Isotopic Study on Energy Store and Consumption in Voluntary Exercising Mice (생쥐내 동위원소 이용에 의한 열량소비 및 축적에 관한 연구)

  • The study was attempted to observe effects of voluntary exercise on energy expenditure and on storage of excessive energy into body fat in adult mice. Mice, body weight 25-30g, were divided into two groups, exercise and sedentary group. Voluntary exercize by running wheel was allowed for former group but not for the mice of the latter group which were restricted by limiting space allowance 13.5$\times$11.5$\times$15.0cm per mouse. During a period of 4 weeks of feeding trial, they were fed ad libitum starch-casein based diet added with $^3\textrm{H}$-glucose (D-[1-$^3\textrm{H}$(N)]-glucose) at a level of 20 nCi per g of diet. Measurements were made to study hematology, lipase activity in epididymal adipose tissue, total contents of body waste and fat, and radiactivity of $^3\textrm{H}$-glucose incoporated into body fat. Dietary intake, body weight gain and amount of voluntary physical activities were also measured. The results obtained ard summarized as follows; 1) Amount of metabolizable energy intake, body weight gain and body fat were not statistically different between the two group. However, mice of the exercise group tended to show lower body weight gain body fat contants, but higher energy expenditure than those of the sedentary group. 2) Radioactivity of $^3\textrm{H}$-glucose incoporated into body fat appeared lower for exercise group expressd on a whole body fat basis. The activity was, however, higher for exercise group when expressed based on per g of fat compared to that of sedentary group. 3) Exercise group showed also higher activity of lipase in epididymal adipose tissue than the other group. 4) Mice whose physical activities were restricted appeared to have lower levels of hemoglobin and hematocrit values than of the exercise group. These results seem to support the theory that turn-over rate of body fat is activated by exercise and to suggest that consumed energy is to be converted primarily into body fat before its use as energy source by oxidation even during a period of continuous energy expenditure by exercise.

A Study on Energy Expenditure in Korean Adolescent (한국 청소년의 에너지 소비량에 관한 연구)

  • A 4-week energy balance study was conducted to estimate the energy expenditure (EE) of 7 high school age men, 16 to 18 year of age, by measurement of energy intakes and changes in body energy (BE) content (intake/balance technique), keeping their normal living pattern and maintenance body weight. Gross energy intake (GE) and fecal energy (FE) loss was measured by bomb calorimetry, Urinary energy (UE) loss was calculated from nitrogen excreted. Fat mass (FM) was determined from body density estimated from skinfold thickness. 1) Mean constitutional ratio of carbohydrate, protein and Int for the total energy intake was $73.7{\pm}0.3%$, $13.5{\pm}0.3%$ and $12.9{\pm}0.5%$, respectively. 2) Fecal energy loss was 2.4% proportion of the gross energy intake. 3) Mean daily metabolizable energy estimated by subtract fecal and urinary energy loss was $2582{\pm}61\;kcal$. 4) Total body energy change estimated from body composition change over 28 days was decreased $4309{\pm}1837kcal$. 5) Mean daily energy expenditure was $2736{\pm}59kcal\;(46{\pm}1kcal/kg$ of body weight).

Improvement of Leptin Resistance (렙틴 저항성의 개선)

  • Leptin, a 16-kDa cytokine, is secreted by adipose tissue in response to the surplus of fat store. Thereby, the brain is informed about the body's energy status. In the hypothalamus, leptin triggers specific neuronal subpopulations (e.g., POMC and NPY neurons) and activates several intracellular signaling events, including the JAK/STAT, MAPK, PI3K, and mTOR pathway, which eventually translates into decreased food intake and increased energy expenditure. Leptin signal is inhibited by a feedback inhibitory pathway mediated by SOCS3. PTP1B involves another inhibitory pathway of leptin. Leptin potently promotes fat mass loss and body weight reduction in lean subjects. However, it is not widely used in the clinical field because of leptin resistance, which is a common feature of obesity characterized by hyperleptinemia and the failure of exogenous leptin administration to provide therapeutic benefit in rodents and humans. The potential mechanisms of leptin resistance include the following: 1) increases in circulating leptin-binding proteins, 2) reduced transport of leptin across the blood-brain barrier, 3) decreased leptin receptor-B (LRB), and/or 4) the provocation of processes that diminish cellular leptin signaling (inflammation, endoplasmic reticulum stress, feedback inhibition, etc.). Thus, interference of the cellular mechanisms that attenuate leptin signaling improves leptin action in cells and animal models, suggesting the potential utility of these processes as points of therapeutic intervention. Various experimental trials and compounds that improve leptin resistance are introduced in this paper.

Development of the Energy Storing Foot (에너지 저장형 인공발의 개발)

  • KESF-1 foot conceptually provides storage of potential energy and is converted to kinetic energy throughout the weight - bearing phase of the gait cycle. This stored energy is progressively released as the foot continues through the toe-off phase to rebound and propel the body forward. A weight deflects the keel through a predetermined range, then the keel "springs back" as weight is removed. Foot designs criteria were selected to guide development beyond the proof-of concept composite material keels; 1) store and return energy (1-3/4 " metatarsal deflection at 435 pounds vertical load), 2) natural feel and stability; 3) useful life of 3-years: 4) lightweight; 5) reduced production costs. The purpose of this study is developed the comfortable stable foot that fitted with Korean lifestyle and road condition. The results produced the realistic cosmetic foot cover with polyurethane form and the keel composed with composite materials of both glass fiber and carbon fiber.

  • This paper describes an analysis of the stability and performance of a large-capacity flywheel energy storage system (FESS) supported by active magnetic bearings. We designed and manufactured the system that can store up to 5kWh of usable energy at the maximum speed of 18,000 rpm. In order to analyze the stability of the systems accurately, we derived a rigid body rotor model, flexible rotor model using finite-element method, and a reduced-order model using modal truncation. The rotor model is combined with those of active magnetic bearings, amplifiers, and position sensors, resulting in a system simulation model. This simulation model is validated against experimental measurements. The stability of the system is checked from the pole locations of the closed-loop transfer functions. We also investigated the sensitivity function to quantify the robustness of the systems to the disturbances such as mass imbalance and sensor noises.

The BIDAS Program : Bioassay Data Analysis Software for Evaluating Radionuclide Intake and Dose (BIDAS프로그램 : 방사성 핵종의 섭취량과 선량 평가용 생물학적분석 자료 해석 소프트웨어 프로그램)

  • A computer software program, called BIDAS (BIoassay Data Analysis Software) is developed to interpret the bioassay measurement data in terms of intakes and the committed effective dose using the human respiratory tract model (HRTM), gastrointestinal tract (GI-tract) model and biokinetic models currently recommended by the International Commission on Radiological Protection (ICRP) to describe the behavior of the radioactive materials within the body. The program consists of three modules; first, a database module to manage the bioassay data, second, another databasee module to store the predicted bioassay quantities of each radionuclide and finally, a computational module to estimate the intake and committed effective dose calculated with the bioassay quantity measurement values from either an acute or chronic exposure of the radionuclies within the body. This paper describes the features of the program as well as the quality assurance check results of the BIDAS software program.

  • Following conclusions were derived from researching various consumption caused after surgical operation. The meaning of surgery in HyungSang medical point of view is a big flaw of the original configuration. Therefore meaning, the loss of container to store something. Changes of the body after surgery can be formation of stagnated blood and abnormal cyst of the removed part and entangled undigested food and stagnant blood. After surgery when the symptoms become chronic or the patients develop side effects as they age, consumption and aging process begins. HyungSang medical treatments of consumption caused after surgery are the followings. We should prevent aging of the framework by using appropriate medication to make up for faults in Gall Bladder, Bladder type and six meridian type by considering its form, color, pulse and symptoms. Stability of emotion that can endure various mental stimulation is needed. Prevent stagnation, indigestion, obstruction and numbness by smoothing up the constructive energy and the defensive energy in the operated part. The Heart and Kidney should be under control and Stomach and Spleen should be reinforced so that food intake is accelerated and produce vital substances and Ki and blood. Eum blood and Yang Ki should circulate around the body well. Examine daily life and if small changes in skin complexion, appetite, sleep and excretion occur, also difference in adaptation of external stimulation and mental health occurs immediate treatment and care is necessary. Treatment of side-effects caused from both aging process and post-surgery in HyungSang's view point is considered better than other ways of treatment and will contribute in expanding the field of medical science.

Use of Human Adipose Tissue as a Source of Endothelial Cells (혈관내피세포 채취의 원천으로 인간 지방조직의 활용)

  • Purpose: Adipose tissue is located beneath the skin, around internal organs, and in the bone marrow in humans. Its main role is to store energy in the form of fat, although it also cushions and insulates the body. Adipose tissue also has the ability to dynamically expand and shrink throughout the life of an adult. Recently, it has been shown that adipose tissue contains a population of adult multipotent mesenchymal stem cells and endothelial progenitor cells that, in cell culture conditions, have extensive proliferative capacity and are able to differentiate into several lineages, including, osteogenic, chondrogenic, endothelial cells, and myogenic lineages. Materials and Methods: This study focused on endothelial cell culture from the adipose tissue. Adipose tissues were harvested from buccal fat pad during bilateral sagittal split ramus osteotomy for surgical correction of mandibular prognathism. The tissues were treated with 0.075% type I collagenase. The samples were neutralized with DMEM/and centrifuged for 10 min at 2,400 rpm. The pellet was treated with 3 volume of RBC lysis buffer and filtered through a 100 ${\mu}m$ nylon cell strainer. The filtered cells were centrifuged for 10 min at 2,400 rpm. The cells were further cultured in the endothelial cell culture medium (EGM-2, Cambrex, Walkersville, Md., USA) supplemented with 10% fetal bovine serum, human EGF, human VEGF, human insulin-like growth factor-1, human FGF-$\beta$, heparin, ascorbic acid and hydrocortisone at a density of $1{\times}10^5$ cells/well in a 24-well plate. Low positivity of endothelial cell markers, such as CD31 and CD146, was observed during early passage of cells. Results: Increase of CD146 positivity was observed in passage 5 to 7 adipose tissue-derived cells. However, CD44, representative mesenchymal stem cell marker, was also strongly expressed. CD146 sorted adipose tissue-derived cells was cultured using immuno-magnetic beads. Magnetic labeling with 100 ${\mu}l$ microbeads per 108 cells was performed for 30 minutes at $4^{\circ}C$ a using CD146 direct cell isolation kit. Magnetic separation was carried out and a separator under a biological hood. Aliquous of CD146+ sorted cells were evaluated for purity by flow cytometry. Sorted cells were 96.04% positivity for CD146. And then tube formation was examined. These CD146 sorted adipose tissue-derived cells formed tube-like structures on Matrigel. Conclusion: These results suggest that adipose tissue-derived cells are endothelial cells. With the fabrication of the vascularized scaffold construct, novel approaches could be developed to enhance the engineered scaffold by the addition of adipose tissue-derived endothelial cells and periosteal-derived osteoblastic cells to promote bone growth.
