Use of 16S-23S rRNA Intergenic Spacer Region for identification in the fish pathogenic Streptococcus iniae (16S-23S rRNA Intergenic Spacer Region을 이용한 어류 병원성Streptococcus iniae의 분자생물학적 동정)

  • This study was performed for the identification of Streptococcus sp. from cultured flounders (Paralichthys olivaceus) showing streptococcosis in the Jeju island. We isolated 10 strains of Streptococcus iniae from the cultured olive flounders with streptococcosis. Isolated strains were identified in S. iniae since they have formed the expected band through performing PCR assay using specific primers, Sin-1 (5'-CTAGAGTACACATGTACT(AGCT)AAG-3') and Sin-2 (5'-GGATTTTCCACTCCCATTAC-3'). In addition to 16S-23S rRNA intergenic spacers (ISR), operon structure of isolated strains showed that all strains had three 16S-23S rRNA ISR band patterns. The 16S-23S rRNA ISR sequence of isolated strains showed 96% sequence identity with S. iniae (GenBank accession number AF 048773). This paper is the first report that S. iniae is associated with streptococcosis of Olive flounder in Korea.

  • Olive flounders which were cultured in commercial fish farm showed abnormal swimming behavior in November 2020. Then, gradual mortality was observed in the fish farm. During the diagnosis, bacterial strain KNCFKW-PN1 was isolated from the kidney of the dead fish. Based on the sequence of gyrase B subunit gene, KNCFKW-PN1 was proved to be Pseudoalteromonas nigrifaciens, showing 99.59% nucleotide identity with that of P. nigrifaciens LMG 2227T. According to the result of antibiotic susceptibility test, P. nigrifaciens KNCFKW-PN1 showed intermediate resistance to ciprofloxacin, and was resistant to ampicillin, cefepime, cefotaxime, ceftazidime, and amikacin. This is the first report of the isolation of multiple-antibiotic-resistant P. nigrifaciens from olive flounder.

  • This study was designed to investigate the biochemical pollutant markers for diagnosis of marine pollutions by changes in cholinesterase activity of the flounder (Pleuronichthys cornutus) in the Yellow Sea of Korea. Acetylchotinesterase (AChE) activities in brain and muscle of wild flounders in the Yellow Sea were significantly lower $(20\~30\%\;and\;10\~40\%,\;respectively)$ than those of wild flounder in Pohang (control) of the last Sea. Butyrylcholinesterase (BChE) activities in brain and muscle of wild flounders in the Yellow Sea were significantly lower $(10\~30\%\;and\;35\~45\%,\;respectively)$ than those of wild flounder in Pohang of the East Sea. lactate dehydrogenase (LDH) activities in serum of wild flounders in the yellow Sea were significantly $(about\;30\%)$ lower than those of wild flounder in Pohang of the East Sea. These results suggest that AChE and BChE activities in brain and muscle of wild flounders of the Yellow Sea may be used as the most effective mean in a biochemical marker for diagnosis of pollutant effects by organophosphorus pesticides.

  • For the recent years, considerable efforts have been made to rationalize the techniques of research and monitoring of biological effects of marine pollutants such as heavy metals, polychlorinate biphenyls (PCBs), organochlorine pesticides, polycyclic aromatic hydrocarbons (PAHs), organophosphorus pesticides. This study was designed as a part of efforts to investigate the biochemical pollutant markers for diagnosis of marine pollutions by the changes in lipid components of the flounder (Paralichthys olivaceus) in Yellow Sea of Korea. Homoglobin levels in serum of cultured and wild flounders in Yellow Sea were significantly lower (5 - 15% and 20 -25 % , respectively) than those of wild flounder in Pohang. Triglyceride (TG) contents in serum of cultured flounders in Yellow Sea were 10 - 40% higher than those of wild flounder in Pohang. Total and low density lipoprotein (LDL)-cholesterol contents in serum of flounders in Yellow Sea were significantly 10-15% and 30-35% (cultured), and 10-20% (wild), respectively, higher than those of wild flounder in Pohang. Atherogenic index (AI) and T-Chol/PL rations in serum of cultured and wild flounders in Yellow sea were significantly 20-45% and 10-!5%, 6-35% and 15-35%, respectively, higher than those of wild flounder in pohang. These results suggest that near-coastal waters as well as neritic waters of the Yellow Sea might be affected by pollutant input.

A Large and Changing U.S. Market for Gadoids and Other Groundfish

  • The United States is the world's leading importer of frozen processed groundfish products, with over two thirds of total world imports. Over 90% of the U.S. groundfish demand is met by inports, while about 2 million mt of groundfish are taken from U, S. waters by joint-venture and foreign fleets. The objective of this paper is to provide descriptive information concerning the U.S. groundfish market and the potential for groundfish resources off Alaska to become a major source of supply to this market. The size of the U.S. market, U.S. imports, trade policies, and catch from U.S, waters are discussed, and a comparison is made between the potential domestic catch of groundfish off Alaska and current U.S. groundfish consumption. The total optimum yield of 2.3 million mt for flounders, cod, and pollock is about four times the round weight equivalent of U.S. imports of these species in 1984.

Identification of Potential Species-Specific Marker in Several Fish Species by RAPD Using Universal Rice Primers (Universal Rice Primer (URP)-RAPD 방법에 의한 어류 종 특이 marker의 동정)

  • Morphologically similar fish species were subjected to the random amplified polymorphic DNA (RAPD) analysis using universal rice primer (URP). The fish species tested were sea basses (Lateolabrax japonicus and L. maculatus), eels (Anguilla japonica, A. bicolor bicolor, A. rostrata, and A. anguilla), and flounders (Limanda yokohamae and L. herzensteinin). Highly reproducible RAPD patterns were observed with several potential species-specific markers. The results indicate that RAPD technique using URP is useful for distinguishing fish psecies in a rapid manner.

Hepatic Response in Cytochrome P45O and De-alkylase Activity of Olive Flounder (Paralichthys olivaceus) Exposed to Water-borne Phenanthrene

  • Olive flounder (Paralichthys olivaceus) was exposed to water-borne phenanthrene for 4 weeks. After the exposure for 2-weeks, hepatic cytochrome P450 contents were significantly elevated. Induction of hepatic ethoxy resorufin-O-deethylase (PROD) activity was significantly increased in flounders treated with 1.0 and 2.0 $\mu$M phenanthrene, compared to untreated group and 0.5 $\mu$M treated group. However, there were no significant changes in pentoxyresorufin-O-deethylase (PROD) activity in hepatic microsome of all the phenanthrene-treated groups, compared to the untreated group. Phenanthrene has the potential to induce cytochrome P450 and EROD enzyme of the olive flounder.

  • To investigate the relationship between metabolic changes in $^1H$-nuclear magnetic resonance (NMR) spectra and fish vaccination, serum was collected from olive flounders treated with a formalin-killed Edwardsiella tarda vaccine and used for $^1H$-NMR metabolite profiling. Principal component analysis and partial least squares were applied to the $^1H$-NMR profile to reduce its complexity and establish class-related clusters. Relative lipid regions were distinguished in vaccinated and non-vaccinated serum. Then, the lipids were extracted from the serum and analyzed. Triolein was identified.

  • Iridovirus is an icosahedral cytoplasmic double-stranded DNA virus with a genome size of 170-200kb. Outbreaks of fish iridovirus infection are characterized by their wide geographic distribution and broad host spectrum, especially in water temperatures of $25-27^{\circ}C$ Recently, the causative agent of high mortalities in flounder (Paralichthys olivaceus) was identified as fish iridovirus in Korea. Iridoviral infection repeatedly occurs in the same area for long periods, suggesting the possibility of viral infection in nursery. To examine this, the existence of iridovirus in juvenile flounders was detected by PCR using virus-specific primers. Antibodies induced against this virus were also examined using ELISA. As a result, juvenile fish in nursery were found to be previously infected with iridovirus, suggesting that proper prevention systems are required.

Effect of Phenanthrene on Gill Biotransformation in Olive Flounder (Paralichthys olivaceus)

  • The potential of phenanthrene to induce xenobiotic responses was investigated. Olive flounder (Paralichthys olivaceus) was exposed to different levels of phenanthrene $(0.5,\;1\;and\;{\mu}M)$ for 4 weeks, Gill CYP450 content and EROD (ethoxyresorufin O-deethylation) activity were found to be significant in the flounders treated with the higher concentration of phenanthrene $(>1.0\;{\mu}M)$, however, there were no significant changes in gill PROD (penthoxyresorufin O-deethylation) activity in all treated group compared to the controls. This study demonstrated that phenanthrene has potential to induce gill cytochrome P450 and EROD enzyme in olive flounder.
