Antioxidant and Antiatopic Effects of Refined Chaff Liquid Smoke (정제 왕겨초액의 항산화 및 항아토피 활성)

  • The refined chaff liquid smoke purified by the second or third filtering, supplemented with Artemisia extract or Perillae folium extract was investigated by measuring antioxidant and antiatopic activities. In DPPH and FRAP assays, three kinds of chaff liquid smokes had potential in antioxidant activity. Additionally, we assessed for antiatopic activity by using C57BLl6 mice with stimulation of dinitrofluorobenzene and observed that their ear swelling was alleviated by various fractions. Overall, all of the chaff liquid smoke reduced the response of swelling on ear epidermis. Especially, refined chaff liquid smoke supplemented with Perillae folium extract was most active than any other fractions.

Effect of Herb Extracts Mixed with Houttuynia Cordata on Antiatopic Dermatitis in DNCB-Induced BALB/c Mouse (DNCB 유도 BALB/c 생쥐에서 어성초 혼합 한방추출물의 항아토피 효과)

  • This study was evaluated the antiatopic activity of cream containing a herb extracts mixed with Houttuynia cordata in DNCB-induced BALB/c atopy mouse. The randomized complete block design was done by BALB/c mouse into two groups, of which the control group with atopy and the cream treatment group. It was found that each level of IgE and histamine in blood was significantly decreased in the cream treatment group, compared with the DNCB-induced atopy control group. When the cream was applied to the atopy mouse, it could be observed that its skin recovered to normal condition with the skin surface being clean and smooth without any horny tissue. The results suggest that the application of herb extracts mixed with Houttuynia cordata has an antiatopic activity through a inhibition of histamin emissions with reducing the levels of blood IgE in BALB/C atopy mouse.

Effect of Hwangryunhaedoktang on Contact Hypersensitivity and Passive Cutaneous Hypersensitivity in Mice

  • During the screening program to discover antiatopic agents from herbal formulas, we investigated the inhibitory effect of Hwangryunhaedoktang (HT) on passive cutaneous anaphylaxis and oxazolone-induced mouse ear dermatitis. HT significantly inhibited PCA reaction in mice at doses of 50 and 200 mg/kg with inhibitory activity of 31 and 53%, respectively. HT at concentration of 0.05 and 0.1% inhibited ear swelling by 23 and 46% at 16 days in oxazolone-induced mouse ear dermatitis. Both HT with anti without human intestinal microflora showed potent inhibitory activity on the ${\beta}$-hexosaminidase release induced by IgE. Based on these findings, HT may be a usable agent for skin disorder contact dermatitis.

Anti-atopic Effects of the Silkworm Feces Extracts in the NC/Nga Mice

  • This study was evaluated the antiatopic activity of silkworm feces extracts in DNCBinduced NC/Nga atopy mice. It was found that each level of IgE and histamine in blood was significantly decreased in the silkworm feces extracts treatment groups, compared with the DNCB-induced atopy control group. Inhibitory Effect of Fermented Red Ginseng against Passive Cutaneous Anaphylaxis Reaction and Scratching behaviors in Mice

  • To evaluate the antiatopic effect of Korea red ginseng (RG, steamed root of Panax ginseng CA Meyer, Family Araliaceae) fermented by Bifidobacterium longum H-1 (FRG), its inhibitory effect on passive cutaneous anaphylaxis (PCA) reaction and itching in mice was measured. FRG and its ingredient saponin fraction (FSF) potently inhibited PCA reaction and scratching behaviors. FRG at a dose of 200 mg/kg and FSF at a dose of 50 mg/kg significantly inhibited the scratching frequency by 45% and 47%, respectively. FRG and FSF also inhibited the degranulation and protein expression of tumor-necrosis $factor-{\alpha}$ and interleukin-4 of RBL-2H3 cells induced by IgE-complex. However, polysaccharide fraction of FRG (FPF) weakly inhibited it, compared with FSF. The inhibitory effect of FRG against PCA reaction and scratching behaviors more potently inhibited than that of RG. Based on these findings, FRG can improve allergic skin disorders atopic dermatitis by the regulation of $TNF-{\alpha}$, and IL-4 produced by mast cells and basophils and its degranulation.

Based on these findings, RG can improve allergic skin disorders atopic dermatitis and contact dermatitis by the regulation of $TNF-{\alpha}$, and IL-4 produced by mast cells and basophils and their membrane stabilization.

Recent Advances in Studies on Chemical Constituents and Biological Activities of Korean Black Ginseng(Panax ginseng C. A. Meyer) (흑삼의 화학성분 및 생리활성에 대한 최근 연구)

  • We have conducted a comprehensive literature review regarding the chemical constituents and biological activities of Korean black ginseng(Panax ginseng C. A. Meyer), three to nine times-steamed and dried ginseng, which shows strong black color through Maillared browning reaction. It has been reported that some chemically deglycosylated and transformed saponins are obtained from black ginseng as artifacts produced during intensive steaming. They have been known to be ginsenosides Rg3, Rg4, Rg5, Rg6, Rh1, Rh2, Rh4, Rk1 and Rk3, quite different from those of red ginseng, among which ginsenosides Rg3, Rg5 and Rk1 are considered to be major components. And also, black ginseng has been recently found to demonstrate anticancer, recovery from learning and memory damages, hypontensive, antidiabetic, antiobesitic, tonic and antiatopic activities, together with antioxidative and exercise performance improving activities, exhibiting their effects to be a little bit stronger than those of red ginseng. These findings suggest that black ginsng might play an important role in the development of promising functional foods and drugs from the viewpoint of the chemical composition and biological activities of black ginseng with a distinction from those of white and red ginsengs. In this review, the authors will survey and evaluate further functions of black ginseng with a focus on its physicochemical properties and biological activities.

Prunus Yedoensis Inhibits the Inflammatory Chemokines, MDC and TARC, by Regulating the STAT1-Signaling Pathway in IFN-γ-stimulated HaCaT Human Keratinocytes

  • Atopic dermatitis (AD) is an inflammatory skin disease commonly characterized by infiltration of inflammatory cells into skin lesions. Keratinocytes produce many chemokines that are involved in the pathogenesis of skin disorders. In particular, macrophage-derived chemokine (MDC/CCL22) and thymus and activationregulated chemokine (TARC/CCL17) are Th2-type cytokines. Serum MDC and TARC levels are increased in AD patients. In this study, we investigated the anti-inflammatory effect and mechanism of action of the active fraction from Prunus yedoensis bark. We evaluated their inhibitory effects on the AD-like inflammatory markers (MDC and TARC) and JAK-STAT pathway (STAT1) in HaCaT keratinocytes. The EtOAc fraction of the crude extract (80% EtOH) and the E5 sub-fraction potently inhibited the induction of MDC and TARC mRNA and protein at 50 ${\mu}g$/mL in HaCaT cells. In addition, the E5 sub-fraction inhibited the phosphorylation of STAT1 protein associated with IFN-$\gamma$ signaling transduction in a dose-dependent manner. Thus, P. yedoensis may have antiatopic activity by suppressing the inflammatory chemokines (MDC and TARC).
