Antioxidative Activity of the Extracts from the Leaves and Fruits of Acer ginnala

  • The antioxidative effect of the extracts from the leaves and fruits of Acer ginnala against free radicals was studied by two different methods using DPPH radical-generating system, and hydroxyl radical-generating system $(Cu^{++}/H_2O_2\;system)$ which induces DNA strand breaking. Compared with well known antioxidative plants, green tea, Scutellaria baicalensis, the Acer ginnala extracts showed excellent radical-scavenging activity in DPPH radical-generating system and inhibited effectively hydroxyl radical induced-DNA strand breaking in a concentration-dependent manner in $Cu^{++}/H_2O_2$ system whereas the green tea extract stimulated the strand breaking at a low concentration. These results suggest that he extracts from the leaves and fruits of Acer ginnula could be good antioxidative agents.

  • Antioxidative effects of soybean paste(SP) on the lipid oxidation were studied with the model systems of ground cooked meat(GCM)-SP or ground cooked fish(GCF)-SF and model systems of linoleic acid mixture(LA)-SP powder (SPP) or LA-SP extract fractions during oxida-tion reaction. SP played a role as an antioxidative substance in the system employed especially in GCM-SP and the antioxidative activity was increased with the increase of SP addition in the system of GCF-SP during storage at 6$^{\circ}C$ SPP also exhibited some antioxidative activity during the oxidation reaction of LA. the activity was increased as the concentrations of SPP increased in the range from 0.1% to 0.5% Considerable antioxidative activities have been observed in both water soluble and lipid soluble fractions from SP on LA reaction system.

Effect of Maengjong-Juk (Phyllostachys Pubescens) Extract Coated Rice Diet on Antioxidative System of C57BL/6 Mice Fed Atherogenic Diet (맹종죽(Phyllostachys Pubescens)추출물 코팅쌀이 Atherogenic 식이를 섭취한 C57BL/6 마우스의 항산화 시스템에 미치는 영향)

  • To evaluate the antioxidative effect of maengjong-juk (Phyllostachys pubescens) extract coated rice in vivo system, maengjong-juk extract coated rice diets were fed to C57BL/6 mice for 16 weeks. Plasma total antioxidative capacity, hepatic lipid peroxidation, protein oxidation, activities of antioxidative enzymes and total glutathione content were measured. Plasma total antioxidative capacity was elevated significantly in maengjong-juk extract diets supplemented group in a dose dependant manner. Hepatic TBARS contents were significantly decreased in maengjong-juk extract diets supplemented group compared to high cholesterol group. Maengjong-juk extract coated rice diets suppressed the protein oxidation significantly in liver. Activities of hepatic antioxidative enzymes such as total SOD, CuㆍZn-SOD, Mn-SOD, GSH-Px and catalase activities of maengjong-juk extract coated rice diets were significantly higher than those of high cholesterol diet. Total hepatic glutathione content was significantly increased by maengjong-juk extract coated rice diets administration. According to this study, numerous antioxidative materials and phytochemicals containing in maengjong-juk extracts appear to protect antioxidative systems in C57BL/6 mice fed bamboo extract coated rice diet. Antioxidative Effects of Ethanol Extract Obtained from Rooibos Tea(Aspalathus linearis) and It's Application of Food (Rooibos Tea(Aspalathus linearis)에탄올 추출물의 항산화 효과 및 식품에 대한 응용)

  • This study was designed the antioxidative effect of the ethanol extract obtained Rooibos tea (Aspalathus linearis) on various kinds of oil, and examined the synergistic effect of Rooibos tea extract by addition of citric acid to the antioxidation activity and also investigate to antioxidation effect of the extract in food production and storage peroids. The antioxidative activity of 0.2mg of Rooibos tea extract was showed similar to same doses of $\alpha$-tocopherol, BHA and BHT in linolieic acid-ethanol system. The antioxidative effect of the mixture with 0.1% Rooibos tea extract on lard was more effective than that of the mixture with same doses of $\alpha$-tocopherol, BHA and BHT. The antioxidative effect of Rooibos tea extract was showed slightly effects on lard or soybean oil. Antioxidative effects of Rooiboe tea extract in addition of citric acid as synergist showed more effective in linoleic acid-ethanol system, but did not showed in the other oils. In the application of Rooibos tea extract to food prodctuction and storage period, the antioxidative effect was more effective in biscult, preparation added lard mixed with 0.1% Rooibos tea extract.

Antioxidative Effect of Persimmon Leaves (감잎의 항산화 효과)

  • The production of malondiadehyde(MDA) was significantly decreased when the methoanol extract of persimmon leaves was added to the system. The methanol extract of persimmon leaves was fractionated by using various solvents such as hexane, chloroform, ethylacetate and butanol. Among the above fractions especially the chloroform fraction, ethylacetate fraction revealed the strong antioxidative activities. The hot water extract of the persimmon leaves was less effective than tannin which was extracted from persimmon leaves in antioxidative activity.

  • Two ellagitannins, pedunculagin and 2,3-(S)-Hexahydroxydiphenoyl (HHDP)-D -glucose and three condensed tannins, (+)-catechin, (-)-epicatechin and procyanidin B-4 which were isolated from Rubus coreanum were evaluated for their antioxidative effects with 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and lipid peroxidation generation system mediated by addition of $H_2O$$_2$to rat liver homogenate (TBARS). The ellagitannins, 2,3-(S)-HHDP-D-glucose and pedunculagin, showed more potent antioxidative activities by DPPH and TBARS than condensed tannins.

  • The antioxidative activity of each molecular weight (MW) fraction of Maillard reaction products prepared from a D-glucose and glycine system, i. e., MW below 1000, MW between 1000 to 3000 and MW above 5000, nondialyzable melanoidins, reduced melanoidins and ozone-treated melanoidins were estimated in a linoleic acid-aqueous system. The antioxidative activity ana reducing ability of Maillard reaction products increased with increasing molecular weight and color intensity. Maillard reaction products of MW above 1000 showed obvious antioxidative activity and reducing ability, whereas the MW fraction below 1000 showed only weak activities. When nondialyzable melanoidins were reduced with sodium borohydride, their color intensity and reducing ability remarkably decreased, but their antioxidative activity did not decrease so much.

  • To investigate and evaluated the scavenging and antioxidative effects of various flavonoids on paraquat induced toxicity, in vivo and vitro tests of eight flavonoids (catechin, epocatechin, flavone, chrysin, apigenin, quercetin, morin and biochanin A) were carried out. The generation of reactive oxygen substances(ROS) in PMS-NADH system $H_2O_2$ induced hemolysis and lipidperoxidation to blood, NADPH dependent lipidperoxidation to liver and lung microsome by paraquat were studied.The results are summerized as follows; 1) In the concentration ranges from 3.3 to 9.8$\mu$M of catechin,epicatechin, quercetin and biochanin A removed the 50% of DPPH radical scavenging effects. 2) In the concentration ranges from 0.60 to 1.86 mM of catechin, epicatechin, quercetin and biochanin A showed the inhibitory and antioxidative activity on superoxide anion which gernerated in PMA-NADH system. 3) In the concentration ranges from 0.12 to 0.49mM of catechin, epicatechin, quercetin and biochanin A showed the inhibitory and antioxidative activity on H202 which generated in PMA-NADH system. 4) In the concentration ranges from 0.6 x10$^{-5}$ to 6.3 x 10$^{-5}$mM of catechin, epicatechin, flavone, chrysin, quercetin and morin showed the inhibitory and antioxidative activity on $H_2O_2$ induced hemolysis to blood 5) All flavonoids tested exhibited inhibitory and antioxidative effects on paraquat induced liver and tung microsomal lipidperoxidation. Therefore, all flavonoids evaluated showed the useful compounds for scavenger and antioxidant on paraquat induced toxicity.

Antioxidative Effect of Crude Anthocyanins in Water-in-Oil Microemulsion System

  • Antioxidative ability of anthocyanins in water-in-oil microemulsion was examined. Microemulsion was prepared by solubilizing crude anthocyanins extracted from grape skin (Cambell early) in organic solvent (hexane) containing anionic surfactant [bis (2-ethylhexyl) sodium sulfosuccinate, AOT] and linolenic acid (10%, w/v). Lipid oxidation significantly decreased with increasing concentration of anthocyanins ($5-20\;{\mu}M$) at micellar phase, and increasing micelle size ($Wo=5-20\;{\mu}M$). At given micelle size (Wo=10), lipid oxidation decreased as number of micelles decreased. These results indicate antioxidative ability of anthocyanins is critically affected by water core and micelle structure formed by surfactant. Interactions between AOT and anthocyanins decreased antioxidative ability of anthocyanins. Antioxidative ability of anthocyanins significantly increased when ${\alpha}$-tocopherol was added into organic phase. This indicates of synergism between the two antioxidants.

Effect of Soybean Saponins on Aflatoxin B1-induced Mutagenicity (대두 사포닌이 Aflatoxin B1으로 유도된 세포돌연변이에 미치는 영향에 관한 연구)

  • Free radicals formed during the metabolism of environmental chemicals are known to induce mutagenicity, while different types of antioxidants suppress this event. The purpose of this study was to determine the antioxidative and antimutagenic effects of soybean saponins, and to examine the relationship between these two effects for the elucidation of mechanisms involved in the anticarcinogenicity of soybean saponins. Also, antioxidative and antimutagenic effects of soybean saponins were compared with those of kinown antioxidants. For the measurement of antioxidative capacity, soybean saponins, L-ascorbic acid, $\alpha$-tocophoerol, and BHT at concentrations between 005 and 1.0mg/ml were tested for their ability to donate hydrogens and to reduce the formation of thiobarbituric substances(TBARS). Antimutagenic activity was examined using the Ames salmonella test system at concentrations of 600, 900 or 1200ug/ml. Study results showed soybean saponins and all of the other antioxidants tested possessed dose-dependent antioxidative activities. The ability of hydrogen-donation to DPPH was in the order of L-ascorbic acid>$\alpha$-tocopherol=>BHT>soybean saponins. TBARS formation was also inhibited by these compounds, in the order of BHT>$\alpha$-tocopherol=L-ascorbic acid>soybean saponins. Soybean saponins and other antioxidants also showed antimutagenicity in a dose-dependent manner. Especially, soybean saponins and BHT were excellent antioxidants compounds, inhibiting near 80% of the mutagenic effects at a concentration of 1200ug/ml. The correlation coefficients between antioxidative capacity and antimutagenicity for each compund was statistically significant at p<0.05. These results indicate that soybean saponins possess antioxidative and antimutagenic capacities. Also, antimutagenicity of saponins and other antioxidats is partly due to their antioxidative activities.
