Real-Time Detection of DNA Hybridization Assay by Using Evanescent Field Microscopy

  • The determination of DNA hybridization reaction can apply the molecular biology research, clinic diagnostics, bioengineering, environment monitoring, food science and other application area. So, the improvement of DNA detection system is very important for the determination of this hybridization reaction. In this study, we report the characterization of the probe and target oligonucleotide hybridization reaction using the evanescent field microscopy. First, we have fabricated DNA chip microarray. The particles which were immobilized oligonucleotides were arranged by the random fluidic self-assembly on the pattern chips, using hydrophobic interaction. Second, we have detected DNA hybridization reaction using evanescent field microscopy. The 5'-biotinylated probe oligonucleotides were immobilized on the surface of DNA chip microarray and the hybridization reaction with the Rhodamine conjugated target oligonucleotide was excited fluorescence generated on the evanescent field microscopy. In the foundation of this result, we could be employed as the basis of a probe olidonucleotide, capable of detecting the target oligonucleotide and monitoring it in a large analyte concentration range and various mismatching condition.

Observation of Strong In-plane End Vibration of a Cylindrical Shell

  • In this paper, the strong in-plane vibration has been experimentally observed at the end of a finite cylindrical shell. The strong in-plane vibration was generated by the evanescent wave field, which was excited along about half the length of the shell. The evanescent waves were generated due to mode conversion of elastic waves at the ends of the cylindrical shells.

  • The propagation characteristic of an optical waveguide was investigated by measuring with a near-field scanning optical microscope (NSOM) the evanescent field formed at the neighbor of its core-cladding interface. For this purpose, the NSOM system was developed specially as a form of Photon scanning tunneling microscope. The evanescent field distributions of several channel waveguides were measured at the wavelength of 1550 ㎚, and the usefulness of the system was verified by comparing experimental results with simulation results. In particular, the interference phenomena of the guided modes during their propagation along a multimode channel waveguide could be observed directly from the measured evanescent field distribution.

  • We examine theoretically the properties of the force on Mie particles induced by evanescent fields at a system of multilayer films (including a metal film), at which the surface plasmon resonance is excited by a p-polarized plane electromagnetic wave. An expression of the surface plasmon-coupled evanescent fields produced in Kretschmann (or Sarid) geometry is expanded in terms of vector spherical wave functions, while multiple reflections between the Mie particle and the metal boundary are taken into account. The Cartesian components of the force on Mie particles by the evanescent fields are analytically formulated and numerically evaluated. The force components are increased by one or two orders of magnitude at metal boundaries over those at dielectric boundaries. As a result, we can confirm the possibility of stable manipulation or rotation of a finite-sized object by forces of surface plasmon-coupled evanescent fields.

Optically induced rotation of combined Mie particles within an evanescent field of a gaussian beam (가우스형 에바네슨트파의 광압에 의한 Mie 입자 결합체들의 회전)

  • We demonstrate a windmill type rotation of combined Mie particles caused by the evanescent field generated at the surface of a Gadolinium Gallium Garnet prism where a linearly polarized Nd:YAG laser beam (of wavelength 532 nm and power 100 mW) is totally reflected. The combined particles (e.g., doublets, triplets or quadruplets) consisting of polystyrene latex (or yeast) spheres with diameters of 3~5 m are reported to rotate at 0.1~1 rpm in water when the particles are slightly displaced from the center of the evanescent field.

  • We report the fiber-optic current sensors composed of a side-polished single-mode fiber with the thermo-optic Polymer layer and the metal wire as a heater. The index change of polymer layers caused by the resistant heat of the metal wires induces the optical attenuation through the evanescent field of the side-polished single-mode fiber. Two types of the sensors are proposed and their characteristics as a current sensor are investigated.

  • Polymer optical waveguides are fabricated with high-index materials deposited to strengthen exciations of evanescent field whose birefringence is utilized for optical sensing. Optical sensing properties are examined as a function of time, using different types of analyte solutions to extract noise-free signal induced by evanescent field birefringence. It is observed that sensing signal can be free of initial noise that may obscure real signal recognition, when glycerol is used for sensing characterization, due to slow accumulation process following adsorption of analyte material onto the sensing surface of the waveguide.

  • We have carried out a basic experiment in order to develope a super high-resolution optical microscope which transcend the limitation of diffraction and the wavelength of lightwave. The image of this scope is composed by measuring the evanescent wave which is localized on the surface of the testing materials. A detecting probe was fabricated with a single mode optical fiber to be sharpened by the chemical etching, and drived by PZT. The standing wave of $0.33\mu\textrm{m}$ wavelength evanescent wave which was generated from the $0.78\mu\textrm{m}$-wavelength semiconductor laser was detected by the $0.5\mu\textrm{m}$-thickness optical fiber probe.

Evanescent-Mode Analysis of Short-Channel Effects in MOSFETs (Evanescent-Mode를 이용한 MOSFET의 단채널 효과 분석)

  • Short channel effects (SCE) of bulk MOSFET with super-steep retrograded channels (SSR), fully-depleted SOI, and double-gate MOSFET have been analyzed using a evanescent-mode analysis. Analytical equations of the characteristics scaling-length (λ) for three structures have been derived and the accuracy of the calculated λ was verified by comparing to the device simulation result. It is found that the minimum channel length should be larger than 5λ and the depletion thickness of the SSR should be around 30 nm in order to be applicable to 70 nm CMOS technology. High-$textsc{k}$ dielectric shows a limitation in scaling due to the drain-field penetration through the dielectric unless the equivalent SiO2 thickness is very thin.
