Purification and Characterization of the Inhibitory Principle aganist Pancreatic Cholesterol Esterase from Ephedra herba (마황으로부터 췌장 Cholesterol Esterase 저해물질 분리 및 규명)

  • Cholesterol esterase(pCEH, pancreas cholesterol ester hydrolase, E.C.3.1.1.13) which is secreted from pancreas has been known as an important lipase for cholesterol uptake. cholesteryl acyl esters from a diet must be hydrolyzed to free cholesterol and fatty acid by cholesterol esterase before the absorption in small intestine. For the development of inhibitory substances from natural source, we screened many extracts of oriental herbs for the inhibition of cholesterol esterase in vitro. The ethanol extract of Ephedra herba showed strong inhibitory activity. Solvent fractionation and silica gel column chromatography with the extract lead to the purification of the inhibitory principle in Ephedra herba. Crystallized inhibitor was identified as ( ) ephedrine by using UV, FT IR, 1H NMR, 13C NMR and GC/Mass. These results suggest that ( ) ephedrine can be used as a potential lead compound for the development of inhibitor for cholesterol uptake by cholesterol esterase inhibition.

Plasma Cholesterol-Lowering Effects of Alpiniae katsumadaii Extract as an Inhibiter of Pancreatic Cholesterol Esterase Activity (췌장 콜레스테롤 에스터레이즈 저해제인 초두구 추출물의 혈중 콜레스테롤 저해효과)

  • Ethanol extract of Alpiniae Katsumadaii semen inhibited potently cholesterol esterase activity in vitro. Chloroform fraction of ethanol extract showed the stronger inhibitory effect than other solvent fractions-ethylacetate fraction, butanol fraction, and aqueous fraction. The chloroform frac ion of Alpiniae katsumadaii semen were studied as a candidator of plasma cholesterol lowering material in high cholesterol-fed rats. In high cholesterol-fed rats, the diet with chloroform fraction of 100 mg/kg and 150 mg/kg lowered not only plasma neutral lipids contents 25.9% and 26.5% but also plasma total cholesterol level 11.8% and 20.8%, respectively. Plasma HDL-cholesterol level and Atherogenic Index(AI) in Alpiniae chloroform fraction-fed rats were recovered as those level of normal rats.

Isolation and Structure Determination of a Cholesterol Esterase Inhibitor from Ganoderma lucidum

  • Bioassay-guided fractionation of a methanol extract of Ganoderma lucidum gave a pure cholesterol esterase inhibitor. On the basis of spectroscopic analysis and comparison with data from the literature, the structure of this compound was identified as $5{\alpha},8{\alpha}$-epidioxyergosta-6,22-dien-$3{\bet}$-ol (compound I). This compound inhibited cholesterol esterase activity with an $IC_{50}$ value of $42{\mu}M$. Lineweaver-Burk plot analysis revealed that compound I is a noncompetitive inhibitor. The findings of this study suggest that compound I may be the active principle of the hypocholesterolemic effect of Ganoderma lucidum.

Purification of Cholesterol Esterase from Aeromoans SP. (Aeromoans SP.가 생산하는 콜레스테롤 에스테라아제의 정제)

  • A cholesterol esterase-producing microorganism, strain CES 506, isolated from soil was identified as Aeromonas sp. This strain produce about 0.023 units of cholesterol esterase per ml of culture broth. The cholesterol esterase produced by this strain was purified, 370 fold to homogeneity in an overall yield of 24% from culture broth. The apparent molecular weight was 64, 000, as estimated by sodium dodecyl sulfate polyacrylamide gel electrophoresis. The enzyme showed a high substrate specificity for cholesterylpalmitate and the Km value for the hydrolysis of cholesterylpalmitate by this enzyme were 0.15mM.

  • Cholesterol-lowering effect of pine nut on mice was investigated by examining changes in body weight, feed intake, and triglyceride, total cholesterol, high density liporotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), and very low density lipoprotein cholesterol (VLDL-C) contents in mice plasma. Changes in body weight and feed intake between pre-designed experimental animal groups fed various contents of cholesterol and pine nut and control group fed basal diet were not significantly different. Food efficiency ratio of experimental groups fed higher than 10% pine nut was significantly higher than that of control group. Apparent differences in atherogenic index representing cholesterol-lowering effect in plasma were not found among all groups fed 0-20% pine nut. Inhibition activities of water and hexane extracts of pine nut on HMG-CoA reductase and cholesterol esterase were examined in vitro. Hexane extract showed 66% inhibition effect on HMG-CoA reductase, whereas none was observed with water extract.

  • Ethanol extract of Cinnamomi cortex inhibited potently cholesterol esterase activity in vitro. Chloroform fraction of ethanol extract showed the stronger inhibitory effect than other solvent fractions - ethylacetate fraction, butanol fraction, and aqueous fraction. The chloroform fraction of Cinnamomi cortex was studied as a candidator of plasma cholesterol-lowering material using high cholesterol-fed rats. In high cholesterol-fed rats, the diet with chloroform fraction of 150 mg/kg lowered not only plasma neutral lipids contents 25.1% but also plasma total cholesterol level 49.6% than only high cholesterol diet. Plasma HDL-cholesterol level in Cinnamomi cortex chloroform fraction-fed rats was recovered as those level of normal rats. LD$_{50}$ of Cinnamomi chloroform extract was calculated as 1,300 mg/kg.

  • To develop a home-version assay system for plasma lipoprotein cholesterol, variables that can control the assay performance were optimized. The system was constructcd by using two major components: nitrocellulose membrane strip with immobilized enzymes (cholesterol esterase, cholesterol oxidase, and horseradish peroxidase); and sample carrier solution containing non-ionic detergent (Triton X-100) and chromogen (3,3'-diaminobenzidine). Once a sample combined with the carrier was absorbed from the bottom of the strip, cholesterol was delivered by capillary action to the immobilized enzymes and a sequential reactions took place. In the final reaction, the chromogen was oxidized and then generated a color as signal that was proportional to the concentration of cholesterol. The signal intensity was enhanced by optimizing conditions for the immobilization of enzymes and the chemical composition of carriel. Under these conditions, a dose-response curve was obtained and revealed a high sensitivity enough to measure the cholesterol in blood.

Development of Total Cholesterol Detection System by Fluorescence Chromatography (형광 크로마토그래피에 의한 콜레스테롤 측정법의 개발)

  • In this study, we have developed a fluorescence chromatographic assay for the quantification of total cholesterol in serum, which is a well-known risk predictor for cardiovascular diseases. The new assay system consists of a chromatographic strip in a cartridge, enzyme buffer containing cholesterol esterase, cholesterol oxidase, horseradish peroxidase, and color developer AEC, and a laser fluorescence scanner. The correlation coefficient (r) between cholesterol concentration and relative fluorescence units was 0.968 in the new assay, showing a reliable linearity through the tested range of cholesterol. Recovery test and comparability with a Hitachi 747 instrument showed 106.5-94% and r = 0.939 (p<0.001), respectively. The new assay system for cholesterol was developed as a pre-POCT platform conducted in clinics since it is fast (8 min) and uses a small volume of sample ($5\;{\mu}l$), and it may be applied for on-site diagnostics to replace expensive automated biochemical analyzer.

Hyperlipidemic Inhibitory Effects of Phellinus pini in Rats Fed with a High Fat and Cholesterol Diet

  • This study evaluated the in vitro and in vivo hypolipidemic effects of the medicinal mushroom Phellinus pini. The methanol extract (ME) of the fruiting body of Ph. pini was active against pancreatic lipase and cholesterol esterase with 99.14% and 67.23% inhibited activity at 1.0 mg/mL, respectively. It also inhibited 81.81% and 55.33% of ${\alpha}$-glucosidase and ${\alpha}$-amylase activities, respectively, at 2.0 mg/mL. Hyperlipidemia as induced by feeding rats with a high fat and cholesterol diet (HFC). HFC supplemented with a 5% fruiting body powder of Ph. pini (HFC + PhP) significantly reduced plasma total cholesterol, low-density lipoprotein cholesterol, and triglycerides in rats compared with HFC. The reduced levels were comparable to rats fed the normal control diet (NC). The atherogenic index of HFC + PhP rats was significantly lower than that of the HFC rats. The excretion of fecal total lipid and cholesterol in the HFC + PhP rats was significantly higher than those in the NC and HFC rats. Histopathological examinations demonstrated scant deposition of lipids in the liver of rats fed HFC + PhP. The dietary supplementation with the fruiting body powder provided natural plasma lipid and glucose lowering effects in experimental rats without adverse effects on the plasma biochemical parameters and liver function related enzyme activities. Therefore, the hypolipidemic effects of Ph. pini may be due to the inhibitory effects on pancreatic lipase, cholesterol esterase, ${\alpha}$-glucosidase, and ${\alpha}$-amylase, and excretion of excess lipids and cholesterol in the feces.

  • Engorgement of macrophages with cholesterol is the defining pathological characteristic of atherosclerotic plaques, the cause of most heart attacks and strokes. Activated human macrophages uptake low density lipoprotein (LDL) by the bulk-phase fluid, and aggregated LDL by patocytosis. After incorporation of LDL and aggregated LDL into macrophages, neutral cholesterol esterase is decreased and cholesterol efflux is also deceased, resulting that macrophages become foam cells.
