  • The cholesterol and fatty acid levels in serum and liver were compared in 4-week-old Sprague-Dawley male fed by addition of polymannuronate (M, 5.0%), polyguluronate (G, 5.0%), and polymannuronate and polyguluronate (MG, 2.5% to each) with by addition of cholesterol (1.0%). Feed efficiency by the addition of M, MG, and G was lower than cholesterol fed group (p<0.01). The liver weights were less in M, MG, and G fed groups than in cholesterol fed group (p<0.01). Triglyceride levels in serum and liver were 58.2∼77.4% and 51.5∼65.5% lower in M, G and MG fed groups than cholesterol fed group, respectively. Total-, LDL-, and free-cholesterol levels in serum and liver in M, MG, and G fed group were significantly lower than cholesterol fed group. The cholesterol levels were the most reduced in M fed group. However, HDL- cholesterol level in serum was increased in M, MG, and G fed group (p<0.01). The of polyene levels were 47% higher in serum and 76% in liver in M fed group than in cholesterol fed group. The activities of GOT and GPT were lower in M, MG, and G fed group than in cholesterol fed group (p<0.01). Above the results demonstrate that supplementation of low molecular polymannuronate in diets improve physiologically lipid composition in serum and liver.

  • The purpose of this study was designed to observe the effects of Zizyphus jujuba seed extract on the concentrations of the lipids and blood glucose in the S.D. rats fed the experimental diets for 4 weeks. Concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, triglyceride (TG), phospholipid (PL) and blood glucose in serum were significantly higher in the cholesterol administration groups (group 2 (cholesterol+water), group 3 (cholesterol+Zizyphus jujuba seed extract)) than those in the control group (group 1, basal diet+water). But the concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, TG, PL and blood glucose in serum ware remarkably lower in the group 3 than those in the group 2. In the ratio of HIDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration, Zizyphus jujuba seed extract administration group was higher percentage than in the group 2. The activities of aspartate aminotransferase (AST), alanine amino-transferase (ALT), lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) in serum were rather lower in the Zizyphus jujuba seed extract administration group (group 3) than in the cholesterol diet group (group 2). From the above research, Zizyphus jujuba seeds were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hyperlipidemic rats. And particularly, Zizyphus jujuba seeds were more effective as a therapeutic regimen for the control of metabolic derangements in adult disease.

  • The present studies were designed to compare the effects of dietary fat sources and levels on plasma and tissue cholesterol. Changes in plasma total cholesterol and HDL-cholesterol, cholesterol concentration in aortas, liver and intestines were determined in young rats fed diets providing dietary fat as corn-oil or butter and levels as 10%, 20% or 30% of calories respectively 1) Plasma total cholesterol and HDL-cholesterol levels were little sensitive to the modification of dietary fat sources and levels. 2) Dietary cholesterol increased the levels of plasma total cholesterol and this effect was accentuated by feeding butter. But dietary cholesterol did not increase the levels of HDL-cholesterol in the butter-fed groups, but decreased in the corn-oil-fed groups. 3) Liver cholesterol concentrations were significantly higher in rats fed diets of corn-oil than those fed diets of butter. On the contrary, cholesterol concentrations of intestines were significantly higher in the butter-fed groups than the corn-oil-fed groups. However, in aortas, no significant differences were found.

  • Cardiovascular diseases (CVDs) are the most common cause of death in patients with nonalcoholic fatty liver disease (NAFLD) and dyslipidemia is considered at least partially responsible for the increased CVD risk in NAFLD patients. The aim of the present study is to understand how hepatic de novo lipogenesis influences hepatic cholesterol content as well as its effects on the plasma lipid levels. Hepatic lipogenesis was induced in mice by feeding a fat-free/high-sucrose (FF/HS) diet and the metabolic pathways associated with cholesterol were then analyzed. Both liver triglyceride and cholesterol contents were significantly increased in mice fed an FF/HS diet. Activation of fatty acid synthesis driven by the activation of sterol regulatory element binding protein (SREBP)-1c resulted in the increased liver triglycerides. The augmented cholesterol content in the liver could not be explained by an increased cholesterol synthesis, which was decreased by the FF/HS diet. HMG-CoA reductase protein level was decreased in mice fed an FF/HS diet. We found that the liver retained more cholesterol through a reduced excretion of bile acids, a reduced fecal cholesterol excretion, and an increased cholesterol uptake from plasma lipoproteins. Very low-density lipoproteintriglyceride and -cholesterol secretion were increased in mice fed an FF/HS diet, which led to hypertriglyceridemia and hypercholesterolemia in Ldlr-/- mice, a model that exhibits a more human like lipoprotein profile. These findings suggest that dietary cholesterol intake and cholesterol synthesis rates cannot only explain the hypercholesterolemia associated with NAFLD, and that the control of fatty acid synthesis should be considered for the management of dyslipidemia.

  • The present study was undertaken to investigate the effects of excess vitamin E supple-mentation (0.5%) and cholesterol (0.5%) on levels of serum and liver vitamin E and cholesterol in two categories of rats, group A and group B. Rats in group A(45-65g) were fed experimental diets for 3 weeks ( I-C, II-E, III-Ch, IV-ECh). On the other hand rats in group B(45g-65g) were first fed control diet for 3 weeks and then fed experimental diets when they were 100-l20g for the subsequent 3 weeks ( I'-C, II'-E, III'-Ch, & IV'-ECh). The levels of serum vitamin E were higher in vitamin supplemented groups as expected. Dietary cholesterol showed a tendency to lower serum vitamin E levels of vitamin E supplemented groups. Serum cholesterol levels tended to stay in a narrow range showing resistancy to dietary cholesterol and were not affected by vitamin E status. Whereas vitamin E supplementation seemed to lower the levels of hepatic cholesterol in both groups A & B( I-C, I'-C vs II-E, II'-E ), simultaneous supplementation of vitamin E with 0.5% cholesterol appeared to increase further the hepatic cholesterol levels which were already increased by cholesterol feeding ( IV-ECh, IV'-ECh, vs III- Ch, III'-Ch). Hepatic vitamin A levels decreased as rats grew older during the experimental period. Even though vitamin A levels did not differ from each other significantly, excess amount of vitamin E supplementation in group B seemed to show a tendency to decrease the vitamin A storage in liver.

  • This experiment was made in order to observe the change in contents of the total cholesterol, free cholesterol and ester cholesterol in the liver and serum by the feeding of glucose and sucrose. The animals used for the experiment were adult male albino rat from a pure strain, weighing 285-332g. The animals were divided into standard, glucose and sucrose diet groups. The glucose and sucrose diet groups were redivided into 10%, 25% and 40% diet groups. Their liver and serum were used as sample after they were fed with the corresponding diets, respectively, for two months. 1. In the liver, the total cholesterol and ester cholesterol contents in the 40% glucose diet group were significantly increased and the free cholesterol contents in all diets were respectively increased, compared with the standard diet group. 2. In the serum, the total cholesterol contents in both diet groups, the 10% glucose and 10% sucrose, were decreased but the content in the 40% sucrose diet group was increased, compared with the standard diet group. The free cholesterol contents in the 10 and 40% glucose diet and 10% sucrose diet group were decreased, compared with the standard diet group. The ester cholesterol contents in the 10% glucose and 10% sucrose diet groups were decreased but the content in the 40% sucrose diet group was increased, compared with the standard diet group. Being taken into consideration of the above facts it was acquired that the cholesterol contents are affected by the amounts and kinds of dietary carbohydrate when the protein contents in each diet were constant.

  • Egg cholesterol has been a hot issue in respect of it's effect on health and diseases in human. There is a general recognition that the cholesterol from eggs is similar to that from the other dietary sources in its biochemical functions. Many evidences show that egg yolk increased plasma cholesterol levels at an average rate of 2.3 mg /100 rnL plasma for every 100 mg cholesterol consumed. The elevation of plasma cholesterol could, however, be altered by various dietary factors such as ratios of polyunsaturated fatty acids and saturated fatty acids. Blood cholesterol levels of Korean adults have been raised up to, on the average, 193 and 187 mg /100 mL plasma for men and women, respectively, from 167/5 mg /100 mL in 1980. Furthermore, proportion of hypercholesterolemics(> 210 mg /dL) has been increased up to 23% of the population. These changes in blood cholesterol levels are attributed to the changes of dietary patterns, suggesting the arguement that Koreans are insensitive to changes of blood cholesterol is not right. Egg cholesterol levels have never been successfully reduced to any significant levels even with tremendous amounts of efforts made during last decades. The alternative and the best way to control blood cholesterol level originating from egg consumption appeares to enrich egg fat with omega-3 series fatty acids. Ingestion of the fatty acid group has specific functions to reduce hypertension and prevent various types of cancer as well as to reduce blood cholesterol. Another way to reduce egg cholesterol directly is to produce smaller egg yolk without reducing whole egg size since cholesterol is located only in the yolk. It is important to keep faith with consumers by providing correct informations on eggs, meanwhile efforts need to focus on to improve the quality of eggs as one of the best protein food items.

  • Gender differences in the effects of gonadectomy and high cholesterol diet on body weight and body cholesterol were investigated by using Sprague Dawley rats. Body weight, plasma and liver levels of cholesterol and triglyceride and platelet aggregation were examined in ovariectomized(OVX) or orchidectomized(ODX) rats with their intacts after feeding diet with or without 0.5% cholesterol. Body weight was significantly increased(p < 0.01) in OVX rats and significantly decreased(p < 0.01) in ODX rats compared to their respective intact rats, and cholesterol diet significantly(p < 0.05) decreased body weight in gonadectomized rats. Liver lobes from rats fed cholesterol diet were opaque and larger than those from rats find control dict, resulting in a significant increase(p < 0.01) in LW/BW ratio. Plasma and liver levels of total cholesterol were significantly increased (p < 0.01) in female rats regardless ovariectomy when find 0.5% cholesterol diet, but those levels in male rats were increased only when they were orchidectomized(p < 0.0l). Plasma HDL-cholesterol was significantly decreased(p < 0.05) in both sexes when find cholesterol diet. HDL-cholesterol were higher in female than male rats regardless treatments(p < 0.05). Liver triglyceride was significantly increased(p < 0.05) in both sexes when find cholesterol dict. Plasma level of triglyceride was not different among groups except significant decrease(p < 0.05) in cholesterol find ODX rats. Maximum platelet aggregation in female rats was significantly lower(p < 0.05) than male, but ovariectomy and cholesterol diet caused an increase te the level of male rats. Microscopic examination showed cholesterol diet caused a lipid accumulation in liver. Results indicate that intact female rats have higher response to hypercholestcrolemic diet than intact male rats and orchidectomy causes male rats more responsive to hypercholesterolemic diet. However, ovariectomy causes an increase female food efficiency ratio to the level of male rats, significantly increasing body weight.

  • This study done to investigate the hypocholesterolemic effect of ow dietary fiber in a high cholesterol diet in aspect of cholesterol absorption and excretion using Sprague Dawley rats. After feeding diets containing 0.5% cholesterol and 5% tangerine pulp, sea tangle or prickly pear cactus for four weeks, we measured the levels of plasma and liver cholesterol and triglyceride, fecal neutral sterols and fecal radioactivity after ingestion 14(sup)C-cholesterol. We also examined platelet aggregation and histological change in liver tissues in association of hypercholesterolemia. The liver to body weight ratio was significantly(p<0.01) lower in rats fed prickly pear cactus than in other groups. The levels of plasma total cholesterol, LDL-cholesterol and triglyceride were decreased significantly(p<0.01) in the prickly pear cactus group compared with the control, while there was no difference in the liver levels of total cholesterol and triglyceride among groups. Fecal corprostanol and cholesterol were significantly(p<0.01) higher groups of control and prickly pear cactus compared to the other two groups. Radioisotope excretion after ingestion of 14(sup)C-cholesterol was higher in the control group than in tangerine pulp group or sea tangle group, with the highest in prickly pear group. Radioisotope excretion was the highest during the 2nd and 3rd days in all groups. Hematocrit and platelet aggregation were decreased in all fiber groups compared to the control, but not statistically different. Microscopic examination showed that cholesterol diet cause a fat accumulation in the liver and prickly pear cactus decreased the fat accumulation. Result indicates that prickly pear cactus has hypocholesterolemic effect by decreasing absorption and increasing excretion of cholesterol, thereby protective effect on fatty liver. Control group fed diet containing high cholesterol and low fiber seems to have a self control system in cholesterol absorption and excretion preventing hypercholesterolemia. • Hurdle technique was used to remove cholesterol efficiently from liquid egg yolk. The quality of the low cholesterol egg products from the process were evaluated. From the 75 % cholesterol reduced egg yolk through $\beta$-cyclodextrin treatment. 2 times weight of soy bean oil was added to the egg yolk and homogenized followed by centrifuged to be maximized to remove cholesterol. When the pH of the yolk was adjusted to 9, 92 % of cholesterol was removed while 95.4 % of cholesterol was removed when 3 times weight of soy bean oil was added to the egg yolk. As the results of application of supercritical carbon dioxide extraction to the 75 % cholesterol reduced egg yolk through ${\beta}$-cyclodextrin treatment, 92.5 % of the cholesterol was removed from the egg yolk at $35^{\circ}C$, 4,500 psi, for 4 hours under co-solvent. The quality characteristics of the produced low cholesterol egg products were analysed. The cholesterol reduced egg yolk produced from ${\beta}$-cyclodextrin and soy bean oil treatment showed the lower emulsion capacity compared with control. The fatty acid composition of the cholesterol reduced egg yolk produced from ${\bet}a$-cyclodextrin and soy bean oil treatment showed increased C18:2 and C18:3 compared with control while decreased C16:1 and C18: 1 compared with control. The saponification method with extracting solvent of hexane showed that cholesterol concentration was 28.1 %. The quantity of hydrolysis solution(95 % ethanol : 33 % KOH = 94 : 6) was varied from 40 to 80 times of sample weights and the cholesterol concentration of 35.7 % was the highest result in the 60 times(v/w) hydrolysis solution. Cholesterol concentration of 35.7 % was recovered at the first trial with saponification method. but it could be improved up to 95.7 % after 4 times repetitive purification.
