Anti-Apoptotic Effects of Catalpol on Preimplantaion Porcine Embryos

  • Catalpol, an iridoid glucoside, isolated from the root of Rehmannia glutinosa Libosch. It possesses a broad range of biological and pharmacological activity including anti-tumor, anti-inflammation and anti-oxidant by acting as a free radical scavenger. Therefore, in this study, the effects of catalpol on blastocyst development, expression levels of reactive oxygen species (ROS) and apoptotic index were investigated in porcine embryos. After in vitro maturation and fertilization, porcine embryos were cultured for 6 days in porcine zygote medium 3 (PZM-3) supplemented with catalpol (0, 100, 200 and $400{\mu}M$, respectively). Blastocyst development not significantly improved in the catalpol treated group when compared with control group. Otherwise, the intracelluar levels of ROS were decreased and the numbers of apoptotic nuclei were reduced in the catalpol ($100{\mu}M$) treated porcine blastocysts (P<0.05). On the other hand, blastocyst development was significantly improved in the catalpol ($100{\mu}M$) treated group when compared with the untreated catalpol group under $H_2O_2$ ($200{\mu}M$) induced oxidative stress (P<0.05). Otherwise, the intracellular levels of ROS in catalpol ($100{\mu}M$) treated group were significantly decreased in the untreated catalpol group under $H_2O_2$ ($200{\mu}M$) induced oxidative stress (P<0.05). Furthermore, the total cell numbers of blastocysts were significantly increased (P<0.05) in the catalpol ($100{\mu}M$) treated group under $H_2O_2$ ($200{\mu}M$) induced oxidative stress, whereas numbers of apoptoic nuclei were significantly reduced (P<0.05). In conclusion, our results indicate that treatment of catalpol may have important implications for improving developmental competence and preimplantation quality of porcine embryos through its anti-oxidant and anti-apoptotic effect.

  • This study was conducted to investigate contents of inorganic constituents, free sugars and catalpol in the Rhizoma of Rehmannia glutinosa as different growth stages. With the approach of harvesting season, the variations of their contents were as follow. The contents of P, K, Ca, Mg, Cu and ash in the Rhizoma decreased, whereas the contents of Fe, water and ethanol extract increased. The contents of total sugar, sucrose and galactose in the Rhizoma decreased, but the fructose and catalpol contents in the Rhizoma were increased.

  • This study was conducted to clarify the relationships between the inorganic constituents contents and the contents of catalpol and free sugars in the rhizoma of Rehmannia glutinosa. Nitrogen contents in rhizoma was positively correlated with the contents of catalpol and ethanol extract, whereas it was negatively correlated with that of ash, galactose and fructose. Calcium contents in rhizoma was highly negatively correlated with the contents of catalpol, ethanol extract and water extract. However, the calcium contents positively correlated with that of ash, acid insoluble ash, fructose and galactose in R. glutinosa. The catalpol cotents was negatively correlated with the contents of ash, galactose and fructose, whereas positively correlated with that of ethanol extract and water extract, and the correlations between extract contents and ash contents showed highly negative significance.

  • Veronicastrum species are perennial herb growing on the moisty land. They involves on species (V. sibiricum, Naeng-Cho), one subspecies (V. sibiricum var. zuccarini, Tuel-Naeng-Cho) and one variety (V. sibiricum var. zuccarini for. albiflora). This paper deals with a comparison of the chemical components between Naeng-Cho and Tuel-Naeng-Cho.) In the specified HPLC condition used with ${\mu}BondapakC_{18}\;column$, the retention time of aucubin, catalpol, minecoside and veratryl catalpol ester were 2.30, 2.65, 3.05 and 3.10 min, respectively, and in the other condition used with ${\mu}Bondapak\;C_8-NH_2$, the retention time of aucubin, catalpol, minecoside and veratryl catalpol were 5.4, 5.4, 4.4 and 3.2 min, respectively. The contents of minecoside in Tuel-Naeng-Cho were found to be 3.5 and 1mg/g of dry plant material and that of catalpol were found to be 5 and 2 mg/g, respectively.

Aucubin, Catalpol, and GABA Contents in Different Plant Parts of Rehmannia glutinosa Cultivars (지황 품종의 부위별 Aucubin, Catalpol, GABA 함량)

  • Background: Rehmannia glutinosa is a perennial herb belonging to the family Scrophulariaceae. Its root has been utilized as a traditional medicine but the aerial parts (flower, flower stalk, leaf) are not used. We aimed to determine the content of three compounds [aucubin, catalpol, and ${\gamma}$-aminobutyric acid (GABA)] in the different organs of R. glutinosa cultivars (Dakang, Tokang, and Suwon 9) Methods and Results: The flower, flower stalk, leaf, and root of R. glutinosa were harvested at the end of August. The aucubin and catalpol contents were analyzed by LC/MS, whereas the GABA content was analyzed by GC/MS. The aucubin content was the highest in the leaf, while catalpol and GABA were the highest in the flower. The aucubin contents of leaf in Dakang, Tokang, and Suwon 9 were 1.43, 0.81, and 1.07 mg/g, respectively. The catalpol contents of flower in Dakang, Tokang, and Suwon 9 were 41.06, 28.78, and 37.48 mg/g, respectively, the GABA contents were 0.79, 0.76, and 0.65 mg/g, respectively. Conclusions: The aucubin, catalpol, and GABA contents were higher in the leaf and flower than that in the root. This study show that R. glutinosa leaf and flower can be used as a potential supplement.

Effects of Soil Properties on the Contents of Catalpol, Sugars and Ash in the Rhizoma of Rehmannia glutinosa (토양이화학성(土壤理化學性)이 지황(地黃)(Rehmannia glutinosa) 근경(根莖)의 Catalpol, 당류(糖類) 및 회분함량(灰分含量)에 미치는 영향(影響))

  • The effects of the soil properties on the contents of catalpol, sugar and ash in the rhizoma of Ji-whang (Rehmannia glutinosa Lib. var. purpurea Makino) were studied to get information on cultivation for higher quality production. Catalpol contents of the rhizoma of Ji-whang ranged from 3.75 to 4.36%. It was shown that the sand contents of soil was positively correlated with the catalpol contents, and negatively correlated with the contents of fructose and galactose in the rhizoma of Ji-whang. The sand contents of subsoil was also negatively correlated with the contents of ash and acid insoluble ash in rhizoma. The clay contents of subsoil was negatively correlated with the catalpol contents, and positively correlated with the contents of fructose, galactose and sucrose in rhizoma, respectively. The clay contents of soil was positively correlated with the ash contents, and negatively correlated with the contents of ethanol extract and water extract in rhizoma, respectively. The organic matter contents of surface soil and subsoil were negatively correlated with the contents of catalpol and extracts (ethanol and water), and positively correlated with the contents of fructose and ash in rhizoma. The total nitrogen contents of surface soil was negatively correlated with the catalpol contents, and positively correlated with the contents of fructose and sucrose in rhizoma. The potassium contents of soil was highly positively correlated with the fructose contents in rhizoma.

Catalpol Content and Antioxidant Activities in Various Cultivars of Rehmannia glutinosa (품종별 지황의 Catalpol 함량 및 항산화능)

  • This study was to investigate catalpol content and antioxidant activities in five cultivars (Kokang, Suwon 10, Suwon 11, Jiwhang 1 and Korea) of Rehmannia Radix. Catalpol content was analysed by HPLC using $C_{18}$ column ($150{\times}4.6\;mm$, $5{\mu}m$), 1% acetonitrile and a UV detector. The standard curve was found to be a linear regression of y=0.0483x-0.0841 ($R^2$=0.999). The catalpol content ranged from 85.2 mg/g to 144.9 mg/g, of which the Korea was the highest among cultivars. The total phenol content in Rehmannia Radix was 0.358~0.459 mg/g, of which the Korea was the highest among cultivars. The antioxidant activities determined by DPPH and hydroxyl radical scavenging activities were the highest in Korea with $IC_{50}$ of 205.8 mg/g for DPPH radical scavenging activity and $IC_{50}$ of 38.8 mg/g for hydroxyl radical scavenging activity. Based on these results, the Korea among Rehmannia Radix cultivars was suggested to be a potent medicinal plant material for antioxidant activities.

Changes in Reducing Sugar and Catalpol Contents of Rehmannia Root Slurry with Aging Treatments (숙성처리에 따른 지황 슬러리의 환원당 및 카탈폴 변화)

  • Rehmannia glutinosa root (R. root) has been used as an traditional medicine, and is important resource for natural medicines and functional foods. However, R. root contains catalpol which is bitter, and undigested sugars, including stachyose and raffinose, which can cause diarrhea. Therefore, this study was performed to identify the changes in reducing sugar from undigested sugars and in catalpol contents in R. root slurry induced by aging treatments. R. root slurry was treated at $10{\sim}70^{\circ}C$ for up to 72 hr; and extracted with a 50% ethanol solution. The catalpol content was analyzed using HPLC-UVD. Reducing sugar content generated from undigested sugars was measured by the Nelson-Somogyi methods, and the reaction rates were calculated from their variation according to aging time and temperature. During the aging treatment, reducing sugar increased and catalpol decreased. Their formation and degradation rates were highest at $50^{\circ}C$ and $30{\sim}40^{\circ}C$, and their rates were $2.05mg/g{\cdot}hr$ and 23.09 to 23.33%/hr, respectively. These results indicated that aging treatment can positively affect the sweetness and digestibility of R. root slurry. Therefore, an aging treatment could be considered for improving the taste and digestibility of R. root.

Characteristics in Tissue Cultured Plants of Chinese foxglove(Rehmannia glutinosa) (지황 조직배양주의 수량성과 성분함량 특성)

  • Chinese foxglove(Rehmannia glutinosa) is receiving much attention as one of the principal medicinal crops and the crud drug. This study was conducted to obtain the basic breeding information of Chinese foxglove derived from tissue culture. To compare plant characteristics between local variety and tissue cultured seedlings, plant growth and root yield has been investigated. In addition, catalpol and free sugar contents were also analized. The ability for the storage of root stock originated from tissue culture seedlings were better than that of local variety. The growth and root yield of in vitro propagated plants were superior to those of conventionally propagated plants. Root yield of 1-year-old and 2-year-old seedlings investigated 112% and 246% respectively than Seacheon local shows 384 kg/10 a. Although there were no significant different between tissue culture seedlings and conventionally propagated one, slight decline of component contents in tissue culture plants were still existed. Investigated sugar content among Seocheon local, 1-year-old tissue culture seedlings, and 2-year-old tissue culture seedlings were 1.86%, 1.21%, and 1.10% respectively. Catalpol content as one of standard materials indicates 0.46% in Seocheon local and 0.40% in tissue cultured ones.

Catalpol and Mannitol, Two Components of Rehmannia glutinosa, Exhibit Anticonvulsant Effects Probably via GABAA Receptor Regulation

  • Epilepsy is a brain disorder that affects millions of people worldwide and is usually managed using currently available antiepileptic drugs, which result in adverse effects and are ineffective in approximately 20-25% of patients. Thus, there is growing interest in the development of new antiepileptic drugs with fewer side effects. In a previous study, we showed that a Rehmannia glutinosa (RG) water extract has protective effects against electroshock- and pentylenetetrazol (PTZ)-induced seizures, with fewer side effects. In this study, the objective was to identify the RG components that are responsible for its anticonvulsant effects. Initially, a number of RG components (aucubin, acteoside, catalpol, and mannitol) were screened, and the anticonvulsant effects of different doses of catalpol, mannitol, and their combination on electroshock- and chemically (PTZ or strychnine)-induced seizures in mice, were further assessed. Gamma-aminobutyric acid (GABA) receptor binding assay and electroencephalography (EEG) analysis were conducted to identify the potential underlying drug mechanism. Additionally, treated mice were tested using open-field and rotarod tests. Catalpol, mannitol, and their combination increased threshold against electroshock-induced seizures, and decreased the percentage of seizure responses induced by PTZ, a GABA antagonist. GABA receptor binding assay results revealed that catalpol and mannitol are associated with GABA receptor activity, and EEG analysis provided evidence that catalpol and mannitol have anticonvulsant effects against PTZ-induced seizures. In summary, our results indicate that catalpol and mannitol have anticonvulsant properties, and may mediate the protective effects of RG against seizures.
