Effects of Garlic Extract on the Antioxidative Activity of Isoflavones (이소플라본의 항산화능에 대한 마늘 추출물의 영향)

  • This study was carried out to elucidate the effect of garlic extracts on the antioxidative activities of three isoflavones. All isoflavone samples showed greater antioxidative activity than butylated hydroxyanisole (BHA). In EDA (electron donating ability) tests, reducing power, SOD-like activity, and hydroxyl radical scavenging activity, all isoflavone samples with garlic extracts added showed significantly greater antioxidative effects than BHA. In conclusion, isoflavones have a potent antioxidative activity and garlic extracts have a big synergistic effect on this antioxidative activity.

Free radical scavenging effect and protective activity from oxidative stress of broccoli flowers and sprouts (Broccoli flower와 Broccoli sprout의 라디칼 소거능 및 산화적 스트레스 개선 효과)

  • In this study, the antioxidative effect and protective potential against oxidative damage of extract and fractions from broccoli were investigated under in vitro and cellular system. The methanol (MeOH) extracts of broccoli flowers and sprouts were partitioned as dichloromethane, n-butanol (BuOH) and aqueous fractions. The comparison of antioxidative effect of broccoli flowers and sprouts showed that broccoli sprouts exerted the more effective protective activity from 2,2'-azobis (2-aminopropane) dihydrochloride (AAPH)-induced oxidative stress in LLC-$PK_1$ porcine renal epithelial cell. In addition, the extract and fractions from broccoli sprouts showed strong scavenging effect of 1,1-diphenyl-2-picrylhydrazyl radical and the BuOH fraction exerted the strongest activity. Therefore, the BuOH fraction was evaluated as the most active fraction with strong radical scavenging activity among the fractions of broccoli flowers and sprouts. The present study suggests the antioxidative potential against free radical-induced oxidative damage of flowers and sprouts of broccoli. In addition, the BuOH fraction of broccoli is considered as the active fraction with antioxidative effect.

Antioxidative effects of Houttuynia cordata root on non-lipid oxidative damage (비 지질 산화손상에 대한 어성초 뿌리의 항산화 효과)

  • Houttuynia cordata root on non-lipid oxidative damage. The antioxidative efects of methanolic (MeOH) extract of Houttuynia cordata rooton non-lipid, including liposome oxidation, oxidation of deoxyribose, protein oxidation, chelating, scavenging,and 2'-deoxyguanosine (2'dG) oxidation were investigated. Houttuynia cordata root exhibited highantioxidative effect in a liposome model system. The inhibitory effect of MeOH extract on deoxyribosedamage exhibited antioxidative effect and it afforded considerable protection against damage to deoxyribose.In addition, MeOH extract at over 300extracts exhibited metal binding ability for hydrogen peroxide. Furthermore, the oxidation of 2'dG to 8-hydroxy-2-deoxyguanosine was inhibited by MeOH extracts, and scavenging activity for hydroxyl radicalexhibited a remarkable effect. The present results on biological model systems showed that MeOH extractswas effective in the protection of non-lipids against various oxidative model systems.

Antioxidative Effect of Persimmon Leaves (감잎의 항산화 효과)

  • The production of malondiadehyde(MDA) was significantly decreased when the methoanol extract of persimmon leaves was added to the system. The methanol extract of persimmon leaves was fractionated by using various solvents such as hexane, chloroform, ethylacetate and butanol. Among the above fractions especially the chloroform fraction, ethylacetate fraction revealed the strong antioxidative activities. The hot water extract of the persimmon leaves was less effective than tannin which was extracted from persimmon leaves in antioxidative activity.

  • Two ellagitannins, pedunculagin and 2,3-(S)-Hexahydroxydiphenoyl (HHDP)-D -glucose and three condensed tannins, (+)-catechin, (-)-epicatechin and procyanidin B-4 which were isolated from Rubus coreanum were evaluated for their antioxidative effects with 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and lipid peroxidation generation system mediated by addition of $H_2O$$_2$to rat liver homogenate (TBARS). The ellagitannins, 2,3-(S)-HHDP-D-glucose and pedunculagin, showed more potent antioxidative activities by DPPH and TBARS than condensed tannins.

Antioxidative Activities of Skipjack Meat Extract (가다랑어 자숙엑스분의 항산화성)

  • To develop cheaper and safer natural antioxidative substance instead of synthesized ones, the extract of skipjack meat was examined for the antioxidative effects and factors. Amino-N of skipjack meat extract was 15.3 mg/100g, and the extract apparently showed inhibitory effect on the oxidation of methyl linoleate. Antioxidative activity of extract revealed a tendancy to reduce slightly in proportion to hydrolyzing. When extract was dialyzed in distilled water, the outer fraction had a strong inhibitory effect on the oxidation of nethyl linoleate, while the inner fraction showed no effect. From the omission test and chemical analysis, the major antioxidative factors of skipjack meat extract were amino acids such as anserine, histidine, carnosine and alanine. As well nucleotides such as AMP, hypoxanthine seemed to act an auxiliary role in antioxidative effect of that.

Antioxidative Effect of Commercial Lecithin on the Oxidative Stability of Fish Oil (어유의 산화안정성에 미치는 레시틴의 산화방지 작용)

  • The antioxidative effects of three kinds of commercial lecithin in fish oil(EPA 25%, DHA 10%) were investigated through active oxygen method (AOM, hrs. at $97.8^{\circ}C$), Oven test, polymer test by gel chromatography and coloring test. Although there were difference of antioxidative effect among commercial lecithins, antioxidative effects of the lecithins added to the fish oil increased with increasing the concentration of lecithin. Lecithin III(acetone insoluble content 65%) had the greatest antioxidative effect and the addition of 1%, 5 and 10% enhanced the oxidative stability to 310%, 620% and 840%, respectively. The results also showed that the polymerization in presence of 10% lecithin III did not occur up to 10 hours at the AOM condition, and the degree of color(Gardner number) increased as storage time went by and was accerated at high temperature.

  • Objectives : The purpose of this study is to investigate the biological Effect of multi-herbal drug 'KOCO-P1' on mouse macrophage Raw 264.7 cells. Methods : Multi-herbal drug 'KOCO-P1' was composed of Ginseng Radix, Astragali Radix, Polygonati Rhizoma, Liriopis Tuber, and Scrophulariae Radix. Cytotoxicity and cytoprotective activity of K0C0-P1 was verificated by MTT assay. And antioxidative effect of K0C0-P1 against EtOH, Nicotine was inspected by Hydroperoxide assay. Results : K0C0-P1 showed no cytotoxicity on RAW 264.7 cells for 24, 48, 72 hours. KOCO-P1 at 200, 100, and 50 ug/mL reduced the production of H202 in Raw 264.7 cells by EtOH. KOCO-P1 at 50 ug/mL reduced the production of H202 in Raw 264.7 cells by Nicotine. Conclusions : KOCO-P1 could be supposed to have antioxidative effect on macrophage with no cytotoxicity.

Antioxidative and Antimutagenic Effects of Ganoderma lucidum Krast Extracts (영지버섯(Ganoderma lucidum Krast) 추출물의 항산화 및 항돌연변이 효과)

  • The antioxidative effect, antimutagenic capacity and inhibitory effect of cancer cell proliferation in ethanol extracts of Ganoderma lucidum were studied for suggestion of prevention and dietetic treatment of chronic diseases and development of antioxidative, antimutagenic and anticancer functional food by employing biological and biochemical assay. The IC/sub 50/ of MDA with BSA conjugation reaction, lipid peroxidation and scavenging effect on DPPH radical in ethanol extracts of Ganoderma lucidum showed 1026.21 mg/mL, 0.152 mg/mL and 0.412 mg/mL, respectively. So, the most effective antioxidative capacity in ethanol extracts of Ganoderma lucidum was the lipid peroxidation, among the method used this study. The indirect and direct antimutagenic effects of ethanol extracts of Ganoderma lucidum were examined by Ames test using Salmonella typimurium TA98 and TA100. The inhibition rates on indirect mutagenicity mediated by 2-Anthramine showed 100% in the Salmonella typimurium TA98 and 98.74% in the Salmonella typimurium TA100 and the direct mutagenicity mediated by sodium azide in Salmonella typimurium TA100 was 82.96%. But, the inhibitory effect on indirect mutagenicity mediated by 2-Nitrofluorene in Salmonella typimurium TA98 was low(7.81%). The inhibitory Effect of Ganoderma lucidum ethanol extracts on cell proliferation in Hela and MCF-7 by MTT test were 74.36% in HeLa and 73.90% in MCF-7 at the 0.50 mg/assay concentration and IC/sub 50/ were 0.163 mg/mL and 0.196 mg/mL respectively. From this result, it is suggested that Ganoderma lucidum is believed to have a possible antioxidative, antimutagenic and anticancer capacities.

A Study on the Physioactivities of Salicornia herbacea L. Grown in Sunchon Bay on Cell Viability and Antioxidative Effect in Cultured C6 Glioma Cells

  • To evaluate the physioactivity of Salicornia herbacea L. (SH), which are obtained from Sunchon bay as wild plants, an SH extract was prepared by freeze drying to obtain SH, and by cold drying to obtain SH. For the evaluation of their bioactivities, cell viability and antioxidative effect were measured. The XTT assay was adopted to measure cell viability after C6 glioma cells were treated with various concentrations of reactive oxygen species (ROS) and hydrogen peroxide ($H_2O_2$) for 8 hours. The DPPH-radical scavenging activity was also measured for the antioxidative effect. In this study, the $XTT_{50}$ value of $H_2O_2$ was determined at $30{\mu}M$ which was highly toxic based on the cytotoxic criteria by Borenfreund and Puerner. The protective effect of SH extract significantly increased cell viability compared with $H_2O_2$-treated group. Its antioxidative effect showed a significant DPPH-radical scavenging activity at concentrations of $1-100{\mu}g/mL$, while SH extract showed highly a DPPH-radical scavenging activity at only $100{\mu}g/mL$. From these results, $H_2O_2$ was highly toxic in cultured C6 glioma cells, and SH extract was effective in the prevention of cell damage by its antioxidative effect.
