Metagenomic analysis of bacterial community structure and diversity of lignocellulolytic bacteria in Vietnamese native goat rumen

  • Objective: In a previous study, analysis of Illumina sequenced metagenomic DNA data of bacteria in Vietnamese goats' rumen showed a high diversity of putative lignocellulolytic genes. In this study, taxonomy speculation of microbial community and lignocellulolytic bacteria population in the rumen was conducted to elucidate a role of bacterial structure for effective degradation of plant materials. Methods: The metagenomic data had been subjected into Basic Local Alignment Search Tool (BLASTX) algorithm and the National Center for Biotechnology Information non-redundant sequence database. Here the BLASTX hits were further processed by the Metagenome Analyzer program to statistically analyze the abundance of taxa. Results: Microbial community in the rumen is defined by dominance of Bacteroidetes compared to Firmicutes. The ratio of Firmicutes versus Bacteroidetes was 0.36:1. An abundance of Synergistetes was uniquely identified in the goat microbiome may be formed by host genotype. With regard to bacterial lignocellulose degraders, the ratio of lignocellulolytic genes affiliated with Firmicutes compared to the genes linked to Bacteroidetes was 0.11:1, in which the genes encoding putative hemicellulases, carbohydrate esterases, polysaccharide lyases originated from Bacteroidetes were 14 to 20 times higher than from Firmicutes. Firmicutes seem to possess more cellulose hydrolysis capacity showing a Firmicutes/Bacteroidetes ratio of 0.35:1. Analysis of lignocellulolytic potential degraders shows that four species belonged to Bacteroidetes phylum, while two species belonged to Firmicutes phylum harbouring at least 12 different catalytic domains for all lignocellulose pretreatment, cellulose, as well as hemicellulose saccharification. Conclusion: Based on these findings, we speculate that increasing the members of Bacteroidetes to keep a low ratio of Firmicutes versus Bacteroidetes in goat rumen has resulted most likely in an increased lignocellulose digestion.

Evaluation of Forage Production and Feed Value of Sasa borealis in the Jeju Area (제주지역 조릿대의 사초생산성 및 사료가치평가)

  • This study was conducted to evaluate the forage production and feed value of Sasa borealis (S. borealis) in Jeju Island in order to improve the utilization of Sasa borealis and to help mitigate the problem of reduced plant species diversity caused by S. borealis in Hanlla Mountain. To investigate the forage production, three quadrat structures were installed in the S. borealis natural community in the middle part of Hanlla Mountain. From May to October 2017, S. borealis in quadrats was cut at a fixed time of each month, and then forage production and regenerated acidity per kg/ha were evaluated. For the evaluation of feed value, compositional analysis was performed on the monthly samples. In vitro digestion experiments were carried out using cannula mounted Hanwoo. In vitro neutral detergent fiber digestibility(IVNDFD) and in vitro acid detergent fiber digestibility(IVADFD) were measured after the experiment. Forage production of S. borealis showed relatively good regeneration ability in May and June, but the regeneration ability decreased as the cutting was repeated. In order to use S. borealis as a forage, it is considered efficient to feed black goats with good fiber decomposition or horses good palatability to S. borealis and relatively good digestibility.

A Comparative Study of Dry Matter Yield and Nutritive Value of Tall type and Turf type Mixtures (상번초 및 잔디형 혼파초지의 건물수량과 사료가치 비교 연구)

  • This study was conducted to investigate the effect of tall type mixtures and turf type mixtures on dry matter yield, botanical composition and quality. The experimental design includes two mixture types : Tall type mixtures; orchardgrass (Potomac) 50+tall fescue (Fawn) 20+ Kentucky bluegrass (Kenblue) 10+red clover (Kenland) 20%) and turf type mixtures; Kentucky bluegrass(Newport) 60%+tall fescue (reboil Jr.) 20+perennial ryegrass (palmer II) 20%. The DM yield was higher obtained in tall type mixture than that of turf type mixture (p<0.05). In the chemical composition, Turf type mixture was higher in crude protein (CP) and dry matter digestibility (DMD), but lower in fibrous compound as NDF, ADF, cellulose and lignin than those of tall type mixture (p<0.05). The crude protein dry matter (CPDM) and digestible dry matter (DDM) yield was higher in tall type mixture than turf type mixture (p<0.05). In this experiment, nutritive value of turf type mixture was higher than that of tall type mixture, but CPDM and DDM yield of turf type mixture were lower than that of turf type mixture due to low dry matter yield. On the other hand, turf type mixture was obtained high feed value, and maintaining the various botanical composition on the 13 th cutting frequency condition. therefore, turf type mixture had a utilization possibility under grazing livestock as sheep and milk goats.

The role of dry land forests for climate change adaptation: the case of Liben Woreda, Southern Oromia, Ethiopia

  • Background: Despite the increasing role of dry forests in climate change adaptation and mitigation, these versatile resources has got less attention in the national and regional planning, their potential to enhance the local and national economy has been overlooked, and their contribution to sustainable environmental management has not been recognized. Hence, the objective of this study was to assess the socioeconomic contribution of dry forests and forest products to climate change adaptation in the Liben Woreda, Southern Oromia region of Ethiopia. Methods: For this study, an integrated qualitative and quantitative approach was used. A total of 74 households from villages in the Bulbul, Boba, and Melka-Guba kebeles were randomly selected for the household survey. Results: Results showed that 75% of the respondents in the area indicated that climate change has become their major sources of vulnerability, where drought has been manifested in the form of crops failure and massive death of livestock particularly cattle species. The main income strategies of the study households include livestock, crop, forests such as gum and resins, firewood and charcoal and non-farm activities such as in the form of petty trade, wage and aid. The average total household income was ETB 11,209.7. Out of this, dry forest income constituted 15% of the total income. In addition to using dry forests as rangeland for livestock, the communities collect wood for construction, fodder, traditional medicine, and forest food both for subsistence and for sale. On the other hand, dry forest products could be considered as less vulnerable, rather resilient livelihood strategies to climate- and environment-related risks compared to livestock and crop production such as in the face of drought periods. More than 48.6% of the households argued that the income generated from dry forests increased substantially due to increment in the level of engagement of family members in forest based income activities. On the other hand, 35.8% of the households responded that livestock production, particularly camels and goats, have been making the livelihood strategies of the respondents more resilient indicating the shift made from grazers browsers to livestock. In general trends show that, the trends of livelihood dependency on dry forest were highly increasing indicating the importance of dry forest income in responsse to frequent droughts. Conclusions: Dry forest income has been becoming crucial livelihood staretgy in response to frequent droughts in the study area and hence, it is important to improve the management of dry forests for livelihood enhancement, while also securing their long-term ecological functions.

The Effects of Ozone Gas on Wound Healing in Skin (오존 가스가 피부창상의 치유에 미치는 영향)

  • This study was performed to determine the optimal dose of ozone for wound healing. Twenty Korean black goats were allocated to 4 groups. The skin wound with $1{\times}1cm$ was induced over the hoof. For 5 minutes per day, 0.1 ppm (Group I), 1 ppm (Group II) and 6 ppm (Group III) ozone gas was applied. In control group, 5 ml of normal saline was applied daily on the wound. Wound square was determined using color image analyzer at day 0, 4 and 14. Soft tissue depth of wound was determined using radiograph. Before the application of ozone gas, $1{\times}1cm$ of skin tissue was resected and observed for histopathologic findings. To determine the healing effect, $2{\times}1cm$ of wound tissue was resected and observed for histopathologic findings at day 14. In mean squares of wound, all experimental groups revealed more reduced mean squares than control group at day 4, and especially Group II (p<0.05) and Group III (p<0.05) revealed significant reduction. At day 14, Group I and Group II (p<0.05) revealed more reduced mean squares than control group, but Group III revealed less reduced mean squares than control group. In soft tissue depth of wound, all experimental groups and control group revealed swelling at day 4, compared with day 0. At day 14, all experimental groups and control group revealed reduction, compared with day 4, and especially Group II (p<0.05) revealed significant reduction. In histopathological findings, inflammation findings in epidermis and dermis, hemorrhagic finding in epidermis were revealed in control group. In Group I, inflammation finding was reduced, compared with control group. Formation of scab and many reepithelialization of epidermis were revealed. In Group II, inflammation findings in epidermis and dermis which were observed in control group were not found, normal skin tissue was revealed. In Group III, inflammation findings in epidermis and dermis were revealed, reepithelialization of epidermis was not founded. Slow healing process was observed, compared with Group I and II. It was concluded that Group II has excellent promotion effect of wound healing.

Comparison of Diluents on Liquid Storage of Korean Native Goat Spermatozoa (희석액의 종류가 재래 흑염소 액상 정액의 생존율에 미치는 영향)

  • This study was to investigate the optimal short-term storage diluents for goat spermatozoa. Semen was collected with electro-ejaculation from two goats. The collected semen was diluted in BTS and centrifuged at 500 g for 5 minutes. The supernatant was discarded and diluted with BTS, Modena or Triladyl$^{(R)}$ and stored at 4 or $17^{\circ}C$ for 8 days. The motility of spermatozoa in BTS and Modena stored at $4^{\circ}C$ was rapidly decreased at day 1. The motility of spermatozoa in BTS at $17^{\circ}C$ was decreased to 30$\sim$45% at day 2. In Modena at $17^{\circ}C$, the inappropriate motility fur artificial insemination (AI) was reached at day 4. The spermatozoa stored in Triladyl$^{(R)}$ at 4 or $17^{\circ}C$ were more viable and higher motility up to day 4. In conclusion, liquid storage of goat spermatozoa in Triladyl$^{(R)}$ at 4 or $17^{\circ}C$ for 4 days showed permissible viability for AI.

Accuracy of Ultrasonography in Early Pregnancy Diagnosis in Doe

  • The present study was undertaken to evaluate the accuracy of ultrasonography in early pregnancy diagnosis in goats. Ultrasonographic scanning with real time B-mode ultrasound machine having 5 MHz linear array transducer was performed on gravid uterus (n=24) obtained from slaughterhouse (Group I). Crown rump length (CRL) measured by ultrasound was found significantly different (p<0.05) with actual CRL measured after dissection in early pregnancy. However, age predicted by ultrasound through the measurement of CRL was found highly correlated (r=0.92) with age measured after dissection through CRL and the weight of fetus. Ages predicted by ultrasound through the measurement of trunk diameter (TD) and uterine diameter (UD) and ages measured after dissection were found highly and equally correlated (r=0.98) and did not differ significantly. Data from six does synchronized (Group II) with PGF2$\alpha$ (Estrumate) at 11 days apart were collected through ultrasound from 17 to 42 days post breding. The correlation between CRL and gestational age was high (r=0.97) in day 30 to 42 post breeding. A high coefficient of correlation (r=0.98) was also observed between predicated age by ultrasound and actual age calculated after kidding. The correlation between CRL and gestational age by the formula Y=(a+bX) i.e. Y=24.42+0.39 X where Y=gestational age and X=CRL, was recorded very high (r=0.99). Accuracy of ultrasonography was lowest on day 17 to 19 (66%) and reached 100% on day 34. Data from 30 does (group III) randomly subjected to only one time ultrasounds scanning to assess the accuracy of pregnancy diagnosis were also obtained. Ages predicted by TD and UD measurements were observed to be non-significantly different with actual age obtained after kidding and correlation between ages predicted by TD and UD measurement with actual age after kidding was found equally and highly correlated (r=0.98). The operator's accuracy in the whole experiment including all three groups was found to be 92%. The sensitivity was 93% and specificity was 86%. From the present study, it was observed that CRL was the most reliable parameter to find out gestational age in early pregnancy and the new formula derived was found very accurate to find out gestational age. TD and UD were also found to be equally reliable parameter to find out gestational age in mid and late stage of pregnancy through ultrasonography. It was concluded that ultrasonography by real time B mode with 5 MHz transrectal transducer was found to be reliable, safe and accurate and practicable means in diagnosing early pregnancy diagnosis as early as 25 days post breeding.

Different Sources and Levels of Copper Supplementation on Performance and Nutrient Utilization of Castrated Black Bengal (Capra hircus) Kids Diet

  • Twenty eight 3-4 month old castrated Black Bengal kids (Capra hircus) were used to determine the effects of source and level of dietary copper (Cu) concentration on their performance and nutrient utilization. Cu was supplemented (0, 10, 20 and 30 mg/kg diet DM) as copper sulfate ($CuSO_4$, $5H_2O$) or copper proteinate (Cu-P). Kids were fed a basal diet containing maize (19.5%), soybean (17.0%), deoiled rice bran (56.5%), molasses (4.0%), di-calcium phosphate and salt (1.0% each) and mineral and vitamin mixture (0.5% each) supplements at 3.5% of body weight to meet NRC (1981) requirements for protein, energy, macro minerals and micro minerals, excluding Cu. The basal diet contained 5.7 mg Cu/kg, 122.5 mg Fe/kg, 110 mg Zn/kg, 0.26 mg Mo/kg and 0.32% S. $CuSO_4$ or Cu-P was added to the basal diet at the rate of 10, 20 and 30 mg/kg. Kids were housed in a well ventilated shed with facilities for individual feeding in aluminum plated metabolic cages. Blood samples were collected from the jugular vein on d 0, 30, 60 and 90 to determine hemoglobin (Hb), packed cell volume (PCV), total erythrocyte count (TEC), total leukocyte count (TLC) and serum enzymes (alkaline phosphatase, alanine transferase and aspertate transferase). A metabolism trial of 6 days duration was conducted after 90 days of experimental feeding. Statistical analysis revealed that source and level of Cu supplementation improved live weight gain (p<0.04) and average daily gain (p<0.01). No significant contribution of source and level of Cu to alter serum serum enzymes was evident. Goats fed Cu-P tended to have higher Hb, PCV and TEC than with $CuSO_4$ supplementation. Cu-P increased digestibility of ether extract (EE, p<0.02) and crude fiber (p<0.05) and showed an increasing trend (p<0.09) for digested crude protein (CP) and crude fiber (CF). Supplemental dose of Cu linearly improved (p<0.02) digestibilities of dry matter (DM), organic matter (OM), EE and nitrogen free extract (NFE). Though the absorption of nitrogen (N) was not affected (p>0.10) by both source and dose of Cu, N retention was affected (p<0.04) and there was a significant $Source{\times}Dose$ interaction (p<0.05). Final body weight (BW) was not influenced (p>0.10) by the source of Cu but increasing dose of Cu increased (p<0.04) the BW of kids. TDN intake (g/kg $W^{0.75}$) was higher (p<0.05) with the increased dose of Cu and there was a significant $Source{\times}Dose$ interaction. It was concluded that supplementation of Cu from different sources and varying dose level in a concentrate based diet may improve performance, nutrient utilization and plane of nutrition in castrated Black Bengal kids. The effects on performance and nutrient utilization are more pronounced with Cu-P than $CuSO_4$ supplementation. Higher dose of Cu showed better result than lower dose.

Effect of Age on Energy Requirement for Maintenance and Growth of Dorper and Hu Crossbred F1 Ewes Weighing 20 to 50 kg

  • This research aimed to define the energy requirement of Dorper and Hu Hybrid $F_1$ ewes 20 to 50 kg of body weight, furthermore to study energy requirement changes with age and evaluate the effect of age on energy requirement parameters. In comparative slaughter trial, thirty animals were divided into three dry matter intake treatments (ad libitum, n = 18; low restricted, n = 6; high restricted, n = 6), and were all slaughtered as baseline, intermediate, and final slaughter groups, to calculate body chemical components and energy retained. In digestibility trial, twelve ewes were housed in individual metabolic cages and randomly assigned to three feeding treatments in accordance with the design of a comparative slaughter trial, to evaluate dietary energetic values at different feed intake levels. The combined data indicated that, with increasing age, the net energy requirement for maintenance ($NE_m$) decreased from $260.62{\pm}13.21$ to $250.61{\pm}11.79kJ/kg^{0.75}$ of shrunk body weight (SBW)/d, and metabolizable energy requirement for maintenance (MEm) decreased from $401.99{\pm}20.31$ to $371.23{\pm}17.47kJ/kg^{0.75}$ of SBW/d. Partial efficiency of ME utilization for maintenance ($k_m$, 0.65 vs 0.68) and growth ($k_g$, 0.42 vs 0.41) did not differ (p>0.05) due to age; At the similar condition of average daily gain, net energy requirements for growth ($NE_g$) and metabolizable energy requirements for growth ($ME_g$) for ewes during late fattening period were 23% and 25% greater than corresponding values of ewes during early fattening period. In conclusion, the effect of age upon energy requirement parameters in the present study were similar in tendency with previous recommendations, values of energy requirement for growth ($NE_g$ and $ME_g$) for Dorper and Hu crossbred female lambs ranged between the NRC (2007) recommendation for early and later maturating growing sheep.

A Textile Analysis of Woolen Tapestry Curtain in Seoul Museum of Craft Art (서울공예박물관 소장 모담방장(毛毯房帳) 직물 분석)

  • A woolen tapestry curtain, owned by the Seoul Museum of Craft Art, is composed of tabby by cotton-warp and wool-weft threads and its patterned part is shown as a tapestry; paint with pigment has been added to it. The chromaticity of this curtain was measured and the substances in the deep red color were confirmed as Hg by an analysis of the ingredients through X-ray fluorescence. This is presumed to be cinnabar or vermilion. Analyses were performed on a total of seven fabric samples, including the warp & weft of the fabric, its trimming, and its back fabric. As a result, the warp of the woolen tapestry curtain was determined to be a cotton fiber with a middle hole or lumen in the cross-section. Furthermore, an infrared peak likewise showed O-H and C-O binding. Wool fibers as wefts were identified with circular and oval cross-sections and IR peaks showed N-H/O-H stretching and amide(-CONH-). The animal hair samples used in the wool fiber are believed to have come from long-tailed goral or goats and the possibility of using easy-to-spin sapsal dog hair is also not to be overlooked. This was determined through a contrast analysis by the Cultural Heritage Administration to identify the animal species used in the tapestry.
