Antioxidative and Anti-proliferative Effects on MCF-7 Human Breast Cancer Cells of Ikiyangyoung-Tang (익기양영탕(益氣養榮湯)의 항산화 및 유방암 세포주 생장 억제 효과에 미치는 영향)

  • Purpose: Breast cancer is the most common disease in Korean women. Despite remarkable improvements in treatment strategies against various cancer during the past 40 years, breast cancer still remains as one of the main causes of cancer mortality among women in the whole world. This study was carried out to investigate antioxidative and anti-proliferative effects on MCF-7 human breast cancer cells of Ikiyangyoung-Tang extract. Methods: We measured a content of polyphenol and flavonoid in the Ikiyangyoung-Tang extract, eliminative ability of DPPH radical, ABTS free radical and hydrogen peroxide, antioxidative effects of linoleic acid, cytotoxicity on MCF-7 human breast cancer cells. MCF-7 cells were cultured in Dulbecco's modified Eagle's medium/F12(DMEM/F12) supplemented with 10 % fetal bovine serum(FBS; Gibco) and antibiotics. Results : The extract of Ikiyangyoung-Tang contains polyphenol of 168.3${\pm}$12.8 ${\mu}$g/mg and flavonoid of 84.3${\pm}$3.4 ${\mu}$g/mg. Above results show profitable abilities of elimination of ${\alpha}$-${\alpha}$-Diphenyl-${\beta}$-picrylhydrazyl(DPPH) radical, ABTS free radical and hydrogen peroxide. Also, the extract of Ikiyangyoung-Tang strongly inhibits the proliferation of MCF-7 cells in a dose ependent manner. And. it has cytotoxicity on NIH3T3 cells. Conclusion : It can be concluded that Ikiyangyoung-Tang extract has an antioxidative effect and antiproliferative effect on MCF-7 human breast cancer cells.

Comparison of Physiological Activity in Different Parts of Dolsan Leaf Mustard (돌산 갓의 부위별 생리활성 작용의 비교)

  • Physiological activity in different parts of Dolsan leaf mustard (DLM) was investigated. To determine the physiological activity, DLM was crushed filtered centrifuged and then the supernatant was used as a sample. of 1%, 3% and 6%. By adding of improved root juice(20th day) with the concentration of 6% the cytotoxicity against HepG2 was the highest about 78.2%. And antioxidative effects in different parts of DLM was measured by using the DPPH method. Antioxidative effects was higher in all leaves than other parts. In particular antioxidative effects was the highest in leaves of traditional DLM at the 20th day of growth about 80.4%. In leaves of improved DLM at the 60th growth angiotensin-I converting enzyme inhibitory effect was the highest about 94.0%. Consequently there was not significant difference of physiological activity between improved and traditional DLM. However the cytotoxicity against HepG2 was the highest in roots of DLM. And the antioxidative and the ACE inhibitory effect in leaves of DLM were higher than those of other parts.

Antioxidative Effect of Typha orientalis L. Extract on the Oxidative Stress Induced by Cytotoxicity of Cadmium Sulfate (황산카드뮴독성의 산화적 손상에 대한 부들 추출물의 항산화 효과)

  • Objectives: This study was carried out to analyze the cytotoxicity of cadmium sulfate ($CdSO_4$) and the antioxidative effect of Typha orientalis L. (TO) extract on the oxidative stress induced by cytotoxicity of $CdSO_4$ in the cultured NIH3T3 fibroblasts. Methods: For this study, the cell viability and the antioxidative effects such as the inhibitory activity of lipid peroxidation (LP) and superoxide dismutase (SOD)-like activity and xanthine oxidase (XO)-inhibitory activity were assessed. Results: The cadmium sulfate significantly decreased cell viability in dose-dependently, and $XTT_{50}$ value was measured at $47.4{\mu}M$ of $CdSO_4$. The cytotoxicity of $CdSO_4$ was determined as highly toxic by Borenfreund and Puerner's toxic criteria. The butylated hydroxytoluene (BHT) as antioxidant significantly increased cell viability injured by $CdSO_4$-induced cytotoxicity in these cultures. In the protective effect of TO extract on $CdSO_4$-induced cytotoxicity, TO extract remarkably increased the inhibitory ability of LP and XO as well as SOD-like ability. Conclusions: From the above results, it is suggested that the oxidative stress is involved in the cytotoxicity of $CdSO_4$, and TO extract effectively protected $CdSO_4$-induced cytotoxicity by antioxidative effects. The natural component like TO extract may be a putative therapeutic agent for treatment of the toxicity induced by heavy metallic compound like $CdSO_4$ correlated with the oxidative stress.

Antioxidative Activity and Anti-inflammatory Effects on the Murine Macrophages of Methanol Extracts of Amphibians

  • Oxidative stress has been reported to be one of causes of neuritis. This study examined antioxidative activities of methanol extracts of six amphibian species known to be medicinal animals (Rana catesbeiana, R. coreana, R. rugosa, R. dybowskii, R. nigromaculata, and Hyla japonica) and investigated their effects of inhibiting nitric oxide (NO) production and cytotoxicity on the murine macrophage RAW264.7 cells. As inflammation is closely associated with reactive oxygen species, assays on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, xanthine oxidase inhibitory activity, superoxide anion radical scavenging activity and NO scavenging activity of the extracts of the six species were performed to investigate their antioxidative activity. The results obtained were as follows; All extracts showed antioxidative activity, and the activity of R. dybowskii was the highest in comparison among those. Anti-inflammatory effects of the extracts were also examined, the five extracts except that of R. rugosa did not show cytotoxicity for RAW264.7 cells at the maximal concentration ($1,000{\mu}g\;mL^{-1}$). Selectivity index, meaning NO scavenging activity compared to cytotoxicity, showed the highest level in the extract of R. dybowskii. These results will be very useful basic data for future studies on prevention and treatment of human diseases to understand the biological roles of amphibian extracts throughout the antioxidative or anti-inflammatory pathways.

  • This study was carried out to investigate the antioxidative activities of domestic plants grown in the area of Kangwon-do province. Through the examination of methanol extracts from 15 plants for radical scavenging effects using DPPH method, the extracts from Pinus densiflora Sieb. et Zucc. And Euphorbia humifusa Willd. Showed strong antioxidative activity. Because of its highest antioxidative activity among 15 domestic plants, radical scavenging effects of four different extract compartments from leaves of P.densiflora, n-Hexane, EtOAc, BuOH and $H_2O$ extracts. Were examined by DPPH method. Antioxidative activities of EtOAc and BuOH extracts were similar or even higher than that of natural (tocopherol) or synthetic antioBfdants (BHA) , suggesting that major fraction for the antioxidative activity of P. densiflora was the EtOAc and BuOH extract compartments.

  • Phenolics as antioxidant were added to linoleic acid to prevent lipid oxidation. Antioxidative effectiveness of them was measured by peroxie value at each 24hour interval in order to compare with 0.02% protocatechuic acid(PRL) and phloroglucinol(PHL) in linloleic acid, contrast tube at 37$^{\circ}C$for 96 hours blowing oxygen into specimen. Perocide values of oxidized linoleic acid, PRL, PHL for 96 hours were 78, 42, 30, From that the effect is more clearly demonstrated by NMR rather than UV and that the effect was dependent on the functional group and geometric molecular structure of phenolics.

  • The ethanol extracts of mixed vegetables (Bioactive V, BV), mixed fruits (Bioactive F, BF) and its liquid formulation (Chungpae Plus$\circledR$) were evaluated for their antioxidative and antigenotoxic activity. They were shown to possess the significant free radical scavenging effect against l,l-diphenyl-2-picryl hydrazine (DPPH) radical generation and were revealed to show the inhibitory effect of lipid peroxidation as measured by malondialdehyde (MDA) formation. Effect of Chestnut and Acorn on Lipid Metabolism, Antioxidative Capacity and Antithrombptic Capacity in Rats (밤과 도토리의 과육 및 내피가 흰쥐의 지방대사, 항산화능 및 항혈전능에 미치는 영향)

  • This study wa performed to investigate the effect of dired powder of chestnut and acorn on lipid metabolism, antioxidative capacity and antithrombotic effect in rats. Fifty-four male Sprague-Dawley rats weighing 199$\pm$17g were blocked into nine groups according to their body weight. Rats were raised with diets containing only flesh or flesh with inner skin of 5% and 10% dried nut powders for four weeks. Food intake, body weight gain, food efficiency ratio and organ weight were no different among the experimental groups. The plasma and liver lipid levels of all the nut diet groups were lover than those of the control group. The nut diets showed hypolipidenic effect in the plasma and liver. Plasma and liver thiobarbituric acid reactive substance (TBARS) concentrations were significantly decreased in all the nut diet groups. The plasma TBARS levels of the inner skin groups were significantly different from the control group dose-dependently. Superoxide dismutase(SOD) activity was significantly different among the experimental groups, and all the nut groups showed higher activity than the control group. There were significant differences in SOD activity between the chestnut and acorn groups and the chestnut groups showed higher erythrocyte SOD activity and the acorn groups showed higher liver SOD activity than the other groups. Whereas catalase and GSH-Px activities in the erythrocyte and liver of both nut groups showed a tendency to increase, they were not significantly different among the experimental groups. The bleeding time and whole blood clotting time tended to be extended by feeding both types of nut but they were not significantly different among the experimental groups. Production of TX $B_2$ and PG $F_{1{\alpha}}$ was no different among the experimental groups. These results suggest that chestnut and acorn diets have the effect of lowering plasma and liver lipid levels, inhibiting lipid peroxide formation and increasing antioxidative enzymes activity. Thus, it is plausible that chestnut and acorn could be recommended in the treatment and prevention of cardiovascular diseases.

Antioxidative Effect of Stachys japonica Miq Extract on Cultured NIH3T3 Fibroblasts Damaged by Ferrous chloride, Mordant (염화제일철 매염제로 손상된 배양 NIH3T3 섬유아세포에 대한 석잠풀 추출물의 항산화 효과)

  • The aim of this study was to examined the dermatoxicity of ferrous chloride (FeCl2) and the antioxidative effect of Stachys japonica Miq (SJ) extract on FeCl2-induced cytotoxicity. For this study, superoxide anion-radical (SAR)-scavenging and superoxide dismutase (SOD)-like abilities with cell viability were done. FeCl2 showed a significant decrease of cell viability in dose-dependent manner, and it was mid-toxic. The caffeic acid showed a significant increase of cell viability against FeCl2-induced cytotoxicity. In the protective effect of SJ extract on FeCl2-induced cytotoxicity, it showed SAR-scavenging and SOD-like abilities with a significant increase of cell viability. From these results, the cytotoxicity of FeCl2 is correlated with oxidative stress, and SJ extract effectively protected the cytotoxicity of FeCl2 by antioxidative effect. Conclusively, the natural resources like SJ extract may be a useful fundamental materials for the development of an alternative antioxidant.
