  • Urease was immobilized with calcium alginate by entrapment method in the form of spherical beads and stored in Tris/acetate buffer (pH 7.3) at $4^{\circ}C$. Urease immobilized at different concentration of alginate beads (3%, 4% and 5%) showed higher apparent $K_{m}$ values than the soluble urease. Furthermore, $K_{m}$ has been shown to be corelated with effective diffusion coefficient (De) at different concentration of alginate gel. The present study showed that diffusion and reaction contribute to control the overall rate.

  • The aim of the present study was to elucidate whether gallic acid could modulate the inflammatory allergic reaction and to study its mechanism of action Gallic acid inhibited compound 48/80- or immunoglobulin E (IgE)-induced histamine release from mast cells. The inhibitory effect of gallic acid on the histamine release was mediated by modulation of cAMP and intracellular calcium. Gallic acid decreased the phorbol 12-myristate 13-acetate plus calcium ionophore A23187-stimulated pro-inflammatory cytokine gene expression and production such as TNF- ${\alpha}$ and IL-6 in human mast cells, and the inhibitory effect of gallic acid was on dependent nuclear factor- ${\kappa}$B and p38 mitogen-activated protein kinase. Our findings provide evidence that gallic acid inhibits mast cell-derived inflammatory allergic reaction by blocking histamine release and pro-inflammatory cytokine expression.

  • The bio-mediated production of calcite crystals by calcinogenic bacteria has great applicable value for the restoration of deteriorated calcareous monuments, because of its high purity and coherency. An investigation of the conditions for calcite production by an alkalophilic Bacillus amyloliquefaciens CMB01 strain was made. Optimal calcite precipitation occurred when the bacterium was cultured at pH 8.0 and 30$^{\circ}C$, and in B4 medium that consisted of 0.4% yeast extract, 0.5% glucose, and 1.5% calcium acetate. Calcium ion of the bacterially induced calcite was analyzed by an inductively coupled plasma (ICP) spectrophotometer. Optical and scanning electron microscopy (SEM) of the calcite revealed a typical rombohedral polycrystalline structure.

  • The biogenic corrosion of mortars adopted in sewage repair by sulfuric acid-producing bacteria was considered in this paper. Calcium aluminate cement (CAC) was known to resist microbiologically-induced corrosion significantly better than portland and blended portland cement. In this study, CAC as well portland cement mortars were tested as main binder to evaluate the corrosion resistance by the chemical immersion test. Replacement ratios of CAC were changed as 0, 20, 40, 50, 60% of OPC binder and 0, 2, 4, 6% of EVA(Ethylene Vinyl Acetate) were also adopted to increase properties of CAC repair mortars in sewage application. Setting time, compressive strength, acid resistance and adhesive strength were measured for various experiments. As a results of the experiments, the proper formulation of repair mortars was found at 40% of CAC and 4% of EVA. Finally, the CAC mortars adopted in field sewer pipe and were demonstrated to superior in adhesion and workability.

  • This study was carried out to investigate the physiological changes induced acutely with the low doses of lead acetate in the synaptosomes from the cerebrum and brain stem of the rat. The general uptake patterns of [$^3$H]-serotonin were observed in synaptosomes, as a model of presynaptic nerve terminal, from the cerebrum and brain stem. And the effects of the low doses of lead acetate on the uptake process were investigated id vitro and in vivo. The Km value of the uptake of the [$^3$H]-serotonin by the synaptosomes was 0.5 $\mu$M in the cerebrum and 0.1 $\mu$M in the brain stem. These low values reveal that the synaptosomes from the cerebrum and the brain stem have a high affinity to [$^3$H]-serotonin, especially in brain stem. The uptake of $\mu$M-serotonin was dependant on the sodium and potassium ions. When being treated with ouabain, the $Na^+$ $-K^+$ ATPase inhibitor, the uptake of [$^3$H]-serotonin was reduced. This supports strongly that the uptake of [$^3$H]-serotonin was sensitive to the changes of the concentrations of the sodium and potassium ions. When the calcium channel blocker, verapamil, was treated, the uptake of [$^3$H]-serotonin was changed only in synaptosomes from the brain stem. The uptake of [$^3$H]-serotonin was reduced by the lead treatment in the synaptosomes from the cerebrum and brain stem in vitro and in vivo. [lead acetate, synaptosomes, $^3$H-serotonin, rat]

  • Recently, we demonstrated that human serum amyloid P component (SAP) specifically recognizes exposed bacterial peptidoglycan (PGN) of wall teichoic acid (WTA)-deficient Staphylococcus aureus ${\Delta}$tagO mutant cells and then induces complement-independent phagocytosis. In our preliminary experiments, we found the existence of human serum immunoglobulins that recognize S. aureus PGN (anti-PGNIgGs), which may be involved in complement-dependent opsonophagocytosis against infected S. aureus cells. We assumed that purified serum anti-PGN-IgGs and S. aureus ${\Delta}$tagO mutant cells are good tools to study the molecular mechanism of anti-PGN-IgG-mediated phagocytosis. Therefore, we tried to identify the intracellular molecule(s) that is involved in the anti-PGN-IgG-mediated phagocytosis using purified human serum anti-PGN-IgGs and different S. aureus mutant cells. Here, we show that anti-PGN-IgG-mediated phagocytosis in phorbol myristate acetate-treated U937 cells is mediated by $Ca^{2+}$ release from intracellular $Ca^{2+}$ stores and anti-PGN-IgGdependent $Ca^{2+}$ mobilization is controlled via a phospholipase C${\gamma}$-2-mediated pathway.

  • The effect of metabolic substrate fructose on the force of contraction of isolated rat atria depressed with lidocaine was determined. Fructose produced dose-dependent increase in the force of contraction of isolated atria depressed by substrate-free Krebs-Ringer bicarbonate medium. The maximally effective concentration of fructose was 30 mM. The isolated atria, suspended in Krebs-Ringer bicarbonate glucose medium aerated with 95% $O_2-5%CO_2$at $30^{\circ}C$ and pH 7.4, were depressed 50% by approximately 2.34 mg/100 ml of lidocaine. Addition of 30 mM fructose to these depressed atria resulted in a marked increase in the contractile force similar to that with pyruvate and acetate. Fructose had no significant effect, however, on atria exposed to low-calcium medium. The results are consistent with a previous report suggesting blockade by lidocaine of the uptake or utilization of glucose in the glycolytic pathway, and further pinpoint the blockade as an early step in the glycolytic sequence prior to the phospho-fructokinase step.

  • Radiation grafting of acrylic acid and 4-vinylpyridine at room temperature has been studied by an impregnation method to improve the hygroscopic properties, the antistatic behavior and the dyeability of polyester fabric. Polyester fabric was impregnated with acrylic acid or aqueous emulsion of acrylic acid-4-vinylpyridine by immersion at 25$^{\circ}$or 7$0^{\circ}C$. The impregnated fabric was irradiated under nitrogen gas with ${\gamma}$-rays from Co-60. When acrylic acid grafted polyester fabric was treated with sodium carbonate, calcium acetate and potassium persulfate, tne rate of water absorption was increased and most parts of polyacrylic acid formed were extracted off from the fabric with 0.1% solution of sodium hydroxide at 10$0^{\circ}C$. In the case of the impregnation of a mixture of acrylic acid and 4-vinylpyridine the petcent of grafting has been shown to be proportional to the ratio of 4-VP/AA and radiation dost. Estimating by contact angle measurements of water on the various polymer surfaces, the antistatic behavior was decreased with the increase of grafting percent. The investigation of electron micrograph disclosed the existence of certain type of discontinuities in the acrylic acid grafted polyester fiber which was treated with various salts.

  • ATP analogs were immobilized or bovine caseins by the action of transglutaminase. The ATP analogs immobilized on the caseins were enzymatically active and interconverten by kinases. The immobilized ATP was dephosphorylated to the corresponding ADP by hexokinase and rephosphorylated to the ATP in solid form by acetate kinase. Under the conditions chosen, about 55% of the immobilized ATP was dephosphorylated and about 80% of the resulted ADP was rephosphorylated. Bovine $\beta$-casein was more useful than $\alpha$sf-casein as a carrier and C8-substituted ATP analognwas more effective than N6-substituted one in immobilization. Michaelis constant of C8-substituted ATP analog immobilized on $\beta$-casein was similar to that of free form of ATP and that of ATP analog. The immobilized ATP was much more stable than free ATP and its analog, while maximum velocity was reduced to 37% of the free ATP analog. The immobilized ATP was recovered almost completely by calcium precipitation.
