Varietal Difference in Salinity Tolerance during Germination Stage of Rice

  • This study was conducted to find out a desirable screening condition for the salinity tolerance in germination of rice. Seeds of 33 rice varieties were tested in NaCl solutions with various concentration levels. The germination percentage had a decreasing tendency with increasing NaCl concentration and inhibition concentration of 50% germination was 320mM. Standard deviation of germination percentage was highest (28.6) under 300mM NaCl. There was a highly significant correlation between the 50% germination concentration and the germination percentage at 20th day after seeding in 300mM NaCl. Also in 300mM NaCl, the germination percentage at 20th day after seeding was significantly correlated with the germination percentage at the 6th day after seeding. The salinity tolerance on the basis of germination percentage at 6th day after seeding in 300mM NaCl, was strong in 'Hyangnambyeo', 'Ilmibyeo', 'Kancheogbyeo', and 'Namwonbyeo', while weak in 'Ansanbyeo', 'Odaebyeo', 'Nonganbyeo', 'Dasanbyeo', and 'Namcheonbyeo'.

Effect of Seed Collection Stage and Temperature on Germination of Deutzia paniculata Nakai, the Korea Native Species

  • In this study, we investigated the effect of seed collection stage and alternating temperature on the germination of Deutzia paniculata (Hydrangeaceae). The seeds were collected from naturally growing D. paniculata populations in Mt. Palgong, Gyeonsangbuk-do, Korea every two weeks over a three month period from August through October, 2014, totaling five collections. We examined seed characteristics such as seed size (length and width) and weight, and germination percentages for each collection stage under four different germination temperature regimes. We also examined germination after 30 and 60 days of cold moist stratification. Each collection phase had a different germination percentage and the lowest germination percentage was observed in the seeds from phase I (average GP of 15.7%). The germination percentage was similar at the later collection phases and in the higher temperatures suggesting that later collection times and higher temperatures are optimal for D. paniculata germination. We found that the optimum collection stage with high viability and germination percentage of for D. paniculata was during the middle of the September.

Changes of Protein and Lipid Composition During Germination of Perilla frutescens Seeds (들깨 종자의 발아에 따른 단백질 및 지방의 조성 변화)

  • To investigate changes in protein and total lipid contents, seed storage protein pattern, and fatty acid composition of germination perilla(Perilla frutescens) seeds. Also, the corresponding value components in cotyledons, hypocotyles and roots were measured according to germination stage. The results were summarized as follows ; During germination, pertein and total lipid contents of Yepsilldalggae and Kwangyang cultivar were decreased continuously. In particular, protein contents rapidly decreased to the 3 days after germination(DAG), and then total lipid contents rapidly decreased between 3 DAG and 10 DAG. In changes of protein and total lipid contents of cotyledons, protein contents of Yeupsildalggae was increased during the germination, but Kwangyang cultivar was decreased during the same periods. The total lipids contents of Yeupsildalggae and Kwangyang cultivar were decreased during the germination. According to SDS-PAGE analysis, there was no detectible polypeptide bands on the gel before seed germination suggesting that this may be due to the rapid degradation of the storage proteins in the mature seed by hydrolyttic enzymes during the stage. During germinatation , the polypeptide band with 27$\sim$28KD of Yeupsildalggae and Kwangyang cultivar were accumulated gradually. In changes of fatty acid composition of total lipid of Yeupsildalggae and Kwangyang cultivar , saturated fatty acids such as palmitic acid and stearic acid increased during the germination. On the other hand, unsaturated fatty acid such as linoleic acid and linolenic acid decreased during the same periods. However, oleic acid increased to the 5 DAG, and then was repidly decreased.

Studies on the Viviparous Germination of Indica $\times$ Japonica Type Varieties in Paddy Rice (수도다수계 품종의 수발아에 관한 연구)

  • This experiment was conducted to find out the factors governing the viviparous germination of In. $\times$ Ja. type rice varieties. Six varieties, i.e., Milyang 21, Raekyeong, Milyang 30, Yeongnamjosaeng, Nopoong and Mansuk, were used for this experiment. The viviparous germination easily occurred between 40 to 45 days after heading, and was easier in yellow-ripe stage or full-ripe stage than dough-ripe stage. The use of exceeding fertilization induced more viviparous germination than ordinary one. The early varieties such as Yeongnam-josaeng and Milyang 21, easily occurred the viviparous germination. The local conditions of viviparous germination were usually poorly drained soils and narrow alluvial valleys where were high humidity prevailed.

  • Toxic effert of water extract from leaves of Pinus rigida and some phenolic compounds on the seeds germination of Raphanus sativus var. hortensis for. acanthiformis Makino has been studied. There was little difference of germination percentage among the pH value of leaf extract (pH3~9). The germination percentage drastically with increased concentration level of leaf extract at about 60 percent. Seeds gemination of Raphanus sativus var. hortensis for. acanthiformis Makino inhibited severely by caffeic acid, ferulic acid and p-coumaric acid at M, but the germination percentage was higher than that of the control group in vanillic acid. In electrophoresis, there was no differences at earlier seedling stage of protein band between allelochemical treated and non-treated group, but in late stage, two protein band near 58kd and 27kd did not appeared in the toxic affected group. In case of caffeic acid treatment, two protein band near 58kd and 27kd did not found at late stage too.

Germination-Induced Changes in Flavoring Compound Profiles and Phytonutrient Contents in Scented Rice (향미벼의 발아 전 후 향기 성분 및 기능성 지질성분 함량의 변화)

  • Although rice has been cultivated as a major food crop for approximately 5,000 years, the interest of customers in 'scented rice' is a recent trend in the Korean market. As a part of developing a germinated scented rice variety, the newly bred scented rice variety 'Cheonjihyang-1 se' was germinated for 24 h, and changes in profiles of flavor-related volatiles, lipophilic phytonutrients, and fatty acids were investigated. The profiling of volatile compounds by using a headspace-gas chromatography-mass spectrometry (HS-GC-MS) revealed a total of 56 odor-active flavoring compounds; 52 at the pre-germination stage, 51 at the post-germination stage, and 47 common at both stages. The major flavoring compounds were nonanol and benzene, which constituted 11.5% and 6.6%, respectively, of the total peak area in pre-germinated rice, and 19.4% and 6.5%, respectively, in post-germinated rice. Germination induced an increase in 13 flavoring compounds, including 3,3,5-trimethylheptane and 1-pentadecene, which increased by 763 and 513%, respectively by germination. However, we observed a germination-induced decrease in most of the other flavoring compounds. Especially, the most important scented rice-specific popcorn-flavoring compound, 2-acetyl-1-pyrroline, showed 89% decrease due to germination. Furthermore, the germination of scented rice induced a decrease in the content of various phytonutrients. For example, the total contents of phytosterols, squalene, and tocols decreased from 207.97, 31.74, and $25.32{\mu}g\;g^{-1}$ at pre-germination stage down to 136.66, 25.12, and $17.76{\mu}g\;g^{-1}$, respectively at post-germination stage. The fatty acid compositions were also affected by germination. The composition of three major fatty acids, linoleic, oleic, and palmitic acids, increased from 36.6, 34.2, and 24.4%, respectively, at the pre-germination stage to 37.9, 36.9, and 20.7%, respectively, at the post-germination stage. All these results suggested significant changes in the flavor-related compounds and phytonutrients of the scented rice variety 'Cheonjihyang-1 se' during the process of germination, and subsequently the need for developing a more precise process of germination to enhance the flavor and nutritional quality of the germinated scented rice products.

Allelopathic Effects of Crimson Clover, Hariy Vetch and Rye on Germination and Radicle Elongation of Several Crops (크림손클로버, 헤어리베치, 호밀 추출물이 몇 가지 작물의 종자 발아와 유근 생장에 미치는 allelopathy 효과)

  • Crimson clover (Trifolium incarnatum), hairy vetch (Vicia villosa) and rye (Secale cereale) are common leguminous cover crops. Because they contain water-soluble allelopathic substances that show a variable level in tissue depending on growth stage of the plants, the weed inhibition effects are dependent on the growth stage of cover crops. This study investigated the allelopathic effects of crimson clover, hairy vetch and rye on soybean, radish, oilseed rape and lettuce seeds germination and seedling growth. We used extract that were prepared from the shoots of crimson clover, hairy vetch and rye at different growth stages, vegetative growth stage, flowering stage, and fruiting stage. Applications of aqueous extracts from three growth stage of cover crops strongly affected to oilseed rape and lettuce seeds germination but the treatment resulted in a slight inhibition of the germination in soybean and radish. Radical length was more sensitive to aqueous extracts than seed germination rate. Especially, three cover crops extracts at vegetative growth stage highly inhibited seedling root growth of oilseed rape and lettuce by over 80% and 90% respectively. Furthermore, the $GR_{50}$ values were lowest in the treatment of extracts from vegetative growth stage and the level of phenolics was decreased by the order of vegetative growth stage, flowering stage and fruiting stage.

The Effects of Optimal Germination of Somatic Embryos Induced from Mature Cotyledon Explants of Panax ginseng C. A. Meyer by Gibberellic Acid (인삼 체세포 배 발아를 위한 $GA_3$의 최적 조건)

  • Somatic embryos on growth regulator-free medium can be produced directly from cotyledon explants of Panax ginseng C. A. Meyer. When the embryo developmental stage was torpedo and cotyledon, the germination rate of embryos was quite high on MS medium supplemented with gibberellic acid ($GA_3$). However, the percentage of plantlet formation at the cotyledon stage was higher than that at the torpedo stage. This result demonstrates that the embryo at the cotyledon stage was the most appropriate for increasing germination by $GA_3$. Embryos cultured on medium including four levels of $GA_3$ concentrations (3, 5, 10, or 20 mg/$\ell$) showed all quite high germination rates (87-91%). When the well-developed embryos were continuously cultured on media including high concentrations of $GA_3$ from 10 to 20 mg/$\ell$, the percentage of formation of normal plantlets was lower than that seen under low concentrations from 3 to 5 mg/$\ell$. This treatment of high concentrations resulted in shoots with abnormal shape. The optimal $GA_3$ treatment provides a basis for the efficient method obtaining healthy plantlets derived from ginseng somatic embryos.

Changes in Physico-chemical Properties and Mineral Contents during Buckwheat Germination (메밀 발아 중 물리화학적 특성과 무기질 함량의 변화)

  • To provide the effective application scheme and basic information of buckwheat (Fagopyrum esculentum Moench), buckwheat was germinated at 10$^{\circ}C$ for 7 days and 100 kernels weight, germination rate, root length, chemical composition and mineral contents were examined at 24 hour Intervals. During the germination period, the 100 kernels weight increased approximately 0.3g per day. The germination rate increased sharply after 2 days and the root length increased greatly after 4 days. The crude protein contents increased with germination time, whereas the carbohydrate contents decreased. The crude ash and fat contents did not differ significantly during the germination period. The Ca contents Increased for the 4th day of gemination, but gradually decreased afterwords. The Na contents increased in the initial stage of germination, but then gradually decreased. However, there were no significant change In the Mg, K. Fe. Mn and Zn contents.

Effects of Sprouting Treatment with Gibberellin to Germination and the Growth of Spring Barley (지벨레린 최아처리가 춘파대맥의 발아생육에 미치는 영향)

  • Germination and field test were made with the spring barley sprouted with gibberellin solution of 5 levels from 5 to 100 ppm. Temperatures of room for germination were classified as ordinary and low. Results obtained will be summarized as follows: 1. Sprouting with Gibberellin promoted the germination by two days in ordinary and three days in low temperature without reducing the germination rate. 2. The higher concentration of gibberellin showed the more promotion of growth in early stage. 3. No retardation of root-growth was found by gibberellin sprouting. 4. No promoting effects to germination by gibberellin sprouting was found in field tests. In high concentration, percentage of germination, number of spike and grain yields reduced remarkably. 5. More remarkable tendency of weak growth and longer duration of recovery were found in the higher concentration of gibberellin, but all plots recovered from weak growth at middle stage of plant growth. 6. In gibberellin concentration of 5 to 10 ppm, retardation of germination, weakness of early growth and reducing of spike number were not remarkable while length of spike and 1000 grain weight were increased. Therefore grain yields showed increasing tendency. 7. Promoting the germination may be expected but hastening of heading and maturing may not be available by gibberellin treatment in sprouting.
