Antioxidant and Antimicrobial Activities of Xanthium sibiricum (창이자의 항산화 활성 및 항균효과)

  • Xanthium sibiricum using extracts antioxidant activity and antimicrobial activity were analyzed and the following results were obtained. Antioxidant activity using DPPH as a VLC column fractionation experiments XS-5 (water fraction of Xanthium sibiricum) concentration in the 500 ${\mu}g/ml$ to 87.53 percent in the highest antioxidant activity was found, VLC fractionation in XS-3-2 (eluted with chloroform/methanol 7~10%) concentration of 500 ${\mu}g/ml$, showed the antioxidant activity of 78.72%. The antioxidant activity measured by PCL XS-1 (n-hexane fraction of Xanthium sibiricum) XS-5 (water fraction of Xanthium sibiricum) and 13.32 nmol open column XS-3-2 (eluted with chloroform/methanol 7~10%) showed antioxidant activity with 14.34 nmol. The antimicrobial activity against Candida albicans XS-2 (methlene chloride fraction fo Xanthium sibiricum) is the concentration of 500 ${\mu}g$/disc 1 mm of the clean zone was formed. XS-3 (ethanol fraction fo Xanthium sibiricum) in the antioxidant activity using DPPH and PCL had higher antibacterial activity was low.

  • There are now increasing evidences that free radicals and reactive oxygen species are involved in a variety of pathological events. Reactive Oxygen Species (ROS) are produced during normal cellular function. ROS lead to lipid peroxidation, massive protein oxdiation and degradation. Under normal conditions, antioxidant are substnaces that either directly or indirectly protect cell against adverse effect of ROS. several biologically important compound include ${\beta}$-carotene, taruine and flavonoids reported have antioxidant function. The various antioxidant either scavange superoxide and free radicals or stimulate the detoxification mechanisms within cells resulting in increased detoxification of free radicals formation and thus in prevention of many pathophysiologic processes. This study carried out to investigate the antioxidant activity of flavonoids, myricetin with other antioxidants, ${\beta}$-carotene and taurine on B16Fl0. In order to investigate the efficacy of antioxidant activity, we measured cell viability, antioxidant enzyme activity (SOD, GPX, CAT) and intracellular reactive oxygen intermediate (ROI). In this results, we show that these flavonoids with other antioxidant substrates are increased antioxidant activity level.

Chlorogenic Acid, an Antioxidant Principle from the Aerial Parts of Artemisia iwayomogi that Acts on 1,1-Diphenyl-2-picrylhydrazyl Radical

  • The antioxidant activity of Artemisia iwayomogi was determined by measuring the radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical. The methanol extract of A. iwayomogi showed strong antioxidant activity, and thus fractionated with several solvents. The antioxidant activity potential of the individual fraction was in the order of ethyl acetate > n-butanol > water > chloroform > n-hexane fraction. The ethyl acetate and n-butanol soluble fractions exhibiting strong antioxidant activity were further purified by repeated sitica get and Sephadex LH-20 column chromatography. Antioxidant chlorogenic acid was isolated as one of the active principles from the n-butanol fraction, together with the inactive components, 1octacosanol, scopoletin, scopolin, apigenin $7, 4^{I}$-di-O-methylether, luteolin $6, 3^{I}$-di-O-methylether (jaceosidin), apigenin methylether (genkwanin), 2, 4-dihydroxy-6-methoxyacetophenone $4-O-{\beta}-$D-glucopyranoside and quebrachitol. The antioxidant activity of chlorogenic acid was comparable to that of L-ascorbic acid, which is a well known antioxidant.

A study on the relations between the color intensity and the antioxidant activity of caramelization products (카라멜화 반응 생성물의 갈색도와 항산화 효과와의 관계)

  • The study was carried out to compare the relation between the color intensity and antioxidant activity of caramelization products using xylose(XY), glucose(GL). sucrose(SU), glucose+citric acid(GLCA), glucose+sodium citrate(GLSC), heated at 80, 120 and 140$\^{C}$ for 24hrs, respectively. The color intensity(absorbance at 490nm) of the browning mixtures increased as the browning temperature and time increased. But the degrees of color intensity of SU and GLCA changed very little. The hydrogen donating ability(HDA) of browning reaction products was generally enhanced as the browning temperature and time increased. When browning mixtures were heated at 80$\^{C}$, the HDA of GLGC was the highest, but the HDA of GLSC was the highest when heated at 120 and 140$\^{C}$. The antioxidant activities for the corn oil substrate containing the anhydrous ethanol extracts from the browning mixtures was inferior to that of SU, but was superior to that of GLCA. The relations among the color intensity, the antioxidant activity, and the hydrogen donating ability(HDA, reducing power) of the browning reaction mixtures were as follows: As the color intensity increased, the antioxidant activity decreased. The correlation coefficient of the color intensity and the antioxidant activity by regression equation was -0.73 ∼ -0.82. As the reducing power increased, the antioxidant activity decreased. The correlation coefficient between the reducing power and the antioxidant activity by regression equation was -0.98 ∼ -0.99. Therefore, the antioxidant activity of browning reaction mixtures seemed not correlated with the color intensity and the reducing power.

Antioxidant Activity and Protective Effects of Extracts from Sambucus williamsii var. coreana on t-BHP Induced Oxidative Stress in Chang cells (접골목 추출물에 의한 항산화 활성이 정상 간세포의 t-BHP 유발 산화스트레스에 미치는 영향)

  • In the present study, antioxidant activity and protective effect of extracts from Sambucus williamsii var. coreana stems (SWC) were evaluated on tert-butyl hydroperoxide (t-BHP) induced oxidative stress in human liver (Chang) cells. Antioxidant activities of the SWC extracts were determined by various radical scavenging activities, such as DPPH, ferric reducing antioxidant power (FRAP), 2,2'-azinobis-(3-ethybenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity and oxygen radical absorbance capacity (ORAC) assay. SWC extracts showed strong antioxidant effect on various assay. To determine the hepatoprotective effects of SWC on t-BHP induced oxidative damage, cell viability was measured using MTT assay. Pretreatment of SWC extracts showed increasing cell viability, decreasing ROS and restoring mitochondria membrane potential on t-BHP induced oxidative stress in Chang cells. Our findings suggest that SWC extracts may be considered a potential agent for therapeutic protective effect from oxidative stress through its antioxidant activity.

Effect of Five Korean Native Taraxacum on Antioxidant Activity and Nitric Oxide Production Inhibitory Activity (국내 자생 민들레 5종의 항산화 활성 및 Nitric Oxide 생성억제 활성)

  • The effect on the antioxidant activity and Nitric Oxide activity production inhibitory activity of Taraxacum has not been known. Therefore, phenolics and flavonoid contents were investigated from the ethanol extracts of five different Taraxacum species. The results showed that, among the five Taraxacum, T. hallaisanensis contains the highest total phenolic and flavonoid contents. When the antioxidant activity was measured by DPPH, $ABTS^+$ and reducing power activity, the free radical scavenging activity of T. hallaisanensis was also the highest among five Taraxacum species. However, measurement by CCK-8 assay in Raw264.7 cells indicated that the extracts of Taraxacum species have no effect on cell viability. Moreover, we also investigated the effect of Taraxacum species on NO scavenging activity in lipopolysaccharide (LPS)-stimulated Raw264.7 cells. The results clearly showed that Taraxacum species inhibited NO production, and the inhibitory effect of T. hallaisanensis was the strongest. The above results suggested that Taraxacum species affected the antioxidant and NO scavenging activity, and among the five species, antioxidant and NO scavenging activity assay of T. hallaisanensis was significantly higher than those of other four Taraxacum species. Therefore, T. hallaisanensis could be used as a potential drug with anti-oxidant and anti-inflammatory effect.

  • The study was aimed at evaluating the antioxidant and free radical scavenging activities of ethanolic extract and its fractions of Cleome rutidosperma. The antioxidant activity, reducing power, total phenolic content, total flavonoid content, superoxide anion scavenging activity, nitric oxide anion scavenging activity, in vitro antilipid peroxidation activity and in vitro non-enzymatic hemoglobin glycosylation were studied. The results obtained in the study indicate that Cleome rutidosperma is a potential source of natural antioxidant. All the parameters were found to be concentration dependent and increased with increasing amounts of sample. Flavonoids, phenolic compound like tannins, terpenoids may be responsible for the antioxidant activity of the plant. Variation of solubility parameters in various models may be attributed to non-linearity of activity of ethanol extract fractions models. Further investigation on the isolation and identification of antioxidant component(s) in the plant may lead to chemical entities with potential for clinical use.

Effect of Red Pepper Seed on Kimchi Antioxidant Activity During Fermentation

  • In this study, the antioxidant activities of red pepper seed kimchi extracts were examined. The extracts were evaluated by various antioxidant assays that included determinations of total phenolic and flavonoid contents, DPPH radical scavenging, superoxide anion radical scavenging, nitric oxide scavenging, superoxide dismutase (SOD) activity, metal chelating activity, and reducing power. All the kimchi extracts showed strong antioxidant activities by the tested methods. The 7% red pepper seed kimchi that was fermented for 6 days possessed the highest activity according to the performed tests. Yet, the highest scavenging activity was exhibited against nitrite, by extracts from kimchi for 0 day of fermentation rather than 6 days. All the activities for the kimchi fermented for 0 day with the addition of 7% red pepper seed increased markedly with an increase in concentration. With the exception of metal chelating and SOD activities, for the antioxidant properties assayed, the red pepper seed kimchi extracts were found to be highly effective.

Antioxidant Activity of Low Molecular Weight Laminarin Prepared with Gamma Irradiation (감마선 조사된 저분자 laminarin의 항산화 활성 연구)

  • In this study, it was investigated the antioxidant activity of laminarin degraded by gamma irradiation. Because the activities of antioxidants have been attributed to various mechanisms, different assay methods have been conducted and compared. 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activity, ferric reducing antioxidant power, and lipid peroxidation inhibitory activity of degraded laminarin were measured and compared with non-degraded. All of these results showed that the antioxidant activity of laminarin degraded by irradiation was increased depending on the absorbed dose. Therefore, gamma irradiation could be an alternative method for the preparation of degraded laminarin with higher antioxidant activity.

  • The antioxidant activities of six isoflavonoids isolated from Sophora japonica wood and bark were examined by DPPH (1,1-diphenyl-2-picrylhydrazyl) free radical scavenging method. This study was focused on the relationship between antioxidant activity of isoflavonoids and their chemical structures. From the results of this study, it could be concluded that the hydroxyl groups that linked at ring B and ring A in isoflavonoids have importance in the antioxidant activity. Additionally, the absence of the 2,3-double bond on the isoflavonoid enhances its antioxidant activity.
