한천분해효소(agarase)는 기초과학영역, 한천유래 고기능성 올리고당의 생산, 해조류를 이용한 바이오에너지 생산 등에 사용될 수 있다. 본 연구진은 2012년에 한천의 분류, 기원, 생산 및 응용에 관하여 총설하였다. 이에 본고에서는 2012년부터의 agarase 재조합 발현에 대해 총설하고자 한다. Agarase의 재조합 발현에 사용된 유전자는 Agarivorans 속(genus) 세균, Flameovirga 속 세균, Pseudoalteromonas 속 세균, Gayadomonas 속 세균, Catenovulum 속 세균, Microbulbifer 속 세균, Cellulophaga속 세균, Saccharophagus 속 세균, Simiduia 속, Vibrio 속 세균 등의 19종의 세균들에서 유래하였다. 47개의 재조합 발현된 agarase 중에서 α-agarase는 2개였고 나머지는 모두 β-agarase 였다. α-Agarase는 모두 agarotetraose (A4)를 생산하였고 β-agarase는 NA2부터 NA12까지 다양한 산물을 생산하였다. 최적온도는 25~60℃, 최적 pH는 3.0~8.5의 범위였다. 50℃ 이상의 최적 온도를 갖는 agarase는 14개로 이들은 한천을 가열한 후에 졸상태가 유지되는 온도에서도 활발한 활성을 보일 것이다. CBM (carbohydrate-binding module)의 조작 등의 인위적 돌연변이로 agarase의 열안정성 증가, 최적온도와 활성의 동시 증가에 관한 연구 사례도 있었다. 재조합발현의 숙주로 E. coli, B. subtilis, S. lividans, S. cerevisiae 등이 활용되었으며, agarase 유전자의 분비신호, 다른 생물의 분비신호 및 riboswitch가 agarase의 재조합 발현에 사용되었다. Agarase를 정제한 후에 아미노산 서열에 기반한 유전자 재조합 이외에도 게놈서열 파악과 유사성 비교를 통해 putative agarase와 메타게놈에서 유래한 agarase의 재조합 발현에 관한 연구도 있다. 이러한 연구들은 향후 agarase 및 agarase를 이용한 한천분해산물의 응용 분야 등에 활발하게 이용될 것으로 기대된다.
L-세린 탈수화효소(LSD)는 L-serine을 피루브산과 암모니아로 전환하는 반응을 촉매하는 iron-sulfur 함유 효소이다. 세균성 아미노산 탈수화 효소 중에서, L-serine에 대한 이들 특정 효소만이 촉매 부위에서 iron-sulfur cluster를 이용하는 것으로 보고되고 있다. 또한, 세균성 LSD는 구조적 특성과 도메인의 배열에 따라 네 가지 유형으로 분류된다. 현재까지, 이 효소들은 소수의 균주로부터 얻어진 LSD 효소에 대해서만 연구되었지만, 다양한 세균성 LSD의 촉매 메커니즘을 이해하기 위해서는 더 많은 자세한 조사가 요구된다. 본 연구에서, Mycobacterium tuberculosis H37Rv로부터 유래된 유형 II LSD (MtLSD) 단백질을 효소 동력학적 방법을 이용하여 생화학적 및 촉매적 특성을 규명하기 위해 발현 및 정제되었다. MtLSD에 대한 L-serine의 포화 곡선은 알로스테릭 협동성(allosteric cooperativity)을 나타내는 전형적인 S자형(sigmoid)의 특성을 보였다. 이때의 $K_m$과 $k_{cat}$ 값은 각각 $59.35{\pm}1.23mM$과 $18.12{\pm}0.20s^{-1}$로 계산되었다. 또한, 고정된 L-serine 농도 하에서 D-serine의 농도 대비 초속도에 대한 그래프는 비선형 쌍곡선 감쇠 형태를 보였고, $k_{cat}$ 값의 변화 없이 $30.46{\pm}5.93mM$의 겉보기 $K_i$ 값으로 D-serine에 대한 경쟁적 억제(competitive inhibition)를 나타내었다. 이들 연구는 MtLSD의 촉매 특성 및 기질 특이성에 관한 통찰력 있는 생화학적 정보를 제공한다.
Most eukaryotic peroxiredoxins (Prxs) are readily inactivated by a high concentration of hydrogen peroxide ($H_2O_2$) during catalysis owing to their "GGLG" and "YF" motifs. However, such oxidative stress sensitive motifs were not found in the previously identified filamentous fungal Prxs. Additionally, the information on filamentous fungal Prxs is limited and fragmentary. Herein, we cloned and gained insight into Aspergillus nidulans Prx (An.PrxA) in the aspects of protein properties, catalysis characteristics, and especially $H_2O_2$ tolerability. Our results indicated that An.PrxA belongs to the newly defined family of typical 2-Cys Prxs with a marked characteristic that the "resolving" cysteine ($C_R$) is invertedly located preceding the "peroxidatic" cysteine ($C_P$) in amino acid sequences. The inverted arrangement of $C_R$ and $C_P$ can only be found among some yeast, bacterial, and filamentous fungal deduced Prxs. The most surprising characteristic of An.PrxA is its extraordinary ability to resist inactivation by extremely high concentrations of $H_2O_2$, even that approaching 600 mM. By screening the $H_2O_2$-inactivation effects on the components of Prx systems, including Trx, Trx reductase (TrxR), and Prx, we ultimately determined that it is the robust filamentous fungal TrxR rather than Trx and Prx that is responsible for the extreme $H_2O_2$ tolerence of the An.PrxA system. This is the first investigation on the effect of the electron donor partner in the $H_2O_2$ tolerability of the Prx system.
Objective: Phellodendron amurense (P. amurense) and Humulus japonicus (H. japonicus) are closely involved in anti-oxidative response and increasing antioxidant enzymes activities. However, the effects of their extracts on development of preimplantation bovine embryos have not been investigated. Therefore, we investigated the effects of P. amurense and H. japonicus extracts on developmental competence and quality of preimplantation bovine embryos. Methods: After in vitro fertilization, bovine embryos were cultured for 7 days in Charles Rosenkrans amino acid medium supplemented with P. amurense ($0.01{\mu}g/mL$) and H. japonicus ($0.01{\mu}g/mL$). The effect of this supplementation during in vitro culture on development competence and antioxidant was investigated. Results: We observed that the blastocysts rate was significantly increased (p<0.05) in P. amurense ($28.9%{\pm}2.9%$), H. japonicus ($30.9%{\pm}1.5%$), and a mixture of P. amurense and H. japonicus ($34.8%{\pm}2.1%$) treated groups compared with the control group ($25.4%{\pm}1.6%$). We next confirmed that the intracellular levels of reactive oxygen species (ROS) were significantly decreased (p<0.01) in P. amurense and/or H. japonicus extract treated groups when compared with the control group. Our results also showed that expression of cleaved caspase-3 and apoptotic cells of blastocysts were significantly decreased (p<0.05) in bovine blastocysts derived from both P. amurense and H. japonicus extract treated embryos. Conclusion: These results suggest that proper treatment with P. amurense and H. japonicus extracts in the development of preimplantation bovine embryos improves the quality of blastocysts, which may be related to the reduction of ROS level and apoptosis.
본 연구에서는 북방전복 (Haliotis discus hannai)의 대용량 염기서열 분석을 통해 GST유전자의 전장 cDNA를 동정하였다. 북방전복 GST 유전자의 총 길이는 1669 bp로 672 bp의 ORF는 총 223개의 아미노산을 코딩하고 있으며 등전점은 5.69, 분자량은 25.8 kDa으로 예측되었다. 북방전복 GST아미노산 서열은 둥근전복과 지중해 담치와 같은 패류의 GSTA와 가장 유사성이 높았으며 계통수 분석을 통해 GSTA와 하나의 그룹을 이루었다. 북방전복 GST에는 GSTA의 특징을 갖는 두 site (N-말단의 G-site, C-말단의 H-site)가 보존되어 있었고 효소활성과 구조 유지에 중요한 잔기가 종간에 매우 보존되어 있었다. 북방전복 GST 유전자의 mRNA는 관찰된 모든 조직에서 발현하고 있었으며, 특히 외투막, 아가미, 간췌장, 소화관에서 높은 발현이 확인되었다. 북방전복의 GST는 비브리오균을 인위감염 시킨 전복의 간췌장에서 감염 후 1시간 뒤 발현이 급격히 증가했다가 3시간까지 증가한 뒤 감소하였고, 혈구세포에서는 감염 3시간 경과 후 발현 정도가 최고로 증가했다가 감소하였다. 따라서 북방전복 GST는 alpha class GST의 특징을 가지며 병원체 감염에 따른 면역반응 조절에 관여할 것이라 생각되며 병원균 감염에 따른 바이오마커로 활용가능 할 것이라 예상된다.

Background: Human leukocyte antigen (HLA) genes have been implicated in cervical cancer in several populations. Objectives: To study the predispositions of HLA alleles/haplotypes with cervical cancer. Materials and Methods: Clinically diagnosed and PAP smear confirmed cervical cancer patients (n 48) and age matched controls (n 47) were genotyped for HLA-A,-B,-DRB1* and DQB1* alleles by PCR-SSP methods. Results: The frequencies of alleles DRB1*04 (OR=2.57), DRB1*15 (OR=2.04), DQB1*0301 (OR=4.91), DQB1*0601 (OR=2.21), B*15 (OR=13.03) and B*07 (OR=6.23) were higher in cervical cancer patients than in the controls. The frequencies of alleles DRB1*10 (OR=0.22) and B*35 (OR=0.19) were decreased. Strong disease associations were observed for haplotypes DRB1*15-DQB1*0601 (OR=6.56; p< $3.5{\times}10^{-4}$), DRB1*14-DQB1*0501 (OR=6.51; p<0.039) and A*11-B*07 (OR=3.95; p<0.005). The reduced frequencies of haplotypes DRB1*10-DQB1*0501 (OR=0.45), A*03-B*35 (OR=0.25) and A*11-B*35 (OR= 0.06) among patients suggested a protective association. HLA-C* typing of 8 patients who possessed a unique three locus haplotype 'A*11-B*07-DRB1*04' (8/48; 16.66%; OR=6.51; p<0.039) revealed the presence of a four locus haplotype 'A*11-B*07-C*01-DRB1*04' in patients (4/8; 50%). Amino acid variation analysis of susceptible allele DQB1*0601 suggested 'tyrosine' at positions ${\beta}9$ and ${\beta}37$ and tyrosine-non-tyrosine genotype combination increased the risk of cervical cancer. Conclusions: Strong susceptible associations were documented for HLA alleles B*15, B*07, DRB1*04, DRB1*15, DQB1*0301, DQB1*0601 and haplotypes DRB1*15-DQB1*0601 and DRB1*14-DQB1*0501. Further, protective associations were evidenced for alleles B*35 and DRB1*10 and haplotypes A*11-B*35 and DRB1*10-DQB1*0501 with cervical cancer in South India.
4,4'-diaminodiphenylmethane (4,4'-MDA)는 일차방향족아민 (primary aromatic amino, PAA)류의 일종으로, 최근 유럽에서는 RASFF를 통하여 나일론 재질의 주방용 국자, 주걱 등에서 4,4'-MDA 이행 사례를 보고하고 있어 국내 유통 합성수지제 조리기구 중 4,4'-MDA 이행량 실태를 조사하였다. 4,4'-MDA의 분석을 위하여 0.3 ppb의 정량한계 및 87.6% 이상의 우수한 회수율을 나타내는 LCMS/MS를 이용한 분석방법을 확립하였다. 식품유사용매 물, 4% 초산, 20% 에탄을 및 n-헵탄에 대하여 4,4'-MDA 이행여부를 조사한 결과, 4% 초산 및 20% 에탄올인 경우에는 분석대상 100 품목 중 나일론 재질 14 품목에서, 물인 경우에는 나일론 재질 11 품목에서 식품유사용매로 4,4'-MDA가 이행되는 것으로 나타났으나, n-헵탄을 사용한 경우에는 1개 품목에서만 미량의 4,4'-MDA가 이행되었다. 이중 유럽연합 기준인 0.01 ppm을 초과하는 제품은 2품목이었다.
Phytocystatins, which are inhibitors of plant cysteine peptidases, are involved in the regulation of protein turnover and in the defense against insect pests and pathogens. Extensive searches in the Brassica rapa genome allowed the prediction of at least eight different phytocystatin genes on seven chromosomes in the B. rapa genome. Structure comparisons based on alignments of the all BrCYS ($\underline{B}$. $\underline{r}apa$$phyto{\underline{cys}}tatin$) proteins using the CLUSTALW program revealed conservation of the three consensus motifs known to interact with the active site of cysteine peptidases. According to the phylogenetic analysis based on the deduced amino acid sequences, the eight BrCYS proteins were divided into several clusters related to the orthologous phytocystatin. The predicted three-dimensional structure models of the eight BrCYS proteins demonstrate that all of these proteins are similar to the reported crystal structure of oryzacystatin-I (OC-I). Digital northern and RT-PCR analyses indicated that the eight BrCYS genes exhibit different expression patterns in B. rapa tissues and respond differently to abiotic stimuli. The differences in gene structure and expression between the eight BrCYS genes suggest that these proteins may play diverse physiological roles in B. rapa and may interact with cysteine peptidases through different mechanisms.
Bone morphogenetic proteins(BMPs) are regarded as members of the transforming growth $factor-{\beta}$ superfamily with characteristic features in their amino acid sequences. A number of studies have demonstrated the biologic activities of BMPs, which include the induction of cartilage and bone formation. Recently there was a attempt to overcome a limitation of mass production, and economical efficieny of rh-BMPs. The method producing PTD by using bacteria have advantages of acquiry a mass of proteins. Hences, a new treatment which deliver protein employed by protein transduction domain(PTD) has been tried. The purpose of this study was to evaluate the bone regenerative effect of TATBMP-2 and TAT-HA2-BMP-2 employed by PTD from HlV-1 TAT protein for protein translocation in the rat calvarial model. An 8mm calvarial, critical size osteotomy defect was created in each of 32 male Spraque-Dawley rats(weight $250{\sim}300g$). The animals were divided into 4 groups of 32 animals each (4 animals/group/healing interval). The defect was treated with TATBMP-2/ACS(Absorbable collagen sponge) (TATBMP-2 0.1mg/ml), TAT-HA2-BMP-2/ACS(TAT-HA2-BMP-2 0.1mg/ml), ACS alone or left untreated for surgical control(negative control). The rats were sacrificed at 2 or 8 weeks postsurgery, and the results were evaluated histologically. The results were as follows: New bone formation were not significantly greater in the TATBMP-2/ACS group relative to negative, and positive control groups. New bone was evident at the defect sites in TAT-HA2-BMP-2/ACS group relative to negative, positive control and TATBMP-2 groups. There were a little bone regeneration in TATBMP-2 groups. While, enhanced local bone formation were observed in TAT-HA2-BMP-2 group. But, The results was not the same in all rat defects. Therefore, further investigations are required to develop a method. which disperse homogenously, and adhere to target cells.
