돼지 삼출성(渗出性) 표피염(表皮炎)에 관한 연구(硏究) : I. 발증돈(發症豚) 및 건강돈(健康豚)으로부터 Staphylococcus hyicus subsp. hyicus의 분리(分離) 및 그 성장(性狀) (Studies on Exudative Epidermitis in Pigs : I. Isolation and Some Properties of Staphylococcus hyicus subsp. hyicus from Diseased and Healthy Pigs)

  • Isolation of Staphylococcus hyicus subsp. hyicus from piglets suffering from exudative epidermitis and healthy pigs was performed, and some properties of the isolates were examined. Twentry-six litters of exudative epidermitis observed in the field were sucking piglets with ages ranging from 1 to 5 weeks and most (73.1%) of these cases occurred in piglets between 2 and 3 weeks of age. Staphylococcus hyicus subsp. hyicus was isolated front 192(38.9%) of 494 healthy pigs. The rate of isolation of these organism from healthy pigs was found to vary greatly among farms at an isolation rate of 33.3% to 45.5% and this organism was isolated more frequently front sucking piglets below the age of 8 weeks than adult pigs. All of the isolates originated from the diseased and healthy pigs were positive in heat-stable DNase, Tween 80 hydrolysis, gelatinase, protease, but negative in clumping factor and hemelysin on sheep blood agar. There was no difference related to origin in the production of extracellular active substances. Typical lesions of the classic exudative epidermitis were produced by inoculation of Staphylococcus hyicus subsp. hyicus isolated from the diseased and healthy pigs. The pathogenic potentials of isolates from healthy pigs were no different from that of the isolates obtained from diseased pigs.

  • To elucidate morphologic lesion of porcine exudative epidermitis which is occurred sporadically in Korea, Staphylococcus hyicus subsp. hyicus isolated from the naturally affected pigs was inoculated to suckling pigs. The infected piglets were observed grossly and histopathologically. Although affected piglets were taking acute, subacute, or chronic course, some piglets suffered from chronic disease showed poor prognosis and marked growth depression. Affected peglets had erythematous skin on the face, ear, and abdomen and these localized lesions appear as brownish spots of exudative epidermitis and fromed crust in the early stage. But, after this stage, the skin were covered by viscous greasy exudate and formed blackish brown crust and appeared fissures and hypertrophy. Grossly, there has been hemorrhage with the removal of crust-like materials of epidermis and edematous subcutis. The superficial lymph nodes were edematous and swollen or congested and hemorrhagic. Some piglets had swollen ureters, cysts in the renal cortex, or polyarthritis. A few cases had mild edematous swelling of kidney, intestinal catarrh and congestion of brain. Microscopically, skin lesions had detachment of keralinized layer and parakeratosis of epidermis, hydropic degeneration of epidermal cell, and retrogressive degeneration of hair root sheath. Dermis had edema, and infiltration of neutrophils and mononuclear cells. As the disease was proceeded, there was marked perivasculitis with lots of mononuclear inflammatory cells. More chronic lesions formed granuloma-like bodies(nodules) due to more mononuclear, perivascular inflammatory cell infiltration and proliferation of fibroblast. Lots of plasma cells and eosinophils were also present in dermis. Epidermis was hyperplastic by proliferation of basal cells stratum germinativum and epidermal pegs often extended into the dermis. In secondary infection, lots of neutrophils could be seen in epidermis and derms. Kidney had neutrophilic infiltration, necrotic and cystic glomeruli, and dilation of renal tubules and ureters. Purulent arthritis was sometimes observed in joints. Three days old mice administrated Staphylococcus hyicus subsp hyicus subcutaneously before had focal congestion and hemorrhage, necrosis, and subcutaneous edema of the skin. This observation was also seen in the study of mice administrated exfoliatin toxin of Staphylococcus which evoked human staphylococcal scalded skin syndrome.

  • A total of 204 strains of Staphylococcus hyicus subsp. hyicus isolated from piglets suffering from exudative epidermitis and healthy pigs were tested for antibiotic susceptibility. The growth of all isolates of Staph. hyicus subsp. hyicus tested was inhibited by cloxacillin(Cx) at a concentration of $0.8{\mu}g/ml$ and by cefazolin (Cf) at a concentration of $0.4{\mu}g/ml$, and by gentamicin and methicillin at a minimum inhibitory concentration of $3.2{\mu}g/ml$. Of total isolates obtained from the diseased and healthy pigs, 69.6% were resistant to penicillin G, 53.4% to tetracycline, 11.8% to kanamycin and 0.5% to chloramphenicol, with considerable variations among farms in the occurrence of resistant strains and resistance patterns to the drugs. By application of Cx or Cf which was active to the organism, piglets exposed to infection were successfully protected from the disease, but piglets suffering from already severe exudative epidermitis did not respond to treatment.

  • The serological typing of staphylococcus hyicus subsp. hyicus isolated from pigs with exudative epidermitis and healthy pigs was attempted by slide agglutination technique with absorbed sera prepared from antisera against swine strains of the organism. Of 204 Staph. hyicus subsp. hyicus isolated, 202 (99.0%) were classified into 6 serotypes tentatively designated as A, B, C, D, E and F. The organisms were found to possess common antigens and a strain specific antigen. In 26 epizootics of exudative epidermitis, type A was found in 4(15.4%), type B in 12(46.2%), type C in 9(34.6%) and type E in 1(3.8%). All of the piglets of each affected litter were infected by serologically identical strain. In 178 strains isolated from the skin of healthy pigs, type A was found in 14.1%, type B in 23.0%, type C in 30.3%, type D in 1.7%, type E in 7.9% and type F in 21.9%, and 2 strains were untypable. There was a difference in the occurrence of serotypes of Staph. hyicus subsp. hyicus among the farms examined.

  • To elucidate pathologic change of skin in porcine exudative epidermitis, immunohistochemical and electron microscopical observations were carried out in the skin of the suckling pigs inoculated with Staphylococcus hyicus subsp hyicus which were isolated from natural case. In immunohistochemistry, ATPase-positive dendritic cells were more populated in epidermo-dermal junctional areas and perivascular area in dermis than in epidermal area as the disease was proceeded. These dendritic cells were identified as Langerhans cell by immunoperoxidase staining and these cells were populated granulomatous bodies. Electron microscopical study showed various retrogressive degeneration and vacuolation of epidermal cell organelles with retention of amorphorous exudates in intercellular space, and cellular seperation. Langerhans cells present in intercellular space of epidermis were populated in epidermo-dermal junctional areas, in dermis, and around granulomatous bodies. Langerhans cells contained decreased Birbeck granules in number but increased lysosome and ribosome. These cells were in contact with lymphocytes. This study was discussed relation between the various immunocytes and the formation of granulomatous bodies, and this inflammation was considered as delayed type hypersensitivity.

돼지에 있어서 Staphylococcus hyicus subsp hyicus의 분포와 분리균의 항균제 감수성 (Prevalence of Staphylococcus hyicus subsp hyicus in pigs with reference to antibiotic susceptibility of isolates)

  • In order to know the prevalence of S. hyicus subsp hyicus in pigs, attempts were made to isolate the organism from 549 healthy adult pigs, 277 healthy suckling piglets and 17 piglets with exudative epidermitis from April 1988 to January 1989 in Chinju, Korea. Also determined was antibiotic susceptibility of the isolates. Isolation rates of S. hyicus subsp. hyicus from adult pigs and suekling piglets were 27.0% and 53.1%, respectively. The organism was isolated predominantly from abdomen (10.9%) of adult pigs and external ear (31.6%) of suckling piglets. Isolation rates of the organism from piglets of different age group were in order of prevalence of 1(82.1%), 3(74.0%), 2(54.7%), 4(52.9%), 5(15.2%) and 6(5.7%) post-natal weeks. All of 489 isolates were sensitive to gentamicin, kanamycin and cephalexin but 23.7~79.6% of the strains were resistant to erythromycin, penicillin G and tetracycline. The 49.2% of these resistant strains exhibited multiple drug resistance. The drug resistant patterns most frequently encountered were PG TC EM(10.7%) in triple pattern, PG TC(23.0%) in double pattern and TC(44.4%) in single pattern.

Real-time PCR을 이용한 원유시료 유래 황색포도상구균의 신속 검출 (SYBR Green I-based Real-time PCR Assay and Melting Curve Analysis for Rapid Detection of Staphylococcus aureus from Raw Milks Samples)

  • 본 연구는 Lightcycler (Roche)를 이용한 Real-Time PCR(LC-PCR)기법을 통하여 원유시료에서 신속, 정확하게 황색포도상구균을 검출하는 기법을 개발하고자 하였다. coagulase 전구체를 coding하는 113 bp의 coa 유전자의 증폭, melting curve 분석 및 DNA염기서열을 분석하여 황색포도상구균 특유의 유전자 검출하는 기법을 개발하였다. 또한 분리된 균주중 메치실린에 내성을 나타내는 균주를 검출하고자 penicillin-binding protein, PBP2a (mecA)를 coding 하는 209 bp의 mecA 유전자의 증폭, melting curve 분석 및 DNA염기서열을 분석하여 메치실린내성 황색 포도상구균을 real-time PCR 기법으로 검출하는 기술을 개발하였다. 본 실험에 따르면 647개의 원유시료중 6개의 시료에서 황색포도상구균이 검출되었으며 이중 2개의 시료에서 분리된 황생포도상구균이 메치실린내성 황색포도상구균임을 확인하였다. 또한 DNA 검출한계는 10 fg으로 기존 PCR에 비해 매우 감도가 우수한 것을 확인하였다. 또한 3개의 원유시료에서 돼지나 소의 삼출성 피부염의 원인균인 Staphylococcus chromogenes가 분리되었다.

API STAPH system을 이용한 돼지, 닭 및 소유래 Staphylococcus hyicus subsp. hyicus의 동정 (Identification of Staphylococcus hyicus subsp. hyicus of swine, poultry and bovine origin with the API STAPH system)

  • The API STAPH system was compared with conventional methods for identification of 214 strains of Staphylococcus hyicus subsp. hyicus isolated from cases of exudative epidermitis in piglets, skin of healthy pigs, skin of healthy chickens and bovine intramammary infections, and biochemical characteristics among the swine, avian and bovine strains were also compared. All of the swine and bovine strains produced acid within 24 hours from fructose, lactose and trehalose by conventional methods, but some of the avian strains showed a delayed positive reaction in these carbohydrates. These delayed positive strains in conventional methods gave usually negative results for them in the API STAPH system. With the API STAPH system, eighteen different profile numbers were encountered in 214 strains of swine, avian and bovine origin. The swine and bovine strains, respectively, were distributed among 4 profiles, while the avian strains were distributed among 17 profiles. The profile number observed most frequently in the strains of each animal species was uniformly 6 516 153. By conventional methods, approximately 96% of the swine strains were positive for ${\beta}$-glucuronidase, but not in any strains from chickens and cattle. For hyaluronidase production determined by degradation of sodium hyaluronidate in a solid culture medium, all the swine and bovine strains were positive, but only 37.5% of the avian strains were positive for it. From these findings, there were differences in the production of extracellular active substances between swine strains of Staphylococcus hyicus subsp. hyicus and those isolated from chickens and cattle.
