Among the 34 strains of Nicotinophiles selected in the previous experiments, strain NCT27 identified with Pseudomonas putida and strain NCT30 identified with Arthrobacter oxydans biotype nan thus were Investigated for optimization of growth conditions for nicotine degradation and other cultural characteristics. The compositions of optimized medium were to be following: $KH_2PO_4$ 2.Ogr, KCI 5.Ogr, $MgSO_4$.$7H_2O$ 20mg, $MnSO_4$.$6H_2O$ 0.2mg, $FeSO_4$.$7H_2O$ 1.Omg, Col$^{++}$ (Cobalt Acetate),2.O$\gamma$, N1$^{++}$ (NiSO4,6H2O) 0.5$\gamma$, and yeast extract 80mg per liter. The optimum initial concentrations of nicotine for growth were 0.4% for Pseudomonas and 0.1% for Arthrobacter, respectively. The optimum temperature and pH were 3$0^{\circ}C$ and 7.0 for both of strains. The pH of culture medium of Pseudomonas was changed from acidic condition to basic one in going from the logarithmic growth phase to the stationary growth phase. In contrast with Pseudomonas, it remained constant in case of Arthrobacter. The growth of Arthrobacter was completely inhibited in the nicotine concentration of 0.7&. However, Pseudomonas could grow even in the nicotine concentration of 1.0%. Moreover, it could grow successfully in the tobacco extract media as well as media containing carbon and nitrogen sources other than nicotine. The maximum rates of nicotine degradation were to be 1.22 gr./hr./liter for Pseudomonas and 0.186 gr./hr./liter for Arthrobacter, respectively.
Although the submentovertex radiograph and surface EMG are not often used due to the difficulty of interpretation, they are accepted as useful diagnostic and analytic aids for skeletal asymmetry. There have been reports which state that they were also useful for the evaluation of vertical skeletal relations. The purpose of this study was to evaluate the correlations between EMG data, measurements from submentovertex radiographs, facial types and facial asymmetry following examination of 60 asymmetric patients. The radiographic corpus length were greater in the nonaffected sides (p<0.001), gonion to interspinosum axis were greater in the affected sides and the mandibular condyle and gonion were located more anteriorly in the non-affected sides than in the affected sides but not significant (p=0.07). The activity of the anterior temporal muscle in rest position was higher in the affected sides than in the non-affected sides (p<0.01). The activity of the masseter muscle at maximum clenching was found to be nonsignificant but it was higher in the affected sides than in the non-affected sides (p=0.09). There was positive correlation between facial index and the intercondylar axes angle (p<0.01). There was positive correlation between masseter muscle activity in maximum occlusion and facial index in the affected and non-affected sides (p<0.05). The results demonstrate that the submentovertex radiograph and EMG can provide useful information for the evaluation of horizontal and vertical skeletal relations.

In this study, antimicrobial activity of oriental herbal medicine extract (OHME) was tested for some organisms and the preventive effects of OHME for the colonization of Campylobacter jejuni on epithelium of small intestine were examined in Korean native broiler chickens fed a forage added 1.0% OHME. The isolated Campylobacter spp were biotyped, serotyped and the susceptiblility of isolates to antimicrobial agent were examined. The growth of Staphylococcus aureus was inhibited in 0.25% OHME. C. jejuni and C. coli were inhibited in 0.1% OHME, and Salmonella spp, Lactobacillus acidophilus and Escheichia coli 0157 were inhibited in 2.0% OHME. For the application of forage added 1.0% OHME in broiler chicken farm, the frequency of Campylobacter spp from feces, liver and spleen sample of chickens were examined during 2 weeks interval. The frequence of Campylobacter spp in feces from chickens fed assorted forage (control group) was increased from 25% in first week to 75% in seventh week. But the frequence of Campylobacter spp in feces sample from chickens 134 forage added OHME was slightly reduced from 25% in first week to 15% in seventh week. The frequence of Campylobacter spp in liver, and spleen was 13.7% and 10% respectively after seventh week in control group, but the Campylobacter spp was not isolated after fifth week in live and spleen from chickens fed forage added OHME. Isolated 56 strains of thermophilic Campyiobacter from Korean native chickens was classified as C. jejuni (76.7%), C. coli (214%) and C. laridis (1.6%). The majority of 43 isolates of C. jejuni was classified on biotype I (60.4%), II (30.2%). Most of 12 isolates of C.coli were biotype I (83.3%). Isolated 31 strains C. jejuni of showed 11 different serotype, and serotype 36 (18.6%), 17 (13.9%)were most frequent. Isolated 10 strains of C. coli showed 5 different serotypes and serotype 31 (33.3%) and 21 (25%) were relatively common. Isolated Campylobacter spp were highly susceptible to nalidixic acid, amikacin, gentamicin, colistin and chloramphenucol.

This study was carried out to evaluate toxicities of 3 registered insecticides to greenhouse whitefly(GWF), Trialeurodes vaporariorum and sweetpotato whitefly(SWF), Bemisia tabaci, B-biotype. Insecticide activities were evaluated by testing systemic action, residual effect in the laboratory, and control efficacy in the greenhouse. All experiments were tested at the recommended concentration(RC), half and a quarter concentrations of RC of each insecticides. Acetamiprid showed 45%, 42% ovicidal effect to greenhouse whitefly and sweetpotato whitefly at 40 ppm, respectively. Acetamiprid showed more than 97% larvicidal activities on the 3rd instars larvae of GWF and SWF at the recommended and its half concentrations. On the adults of the two whitefly species, acetamiprid and acetamiprid+ethofenprox showed more than 92% mortality even at half of recommended concentrations. Acetamiprid and acetamiprid+ethofenprox showed both residual effect and systemic activity. In the control efficacy test on GWF and SWF, 90% control values were obtained at the 3th day after treatments of acetamiprid and acetamiprid + ethofenprox by application with recommended concentration. These results indicate that acetamiprid and acetamiprid+ethofenprox can be used in the control of the two whitefly species in field.
Sulfonylurea herbicide-resistant weeds are spreading widely and distributed about 106,951 hectare in paddy rice fields in Korea. Morever all biotype of Scirpus juncoides which were collected at 69 spots all over paddy rice fields in 2008 were identified biotype of sulfonylurea herbicide-resistant. Benzobicyclon is a p-hydroxyphenylpyruvate dioxygenase (HPPD) inhibitor, which is absorbed through root and basal stem of weeds so cause bleaching of newly developing leaves. Benzobicyclon was very effective to control Scirpus juncoides, Monochoria vaginalis, sedges and broadleaves weeds, so it have been developed various formulation like a suspension concentrate (SC), a water dispersible granule (WG), a granule (GR) and a DT (tablet for Direct application). During recently 6 years, benzobicyclon-mixtures herbicides have been registered over than 54 products in paddy fields. Herbicidal efficacy by formulations of benzobicyclon and its mixture herbicides were highest in DT, followed by SC and GR. Herbicidal efficacy of the kaolin and $CaCO_3$ carrier of GR was better and stable than that of talc and bentonite carrier. Growth and yield of rice were not affected much by formulations, application rates and rice cultivation methods.

We describe two new Trichoderma species associated with oyster mushroom in Korea. Trichoderma green mould has been one of the most serious diseases of oyster mushroom in Korea. Of these the predominant species are two unrecorded species. We designed as Trichoderma sp. Korean type 1 (Th K1) and Trichoderma sp. Korean type 2 (Th K2), respectively. Th K1 and Th K2 can be distinguished from previously reported Trichoderma species as well as each other in morphological characteristics including growth rate at 35$^{\circ}C$, colony morphology, conidia shape and branch pattern of phialides. Sequence of the ITS region of rDNA, the protein coding translation elongation factor gene(EF-1${\alpha}$), and RNA polymeraseII (RPB2) not only clearly separated Trichoderma sp. Korean types from their closely related T. harzianum biotype but also distinguished them from each other. Analyses of the EF-1${\alpha}$ and RPB2 sequences were found to be more useful for establishing systematic relationships among Trichoderma isolates than those of the ITS sequence. This paper deals with the distribution, reservoir and mode of spread of infection on 6 hatcheries in Taegu, Kyungpook and 5 broiler farms in Kyungpook during the period from June 1991 to June 1992. Isolated Salmonella were examined for serotypes, biotyping of Salmonella(S) typhimurtum, antibiotic susceptibility and some biochemical characteristics. Forty two Salmonella strains were isolated from 42(2.7% of 1,577 caecal samples of chicks, and their serotypes were S typhimurium 10, S typhimurium var Copenhagen 5, S infantis 4, S thompson 3, and untypable 20. The isolation rate of Salmonella varied from 0 to 5.1% in 6 hatcheries and that of Salmonella from 5 broiler farms was 10. 5%. Biotypes of 10 S typhimurium and 5 S typhimurium var Copenhagen strains isolated from chicks of hatcheries and broiler farms were biotype 2(86.6%), 8(6.7%), and 10a(6.7%), and 26i(6.7%) according to Duguid's scheme. Antibiotic susceptibility test of Salmonella isolated were performed by agar dilution method, using 9 antibiotics as follows: ampicillin(Am), chloramphenicol(Cm), gentamicin(Gm), kanamycin(Km), nalidixic acid(Na), rifampicin(Rf), streptomycin(Sm), sulfadimethoxine(Su), and tetracycline(Tc), All the strains were sensitive to RF. But 8 strans(23.8%) were resistant to one or more drugs and the most common resistance patterns of transferred R plasmids were SmSuGm and SmSu. Among 42 isolates, one had transferable citrate utilizing plasmid. S typhimurium and S typhimurium var copenhagen strains were resistant to killing by 90% normal guinea pig serum.
Papaya ringspot virus (PRSV) causes the most widespread and devastating disease in papaya. Isolates of PRSV originating from different geographical regions in south India were collected and maintained on natural host papaya. The entire coat protein (CP) gene of Papaya ringspot virus-P biotype (PRSV-P) was amplified by RTPCR. The amplicon was inserted into pGEM-T vector, sequenced and sub cloned into a bacterial expression vector pRSET-A using a directional cloning strategy. The PRSV coat protein was over-expressed as a fusion protein in Escherichia coli. SDS-PAGE gel revealed that CP expressed as a ~40 kDa protein. The recombinant coat protein (rCP) fused with 6x His-tag was purified from E.coli using Ni-NTA resin. The antigenicity of the fusion protein was determined by western blot analysis using antibodies raised against purified PRSV. The purified rCP was used as an antigen to produce high titer PRSV specific polyclonal antiserum. The resulting antiserum was used to develop an immunocapture reverse transcription-polymerase chain reaction (IC-RT-PCR) assay and compared its sensitivity levels with ELISA based assays for detection of PRSV isolates. IC-RT-PCR was shown to be the most sensitive test followed by dot-blot immunobinding assay (DBIA) and plate trapped ELISA.
One hundred and nine Agrobacterium isolates were recovered from 68 samples(51 plant tumor and 17 soil) that were collected from different habitats in Northern Jordan. The isolated cultures were grouped into 3 biovars based on their biochemical characteristics and biovar I, II, and III comprised a total number of 46, 41, and 22 isolates, respectively. Isolates of biovar I were obtained primarily from the diseased peach, oak and rose plants, whereas isolates of biovar II and ill were obtained mostly from apple and grape plants, respectively. Twenty-nine isolates were found to be virulent to at least one of the tested hosts such as carrots, chickpeas, garden peas and tomato plants with a response of tumor formation or tumor with roots induction. Our result suggested that A. tumefaciens strains from tumor of various plants and soil of Jordan were diverse and they have a variation in their virulence.
The majority of soil microorganisms can derive ethylene from L-methionine (L-MET), while some rhizobacteria can hydrolyze 1-aminocyclopropane-1-carboxylate (ACC) due to their ACC-deaminase activity. In this study, three strains having either ACC-deaminase activity (Pseudomonas putida biotype A, $A_7$), or the ability to produce ethylene from L-MET (Acinetobacter calcoaceticus, $M_9$) or both (Pseudomonas fluorescens, $AM_3$) were used for inoculation. The highly ethylene specific bioassay of a classical 'triple' response in pea seedlings was used to investigate the effect of the inoculation with the rhizobacteria in the presence of 10 mM ACC or L-MET. The exogenous application of ACC had a concentration-dependent effect on the etiolated pea seedlings in creating the classical 'triple' response. The inoculation with P. putida diluted the effect of ACC, which was most likely due to its ACC-deaminase activity. Similarly, the application of $Co^{2+}$ reduced the ACC-imposed effect on etiolated pea seedlings. In contrast, the inoculation of A. calcoaceticus or P. fluorescens in the presence of L-MET caused a stronger classical 'triple' response in etiolated pea seedlings; most likely by producing ethylene from L-MET. This is the first study, to our knowledge, reporting on the comparative effect of rhizobacteria capable of utilizing ACC vs L-MET on etiolated pea seedlings.
