Effect of Growth Stimulating Agent in Lactic Acid Bacteria Fermented Food Prepared from Egg White Powder and Casein (난백분말과 카제인으로 만든 젖산균 발효식품에서 생육촉진물질의 효과)

  • Lactic acid bacteria (LAB) fermented foods were prepared from egg white powder (EWP), casein and growth stimulating agents (GSA). The effects of GSA on acid production and growth of Lactobacillus were studied. The effects of GSA on sensory properties and viscosity of LAB fermented foods were also studied. Acid production by Lactobacillus was stimulated by addition of GSA (0.3% or 1%, W/V). Although stimulating effect differed among each GSA, some GSA increased the acidity up to the level of fermented milk. However, stimulating effect of GSA on viable cells was not noticeable. Acid production by L. acidophilus was generally higher than other Lactobacilli. The optimum concentration of GSA added to substrate was 1% (W/V). Sensory evaluation showed that the optimum fermentation time was 18hr. The sensory properties of GSA samples were evaluated as slightly lower than that of fermented milk because GSA samples showed whey separation and taste and smell of GSA. Apparent viscosity of GSA samples was significantly lower than that of fermented milk and control sample (p<0.05). There was no significant difference of apparent viscosity among GSA samples. GSA samples, fermented milk and control sample showed thixotropic flow characteristics.

Dual Effect of the Cubic Ag3PO4 Crystal on Pseudomonas syringae Growth and Plant Immunity

  • We previously found that the antibacterial activity of silver phosphate crystals on Escherichia coli depends on their structure. We here show that the cubic form of silver phosphate crystal (SPC) can also be applied to inhibit the growth of a plant-pathogenic Pseudomonas syringae bacterium. SPC pretreatment resulted in reduced in planta multiplication of P. syringae. Induced expression of a plant defense marker gene PR1 by SPC alone is suggestive of its additional plant immunity-stimulating activity. Since SPC can simultaneously inhibit P. syringae growth and induce plant defense responses, it might be used as a more effective plant disease-controlling agent.

Anti-Oxidant and Hair-Growth-Promoting Effect of Pectin Lyase-Modified Red Ginseng Extract (GS-E3D) (홍삼가수분해추출물(GS-E3D)의 항산화 및 양모 효과)

  • To develop new therapy for prevention and treatment of hair loss is very important according to increase of the number of hair loss people. The aim of this study was to investigate the hair growth promoting effects of pectin lyase-modified red ginseng extract (GS-E3D). We examined antioxidant and anti-inflammatory effects, human hair dermal papilla cells (HHDPC) proliferation, and testoterone-induced $5{\alpha}$-reductase inhibitory effects. GS-E3D show not only 1,1-diphenyl-2-picryhydrazyl (DPPH) radical scavenging activity and xanthine oxidase inhibitory effects as an anti-oxidant property, but also lip-oxygenase and hyaluronidase inhibitory effects as an anti-inflammatory property. Human hair dermal papilla cells proliferation by GS-E3D was higher than those of minoxidil or finasteride, using the positive controls. Moreover, GS-E3D exhibited $5{\alpha}$-reductase inhibitory activities after stimulating by testoterone. The present results indicate that GS-E3D has a potential to be as an hair growth promoting agent for cosmetic materials.

Freeze Drying of Lactic Acid Bacteria Fermented Food Prepared from Egg White Powder and Casein Supplemented with Growth Stimulating Agent (생육촉진물질이 첨가된 난백분말과 카제인으로 만든 젖산균 발효식품의 동결건조)

  • Lactic acid bacteria fermented foods were prepared from egg white powder, casein and growth stimulating agent. pH change and growth of Lactbacillus acidophilus(KCTC 2182) during freeze drying were studied. The effects of freeze drying on sensory evaluation. hardness and volatile aroma compounds in freeze dried sample or reconstituted sample were also studied. Freezing and freeze drying did not affect pH of fermented samples. Number of viable cells in original fermented samples was markedly reduced during freezing or freeze drying. When number of viable cells in original fermented sampler was considered at 100%. survival ratio of viable cells after freezing was $72.0{\sim}82.4%$ and that after freeze dying $10.0{\sim}20.4%$. When sensory properties of original fermented samples were compared with those of freeze dried/reconstituted samples, sensory properties of original samples were generally better than those of freeze dried/reconstitute samples. However, the reconstitution property and the acceptability of freeze dried samples were good. Volatile aroma compounds in original fermented samples were reduced during freeze drying. The reduction degree of volatile aroma compounds varied with sample.

  • Human granulocyte colony-stimulating factor (hG-CSF) is a hematopoiesis agent that principally affects the differentiation of neutrophils in the bone marrow. At present, recombinant hG-CSF is used successfully in the treatment of chemotheraphy-induced neutropenia and its indication has been expanded to bone marrow transplantation and aplastic anemia. In this study, we have constructed rhG-CSF secretion plasmid pYRC1 in which OmpA signal sequence/hG-CSF gene was expressed under the control of the T7 promoter. rhG-CSF produced in E. coli BL21 (pYRC1) grown at $37{\circ}C$ was found in aggregates. However, 15% of the periplasmic protein was soluble rhG-CSF when the E. coli BL21 (pYRC1) was cultured at $25^{\circ}C$ for 7 h in the modified MBL medium containing 10 g/$\ell$ glucose with 10 $\mu$M IPTG induction. The production of soluble rhG-CSF in E. coli BL21 (pYRC1) using fed batch culture was also studied. In the fed batch culture system, the final yield of rhG-CSF produced from E. coli BL21 (pYRC1) was increased from 4.4 mg/$\ell$to 24 mg/$\ell$by controlling the specific growth rate from $0.43 h^{-1}$ to $0.14 h^{-1}$, and optimizing the time of induction.

The Experimental Studies of YangHyulEum Gami-Bang Extracts on the Hair Growth Effect (양혈음가미방(養血飮加味方) 추출물의 발모효과에 대한 실험적 연구)

  • Objectives : YangHyulEum Gami-Bang(YHEG) is a hair care extracts which is composed of fourteen plant extracts used in oriental medicine. The purpose of this study is to investigate the effect of YangHyulEum Gami-Bang(YHEG) on the alopecia and hair growth.Methods & Results : The herbal extracts from YangHyulEum Gami-Bang(YHEG) was tested using in vivo and in vitro test models. 1. The YHEG extracts showed effect on the DNA proliferation of the hair dermal papilla cells measured by [3H]thymidine incorporation. 2. YHEG showed promoting on the expression of growth factors such as IGF-1, KGF-1 and inhibiting on the expression of inhibitory hair growth factor such as TGF-β1, BMP-2 estimated by qPCR. 3. The YHEG extracts showed effect on the activation of β-catenin in the dermal papilla cells. 4. YHEG showed inhibitory effects of NO synthesis at 0.2% concentrations. 5. YHEG showed effects in the expression of IL-1β, TNF-α, IL-6, COX-2 and iNOS gene in the LPS stimulated RAW 264.7 cells. 6. The hair growth index of the YHEG extracts ranked at over 2 when compared to control group which was ranked at 0. 7. The hair follicle number, length and size of the experimental group were remarkably higher than the control group in the histological observation.Conclusions : These results suggest that YangHyulEum Gami-Bang(YHEG) has hair growth promoting activity and it can be used as a potent treatment agent for preventing hair loss and stimulating hair growth for treatment of alopecia.

Development of anti-wrinkle agent with Artificial Skin Culture Broth (DA-3711) (인공피부배양물(DA-3711)을 이용한 주름개선제 개발)

  • Artificial skin culture broth (DA-3711) was developed by using Dong-A pharmaceutical's artificial skin culture technique, and it contains plenty of nutrients, natural extracellular matrix proteins and human growth factors, which may improve aged skin conditions. The anti-aging effect of DA-3711 was investigated both in vitro and in vivo. Furthermore the clinical studies showed that DA-3711 had been very effective in improving firmness and reducing wrinkles. The results suggest that the novel anti-wrinkle agent. DA-3711, may have anti-aging and anti-wrinkle effects by stimulating skin regeneration.

  • The treatment of pediatric patients with chronic renal disease comprises management of nutritional imbalance, fluid, electrolyte, and acid-base disturbances, mineral bone disease, anemia, hypertension, and growth retardation. The treatment also includes administration of appropriate renal replacement therapy, if required. Adequate dietary intake of carbohydrates, fats, and proteins and caloric intake must be encouraged in such patients to ensure proper growth and development. In addition, fluid, electrolyte, and acid-base status must be regularly monitored and should be well maintained. Serum calcium, phosphorus, and parathyroid hormone levels must be maintained at their target range, which are determined on the basis of the glomerular filtration rate, to avoid the development of mineral bone disease. This can be achieved by using phosphorus binders and vitamin D analogues. An erythropoiesis-stimulating agent must be administered along with iron supplementation to maintain the hemoglobin level of the patients between 11-12 g/dL. Hypertension must be controlled with adequate water and sodium balance and appropriate antihypertensive agents. Administration of recombinant human growth hormone is recommended to improve the final adult heights.

Development of Wound-treatment Formulation Using Plant Growth Regulators for Wound Healing of Some Tree Species (수피상처치료를 위한 식물생장조절물질 기반 상처유합제 개발)

  • New tree wound-treatment formulations (WTF) were developed in this study. Stimulating effects of plant growth regulating substance on callus formation were evaluated in vivo twig disc culture of Zelkova serrata, Ziziphus jujuba, Pinus densiflora, Ginkgo biloba. and Aesculus turbinata. Based on the evaluation, WTF were prepared with 2,4-D, NAA, and IBA using xanthan gum as an extending agent. WTF were pasted on $2{\times}10cm$ artificial wound on the stem and the effects were evaluated 12 weeks later with the thickness of new callus-tissues. The effects varied with tree species and the growth regulators. In Z. serrata and A. turbinata, 2 mg/L of 2,4-D was the best WTF. In P. koraiensis and G. biloba, 8 mg/L of NAA and 1 mg/L of IBA were the best, respectively. Callus formation was quite lower in G. biloba compared to other species. Stimulating effect of thiophanate-methyl paste was excellent only in Z. serrata and lower than certain WTF in other species. Vaseline, which was used generally as an WTF, remained longer on the wound and causing decay. From the results, it could be recommended that 2,4-D, NAA and IBA were the best WTF for Z. serrata/A. turbinata, P. koraiensis and G. biloba, respectively.

Acacia ferruginea Inhibits Tumor Progression by Regulating Inflammatory Mediators-(TNF-α, iNOS, COX-2, IL-1β, IL-6, IFN-γ, IL-2, GM-CSF) and Pro-Angiogenic Growth Factor-VEGF

  • The aim of the present investigation was to evaluate the effect of A ferruginea extract on Dalton's lymphoma ascites (DLA) induced tumours in BALB/c mice. Experimental animals received A ferruginea extract (10 mg/kg.b.wt) intraperitoneally for 14 consecutive days after DLA tumor challenge. Treatment with extract significantly increased the life span, total white blood cell (WBC) count and haemoglobin (Hb) content and decreased the level of serum aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), gamma glutamyl transferase (${\gamma}$-GT) and nitric oxide (NO) in DLA bearing ascites tumor models. In addition, administration of extract significantly decreased the tumour volume and body weight in a DLA bearing solid tumor model. The levels of pro-inflammatory cytokines such as tumor necrosis factor-alpha (TNF-${\alpha}$), interleukin-1 beta (IL-$1{\beta}$), interleukin-6 (IL-6) and granulocyte monocyte-colony stimulating factor (GM-CSF), as well as pro-angiogenic growth factors such as vascular endothelial growth factor (VEGF) and inducible nitric oxide synthase (iNOS) were elevated in solid tumour controls, but significantly reduced by A ferruginea administration. On the other hand, the extract stimulated the production of interleukin-2 (IL-2) and interferon-gamma (IFN-${\gamma}$) in animals with DLA induced solid tumours. Increase in $CD4^+$ T-cell population suggested strong immunostimulant activity for this extract. GC/MS and LC/MS analysis showed quinone, quinoline, imidazolidine, pyrrolidine, cyclopentenone, thiazole, pyrazole, catechin and coumarin derivatives as major compounds present in the A ferruginea methanolic extract. Thus, the outcome of the present study suggests that A ferruginea extract has immunomodulatory and tumor inhibitory activities and has the potential to be developed as a natural anticancer agent.
