Pharmacokinetics of Ethanol After Oral Administration of Aspartate-Containing Compositions (Aspartate함유 복합성분과 Ethanol의 약물동태학적 거동)

  • The purpose of this work was to investigate pharmacokinetics of alcohol as a function of dose and time of administration of ethanol. The pharmacokinetics of alcohol 15 min after and before oral administration of aspartate-containing compositions to rats were also evaluated. The retention time of acetaldehyde, alcohol and isopropyl alcohol an internal standard in gas chromatogram was 3.6, 6.0 and 10.5 min, respectively. The maximum concentration of alcohol $(C_{max})$ and area under the blood concentration (AUC) were significanly increased as a function of ethanol dose in a nonlinear fashion. The significant diurnal variation of alcohol pharmacokinetics was also noted, showing fast metabolism and elimination when given orally in the night time. When APAP was given after administration alcohol (1g/kg) to rats, AUC and $C_{max}$ were increased when compared to alcohol only. However, AUC and $C_{max}$ were decreased when aspartate or standard complex compositions containing aceaminophen (APAP, 250mg). sodium L-aspartate(25 mg), dl-methionine (125 mg) and anhydrous caffeine (25 mg) was orally given by coupling malate/asparate shuttle in hepatocyte. The blood alcohol concentration profiles between aspartate and standard complex compositions were similar when given before or after administration alcohol (1g/kg) to rats. No significant difference of administration sequence was observed. However, it was noted that AUC and $C_{max}$ of standard complex compositions given before alcohol administration were significantly lower when compared with alcohol only. Based on these findings, dose, time of administration and composition of drugs to improve alcohol metabolism and elimination were considered to be important in the pharmacokinetics of alcohol. The administration sequence of drug compositions and alcohol might be also considerd.

  • Alcohol is a major risk factor for several diseases and excessive, long-term alcohol consumption are caues physical alteration-fatty liver, hepatitis, cirrhosis, breaking down, Wernicke-karsakoff's syndrome, weight loss, and poor immunity-in virtually all organ and tissue. This study was observed that liver, kidney, and stomach were altered in mouse by the effect of chronic alcohol administration. The mouse were sacrificed to obtain the tissue after mouse were orally injected with 25 % ethanol $18m{\ell}/kg/day$ for 120days. The tissue were stained by hematoxylin and eosin and then observed by light microscope. The results of this study were as follows : 1. The congestion was appeared in liver after 120days alcohol admistration. 2. The destruction of glomerulus were increased and the parietal cell of Bowman's capsule were swelled such as cuboidal cell after 120days alcohol administration. The congestion was appeared in alcohol administrated group. 3. The mucosa and gastric pit were destructed and the ulceration was appeared in stomach after 120days administration. The parietal cells and chief cells were damaged. Above results were shown that the tissue were damaged by chronic alcohol administration.

Aspect of Blood Pressure after Oral Administration of Alcohol : Effect of Dissolved Oxygen on Blood Pressure (알코올 투여에 따른 혈압의 변화 양상 : 용존산소량에 따른 혈압 변화)

  • Purpose: The purpose of this study was to elucidate the effect of dissolved oxygen in alcohol to blood pressure of healthy persons. Methods: Subjects (n=30) were randomized in a double blind crossover study to receive 120 mL, 240 mL, 360 mL of alcohol (Korean spirit, 19.59 v/v%, dissolved oxygen is 8 ppm and 20 ppm). Blood alcohol concentration (BAC) and blood pressure were measured applying Lion SD-400 Alcolmeter$^{(R)}$ Breathalyser and Tensoval duo control. Pharmacokinetic parameters ($C_{max}$, $T_{max}$, $AUC_{last}$, $K_{el}$, $V_d$, Clearance) were calculated using Winnonlin$^{(R)}$ program. The difference of parameters and values were analysed by student t-test using Microsoft$^{(R)}$ Excel program. Results: The $AUC_{last}$ values of 8 ppm group and 20 ppm group in 240 ml administration were $6.15{\pm}2.60cg{\cdot}min/ml$, $5.33{\pm}1.84cg{\cdot}min/ml$ (p<0.05) and those in 360 mL were $11.93{\pm}5.70cg{\cdot}min/ml$, $10.33{\pm}4.60cg{\cdot}min/ml$ (p < 0.01), respectively. Thus, the $AUC_{last}$ was significantly decreased. On the other hands, there was a significant change in systolic blood pressure (SBP) after alcohol administration. All measured value after 360 mL of alcohol administration was significantly decreased (p < 0.01). Conclusions: The dissolved oxygen in alcoholic beverage has no effect on blood pressures but the alcohol administration has an effect on blood pressure. Thus, SBP can be used as a biomarker of alcohol administration and utilized in PK/PD modeling of alcohol.

Effect of Herbal Composition, DTS20 on Alcohol Degradation and Anti-inflammatory Activity (생약 조성물, DTS20의 알코올 분해 및 항염증 효과)

  • These studies were conducted to investigate the alcohol degradation effects of the extract of herbal composition (DTS20) containing Viscum album L., Lycium chinense L., Inonotus obliquus and Acanthopanax senticosus H., on the alcohol administered mice. To investigate anti-hangover effect, alcohol and alcohol dehydrogensae (ADH) concentration of blood were measured after oral administration of ethanol. The administration of DTS20 (200-500 mg/kg) had beneficial actions toward alcohol degradation in acute alcohol treated mice model. The oral administration of DTS20 showed decreased gastric mucous membrane damage produced in ethanol treated mice. In addition, intraperitoneal administration of DTS20 showed anti-inflammatory effects in inhibition tests of vascular permeability produced by acetic acid. DTS20 also reduced the concentration of nitric oxide (NO), tumor necrosis factor (TNF)-${\alpha}$ in macrophages that were activated by LPS. These results demonstrate that DTS20 possesses potential to stimulate the alcohol degradation and inhibit the inflammatory effects in mice.

Effect of Acanthopanax senticosus Extracts on Alcohol Degradation and Anti-Inflammatory Activity in Mice (가시오가피 추출물의 알코올 분해 및 항염증 효과)

  • This study was conducted to investigate the effects of Acanthopanax senticosus extracts(ASE) on alcohol administered mice. The administration of Acanthopanax senticosus extracts(60 mg/kg) had beneficial actions toward alcohol degradation in acute alcohol treated mice. In the acute alcohol degradation experiment, serum alcohol concentration were lower 3 and 6 hours after taking ethanol(5 g/kg) in ASE treated mice. The oral administration of ASE showed decreased gastric mucous membrane damage produced in ethanol treated mice. In addition, intraperitoneal(i.p.) administration of ASE showed antiinflammatory effects in inhibition tests of vascular permeability produced by acetic acid. ASE also reduced concentrations of nitric oxide(NO), tumor necrosis alpha(TNF)-${\alpha}$ and interleukin(IL)-6 in macrophages that were activated by LPS. These results demonstrate that Acanthopanax senticosus extracts possesses the potential to stimulate alcohol degradation and inhibit inflammatory effects in mice.

Effect of Fermented Blackberry Drinks Formed from Radiation-induced Mutant on Alcohol Metabolism and Hangover in Rats (방사선 유도 돌연변이체 블랙베리 발효음료의 알코올 대사 및 숙취 억제 효과)

  • This study was designed to elucidate the effect of fermented blackberry drinks (BD) on alcohol metabolism and hangover in alcohol-treated rats. We showed that the administration of BD increased the activity of alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) in alcohol-treated rats. Moreover, the administration of BD reduced the serum alcohol and acetaldehyde concentrations in alcohol-treated rats. Furthermore, the administration of BD attenuated the levels of serum aspartate aminotransferase and alanine aminotransferase in alcohol-treated rats. Taken together, these results suggest that BD plays an important role in alcohol metabolism and liver function by reducing blood alcohol and acetaldehyde through the activation of ADH and ALDH in alcohol-treated rats and could be used as a functional anti-hangover drinks.

Effects of BCL oral administation and herbal acupuncture at BL18, BL19 on Liver function changes induced by Alcohol in the mice (죽력(竹瀝)(죽초액(竹酢液)) 경구투여(經口投與)와 간유(肝兪) 담유(膽兪) 약침(藥鍼)이 Alcohol 대사(代謝) 및 간기능(肝機能)에 미치는 영향(影響))

  • This dissertation was designed to evaluate the effect of BCL(refinded Bambusae Caulis in Liqua-men) oral administration and herbal acupuncture on alcohol metabolism and liver function. For this study. mice were damaged by a large quantity of alcohol and received treatment of either BCL 1 mg/kg in oral or BCL 250㎍/kg in herbal acupuncture-BL18 . BL19 bilateral. and then such parameters as GOT. GPT. catalase and superoxide dismustase(CuZn-SOD, Mn-SOD) were measured. The results of the experiments were summarized as follows. 1. Compared with control group, the proper degree of alcohol in serum was not significantly differ from oral administration group and herbal acupuncture group. 2. Compared with control group. the activity of GOT in serum was significantly reduced both oral administration and herbal acupuncture group. 3. Compared with control group. the activity of GPT in serum was significantly reduced both oral administration and herbal acupuncture group. 4. The activity of catalase in liver cell tissue, compared with control group. was not sigificantly affected either by oral administration and herbal acupuncture group. 5. The activity of CuZn-SOD in liver cell tissue was not significantly change in herbal acupuncture and oral administration group. The activity of Mn-SOD was significantly increased in oral administration group. while it was not the case in acupuncture group. In conclusion. we consider that BCL oral administration and herbal acupuncture is highly effetive in recovering alcohol metabolism and liver disfunction induced by alcohol.

Effect of Pepino Extract on Alcohol Metabolism in Rats (페피노 추출액이 흰쥐의 알코올 대사에 미치는 영향)

  • This study was conducted to investigate the effect of Pepino extract on alcohol metabolism in male Sprague-Dawley rats. When the rats were given Pepino extract 30 min before 60% alcohol (4 g/kg B.W) administration, alcohol concentration in blood was significantly reduced, but acetaldehyde concentration was not significantly different, compared with the control group after 3 hrs of alcohol administration. When the rats were given Pepino extract ($1^{\circ},\;5^{\circ},\;10^{\circ},\;&\;15^{\circ}$ Brix) 30 min before 60% alcohol administration, alcohol concentration in blood with $1^{\circ}$ Brix Pepino extract was 44% after 3 hrs of alcohol administration, compared with the control group. When the rats were given with $1^{\circ}$ Brix Pepino extract at 30 min before 60% alcohol administration, alcohol concentration in blood was significantly reduced after one hour and acetaldehyde concentration was reduced by 19% after 5 hrs of alcohol administration, compared with the control group. Glutamate-oxaloacetate transaminase and glutamate-pyruvate transaminase activities were not significantly different in all experimental groups, compared with the control group. These results suggest that Pepino extract can be effective in alcohol metabolism in the alcohol-treated rats.

Effect of Leaf Mustard (Brassica juncea) on Lipid Metabolism of Rat with Chroethanol Administration (갓(Brassica juncea) 첨가식이가 만성적인 알콜 투여시 흰쥐의 지방대사에 미치는 영향)

  • This study was undertaken to investigate the effects of mustard leaf diet on the lipid metabolism under chronic alcohol administration. Sprague-Dawley male rats were fed either AIN-76 diet(control), control with ethanol, AIN-76 diet plus 5% mustard leaf(mustard leaf), or mustard leaf with ethanol lot 30 days. On the 21st day, all of the rats were given an oral dose of ethanol and blood-ethanol concentration were monitored for the next 5 hours. Lipid and enzyme determinations in serum and liver were carried out after 30 days. The results obtained were summarized as following: 1) Supplementing 5% of mustard leaf did not recover the body weight loss due to chronic alcohol administration. 2) There were no significant differences in blood ethanol concentrations among the experimental groups. 3) Mustard leaf diet decreased the plasma total cholesterol, triglyceride levels increased due to the chronic alcohol administration, but not HDL-, LDL-cholesterol, and liver lipids. 4) Mustard leaf diet decreased ${\gamma}$ -GTP level increased by chronic alcohol administration. Overall, these data suggest that mustard lear can have a recovery function, which was not via ethanol metabolism on the symptoms of alcohol related diseases.(Korean J Human Ecology 1(1) : 94~102, 1998)

Effects of Gastrodiae rhizoma on the Liver Function and Alcohol Metabolism in Alcohol Treated Rats (천마 추출물이 알코올을 투여한 흰쥐의 간 기능 및 알코올 대사에 미치는 영향)

  • Purpose: This study was conducted to investigate the effect of freeze-dried Gastrodiae rhizoma powder (GR) on the liver function and alcohol metabolism in alcohol treated rats. Methods: The rats were administered various concentrations of GR (100 mg, 200 mg, 300 mg/kg B.W.) for 3 days 1 hour before 50% (v/v) ethanol (3 g/kg B.W.) administration. Two tests focusing on liver function and alcohol metabolism in acute alcohol treatment were carried out. Results: Glutamic oxaloacetic transaminase activity was significantly increased by alcohol treatment, and was decreased by 100 mg GR administration. Acute ethanol treatment led to significant increase in alcohol and acetaldehyde levels of serum and liver. However, 100 mg GR administration led to a significant reduction in increased alcohol level in the serum with decreased alcohol dehydrogenase (ADH) activity and increased acetaldehyde level in liver was significantly reduced by three levels. Conclusion: These results suggest that GR can be effective in enhancing liver function and alcohol metabolism in the alcohol-treated rats. Studies on the appropriate dosage of GR should further be developed to treat alcohol detoxification and stimulate liver function.
