Isolation and Identification of Terpenoids from the Leaf of Chrysanthemum coronarium L. (쑥갓 잎에서 터펜노이드계 화합물의 분리 및 동정)

  • Three terpenoids were isolated from the leaves of Chrysanthemum coronarium L. by silica gel chromatography and recrystalization. The structure of compounds 1, 2, and 3, using various spectroscopic data, were identified as to be terpenoid derivatives of dihydrotulipinolide, loliolide, and $5{\alpha},8{\alpha}-peroxyergosterol$, respectively. The presence of these 3 is the first time reported in this plant. The cytotoxic activity of $5{\alpha},8{\alpha}-peroxyergosterol$ showed strong activities against human cancer cell lines such as A549, PC-3, and HCT 116.

  • On the basis of potent HDAC-inhibitory activity and anticancer activity of SAHA, novel SAHA derivatives 3a-d and 7 with a bulky cap such as p-dimethylaminophenyl, 4-phenylaminophenyl, 4-phenyloxyphenyl, 9H-fluorenyl or naphthalenyl ring were synthesized starting from the corresponding aryl amines or naphthalenyl acetic acid using an EDC-mediated amide coupling reaction in the presence of HOBt followed by a nucleophilic addition-elimination reaction with hydroxylamine. Compounds 3b, 3c and 3d showed more potent inhibitory activity on total HDACs (14~27-fold), HDAC1 (8~15-fold), HDAC2 (1.3~25-fold) and HDAC7 (1~3-fold) and more potent anticancer activity (2~22-fold) against MCF-7, MDA-MB-231, MCF-7/Dox, MCF-7/Tam, SK-OV-3, LNCaP and PC3 human cancer cell lines than SAHA.

Ethanol Eluted Extract of Rhus verniciflua Stokes Showed both Antioxidant and Cytotoxic Effects on Mouse Thymocytes Depending on the Dose and Time of the Treatment

  • For a long time Rhus verniciflua Stokes (RVS) has traditionally been used as a herbal plant. It is known to contain various biological activities. Previously, a crude ethanol extract from RVS was reported to have antioxidant effects, and antiproliferative activities, on human cancer cell lines. In this report, we prepared a highly purified ethanol extract from RVS, which did not contain the urushiol derivatives, named REEE-1 ($\underline{R}$hus $\underline{e}$thanol $\underline{e}$luted $\underline{e}$xtract-1), to investigate the mechanisms of the scavenging activity of hydroxyl radicals using mouse thymocytes. The results from the deoxyribose, DNA nicking, and glucose/glucose oxidase enzyme assays showed that REEE-1 contained a strong scavenging activity of oxygen free radicals, especially of hydroxyl radicals. However, interestingly, REEE-1 also showed cytotoxicity against the thymocytes, although the effect was variable, depending on the concentrations and times of treatment. The REEE-1-mediated cytotoxicity against thymocytes, which has been used as one of the well-characterized models for apoptosis studies, was verified to be apoptotic. This was proven by the following: the appearance of DNA laddering, increases in DNA fragmentation, low fluorescence intensity in the nuclei after propidium iodide staining, and positive Annexin V staining of the cells. These results suggested that REEE-1 had both antioxidative activity and cytotoxicity against the thymocytes, although the effect of the cytotoxicity was variable, depending on the dose and time of the treatment.

Antiviral Activity of Antibiotic Peptaibols, Chrysospemins B and D, Produced by Apiocrea sp. 14T against TMV Infection

  • A total of about 300 fungal isolates from forest havitats were screened for inhibitors of tobacco mosaic virus (TMV) infection using its local lesion host, Nicotiana tabacum cv. Xanthi nc. Ine of the isolates, 14T, showed a strong activity against TMV infection, and was identified as an Apiocrea sp. based on its morphological characterstics. Rice was an optimum culture medium for its fermentation, and two antiviral compounds, KGT 141 and KGT 142, were resolved from the rice culture through column chromatography, TLC, and HPLC. By NMR and FAB-MS, the two compounds were identified as chrysospermins B (KGT 141) and D (KGT 142), both of which are peptaibols with 19-mer amino acids possessing an acetylated N-terminus and a hydroxy-amino acid (tryptophanol) at the C-terminus. Both compounds showed inhibitory activities against TMV infection, but chrysospermin D showed the stronger activity than chrysospermin B. The former of $100{\;}\mu\textrm{g}/ml$ and 54.7% at $10{\;}\mu\textrm{g}/ml$, respectively. Furthermore, the chrysospermins were highly cytotoxic toward cancer cell lines of PC-3 (prostrate) and K562 (leukemia), and inhibited growth of the Gram-positive bacteria tested, especially the plant pathogenic bacterium Corynebacterium lilium. To the best of our knowledge, this is the first report on the inhibition of plant virus infection by antimicrobial peptaibols.

  • The present study was conducted to evaluate of the 1,1-Diphenyl-2-Picrylhydrazyl (DPPH) radical scavenging effect, cytotoxicity and tyrosinase inhibition activities using methanol extracts from different parts of four herb plants. The results showed that whole and root extracts of yarrow the highest total polyphenol and flavonoid contents as well as whole of yarrow revealed the highest DPPH radical scavenging effect. In cytotoxicity test against three cancer cell lines, HeLa (uterus), SK-Hep-1 (liver), and YD-15 (oral), the whole extract of feverfew showed the highest toxicity with $IC_{50}$ values of $102.58-138.68{\mu}g/mL$. Also, mallow root extract ($71.24{\mu}g/mL$) exhibited potent tyrosinase inhibitory activity comparable to arbutin ($69.56{\mu}g/mL$), which was used as the control.

Phenolic constituents and biological activities of leaf extracts of traditional medicinal plant Plectranthus amboinicus Benth (Lamiaceae)

  • Plectranthus amboinicus Benth (Lamiaceae) is a medicinal plant native to India, and its leaves are widely used in several traditional medicinal preparations. The purpose of this study was to detect and quantify phenolics present in ethyl acetate and acetone extracts of P. amboinicus leaves, and evaluate their antioxidant, antibacterial, antimutagenic and anticancer activities. The HPLC chromatograms of crude leaf extracts indicated the presence of phenolics like caffeic acid, coumaric acid, rutin, quercetin and gallic acid, which were present in the range of 0.01 - 1.41 mg/g in ethyl acetate and 0.03 - 1.93 mg/g in the acetone extract. The acetone extract showed statistically (p < 0.05) higher antioxidant activity ($IC_{50}$, 99.59 ${\mu}g/ml$) than ethyl acetate extract ($IC_{50}$, 149.96 ${\mu}g/ml$). Statistically (p < 0.05) higher antimutagenicity was shown by acetone extract (46.16%) as compare to ethyl acetate extract (12.16%) at 500 ${\mu}g/plate$ concentration. The acetone extract showed higher antibacterial activity than ethyl acetate extract, and both the extracts showed highest activity against B. cereus (375 and 625 ${\mu}g/ml$, respectively) and lowest activity against Y. enterocolitica (1000 and 1125 ${\mu}g/ml$, respectively). Both the extracts also showed inhibitory effect on cancer cell lines HCT-15 and MCF-7. These results suggest that the leaves of P. amboinicus possess various biological activities, and validate the traditional use of the leaves of P. amboinicus against cold, infection and ulceration.

Antioxidant and Cytotoxic Activity of Compounds from the Stem of Juncus effusus (등심초로부터 분리된 화합물의 항산화 활성 및 암세포 성장 억제 효과)

  • The stems of Juncus effusus were extracted with 70% aqueous ethanol and the concentrated extract was partitioned with ethyl acetate, n-butanol and $H_2O$, successively. Two compounds were isolated from the ethyl acetate fraction through the repeated silica gel and Sephadex LH-20 column chromatographies. According to the results of physico-chemical and spectroscopic data including NMR and IR, the chemical structures of the compounds were determined as dehydroeffusol (1) and effusol (2). Dehydroeffusol and effusol exhibited potent scavenging activity for DPPH and ABTS radicals with the $IC_{50}$ values as $130{\pm}3.21$ and $79{\pm}1.53{\mu}M$ in DPPH assay, and as $39{\pm}3.51$ and $24{\pm}2.73{\mu}M$ in ABTS assay, respectively. The compounds also significantly inhibited the proliferation of human cancer cell lines, AGS and A549.

Chemical Characteristics and Biological Activities of Herbimycin A and Dihydroherbimycin A Produced by a Soil Isolate Streptomyces sp. AO-0511 (Streptomyces sp. AO-0511이 생산하는 Herbimycin A 및 Dihydroherbimycin A의 이화학적 특성 및 생물 활성)

  • A streptomycete strain was isolated from the soil samples from Korea. The chemotaxonomy and 16S rDNA sequencing confirmed that the strain belonged to the genus Streptomyces and we named it Streptomyces sp. AO-0511. Two antibiotics, herbimycin A and dihydroherbimycin A produced by this strain were tested for their physico-chemical and biological characteristics. Both compounds were stable under acidic pH. Dihydroherbimycin A was more heat-stable and polar compared with herbimycin A. Only weak antibacterial activities were detected against Bacillus subtilus ATCC 6633 and Micrococcus luteus ATCC 9341. However, herbimycin A and dihydroherbimycin A showed strong inhibitory activities on lung cancer cells (A549 cells) and leukemia cells (HL-60). The cytotoxicity was determined using L5178Y and P388 cell lines. The results showed that herbimycin A and dihydroherbimycin A had lower toxic effects on the cells compared with the standard compounds, comptothecin and cyclosporin A. Therefore, both compounds could be good candidates for the development of new anticancer drugs.

In Vitro Studies on the Anticancer Effect and Topoisomerase I Inhibition Activity of Caesalpinia sappan L. Extract (소목 추출물의 세포독성 효과와 Topoisomerase I 억제 활성에 관한 연구)

  • To evaluate cytotoxic effect and topoisomerase I inhibition activity of Caesalpinia sappan L., both water and methanol extracts were examined using in vitro assay. The cytotoxic effect of Caesalpinia sappan L. examined using MTT and SRB assay and $IC_{50}$ values were measured against U937, HL60, HepG2, SNU-1, SNU-16 cancer cell lines. Among them the representative cytotoxic results are shown as follows; water extract (U937=13.39 ${\mu}g/ml$, HL60=8.65 ${\mu}g/ml$, HepG2=38.48 ${\mu}g/ml$, SNU-1=7.72 ${\mu}g/ml$, SNU-16=25.49 ${\mu}g/ml$), methanol extract (U937=13.35 ${\mu}g/ml$, HL60=9.43 ${\mu}g/ml$, HepG2=25.67 ${\mu}g/ml$, SNU-1=8.37 ${\mu}g/ml$, SNU-16=28.64 ${\mu}g/ml$). The inhibitory concentration of DNA topoisomerase I activity against water extract was 100 ${\mu}g/ml$ and the inhibitory concentration of DNA topoisomerase I against methanol extract was 400 ${\mu}g/ml$.

  • Far-infrared radiation therapies are becoming more popular for blood circulation disorders, cardiovascular disease, skin diseases, inhibit cancer cell, etc replacing conventional operations. In this research, thermal characteristics of heating part in panel radiators, which is effective on the blood circulation disorders were experimentally analyzed. The heating line supplies heat energy to insulation coatings with heat flux of $150mW/m^2$ in normal status and as a result the coatings reached 20% of the heating line temperature. In other words, the insulation itself could increase surface temperature of heating plates by 20% and raise thermal time constant promote blood circulation effect. We also found that space arrangement of the heating lines was an important factor in designing heating parts and both coefficient of heat conduction and density of the heating plate should be also considered for superimpose of thermal diffusion.
