Transferable R plasmid of Edwardsiella tarda isolated from diseased flounders, Paralichithys olivaceus (넙치에서 분리된 Edwardsiella tarda의 약제내성 전달성 R plasmid)

  • MIC test of 16 chemotherapeutic agents was performed on 24 isolates of Edwardsiella tarda collected from flounders. They revealed resistance against combinations of ampicillin, amoxicillin, erythromycin, flumequine, doxycycline(DOXY), nalidixic acid, novobiocin, oxolinic acid, oxytetracycline(OTC), thiamphenicol(TP) and sulfonamide. Two strains carried transferable R plasmid encoding Otc Kanamycin Tp and Otc chloramphenicol Doxy Tetracycline Tp, respectively. The R plasmids were not similar each other on the basis of their digestion pattern of restriction endonuclease, suggesting distribution of different transferable R plasmid among E. tarda from flounders.

Isolation of vibrio species from cultured flounders (paralichthys olivaceus) with uicers and ascites in the southern coast of Korea during the winter season (동절기 한국 남해안의 궤양증 및 복수증 양식 넙치로부터 vibrio종의 분리)

  • this study was performed for the purpose of surveying the distribution and characteristics of the Vibrio species in the diseased farm flounders (Paralichthys olivaceus) during the winter season in Korea. Samples were collected along the southern coast in the Sinam, Keoje Island, and Teosu areas from November '90 to Feburary '91. Nine species of Vibrio were identified as V. tubiashii (85 strains), V. damsela (78 strains), V. anguillarum (21 strains), V. compbelli (9 strains), V. fluvialis (9 strain), V. costicola (1 strain), V. alginolyticus (1 strain), V. gazogenes (1 strain), V. marinus (1 strain), and unidentified 3 groups of bacteria (154 strains) which may have related with V. tubiashii were isolated. Eleven kinds of discs (BBL Co.) were used for antibiotic susceptibility test and 116 strains were selected for the experiment. Most bacteria were powerfully inhibited their growth by 4 antibiotics such as cephalothin, chloramphenicol, colistin, and tetracycline, but they were poorly inhibited by 4 antibiotics such as carbenicillin, kanamycin, penicillin G, and streptomycin.

Optimum Rearing Temperature for Fingerling Production Between Stone Flounder Platichthys bicoloratus ♀ and Starry Flounder P. stellatus ♂ (돌가자미(Platichthys bicoloratus) 암컷과 강도다리(P. stellatus) 수컷 간 교잡종 종자 생산을 위한 최적 사육 수온)

  • Interspecific hybridization is a beneficial strategy in aquaculture. In this study, diploid hybrids were produced through interspecies hybridization of flounder Platichthys bicoloratus ♀ × starry flounder P. stellatus ♂, which became commercial within a year. These hybrids exhibited an advantage in growth ability, with female hybrids growing twice as large as males at 11 months. Furthermore, hybrid flounders exhibited faster growth rates than starry flounders and reciprocal hybrids. The average growth rate of hybrids was 1.37 and 1.18 times greater than that of starry flounder and reciprocal hybrids at 5.5 months, respectively; Additionally, feed efficiency was 1.21 and 1.10 times higher than that of the experimental groups. The experiments were conducted at five temperatures to determine the optimal fry production method. Six weeks of culture resulted in a significant temperature effect, with maximum growth at 27℃. However, over 90% of the males occurred in the groups at temperatures above 24℃. Therefore, the optimum temperatures for survival, growth, condition factor and frequency of the best-sized fingerling at 13 weeks of age were estimated at 21℃. Thus, hybrid flounders are of great potential for use in the flounder aquaculture industry.

  • This study was designed to investigate the biochemical pollutant marker for diagnosis of marine pollutions by the changes in lipid components of the flounder (Pleuronichthys cornutus) in the Yellow Sea of Korea. Hemoglobin levels in serum of wild flounders in the yellow Sea were significantly lower $(15\~30\%)$ than those of wild flounder in Pohang (control) of the East Sea. Triglyceride (TG) contents in serum of wild flounders in the Yellow Sea were higher $(15\~50\%)$ than those of wild flounder in Pohang. Total and low density lipoprotein (LDL)-cholesterol contents in serum of flounders in the Yellow Sea were significantly higher $(20\~25\%\;and\;15\~35\%,\;respectively)$ than those of wild flounder in Pohang. Atherogenic index (Al) in serum of wild flounders in the Yellow Sea were significantly higher $(10\~60\%)$ than these of wild flounder in Pohang but T-Chol/PL ratios showed almost no change in serum of flounders in the Yellow Sea we examined. These results suggest that near-coastal water as well as neritic water of the Yellow Sea might be affected by pollutant input.

Metabolism of Dietary Carotenoids and Effects to Improve the Body Color of Cultured Flounder and Red sea bream (양식 넙치, 참돔의 사료 Carotenoids 대사와 체색개선에 미치는 영향)

  • To investigate the effects on pigmentation and carotenoid metabolism of red sea breams Pagrus major and flounders Paralichithys olivaceus by the supplemented carotenoids, fishes wire fed the diet each containing ${\beta}$-carotene, lutein ester, astaxanthin, astaxanthin monoester, astaxanthin diester and ${\beta}$-apo-8'-carotenal for 8 weeks. Carotenoids in the integuments were analyzed. In cultured red sea breams with supplemented carotenoids, carotenoid deposition and pigmentation were higher in order of astaxanthin diester group, ${\beta}$-apo-8'-carotenal group and astaxanthin monoester group. The main carotenoids of red sea breams were astaxanthin diester, tunaxanthin and ${\beta}$-carotene. Difference in the content of astaxanthin diester and ${\beta}$-carotene was observed from natural and cultured red sea breams. In cultured flounders with supplemented carotenoids, carotenoid deposition and pigmentation were higher in order of ${\beta}$-carotene group and lutein ester group. The main carotenoids of flounders were zeaxanthin and lutein. Difference in lutein and ${\beta}$-carotene contents was observed from the natural and cultured flounders. Based on the contents and composition of carotenoids in each group after feeding experimental diet, carotenoid metabolism in red sea breams were presumed the reductive metabolic pathway, astaxanthin to tunaxanthin, and likewise, in flounders, lutein to tunaxanthin.

  • This study estimated the genetic parameters and breeding values for the growth-related traits of the 1st generation produced in 2005. The heritability of body weight, total length, body height, body shape and condition factor of 180 days old olive flounders Paralichthys olivaceus, the 1st generation of selection, was estimated as 0.564, 0.590, 0.588, 0.306 and 0.332, respectively. And reproductivity of genetic evaluation for crossing superior flounders and inferior ones was tested using the subsequent generation produced in 2006 based on the breeding values of 1st generation. In the least-squares means of body weight and total length for each group of crossing, the values of crossing group between superior flounders ($S{\times}S$) showed $145.6{\pm}1.8\;g$ and $22.4{\pm}0.1\;cm$, respectively. The values of crossing group between superior and inferior flounders ($S{\times}I$) showed $133.2{\pm}2.5\;g$ and $22.1{\pm}0.1\;cm$, respectively. The values of crossing group between inferior flounders ($I{\times}I$) showed $114.0{\pm}2.1\;g$ and $21.08{\pm}0.12\;cm$, respectively. In the results, flounders are determined as appropriate selective breeding fish with the high heritability of flounders in early ages at 180 days old, and the reproductivity of genetic evaluation was also high.

Development of a Waterless Container Utilizing Thermoelectric Modules for Live Fish Transportation (열전소자를 이용한 활어 수송용 무수 컨테이너의 개발)

  • The purpose of this study is to develop a low temperature waterless container for live fish transportation which is economical and efficient The principle of the waterless transportation is that a live fish becomes asphyxial at about $5^{\circ}C$can survive without water for a long time. A low temperature waterless container is developed for this purpose, which utilizes thermoelectric modules for rather smaller and lighter cooling system with precise temperature control devise compared to the existing mechanical system. At first, we succeeded in making flounders alive in the waterless container for 24 hours. Also when flounders were transported in a round trip from Inchon to Pusan in the waterless container, carried in a car, they survived in the waterless container for over 21 hours.

Prevalence of Antibiotic Resistant Foodborne Bacteria Isolated in Korea

  • This study was performed to determine the antibiotic susceptibility profiles of Salmonella spp., coliforms, Listeria monocytogenes, Staphylococcus aureus and Vibrio spp. isolated from broiler carcasses, aquacultured flounders, hamburgers, and lettuce, which are foods consumed in large quantities in Korea. Salmonella spp. and L. monocytogenes were isolated only from broiler carcasses and Salmonella spp. had a high multidrug resistance rate of 61.1%. Meanwhile, coliforms and S. aureus were isolated from all four foods tested in this experiment. The multidrug resistance rate of coliforms from broiler carcasses was 50%, and that of Vibrio spp. from flounders was 71.4%. The resistance to tetracycline, streptomycin, ampicillin or carbenicillin was common regardless of the kind of food or isolate.

A Study on the Flounder Consumer Market in the US (미국의 넙치 소비 시장에 관한 연구)

  • Flounder was selected as one of the 10 strategic export aquaculture products for seafood export expansion in 2013. The flounder aquaculture industry has promoted export market diversification and product diversification from live to processed goods as a it's main strategy. The purpose of this study is to find an improvement plan for export expansion to the United States, as it emerged as a new target export market for the flounder. A summary of the key findings is as follows. First, the western region of the United States prefers to consume live and fresh flounder, whereas the eastern region prefers to consume fresh flounder. Second, because of it's high quality, Korean flounder is favored in the western region of the United States despite it's high price, whereas in the eastern region of the United States, where production volume is high, Korean flounder has to compete with US flounders because of it's high price. Third, according to the survey results, US consumers tend to enjoy seafood, as well as flounder cuisines. Fourth, the main consumption place of flounders by US consumers are restaurants, and they prefer to consume them in the form of sashimi and sushi. Fifth, 70% of US consumers expressed willingness to consume flounder when eating out. which shows great market potential. However, the high price of Korean flounder and limited size of the live fish market act as major obstacles to expanding export volume. To expand exporting Korean flounder, continuos efforts such as price reduction, exporting highly fresh fish, the co-development of processed food with the US are needed.

Effects of Temperature on the Pharmacokinetics of Ciprofloxacin in the Cultured Black Rockfish (Sebastes schlegeli) and Olive Flounders (Paralichthys olivaceus)

  • Temperature-dependent pharmacokinetics of ciprofloxacin (CIP) was studied in the cultured olive flounders, Paralichthys olivaceus, and black rockfish, Sebastes schlegeli using high performance liquid chromatography (HPLC) originally developed for quinolone determination from livestock. Pharmacokinetics of CIP was apparently affected by ambient water temperature. In a two-compartment model for flounders after oral dosage of 20 mg/kg, $K_{01},\;at\;13^{\circ}C$ and $23^{\circ}C$ were 4.18 and 1.20/hr, respectively. The $K_{10},\;T_{max}\;and\;C_{max}\;at\;13^{\circ}C$ were 5.574/hr, l4.37${\mu}g/mL\;and\;3.15{\mu}g/mL,$ respectively. The corresponding values at $23^{\circ}C$ were l2.84/hr, 15.39${\mu}g/mL\;and\;6.38{\mu}g/mL$, respectively. The AUC, $T_{1/2} (\alpha)\;and\;T_{1/2}\;(\beta)$ were 278.23 ${\mu}g \cdot hr/mL$, 0.24hr and 47.02hr at $13^{\circ}C$ and 3l7.8l${\mu}g \cdot hr/mL$, 0.30 hrs and 60.78hrs at $23^{\circ}C$ for the flounder, respectively. Similar CIP pharmacokinetics were revealed in the black rockfish after oral dosage of 20 mg/kg under the two water temperature regimes. These pharmacokinetical results have some implication in the optimal usage of recently introduced antibacterials in the farmed fish, which were primarily adapted for poultry and mammalian species.
