Growth Performance of Transgenic Mud Loach Misgurnus mizolepis Carrying a GH Transgene Driven by Mud Loach C-Type Lectin Regulator

  • Growth hormone (GH) transgenesis in fish has the potential to improve aquaculture efficiency and capacity. However, many fast-growing transgenic fish have experienced side effects caused by excess GH expression. To overcome this unwanted issue associated with several GH transgenic mud loach Misgurnus mizolepis lines carrying GH construct driven by a strong ${\beta}$-actin regulator ($pml{\beta}$-actGH), we performed an alternative version of GH autotransgenesis using a weaker but more stable regulator, the mud loach lectin promoter. GH transgenesis with a pmlectGH construct consisting of the mud loach GH gene driven by the 2.3-kb lectin promoter exhibited significant growth stimulation. However, the extent of the growth acceleration in pmlectGH transgenics (six times maximum when assessed 2 months post hatching) was much less than that in transgenic individuals carrying the $pml{\beta}$-actGH construct. Additionally, the extraordinary gigantism that was common in $pml{\beta}$-actGH-transgenic mud loaches was diminished in transgenic loaches harboring the pmlectGH construct. Transgenic founders (pmlectGH) successfully transmitted their transgene into the next generation with up to 41% frequency. Growth stimulation also persisted in the transgenic F1 strains, with a seven-fold increase in maximum body weight at 6 months of age.

  • In order to investigate the effects of growth regulators on the formation of capsaicinoids in callus of Capsicum annuum L. tissues were cultured in the Linsmaier and Skoog RM 1964 medium containing various growth regulators. Production of capsaicinoids during culture was monitored by gas chromatography. In the presence of $10^{-6}M$ of 2,4-D and kinetin in the medium, $1182{\mu}g$ of capsaicinoids were formed per 100g dry wt. of tissue, of which was greater than with any of three other growth regulators. IAA, NAA, and kinetin of same concentrations had 65%, 38%, 68% effect of 2.4-D in capsaicinoids formation, respectively. Production of capsaicinoids increased gradually in the presence of 2,4-B as culture period was proceeded. Of phenylpropanoids formed, cinnamic acid and coumaric acid were not significantly different in their levels, although growth regulators were varied. On the other hand, caffeic acid and ferulic acid formation were highest in the presence of 2,4-D. Effects of kinetin and IAA were about 70 percent of that of 2,4-D, whereas NAA had only about 30 percent effect. Phenylalanine ammonia-lyase activity in cultured tissue was increased during the periods; 52, 81, and 209 n moles of cinnamic acid per g fresh wt. were formed after 5, 15, and 25 days of culture, respectively.

Benzaldehyde as a new class plant growth regulator on Brassica campestris

  • Plant growth regulator is an essential pesticide to date while the available active ingredient is not well understood unlike fungicide, insecticide and herbicide. This study was aimed to evaluate a new chemical class of plant growth regulator, and the total of 92 benzene derivatives were screened for their germination and early stage of the root growth regulation on Brassica campestris. Thirty benzaldehydes, nine acids, one amide, and one ester showed potent root growth inhibitory activity (>70 % inhibition) while only salicylaldehyde showed potent germination inhibition ($IC_{50}=81.2mg/L$) suggesting that benzaldehyde was a key module candidate for the growth inhibition. Benzaldehydes were further evaluated for root growth inhibition. 2,3-Dihydroxybenzaldehyde and salicylaldehyde showed $IC_{50}$ values of 8.0 and 83.9 mg/L, respectively. On the other hand, salicylaldehyde, and 2,4,5-trihydroxybenzaldehyde were found to have root growth promotion effects less than 10 mg/L. This result suggests that the benzaldehyde is a new class candidate for plant growth regulator.

Effects of Bedsoil and Growth Regulator on Cutting Propagation of Cudrania tricuspidata Bureau (상토 및 생장조절제 처리가 꾸지뽕나무의 삽목번식에 미치는 영향)

  • This Study was carried out to select bed soil and growth regulator which are effective for the macro propagation by cutting of Cudrania tricuspidata Bureau. The rooting of Cudraniae tricuspidata showed good result when the bed soil was coarse soil, rooted hardwood cutting 56.9%, and softwood cutting 62.3%. The rooting of Cudraniae tricuspidata showed good result when treated by growth regulator IBA 100 ppm, rooted hardwood cutting 57.9%, and softwood cutting 89.2%.

  • The effects of insect growth regulator Dimilin which interfere with the synthesis of chitin in the cuticle of insect larvae were investigated at various concentrations using the crab larvae of Hemigrapsus sanguineus. The larvae were cultured at control, 0.5, 1, 5 and 10 $\mu g$ Dimilin solutions and three replicate experiments were carried out to give correct analysis. Significant differences in percent mortality have occurred between control and 10$\mu g$ when the larvae were exposed to Dimilin whereas no differences were found between 5 and 10$\mu g$ Dimilin concentrations. If lethal concentration is defined as concentration at which less than 10 percent of crab larvae reach to the last zoeal stage from hatching it can be concluded that insect growth regulator Dimilin is lethal to the larvae of Hemigrapsus sanguineus at 5 and 10$\mu g$ Dimilin.

  • Effects of the plant growth regulator (P. G. R.)on the reaction of proteinase, $\gamma-amylase$ and acid phosphatase were investigated, and also were the conditions of production of P. G. R. by Stroptomyces sp. 445. The P. G. R. had no effect on the act ivities of such enzymes in mung bean seedling. But in germinating seed previously treated with P. G. R. it effected the activity of protease in cotyledon. In the conditions of production of P. G. R., the maxim, activity was appeared in shaking cutlure at $30^{\circ}C$ for 5 days, and by the addition of peptone or casein hydrolysate as nitrogen source, soluble starch as carbon source, and sulfur as metal ion.

  • The gene bolA was discovered in the 80's, but unraveling its function in the cell has proven to be a complex task. The BolA protein has pleiotropic effects over cell physiology, altering growth and morphology, inducing biofilm formation, and regulating the balance of several membrane proteins. Recently, BolA was shown to be a transcription factor by repressing the expression of the mreB gene. The present report shows that BolA is a transcriptional regulator of the dacA and dacC genes, thus regulating both DD-carboxypeptidases PBP5 and PBP6 and thereby demonstrating the versatility of BolA as a cellular regulator. In this work, we also demonstrate that reduction of cell growth and survival can be connected to the overexpression of the bolA gene in different E. coli backgrounds, particularly in the exponential growth phase. The most interesting finding is that overproduction of BolA affects bacterial growth differently depending on whether the cells were inoculated directly from a plate culture or from an overnight batch culture. This strengthens the idea that BolA can be engaged in the coordination of genes that adapt the cell physiology in order to enhance cell adaptation and survival under stress conditions.

Effects of Plant Growth Regulator on Seedling Growth in Onion Seed(Allium cepa L.) (식물 生長調整劑處理가 양파(Allium cepa L.)의 幼苗生長에 미치는 영향)

  • This study was conducted to stable direct sowing cultivation with seedling growth promotion by plant growth regulator treatment in onion seed(Allium cepa L.) The emergence percentage of soaked seed in BA, GA$_3$, and kinetin solution were higher than control seed, and those were 93.0, 94.3, 93.8%, respectively. The plant height was significant elongation in growth regulators treated seed, and those effects were high GA$_3$, kinetin and BA in the order, and the extend were high as increasing the growth regulator solution concentration in GA$_3$ and BA, and that was reverse in kinetin. The No. of leaf was increased in growth regulators treated seed, and the extend were similar among the growth regulators. The No. of root was increased significantly in growth regulators treated seed, and the extend were high in GA$_3$, BA and kinetin in the order.

Vertical Shoot Growth of Korean Lawngrass (Zoysia japonica Steud.) Influenced by Trinexapac-ethyl, Amidochlor, and Mefluidide (Trinexapac-ethyl, Amidochlor 및 Mefluidide가 들잔디 직립생장에 미치는 효과)

  • Research was initiated to evaluate plant growth regulator effects on the vertical shoot growth of Korean lawngrass and to determine desirable growth regulator and its rate. The experiments were conducted twice at different sites in 1995. All the tested growth regulators inhibited the growth, but the inhibition period was variable among the product in Experiments I and II. During the first week after treatment, there was approximately 10 to 20% growth reduction in most of the treated plots. In the amidochlor-treated plots, growth suppression was effective for 3 to 4 weeks at low to medium rates ($0.60mL{\cdot}m^{-2}$). A Type II growth regulator, trinexapac-ethyl exceeding the medium rate of $0.08mL{\cdot}m^{-2}$ consistently tended to suppress vertical shoot growth for 8 weeks, being above 35% reduction in both experiments. In the plots applied with mefluidide, growth suppression appeared with foliar discoloration 3 or 4 days earlier than the other growth regulators and continued to work till the 8 weeks after treatment. Suppression intensity on vertical shoot growth increased with time after treatment up to a certain period of time, depending on growth regulators. Generally, the higher the application rate, the greater the suppression intensity. Seasonal variation of activity and effectiveness of growth regulators was observed, resulting in lower suppression intensity in July than in June. It is expected to reduce mowing requirements by 30 to up to 60% for a certain period with a specific growth regulator. In low to medium maintenance of Korean lawngrass turf, a long-term suppression may be more effectively accomplished with trinexapac-ethyl rather than mefluidide and amidochlor in terms of vertical shoot growth inhibition. Therefore, turf managers will need to select proper growth regulator and determine optimum rate of application for turfgrass management, based on a defined period of mowing reduction.

  • This experiments were conducted to investigate growth regulator concentrations and a period of chilling treatment for increasing the size of bulblets of Narcissus pseudo-narcissus cv King Alfred. Combination of 2.5 mg/L BA and 2.5 mg/L NAA remarkedly increased the diameter and growth of regenerated bulblets. Chilling period favorable for the growth of regenerated bulblets was found to be 8 weeks, and addition of 1.0 mg/L NAA alone to MS medium was the most effective for increasing the size of bulblet for the culture after the chilling treatment.
