  • This study was designed to evaluate the effects of anesthetics on waveform of SEPs and to authorize possible anesthetic protocol for measurement of the somatosensory evoked potentials (SEPs). Thirteen anesthetic methods were used. The SEPs were recorded on two channels (between the 5th and 6th lumbar vertebra as the channel 1 and between the 11th and 12th thoracic vertebra as the channel 2) following stimulation of posterior tibial nerve. ID analyze SEPs wave, latency and conduction velocity were measured. Among thirteen anesthetic methods, standard SEPs waveforms were observed in dogs anesthetized with following six methods: Acepromazine + Thiepfntal Na + Isoflurane, Acepronazine + Propofol + Isoflurane, Diazepam + Xylazine, Xylazine + Ketamine, Acepromazine + Propofol infusion and Propofol infusion. Above six methods could be used with sufficient anesthetic depth. The differences of latency and conduction velocity among six groups were minimal compared to general waveform of SEPs. These results indicate that the six anesthetic methods can be used for recording SEPs in the dog. In particular, Diazepam + Xylazine and XylaBine + Ketamine as injectable anesthesia are considered more convenient than other four methods in veterinary medicine.

  • The aim of this study was to develop an analytical method for the quantification of diazepam residues in fishery products, using liquid and gas chromatography-tandem mass spectrometry (LC-MS/MS and GC-MS/MS). The sample utilized in the study was extracted from the fish sample (crucian carp) using 0.1% formic acid in acetonitrile. For the utilization of the purification process, the dispersive solid phase extraction (dSPE) was used for LC-MS/MS, dSPE and SPE was used for GC-MS/MS, respectively. To be sure, the standard calibration curves showed a good linearity as the noted correlation coefficients, $r^2$ was > 0.99. The average recoveries for accuracy ranged in 99.8~124% for the samples which were fortified at three different levels (0.001, 0.002 and 0.010 mg/kg). The correlation coefficient for the precision effect was measured at a range of 4.01~11.8%. The limit of detection (LOD) for the diazepam analysis was 0.0004 mg/kg, and the limit of the quantification (LOQ) was 0.001 mg/kg. The proposed analytical method was characterized with a high accuracy and acceptable sensitivity to meet the established Codex Alimentarius Commission (CAC/GL71-2009) guideline requirements. We therefore established the optimal analysis method for the determination of diazepam in the fishery products using LC-MS/MS and GC-MS/MS. It would be applicable to analyze the diazepam residues in fishery products in further studies on this subject.

  • Diazepam (DZ) is one of the most frequently prescribed drugs as an antianxiety agent. muscle relaxant. and anticovulsant and sometimes causes intoxication due to accidental overdose, misuse or abuse. DZ is metabolized to nordiazepam (NDZ, desmethyldiazepam), oxazepam (OX) and temazepam (TM) which are also pharmacologically active, although OX and TM do not accumulate in blood or plasma to an appreciable extent. • The roots of the plant Korean ginseng have been extensively used in the traditional Chinese herbal medicine. The effects of chronic administration of Korean ginseng extract (KGE) were investigated on two different anxiety models: the elevated T-maze (for inhibitory avoidance and escape measurements) and the open field test (OFT). Diazepam (1 mg/kg), KGE (10, 30 and 100 mg/kg) were administered orally for 15 days. On the 14th day, mice were previously exposed for 30 min to one of the open arms of the T-maze, 24 h before the test. On 15th day, mice had two exposures to the enclosed and open arm of the elevated T-maze followed by exposure to the open field apparatus. The number of line crossings in the apparatus was used to assess locomotor changes. Cumulative Concentration Response Curve of 5-HT was plotted using rat fundus which were pre-treated in a similar way. Treatment with Diazepam (1 mg/kg) and KGE (10, 30 and 100 mg/kg) significantly (P < 0.05) impaired inhibitory avoidance performance but did not impair escape latency. In OFT, diazepam facilitated locomotion as compared to vehicle and other treatment groups. KGE at any of the selected doses did not impair locomotion. Concentration response curve of 5-HT was shifted towards the right with suppression of maxima in rats treated with KGE. The results suggest that KGE exerts anxiolytic like behaviour in a specific subset of defensive behaviour, particularly those related to generalized anxiety disorder.

  • Zizyphi Spinosi Semen (ZSS), a traditional Chinese folk medicine, has been used for treatment of insomnia and anxiety. This experiment was performed to investigate the anxiolytic-like effect of methanol extract of ZSS (MEZSS) in mice by using the experimental paradigms of anxiety and compared with that of a known anxiolytic, diazepam. In the elevated plus-maze test, it showed that MEZSS (100 mg/kg, p.o.) and diazepam (2.0 mg/kg, p.o.) increased the percentage of time spent on the open arms and the number of open arms entries. MEZSS (50, 100 and 200 mg/kg, p.o.) and diazepam (0.5 mg/kg, p.o.) significantly increased the number of head dips compared with that of control group in the hole-board test. However, MEZSS has no effect on decreasing the locomotor activity, while diazepam (2.0 mg/kg, p.o.) significantly inhibited locomotor activity. MEZSS did not decrease the strength force in the grip strength test, either. In addition, GABAergic involvements were also investigated to understand the possible mechanisms. $GABA_{A}$ receptors subunits and glutamic acid decarboxylase (GAD) were not over expressed, compared with that of the saline group. We also found that MEZSS did not increase chloride influx in cultured cerebellar granule cells. It is concluded that MEZSS might have anxiolytic-like effects, but these effects might not be mediated by GABAergic transmission.

  • This study developed effective assay method of pharmaceutical quality control was developed by near-infrared spectroscopy (NIRS). The calibration equation model of assay was developed by 2nd deriviative PLS(Partial Least Squares) regression method with NIRS over the wavelength range from 1100 to 1400nm using diazepam tablets (2mg, 5mg). Although diazepam tablets are made by 5-different manufacture, they have similar formulation. When the correlation was compared with values by NIRS and HPLC, the R-2s and standard error of calibration (SEC) for 2mg were 0.9300 and 0.98%, the R-2s and SEC for 5mg were 0.9165 and 0.63%. The validation of the calibration equation model yield that the R-2s and standard error of prediction (SEP) for 2mg were 0.9611 and 0.995%, the R-2s and SEP for 5mg were 0.9114 and 0.842%. The method was validated on assay method for diazepam tablets by the calibration equation.

  • To detect anxiolytic activity from Sanguisorbae Radix we used various animal models of fear or anxiety that are sensitive to known anxiolytic drugs. While diazepam showed significant anxiolytic activities in all five animal models empolyed in this study, $5-HT_3$ antagonist ondansetron and ethylacetate fraction of Sanguisorbae Radix did show anti-anxiety effects in social interaction and two compartment exploration tests. Ethylacetate fraction of Sanguisorbae Radix and 5-HT related drugs like ondansetron and buspirone, however, seem to have merit over diazepam in terms of not causing drowsiness. Among ten subfractions obtained from ethylacetate fraction of Sanguisorbae Radix by silica gel chromatography, subfraction I showed higher anxiolytic activities than subfraction DEF in two animal models, social interaction and two compartment exploration tests. There is growing evidence for the role of 5-HT in the control of anxiety. We hope that new compound(s) will be found from the active fractions of Sanguisorbae Radix as a potential anxiolytic agent in the future.

  • Murraya koenigii leaves commonly known as 'curry patta' are routinely added to Indian gravy and vegetarian dishes by south Indian as a favourate condiment. The present study was undertaken to investigate the effects of Murraya koenigii leaves (MKL) on memory in rats. Elevated plus-maze and Hebb-Williams maze served as the exteroceptive behavioral models for testing memory. Diazepam-, scopolamine- and ageing-induced amnesia served as the interoceptive behavioral models. MKL fed orally to various groups of young and aged rats with diet containing 2, 4 and 8% w/w of MKL for 30 days consecutively were investigated. The MKL diets produced a significant dose-dependent improvement in memory scores of young and aged rats and significantly reduced the amnesia induced by scopolamine (0.4 mg/kg, i.p.) and diazepam (1 mg/kg, i.p.). Therefore, it would be worthwhile to specifically investigate the therapeutic potential of MKL in the management of dementia patients.
