonia moutan the flower of Artemisia princeps var. Orientalis, Allium sativum, were not mutagenic. This result suggested that antimutagenicity assay on the essential oil is necessary for the biological available substances.

  • Various N'-substituted anilino-N-methyl-N'-nitrosoureas(2a-n) were easily prepared from the reaction of substituted phenylhydraines $(3, 4-CH_3, {\;} 3-, {\;} 4-OCH_3, {\;} 3-, {\;} 4-F, {\;} 3, {\;} 4-Cl, {\;} 4-Br, {\;} 2-, {\;} 3-, {\;} 4-NO_2, 4-(NO_2)_2)$ with methyl isocyanate, followed by the nitrosation with 99% HCOOH and dry sodium lnitrite powder. Surprisingly, of these series of analogus, the anilino-nitrocosureas substituted with eletron-withdrawing nitro groups (2k-a) showed significantly low $ED_{30}$ values of $1.4-3.4 {\mu}g/ml.$ In addition, none of these copounds subtituted with electron-donating groups exhibited cytotoxicities.

  • To explore the effect of monosodium-L-glutamate(MSG) on CCI$_{4}$-damaged liver in Wister male rat, 5% MSG solution as drink water were administered after S.C. injection of 0.1 mg/kg CC1$_{4}$ twice a week for 4 weeks. After last administration of MSG, heptic glutathione(GSH) dependent system was assayed. It showed that MSG increased significanly hepatic glutathione(GSH) and glutathione peroxidase(GSH$_{px}$), but decreased glutathione-S-transferase(GST) acivity in normal rats. MSG increased significantly the GSH$_{px}$ and GST activities in rats with CCI$_{4}$-induced liver damage. These results indicate that decrease of GSH dependent systems in CC1$_{4}$ liver injury might be partially elevated by coadministration of MSG.

  • The methanol extract of Arachis hypogaea shell showed antioxidative and antimicrobial activity. The methanol extract was successively purified by solvent fractionation, silica gel adsorption column chromatography, Sephadex LH-20 column chromatography and octadecylsilane column chromatography. The purified active substances were isolated by high performance liquid chromatography, and were identified as 3-methoxy-4-hydroxybenzoic acid and 4-hydroxybenzoic acid by LC-MS and GC-MS. The amount of 3-methoxy-4-hydroxybenzoic acid and 4-hydroxybenzoic acid were 3.8mg and 9.8 mg per kg of shell, respectively.

Antioxidant Compounds from Twig of Morus alba

  • The MeOH extract of the twig of Morus alba L. (Moraceae) inhibited strong lipid peroxidation activity. In order to find out active principle from the plant, acivity-guided fractionation was performed and five antioxidant compounds were isolated. Their chemical structures were identified as 6-geranylapigenin (1), 6-geranylnorartocarpetin (2), resveratrol (3), oxyresveratrol (4) and quercetin (5) by physicochemical and spectrometric methods. Compounds 1-5 significantly inhibited lipid peroxidation in rat brain homogenate $(IC_{50}\;values\;of\;3.37,\;3.74,\;0.23,\;0.29\;and\;0.06\; {\mu}M,\;respectively)$.

A Study on Antimutagenic acitivities of from Puerariae Flos Extracts (갈화(葛花)의 항돌연변이(抗突然變異) 활성(活性)에 관한 연구(硏究))

  • Antigenotoxicity test (SOS chromotest), antimutagenecity test (Ames test), and antioxidant test (NBT method and xanthine-xanthine oxidase method) were carried out using water-soluble and methanolic extracts from Puerariae Flos. Against the mutagens MNNG and NQO, antigenotoxic activity of methanolic extracts were much more effective than that of water-soluble ones. When the methanolic extract was added to the certain concentration $(100{\mu}{\ell}/tube)$, antigenotoxic acivity against the mutagen MNNG was enhanced. Contrary to the water-soluble extract, the methanolic extract showed high antigenotoxicity against the mutagen NQO with increment of the extract. Against the mutagen MNNG with Ames test, antimutagenic activity of the methanolic extract at $300{\mu}{\ell}/tube$ was 96% as an inhibition ratio of revertant forming CFU/plate. The antioxidant activity of water-soluble extract was comparatively higher than that of the methanolic one.
