Study on anti-oxidant and cosmeceutical activities of Isatis tinctoria L. (판람근(Isatis tinctoria L., 板藍根)추출물의 항산화활성 및 화장품약리활성에 관한 연구)

  • Objective : In this study, anti-oxidant and cosmeceutical activities of Isatis tinctoria L. extracted from water, ethanol and supercritical fluid condition were confirmed to investigate cosmeceutical activities for utilization as cosmetic ingredient. Methods: Anti-oxidant and cosmeceutical activities were investigated by using electron donating ability, xanthine oxidase, tyrosinase, astringent effect. Result : Isatis tinctoria L. extracts by supercritical fluid, water and ethanol showed good electron donating ability which were 82.7%, 62.6% and 44.8% at the concentration at 1,000ppm, respectively. Xanthine oxidase activity related with purine metabolism was inhibited by ethanol extract about 52.3% at the concentration at 1,000ppm. Tyrosinase inhibition effects, by supercritical fluid extract, ethanol extract and water extract, were 83.3%, 52.9% and 41.2% respectively at 1,000ppm. In the measurement of astringent effect, supercritical fluid extract at the concentration to 5,000ppm showed 85.7% in related activity. The water extract showed 95.9% nitrite scavenging activity at 5,000ppm. Conclusion : According to these results, it is possible that the extract of Isatis tinctoria L. can be used as a new natural material of cosmetic industry.

  • Objectives : In this study, anti-oxidant activity of Taraxacum platycarpum is confirmed to investigate cosmeceutical activity for utilization as a cosmetic ingredients. Methods : In the anti-oxidant and whitening effect experiment, the electron donating ability of Taraxacum platycarpum extracts, their SOD-like activity, and the inhibitory activity of xanthine oxidase, tyrosinase were examined. Results : Radical-scavenging activity of water and ethanol extract was examined using ${\alpha},{\alpha}-diphenyl-{\beta}-picrylhydrazyl$ (DPPH). Water and ethanol extract from Taraxacum platycarpum showed 62.9% and 54.7% at 500 ppm concentration in DPPH radical inhibition, respectively. In the test of superoxide dismutase (SOD)-like activity, water extract showed 63.4% at 1,000 ppm concentration, while ethanol extract showed 49% at 1,000 ppm concentration. In the test of xanthine oxidase inhibition, ethanol extract showed 66.8% at 1,000 ppm concentration. Tyrosinase inhibition effect related to whitening effect showed 36.3% and 54.2% in ethanol extract and water extract at 1,000 ppm concentration, respectively. Conclusion : According to these results, it is possible that the extract of Taraxacum platycarpum can be used as a new natural material of cosmetic industry.

  • Objectives : The cosmeceutical activity of Cuacuta japonica Choisy were investigated for cosmetics industrial application. Methods : Cosmeceutical activities include antioxidant, tyrosinase inhibition effects and nitrite-scavenging ability were examined through the Cuacuta japonica Choisy Results : The result were obtained as follows : The electron donating ability of water and ethanol extract was 69.5%, 78.3% at 1,000ppm. In SOD-like activity, water extract showed low effect, but ethanol extract scored 63.4% at 1,000ppm. In inhibition activity of xanthine oxidase, water extract showed low effect, which is 27.2% at 500ppm, in contrast to ethanol extract high effect: 99.5% at 500ppm, in nitrite-scavenging ability test, 500ppm of C. japonica Choisy water extract showed the effect of 51.3%, while 500ppm of C. japonica Choisy ethanol extract showed the effect of 72.7%. From the test of inhibitory effect of tyrosinase, both water and ethanol extract showed insufficient inhibitory rate. Conclusion : According to all the test results that have been given, it is possible that the extract of C. japonica Choisy can be used as a new natural material of the cosmeceutical industry.

  • Purecell, hydroxyethyl-cellulose-2-hydroxypropyltrimethyl ammonium chloride ether, was developed as a thickener and evaluated its functions for stability and safety when it was added in the cosmetic composition. Compared with a commercially available thickener Natrasol, Purecell have a basic chemical backbone of hydroxyethylcellulose which has been widely used in the cosmetic industry. pH and viscosity of Purecell was lower than those of Natrasol, however, a similar pH value to was shown when the Purecell was added into a shampoo or cream. The Purecell was stable to temperature and light. When 0.5% of the Purcell was added to a shampoo having cationic surfactant, a separation occurred due to the same cationic surfactant of the Purecell and shampoo. However, the shampoo and cream made with Purecell improved a smoothness and adaptability of hair. It also showed a negative response for Patch test of human, indicating the Purecell was safe for use.

  • Objectives : The physiological activities of Coptidis Rhizoma investigated for application as a cosmetic ingredients. Methods : We were experimented anti-oxidation effect, growth inhibition ability on cancer cells, antibacterial activity on various kinds of bacteria of skin. Results : The results were obtained as follows : Electron-donating ability(EDA) shows that Coptidis Rhizoma extracted with ethanol(94.51%) gives higher EDA in comparison to that extracted with water in 1000 ppm(63.24%). In the test of SOD-like activity, ethanol extract showed quite more activity with 81.01% in 1000 ppm, while water extract was low in 18.22%. In the inhibition activity of Xathine oxidase, ethanol extract showed higher inhibition activity compared to water extract. In measuring lipid oxidation inhibition ability using fish oil, ethanol extract in 100 ppm showed prominent ability. In the oxidation inhibition effect added with $Cu^{2+}\;ion\;and\;Fe^{2+}$ ion as a oxidation promoter, ethanol extract in 1000 ppm, each in the proportion of 90% and 92%, showed high oxidation inhibition effect compared with water extract. Conclusions : The results indicated that, ethanol extract which is superior in its anti-oxidation and antibacterial effect is useful to be applied in cosmetic industry.

  • The aim of this study was to assess the cosmeceutical activies of four kinds of Korea herb medicine extracts using in cosmetics and related industries. The cosmeceutical activities of extracts were investigated by tyrosinase inhibition, astringent, anti-bacterial and MTT assay for cell viability. In the whitening effect, PA that the highest tyrosinase inhibition activity showed 56% at 10 ppm in ethanol extract. Also water and ethanol extract of RE showed 54%, 68% at 1,000 ppm, respectively, but LE and CA showed lower effect. Astringent effect of water and ethanol extract of PA appeared over 60% at 1,000ppm concentration but other extracts showed no astringent effect. In the anti-bacterial test, water and ethanol extract of PA showed no anti-bacterial effect against all microorganisms. But water and ethanol extract of RE showed anti-bacterial effect on Staphylococcus aureus, ethanol extract of showed on Staphylococcus aureus and Propionibacterium acnes, ethanol extract of CA showed on Candida albicans. The resUlt of stability test showed that the emulsion of containing PA were very stable at various temperature and sun-light test. Viscosity and pH of emulsion did not change. From the results of human patch test to assess the safety of cosmetics containing PA there was no negative reaction on skin was found.

Cosmeceutical activities and Anti-inflammatory effects of Shell from Persicae semen (도인(桃仁) 껍질의 화장품약리활성 및 항염 효과)

  • Objectives : In this paper, we tested the applicability of shell from Persicae semen in cosmetics through cosmeceutical activities including anti-oxidant, tyrosinase inhibition and anti-inflammatory effects. Methods : Persicae semen, which had been extracted, concentrated, and freeze drying with water and ethanol, have been used for the experiment. The effects on electronic donating ability, SOD-like activity, xanthine oxidase inhibition, whitening effect have been investigated in the cosmeceutical activity measurement of function experiment. Results : In the electron donating ability test, 1,000ppm of EPS (ethanol extract of shell from Persicae semen) showed an effect of 87%. SOD-like activities showed 93% at the 10,000ppm of WPS (water extract of shell from Persicae semen). In the xanthine oxidase inhibition test, 1,000ppm of BHA showed an effect of 27%, while EPS showed an effect of 62%. We were able to get an effect of 95% from EPS at 10,000 ppm in the tyrosinase inhibition test. In the anti-inflammatory test, the EPS inhibited the generation of nitric oxide. In the case of the EPS, there were no signs of cytotoxicity against raw 264.7 and anti-inflammatory effects could be identified when the manifestation of iNOS was decreased. Conclusion : Therefore, the EPS has potential as an effective raw materials for cosmetic.

  • Objectives : This study aimed to establish cosmeceutical activities of Perilla frutescens var. acuta. Methods : Folium Perillae, which had been extracted, concentrated, and freeze drying with water and ethanol, have been used for the experiment. The effects on electronic donating ability, SOD-like activity, xanthine oxidase inhibition, whitening effect have been investigated in the cosmeceutical activity measurement of function experiment. Results : Both water and ethanol extracts of Perilla frutescens var. acuta showed relatively high electron donating ability of more than 70% at over 500ppm. Also, SOD like activity increased in a dose dependent matter of more than 80% at 5,000ppm, while tyrosinase showed insufficient inhibitory rate. Xanthine oxidase showed a meaningful inhibitory effect of 72.4% in water extract and 55.3% at 1,000ppm in ethanol extract. The addition of $Fe^{2+}ion\;and\;Cu^{2+}ion$ showed relatively high oxidizing inhibitory effect of 57% and 41%, respectively, at 500ppm in water extract and, when $Fe^{2+}$ ion was added in ethanol extract, the effect of 64% at 1,000ppm was achieved. Also, higher oxidizing inhibitory effect was shown against $Fe^{2+}$ rather than $Cu^{2+}$ in both water and ethanol extracts. For human cancer cells, relatively high growth inhibition ability was shown against melanoma G361 in both water and ethanol extracts. Conclusion : From the above results, it was confirmed that Perilla frutescens var. acuta could be used in functional cosmetics.
