Phosphorylation of Elongation Factor-2 And Activity Of Ca2+/Calmodulin-Dependent Protein Kinase III During The Cell Cycle

  • Phosphorylation of the eukaryotic elongation factor 2 (eEF-2) blocks the elongation step of translation and stops overall protein synthesis. Although the overall rate of protein synthesis in mitosis reduces to 20% of that in S phase, it is unclear how the protein translation procedure is regulated during the cell cycle, especially in the stage of peptide elongation. To delineate the regulation of the elongation step through eEF-2 function, the changes in phosphorylation of eEF-2, and in activity of corresponding $Ca^{2+}$/calmodulin (CaM)-dependent protein kinase III (CaMK-III) during the cell cycle of NIH 3T3 cells, were determined. The in vivo level of phosphorylated eEF-2 showed an 80% and 40% increase in the cells arrested at G1 and M, respectively. The activity of CaMK-III also changed in a similar pattern, more than a 2-fold increase when arrested at G1 and M. The activity change of the kinase during one turn of the cell cycle also demonstrated the activation at G1 and M phases. The activity change of cAMP-dependent protein kinase (PKA) was reciprocal to that of CaMK-III. These results indicated: (1) the activity of CaMK-III was cell cycle-dependent and (2) the level of eEF-2 phosphorylation followed the kinase activity change. Therefore, the elongation step of protein synthesis might be cell cycle dependently regulated.

Improved Production of Long-Chain Fatty Acid in Escherichia coli by an Engineering Elongation Cycle During Fatty Acid Synthesis (FAS) Through Genetic Manipulation

  • The microbial biosynthesis of fatty acid of lipid metabolism, which can be used as precursors for the production of fuels of chemicals from renewable carbon sources, has attracted significant attention in recent years. The regulation of fatty acid biosynthesis pathways has been mainly studied in a model prokaryote, Escherichia coli. During the recent period, global regulation of fatty acid metabolic pathways has been demonstrated in another model prokaryote, Bacillus subtilis, as well as in Streptococcus pneumonia. The goal of this study was to increase the production of long-chain fatty acids by developing recombinant E. coli strains that were improved by an elongation cycle of fatty acid synthesis (FAS). The fabB, fabG, fabZ, and fabI genes, all homologous of E. coli, were induced to improve the enzymatic activities for the purpose of overexpressing components of the elongation cycle in the FAS pathway through metabolic engineering. The ${\beta}$-oxoacyl-ACP synthase enzyme catalyzed the addition of acyl-ACP to malonyl-ACP to generate ${\beta}$-oxoacyl-ACP. The enzyme encoded by the fabG gene converted ${\beta}$-oxoacyl-ACP to ${\beta}$-hydroxyacyl-ACP, the fabZ catalyzed the dehydration of ${\beta}$-3-hydroxyacyl-ACP to trans-2-acyl-ACP, and the fabI gene converted trans-2-acyl-ACP to acyl-ACP for long-chain fatty acids. In vivo productivity of total lipids and fatty acids was analyzed to confirm the changes and effects of the inserted genes in E. coli. As a result, lipid was increased 2.16-fold higher and hexadecanoic acid was produced 2.77-fold higher in E. coli JES1030, one of the developed recombinants through this study, than those from the wild-type E. coli.

Fatigue characteristics of distributed sensing cables under low cycle elongation

  • When strain sensing cables are under long-term stress and cyclic loading, creep may occur in the jacket material and each layer of the cable structure may slide relative to other layers, causing fatigue in the cables. This study proposes a device for testing the fatigue characteristics of three types of cables operating under different conditions to establish a decay model for observing the patterns of strain decay. The fatigue characteristics of cables encased in polyurethane (PU), GFRP-reinforced, and wire rope-reinforced jackets were compared. The findings are outlined as follows. The cable strain decayed exponentially, and the decay process involved quick decay, slow decay, and stabilization stages. Moreover, the strain decay increased with the initial strain and tensile frequency. The shorter the unstrained period was, the more similar the initial strain levels of the strain decay curves were to the stabilized strain levels of the first cyclic elongation. As the unstrained period increased, the initial strain levels of the strain decay curves approached those of the first cyclic elongation. The tested sensing cables differed in the amount and rate of strain decay. The wire rope-reinforced cable exhibited the smallest amount and rate of decay, whereas the GFRP-reinforced cable demonstrated the largest.

Creep-Rupture and Fatigue Properties of Transient Liquid Phase Bonded Joints of Ni-Base Single Crystal Superalloy (액상확산접합한 Ni기 단결정 초내열합금의 크리프 파단 및 피로특성)

  • The creep-rupture and low cycle fatigue properties of transient liquid phase bonded joints of Ni-base single crystal superalloy, CMSX-2 was investigated using MBF-80 insert metal. The (100) orientation of bonded specimen was aligned perpendicular to the joint interface. CMSX-2 was bonded at 1523K for 1.8ks in vacuum, optimum bonding condition. The creep rupture strength and rupture lives of the joints were the almost identical to ones of the base metal. SEM observation of the fracture surfaces of joints after creep rupture test revealed that the fracture surfaces classified three types of region, ductile fracture surface, cleavage fracture surface and interfacial fracture surface. The low cycle fatigue properties of the joints were also the same level as those of base metal. The elongation and reduction of area values of joints were comparable to those of base metal while fell down on creep rupture condition of high temperature.

  • Alloy 600 having a superior resistance to a corrosion is used as a steam generator tubing in nuclear power plants. In spite of its high corrosion resistance, there are many tubings which experience corrosion problems such as a SCC under the high temperature and high pressure environments of nuclear power plants. The Alloy 600 tubing can be repaired by using a Ni electroplating having an excellent SCC resistance. In order to carry out a successful Ni electrodeposition inside a steam generator tubing, the effects of various parameters on the material properties of the electrodeposit should be elucidated. Hence this work deals with the effects of an applied current density, duty cycle($T_{on}/(T_{on}+T_{off})$) of a pulse current, bath temperature and solution pH on the material properties of Ni electrodeposit obtained from a Ni sulphamate bath by analyzing the current efficiency, potentiodynamic curve, hardness and stress-strain curve. Hardness, YS(yield strength) and TS(tensile strength) decreased whereas the elongation increased as the applied current density increased. This was thought to be by a concentration depletion at the interface of the electrodeposit/solution, and a fractional decrease of the hydrogen reduction reaction. As the duty cycle increased, the hardness, YS and TS decreased while the elongation increased. During an off time at a high duty cycle, the concentration depletion could not be recovered sufficiently enough to induce a coarse grain sized electrodeposit. With an increase of the solution temperature and pH, the YS and TS increased while the elongation decreased. The experimental results of the hardness and the stress-strain curves can be supplemented by the results of the potentiodynamic curve.

Phosphorylation of Eukaryotic Elongation Factor 2 Can Be Regulated by Phosphoinositide 3-Kinase in the Early Stages of Myoblast Differentiation

  • We have previously reported that phosphorylation of eukaryotic elongation factor 2 (eEF2) is related to the differentiation of chick embryonic muscle cells in culture. In the present study, we found that eEF2 phosphorylation declined shortly after induction of differentiation of L6 myoblasts, when the cells prepare for terminal differentiation by withdrawing from the cell cycle. This decrease in phosphorylation was prevented by inhibitors of phosphoinositide 3-kinase (PI3-kinase) that strongly inhibit myoblast differentiation. We hypothesized that PI3-kinase plays an important role in myoblast differentiation by regulating eEF2 phosphorylation in the early stages of differentiation. To test this hypothesis, myoblasts were synchronized at in $G_2/M$ and cultured in fresh differentiation medium (DM) or growth medium (GM). In DM the released cells accumulated in $G_0$/$G_1$ while in GM they progressed to S phase. In addition, cyclin D1 was more rapidly degraded in DM than in GM, and eEF2 phosphorylation decreased more. Inhibitors of PI3-kinase increased eEF2 phosphorylation, but PI3-kinase became more activated when eEF2 phosphorylation declined. These results suggest that the regulation of L6 myoblast differentiation by PI3-kinase is related to eEF2 phosphorylation.

A Study on Low-Cycle Fatigue Behavior at Elevated Temperature of High Carbon Steel Used For Structural Purpose (構造용高炭素鋼材 의 高溫 低 사이클 피勞擧動 에 關한 硏究)

  • This study was undertaken to determine tensile properties and low-cycle fatigue behavior of 0.6%C high carbon steel used of structural purposes at temperatures up to 500.deg.C. In the low-cycle fatigue test the upper limit was decided by elongation(i.e. the total strain range), while the lower limit was defined by the load (i.e. zero load). The following results were obtained. Both, the ultimate tensile strength and low-cycle fatigue resistance attain the maximum values near 250.deg.C. Above this temperature the values decrease rapidly as the temperature increases. The low-cycle fatigue resistance decreases whenever there is an increase of the total strain range. Because the hardness of cycle fatigued specimen correlates cyclic hardening and cyclic softening, therefore the hardness of cycle fatigued specimen is smaller than that of the nonfatigued specimen at room temperature and 500.deg.C but much larger than the hardness of the nonfatigued specimen near 250.deg.C.

Evaluation of the Herbal Extract Mixture for the Effects of Hair-Regrowth Compared to 3% Minoxidil; Elongation of Anagen Period on C3H Mice (모발생장기 유도 C3H 생쥐에 있어서 미녹시딜과 생약추출 혼합 조성물의 모발 재성장 유도 효능)

  • The hair cycle consists of three phases, growth (anagen), involution (catagen) and quiescence (telogen) phases. In order to evaluate hair re-growth effect of herbal extracts mixture containing the 70% ethanol extracts of Polygoni Multiflori Radix, Mori Cortex Radicis, Gingko Biloba Folium and Pine bud, we have examined the induction of the anagen phase and/or elongation of the anagen period using C3H mice. Morphological examination was done by Hattori' and Ogawa's method. Enzyme activities of ${\gamma}$-glutamyl transpeptidase (${\gamma}$-GT) and alkaline phosphatase (ALP) was detected by Bessey-Lovry-Brock's method. Enzyme activity as a biochemical marker of hair cycle was investigated in the third hair cycle period of C3H mice after depilation. 3% Minoxidil treated group and herbal extract mixture treated group were shown 3 days earlier initiation of anagen than control group. In cycling mouse skin, ${\gamma}$-GT activity is pronounced during anagen and greatly diminished during telogen. Herbal extract mixture has shown promising hair re-growth effect on hair follicular cycles of C3H mice.

Effect of Thermo-mechanical Treatment on the Tensile Properties of Fe-20Mn-12Cr-3Ni-3Si Damping Alloy (Fe-20Mn-12Cr-3Ni-3Si 합금의 인장성질에 미치는 가공열처리의 영향)

  • This study was carried out to investigate the effect of thermo-mechanical treatment on the tensile properties of Fe-20Mn-12Cr-3Ni-3Si alloy with deformation induced martensite transformation. ${\alpha}^{\prime}$ and ${\varepsilon}$-martensite, dislocation, stacking fault were formed, and grain size was refined by thermo-mechanical treatment. With the increasing cycle number of thermo-mechanical treatment, volume fraction of ${\varepsilon}$ and ${\alpha}^{\prime}$-martensite, dislocation, stacking fault were increased, and grain size decreased. In 5-cycle number thermo-mechanical treated specimens, more than 10% of the volume fraction of ${\varepsilon}$-martensite and less than 3% of the volume fraction of ${\alpha}^{\prime}$-martensite were attained. Tensile strength was increased and elongation was decreased with the increasing cycle number of thermo-mechanical treatment. Tensile properties of thermo-mechanical treated alloy with deformation induced martensite transformation was affected to formation of martensite by thermo-mechanical treatment, but was large affected to increasing of dislocation and grain refining.

The Effects of Cyclic Heat Treatment Process for Fine Microstructure of TiAl Cast Alloy (주조용 TiAl 합금의 조직 미세화를 위한 반복열처리 공정 조건에 관한 연구)

  • For expanding the applications and workability of TiAl alloy, elongation is very important property. Fine microstructure is needed for elongation and physical properties of TiAl alloys. In this study, The effects of cyclic heat treatment process for fine microstructure of Ti-46Al-Nb-W-Cr-Si-C alloy, which was made by VAR (vacuum arc remelting) and VIM(vacuum induction melting) centrifugal casting process, was investigated. Cycle heat treatment process was very effective for recrystallization of this TiAl system, which has microstructure size of $50{\sim}100{\mu}m$ through pre-heat treatment, cyclic heat treatment in ${\alpha}+{\gamma}$ phase region and solution heat treatment respectively. Refined grain size was finally confirmed by photos of optical microscope and scanning electron microscope.
