Two Ethylene Signaling Pathways in Senescing Carnation Petals: Exogenous Ethylene-induced Expression of Genes for 1-Aminocyclopropane-1-Carboxylate (ACC) Synthase and ACC Oxidase is Different from That of the Gene for Cysteine Proteinase

  • Carnation petals exhibit autocatalytic ethylene production and wilting during senescence. The autocatalytic ethylene production is induced by the expression of 1-aminocyclopropane-1-carboxylate (ACC) synthase and ACC oxidase genes, whereas the wilting of petals is related to expression of the cysteine proteinase (CP) gene. Until recently, it has been believed that these two phenomena, autocatalytic ethylene production and wilting, are regulated in concert in senescing carnation petals, since the two phenomena occurred closely in parallel. Our studies with petals of a transgenic carnation harboring a sense ACC oxidase transgene and petals of carnation flowers treated with 1,1-dimethyl-4-(phenylsulfonyl) semicarbazide showed that the expression of ACC synthase and ACC oxidase genes and that of CP are regulated differently in carnation psanetals. Interestingly, in the petals of transgenic carnation, the transcript for CP was accumulated but the transcripts for ACC synthase and ACC oxidase were not accumulated in response to exogenous ethylene. Based on these results, we hypothesized that two ethylene signaling pathways, one leading to the expression of ACC synthase and ACC oxidase genes and the other leading to the expression of CP gene, are functioning in senescing carnation petals.

  • To clarify the influence of ethylene on flowering and senescence of carnation flowers and the treatment of the floral preservative solution for reducing their ethylene production, bud-cut carnations (Dianthus caryophyllus L. cv. Coral) were sprayed with/without 500 ppm ethephon [(2-chloroethyl) phosphonic acid] and then held in distilled water or preservative solution (5% sucrose+50 ppm $AgNO_3$+300 ppm 8-hydroxyquinoline). 1. Ethylene inhibited to bloom carnation flowers because of causing sleepiness. 2. Carnation flowers were led to senescence after a lot of ethylene was produced by them. Therefore, ethylene seems to act as a trigger of senescence in carnation flowers. 3. The features of senescence in carnation flowers were different in accordance with the amount of ethylene produced by the flowers. 4. Exposure to ethylene decreased fresh weight, flower diameter, and water uptake of carnations. 5. The floral preservative extended vase life markedly by inhibiting ethylene production of cut flowers. The flowers exposed to lots of ethylene, however, were not restored even though they were held in the preservative solution.

  • A blight disease of carnation (Dianthus caryophyllus L.) caused by Alternaria dianthi was found in greenhouses and vinylhouses in southern Korea. In severely affected beds, many carnation plants were blighted and died. The growth of the fungus id vitro was optimum between 24 and $28^{\circ}C$. In controlled inoculations, A. dianthi caused lesions on leaves and stems of carnation plants.

  • Carnation is considered to be one of the top three cutting flowers in the world, which is a main crop with 21 billion annual volume of manufacture. The four carnation items such as cuttings, seed, plant and unrooted cuttings are imported and exported. Viruses can be easily transmitted during vegetative propagation of carnation. Carnation necrotic fleck virus (CNFV) and Carnation ringspot virus (CRSV) are designated as Korea plant quarantine viruses and inspected. This study was aimed to develop specific primer sets for easy and rapid detection of CNFV and CRSV. Two RT-PCR primer sets were efficiently amplified 288 and 447 bp fragments for CNFV and 503 549 bp fragments for CRSV. Furthermore, developed nested primer sets make possible to high sensitive detection and verification. CNFV nested PCR primer sets all produced band of 147 bp and CRSV nested PCR primer sets did bands of 395 and 347 bp. In addition, plasmid inserted 6 sequences in amplicon were used as a positive control to improve inspection confidence. The successful application of PCR module newly developed in this study will be highly useful for detect of CNFV and CRSV for quarantine inspections.

First Report on Carnation vein mottle virus in Dianthus barbatus in Korea

  • A potyvirus causing chlorotic mottle and yellow spots on leaves of Dianthus barbatus was isolated and identified as an isolate of Carnation vein mottle virus (CVMV). Purified preparations of Chenopodium quinoa infected with CVMV-K showed filamentous particles between 695 and 785 om long. Many cytoplasmic inclusions were observed, and these consisted of pinwheels, dense bands, loops, and circles. The coat protein of CVMV-K was about 32 KDa in western blot analysis using a CVMV antibody. The nucleotide sequence of coat protein gene showed 97.6% homology with a Japanese isolate. The genome size of CVMV-K was about 9.0 kb by dsRNA analysis. These results indicate that the virus is an isolate of CVMV. This is the first report on CVMV in Korea.

Biological Control of Fusarium Wilt of Carnation Plants by Antagonistic Microorganism in Greenhouse (카네이션의 시설재배에서 길항성 세균을 이용한 Fusarium Wilt 의 생물학적 방제)

  • This study was carried out to screen and select the effects of antifungal bacterial strains which inhibit the growth of plant pathogen, Fusarium oxysporum(fusarium wilt) occurred in carnation plants in greenhouse. We isolated an effective bacterial strains and investigated into the antifungal activity of the antagonistic microorganism and it's identification. Twenty bacterial strains which strongly inhibited Fusarium oxysporum were isolated from roots of carnation plants and the soil in greenhouse, and the best antifungal bacteria designated as C121, was finally selected. Antagonistic bacterial strain, C121 was identified to be the genus Bacillus sp. based on the morphological, biochemical and cultural characterizations. The Bacillus sp. C121 showed 58.1% of antifungal activity against the growth of Fusarium oxysporum. By the bacterialization of the cultural broth and the heat bacterialization culture filtrate of it, Bacillus sp. C121 was shown 92.1% and 21.0% of antifungal activity, respectively.

  • In order to carnation productivity improvement, ozonizer and negative ionizer has been designed and manufactured. The ozone is generated by DC positive streamer discharge and the negative ion is generated by DC negative discharge. When the ozone and negative ion are supplied growth room and farm to carnation cultivation, the carnation productivity is improved.

  • For carnation productivity improvement, compact-low cost discharge lamp type ozonizer using pulse power source is designed and manufactured. This paper describes the discharge characteristics and the ozone generation characteristics with variation of the output voltage of pulse power source, the quantity of flowing air and the operating number of ozonizer. The important conclusions are as follows. The discharge voltage and current values are proportional to the output voltage of pulse power source. The ozone concentration and generation are proportional to the output voltage of pulse power source and the number of ozonizer. The maximum value of the ozone concentration and generation of the discharge lamp type ozonizer can be obtained 890[ppm] and 59.7[mg/h] respectively. In the carnation productivity house, when the discharge lamp type ozonizer is used the carnation is good quality in the uniformity of the carnation stalk thickness and increased the productivity through improvement of the air and the soil.

Chemical Properties of the Greenhouse Soil and Nutrient Contents in Leaves and Stems of Carnation, Lily, and Rose. (카네이션, 백합, 장미 시설재배지 토양중 양분함량 과 품종별 경엽중 양분함량)

  • This study was conducted to investigate tissue nutrient contents and salt accumulation in plastic house soils cultivating lily, rose and carnation. The soil tested had high total salts, available phosphate and exchangeable potassium. The soil cultivating rose had highest salt concentvation followed by chose of carnation and lily. Tissue nutrient contents of lily were higher than chose of carnation and rose. In comparison among cultivars, the nutrient contents were as follows; 'Snow Qeen'>'Le Reve'>'Casa Blanca' in lily; 'Marina'>'Super star'>'Mary Devor'>'Madelon' in carnation; and 'Cocktail'> 'Marina'>'Maderon' in rose. The range of the nutrient contents were: T-N: $1.66\;{\sim}2.35%$, K: $1.73{\sim}2.23%$, Zn: $2.13{\sim}6.43\;mg/kg$, Cu: $3.79{\sim}13.89\;mg/kg$ in carnation; T-N: $0.79{\sim}1.65%$, P: $0.18{\sim}0.44%$, Ca: $0.59{\sim}1.26%$, Mg: $0.21{\sim}0.46%$, Zn: $23.65{\sim}90.30\;mg/kg$, Cu: $0.99{\sim}4.62\;mg/kg$ in lily; and T-N: $0.75{\sim}1.62%$, P: $0.17{\sim}0.30%$, K: $1.60{\sim}2.91%$, Ca: $0.64{\sim}0.94%$, Zn: $24.57{\sim}48.31\;mg/kg$, Cu: $3.10{\sim}9.08\;mg/kg$ in rose. The amount of nutrients uptake per plant was high in order of: K > T-N > Ca > Mg in lily; and T-N > K > Ca > P > Mg in rose.
