천년초선인장으로부터 분리한 페놀성 화합물의 생리활성 효과 (Biological Activity of Phenol Compound from a Cactus Cheonnyuncho (Opuntia humifusa) in Korea)

  • 천년초는 한국에서 자생하는 손바닥선인장으로 예전부터 민간에서 약재로 사용되어져 왔다. 천년초를 70% 에탄올로 추출한 후 이를 헥산, 클로로포름, 에틸아세테이트, 부탄올, 물 층으로 순차 분획하여 항산화 활성이 우수한 에틸아세테이트 획분을 얻었다. 에틸아세테이트 획분을 가지고 TLC를 실시하여 10개의 fraction으로 분리하였고 이중 항산화 활성이 가장 우수하게 나온 3번째 fraction을 취한 후 silica gel chromatography를 사용하여 정제하였다. 총 10개의 분획을 채취한 후 항산화 활성이 우수한 4번째 분획을 채취하여 HPLC를 사용하여 정제하였다. 정제물은 EI-MS를 사용하여 분자량을 측정하였고 $^1H$-NMR과 $^{13}C$-NMR spectrum을 측정하여 검증한 결과 taxifolin으로 확인되었다. 이를 가지고 항산화 활성을 검증한 결과 $\alpha$-tocopherol, BHA와 비교하여 뒤지지 않는 효과를 보여주었고 항균 활성을 검증한 결과 그람 양성, 음성의 식중독균에 모두 benzoic acid보다 우수한 활성을 보여주었다.

Effects of Different Growing Regions on Quality Characteristics, Bioactive Compound Contents, and Antioxidant Activity of Aronia (Aronia melanocarpa) in Korea

  • The objective of this study was to determine the effects of different growing regions on quality characteristics, total bioactive compound contents, and in vitro antioxidant activity in aronia. Aronia grown in 3 different regions (Sangjoo, Ulju, and Youngcheon) in Korea was obtained and used fresh or as a freeze-dried powder. No statistically significant differences were observed for moisture, ash, crude lipid, and crude protein contents in aronia sampled from the 3 different regions. Aronia grown in Sangjoo had the highest total acid content and the lowest sugar content and pH value. Conversely, aronia grown in Youngcheon possessed the lowest total acid content and the highest sugar content and pH value. Aronia grown in Sangjoo possessed relatively high levels of polyphenols, flavonoids, and anthocyanins, as well as high antioxidant activity in comparison with aronia produced in other regions. Aronia grown in Youngcheon scored the highest for taste and overall acceptability in sensory evaluations, which may be related to the high sugar content and pH, and the low total acidity of the fruits. It is possible that higher sugar contents and pH, and lower total acidity in the aronia grown in Youngcheon result in more preferable sensory characteristics. However, they also contain relatively low levels of total polyphenols, flavonoids, and anthocyanins, and have low antioxidant activity as measured by 2,2-diphenyl-1-picrylhydrazyl and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging assays.

전통장류 유래 GB-07균주에 의해 생산된 Compound K 함유 발효인삼의 항산화 및 항염증 활성 연구 (Study on antioxidant and anti-inflammatory activity of compound K extract produced by Saccharomyces servazzii(GB-07) strain derived from traditional soy)

  • 인삼은 약용으로 수 천년 동안 소중하게 사용되어 왔으며 근대에 들어와 단일 성분의 분리와 각 성분에 대한 효능의 과학적인 연구가 활발히 이어져 오고 있다. 최근에는 인삼 성분 중에 Rg3, compound K 등이 주목 받고 있으며 기능성을 갖는 다양한 소재들이 연구 개발되어 제품화 되고 있다. 본 연구에서는 전통 장류에서 유래한 효모균 (Saccharomyces servazzii, GB-07)을 이용하여 인삼 추출물을 발효하여 compound K를 생산하고 기능성을 확인하여 상업적 이용 가능성을 확인하였다. Compound K 함유 ($20{\mu}g/g$ 함유) 인삼 발효 추출물을 이용하여 항산화 및 항염증 활성을 측정하였다. 발효 인삼추출물은 자유 라디칼 DPPH (di(phenyl) -(2,4,6-trinitrophenyl)를 농도 의존적으로 소거하였으며. macrophage(RAW 264.7 cell)에서 ROS의 생성을 농도 의존적으로 억제하였다. 또한 LPS에 의해 유도된 염증 사이토카인 $IL-1{\beta}$와 IL-6 그리고 $TNF-{\alpha}$의 생성을 억제하는 것으로 나타났다. 본 연구 결과는 향후 공정개발과 다양한 효능 시험을 통해 compound K를 함유한 제품의 개발을 통한 산업화의 가능성 시사하고 있다.

  • Indonesia is a tropical country having a temperature range of 25-35$^{\circ}C$ which can affect the skin and causes damages like aging. This aging process is due, at least, to free radical reactions. For this reason, many attempts had been done to find out creams containing natural antioxidant compound which have a potential of free radical scavenger. Kluwek, a fermented form of foot ball fruit or picung (Pangium edule.Reinw), had been proved to contain antioxidant compound in its methanol fraction oil to which antiaging cream was formulated. Stability evaluation was conducted for cream with Kluwek oil compared to base cream, including organoleptic (colour and odour), pH, viscosity, particle size, centrifugation test and flow characteristics either in room temperature (27$^{\circ}C$) or stress condition (4$^{\circ}C$ and 5$0^{\circ}C$) for 8 weeks continuously, and six times cycling test at 4$^{\circ}C$ and 5$0^{\circ}C$ every 24 hours. The results showed that cream with Kluwek oil and base cream were stable at temperature 27 and 4$^{\circ}C$, cycling test and centrifugation test, but not stable at 5$0^{\circ}C$. Free radical evaluation was done by Electron Spin Resonance and the result showed that cream with Kluwek oil had less free radicals compared to base cream.

Isolation and Identification of the Antioxidant DDMP from Heated Pear (Pyrus pyrifolia Nakai)

  • We evaluated antioxidant activities of heated pear juice (HPJ) exposed to 120, 130, and $140^{\circ}C$ for 2 hr. HPJ was partitioned using n-hexane, chloroform, ethyl acetate, n-butanol, and water. The ethyl acetate fraction treated at $130^{\circ}C$ for 2 hr showed strong antioxidant activity; thus, this extract was isolated and purified using silica gel column chromatography and preparative high performance liquid chromatography. The structure of the purified compound was determined using ultraviolet and mass spectrometry, $^1H$-nucelar magnetic resonance (NMR), and $^{13}C$-NMR. Antioxidant activities of the isolated compound were evaluated and compared with ${\alpha}$-tocopherol, ascorbic acid, and butylated hydroxytoluene (BHT) using DPPH and ABTS assays. The isolated compound was identified as 2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one (DDMP). The DPPH radical-scavenging activity ($IC_{50}$) of DDMP occurred in the following order: ascorbic acid ($45.3{\mu}g/mL$) > ${\alpha}$-tocopherol ($69.2{\mu}g/mL$) > DDMP ($241.6{\mu}g/mL$) > BHT ($268.0{\mu}g/mL$). Furthermore, DDMP showed strong ABTS radical-scavenging activity (569.0 mg AA eq/g).

Radical Scavenging Activity and Content of Cynarin (1,3-dicaffeoylquinic acid) in Artichoke (Cynara scolymus L.)

  • The contents of total phenol and total flavonoid of artichoke (Cynara scolymus L.) were measured. The antioxidant activity of the artichoke was evaluated based on its potential as a scavenging the ABTS radical. These results showed the antioxidant activity of artichoke has a close relationship with the total flavonoid content. The compound showing antioxidant activity was isolated from the artichoke by repeated column chromatography and recrystallization. Based on the spectrometric studies, the compound was identified as 1,3-dicaffeoylquinic acid, known as cynarin. The content of cynarin from heads and leafs of the artichoke determined by $C_{18}$ reversed phase HPLC (high-performance liquid chromatography) coupled with photodiode array detector was 10.15 and 0.67 mg/g, respectively. This compound showed potent antioxidant activities against DPPH and ABTS radicals ($EC_{50}$ = 14.09 and 28.85 ${\mu}M$, respectively).

감마선 조사가 결명자의 생육과 항산화 활성에 미치는 영향 (Effect of Gamma Ray on Germination, Growth and Antioxidant Activity of Senna tora)

  • Background: Senna tora is a flowering plant in the legume family Fabaceae. Its seeds are roasted and consumed as tea in Asia, to reduce inflammation in the liver and improve eyesight. Thus, it has been considered as an important medicinal crops in Asia. However, breeding trials to improve its genetic properties are rare. Mutation breeding by gamma ray is known to be an effective and highly successful approach for the generation of agronomically useful cultivars. Here we analyzed the effects of several dosages of gamma ray on the biological conditions of Senna tora seeds. Methods and Results: The germination rate and growth patterns of Senna tora were examined following irradiation with gamma ray at 100, 200, 300 and 400 Gy. The total phenolic compound contents and antioxidant activities of Senna tora were analyzed. Germination increased at 100 and 200 Gy in the M1 and M2 generations compared with that of the control (M0). The total phenolic compound contents and antioxidant activity of the seeds significantly decreased as the radiation dosage increased above 100 Gy in the M1 generation. Conclusions: Senna tora, irradiated with gamma ray at dosages 100, 200, 300, and 400 Gy, showed maximum germination rate at 200 Gy in the M2 generation. Plant height and leaf size gradually decreased with increasing gamma ray intensity in the M2 generation. The total phenolic compound contents decreased significantly at 400 Gy, and the related antioxidant activity was also decreased as the radiation dosage increased.

진세노사이드 Rd와 사포닌 대사물인 compound K의 항지질과산화 효과 (Anti-lipid Peroxdation Effect of Ginsenoside Rd and Its Metabolite Compound K)

  • To study on antioxidant effects in the liver of 40-week-old mouse, the sample were orally pretreated 5mg/kg/day for 5 days with red ginseng saponin components(total saponin, protopanaxadiol saponin, protopanaxatriol saponin, ginsenoside-Rd, ginsenoside-Re, compound-K) for 5 days. The ability of saponin to protect the mouse liver from oxidative damage was examined by determining the activity of superoxide dismutase(SOD), glutathione peroxidase(GPx) and the contents of glutathione, the level of malondialdehyde, The only protopanaxadiol among the ginseng saponin fractions was significantly increased the hepatic SOD activity(p<0.01). The red ginseng saponin induced a slight increase of GPx activity, especially ginsenoside Rd, compound K and protopanaxatriol treatments significantly increased its activity. The content of glutathione was significantly increased by total saponin, protopanaxadiol and ginsenoside Rd(p<0.01), but the oxidized glutathione level was lowered in all the red ginseng saponin. Finally, the level of malondialdehyde was significantly decreased by ginsenoside Rd and protopanaxadiol. In conclusion, protopanaxadiol and ginsenoside Rd among the saponin fraction were especially increased in the activity of hepatic antioxidative enzyme and decreased the lipid peroxidation that was expressed in term of MDA formation. This comprehensive antioxidant effects of red ginseng saponin seems to be by a certain action of saponin other than a direct antioxidant action.

  • A chemical investigation of the marine-derived fungi Aspergillus versicolor led to the isolation of a new aromatic polyketide (1), The structure was elucidated by spectroscopic analysis, and its radical-scavenging activity, reducing power, and inhibitory activity to lipid oxidation were investigated. Those activities of compound 1 were compared with standard antioxidants such as butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT), tertiarybutylhydroquinone (TBHQ), and ascorbic acid ($V_C$). Compound 1 showed antioxidant activity comparable to that of BHA, and siginificantly higher than that of BHT.

Volatile Compound, Physicochemical, and Antioxidant Properties of Beany Flavor-Removed Soy Protein Isolate Hydrolyzates Obtained from Combined High Temperature Pre-Treatment and Enzymatic Hydrolysis

  • The present study investigated the volatile compound, physicochemical, and antioxidant properties of beany flavor-removed soy protein isolate (SPI) hydrolyzates produced by combined high temperature pre-treatment and enzymatic hydrolysis. Without remarkable changes in amino acid composition, reductions of residual lipoxygenase activity and beany flavor-causing volatile compounds such as hexanol, hexanal, and pentanol in SPI were observed after combined heating and enzymatic treatments. The degree of hydrolysis, emulsion capacity and stability, 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity, and superoxide radical scavenging activity of SPI were significantly increased, but the magnitudes of apparent viscosity, consistency index, and dynamic moduli (G', G") of SPI were significantly decreased after the combined heating and enzymatic treatments. Based on these results, it was suggested that the enzymatic hydrolysis in combination with high temperature pre-treatment may allow for the production of beany flavor-removed SPI hydrolyzates with superior emulsifying and antioxidant functionalities.
