Enantioselective Resolution for the Preparation of Chiral para-Nitrostyrene Oxide by Microbial Epoxide Hydrolase in an Organic Solvent (미생물 에폭사이드 가수분해효소 활성을 이용한 유기용매에서의 광학활성 para-Nitrostyrene Oxide 생산)

  • Enantioselective resolution of racemic para-nitrostyrene oxide was investigated using epoxide hydrolase activity of Aspergillus niger LK for the production of optically pure (S)-para-nitrostyrene oxide. To overcome the poor solubility of the substrate, enantioselective hydrolysis in an organic solvent was attempted under optimized reaction conditions including reaction temperature and water content. (S)-para-Nitrostyrene oxide with high optical purity (> 99% ee) was obtained at 37% yield using fungal epoxide hydrolase-catalyzed enantioselective resolution.

  • An enantioselective lipase gene (esf) for the kinetic resolution of optically active (S)-flurbiprofen was cloned from the new strain Serratia marcescens ES-2. The esf gene was composed of a 1,845-bp open reading frame encoding 614 amino acid residues with a calculated molecular mass of 64,978 Da. The lipase expressed in E. coli was purified by a three-step procedure, and it showed preferential substrate specificity toward the medium-chain-length fatty acids. The esf gene encoding the enantioselective lipase was reintroduced into the parent strain S. marcescens ES-2 for secretory overexpression. The transformant S. marcescens BESF secreted up to 217kU/ml of the enantioselective lipase, about 54-fold more than the parent strain, after supplementing 3.0% Triton X-207. The kinetic resolution of (S)-flurbiprofen was carried out even at an extremely high (R,S)-flurbiprofen ethyl ester [(R,S)-FEE] concentration of 500 mM, 130 kU of the S. marcescens ES-2 lipase per mmol of (R,S)-FEE, and 1,000 mM of succinyl ${\beta}-cyclodextrin$ as the dispenser at $37^{\circ}C$ for 12h, achieving the high enantiomeric excess and conversion yield of 98% and 48%, respectively.

  • Enantioselective macromer of trimesic acid was prepared using S(-) menthol with trimesoyl chloride on polyimide (PI) ultrafiltration membrane. The chemical composition of macromer as well as polyimide ultrafiltration membrane was determined by ATR-FTIR Spectroscopy. The optical resolution of chiral alcohols was performed in pressure driven process. The effect of monomer solutions concentration, effect of air-drying time of S(-) menthol solution, effect of reaction time, effect of operating pressure, effect of feed concentration of racemate on the performance of macromer was studied. The synthesised material exhibits separation of chiral alcohols (menthol ~23% and sobrelol ~21%).

  • Xanthorrhizol is a bisabolane type of natural sesquiterpene, the major component of essential oils of Curcuma xanthorrhiza. 2-(3-Methoxy-4-methylphenyl)propan-1-ol and 2-(3-hydroxy-4-methyl phenyl)propan-1-ol could be essential building block for enantioselective synthesis of xanthorrhizol. Enantioselective (c = 53%, E = $80{\pm}3$) for R-(+)-2-(3-hydroxy-4-methylphenyl) propan-1-ol and (c = 58%, E = $27{\pm}1$) for R-(+)-2-(3-methoxy-4-methylphenyl) propan-1-ol resolution processes were developed via lipase-catalyzed reaction. We found lipase Aspergillus oryzae (AOL) and Porcine pancreas (PPL) are selective to transesterification and hydrolysis in organic and aqueous phase. Modified demethylated substrate is appropriate for enantioselective hydrolysis reaction without any additives. Enantiopure chiral alcohol was crystallized from ethyl acetate/n-hexane co-solvent system. Gram scale resolved chiral intermediate will facilitate the synthesis of the unnatural S-(+)-xanthorrhizol, the corresponding isomer of the natural one.

  • Chiral epoxides are highly valuable intermediates, used for the synthesis of pharmaceutical drugs and agrochemicals. They have broad scope of market demand because of their applications. A major challenge in modern organic chemistry is to generate such compounds in high yields, with high stereo- and regio-selectivities. Epoxide hydrolases (EH) are promising biocatalysts for the preparation of chiral epoxides and vicinal diols. They exhibit high enantioselectivity for their substrates, and can be effectively used in the resolution of racemic epoxides through enantioselective hydrolysis. The selective hydrolysis of a racemic epoxide can produce both the corresponding diols and the unreacted epoxides with high enantiomeric excess (ee) value. The potential of microbial EH to produce chiral epoxides and vicinal diol has prompted researchers to explore their use in the synthesis of epoxides and diols with high ee values.

  • Chiral epoxides are useful chiral synthons in organic synthesis, and various biological methods have been investigated for their production. In this work, the enantioselective resolution of racemic styrene oxide was investigated using Aspergillus niger sp. for the production of optically pure (S)-styrene oxide. The enantioselectivity and initial hydrolysis rates of the racemic substrate were highly dependent of the pH, temperature, and the volume ratio of cosolvent. Experimental sets of pH, temperature, and the volume ratio of cosolvent were investigated using a central composite experimental design, and reaction conditions were optimized by response surface analysis. The optimal conditions of pH, temperature, and the volume ratio of cosolvent were determined to be 7.78, $28.32^{\circ}C$, and 2.4%(v/v), respectively, and optically pure (S)-styrene oxide (>99% ee) was obtained at 35% yield using this microbial enantioselective hydrolysis reaction.

  • An enantioselective ring opening of racemic terminal epoxides has been achieved by using heterobimetallic cobalt salen complexes with variety of nucleophiles. They were proven to be highly enantioselective and reactive for the synthesis of valuable chiral building blocks in enantio-riched forms up to 98% ee.

  • A newly isolated Aspergillus niger possessing the novel epoxide hydrolase(EHase) activity was investigated for the enantioselective hydrolysis of racemic aromatic epoxides. The gene encoding EHase was cloned by RT-PCR, and molecular characteristics of the EHase gene were compared with other microbial EHases. The cloned gene encodes 398 amino acids with a deduced molecular mass of 44.5 kDa and pI of 4.83, and sequence homology with other microbial EHase was low. Functional recombinant EHase could be obtained by heterologous expressions in E. coli. Enantioselectivity of recombinant EHase was tested for valuable aromatic epoxide intermediates. Reaction conditions of EHase-catalyzed asymmetric resolution were optimized for the production of chiral styrene oxide.

  • The chiral B-isopinocampheyl-9-borabicyclo[3.3.1]nonane-potassium hydride (IPC-9-BBN-KH) and potassium B-isopinocampheyl-9-boratabicyclo[3.3.1]nonane (K IPC-9-BBNH) systems were applied to the enantioselective reduction of representative racemic epoxides, namely 1,2-epoxybutane, 1,2-epoxyoctane, 3,3-dimethyl-1,2-epoxybutane and styrene oxide. In the case of IPC-9-BBN-KH system, the optical yields are in the range of 8.3-37.4$\%$ ee. However, the system of K IPC-9-BBNH provides significantly lower optical yields, showing 7-22.5$\%$ ee. These results strongly suggest that the enantioselective coordination of chiral organoborane to the epoxy oxygen of racemic epoxides plays an important role in this resolution.
