The Effect of 5-Oxohexyl-3,7-dimethylxanthine on Carbon Tretrachlroride-Induced Hepatotoxicity in Rats (5-Oxohexyl-3,7-dimethylxanthine이 사염화탄소로 유발된 흰쥐의 간손상에 미치는 영향)

  • A series of new derivatives of 1-(5-oxohexyl)-3,7-dimethylxanthine has been prepared for the treatment of the vascular dementia. To investigate hepatoprotective effect of these derivatives, the serum biochemical activity and the histological change of liver tissue were evaluated in carbon tetrachloride ($CCl_4$) treated rats. The activities of aspartate aminotransferase and alanine aminotransferase and the activities of total cholesterol, triglyceride and total bilirubin in 5-oxohexyl-3,7-dimethylxanthine derivatives pretreated rats were significantly decreased compared to the $CCl_4$ only treated rats but the content of glutathione S-transferase was increased compared to the $CCl_4$ only treated rats. These results indicated that 5-oxohexyl-3,7-dimethylxanthine derivatives have hepatoprotective effect in $CCl_4$ intoxicated rats.

Saucerneol B with Hepatoprotective Effect of the Roots of Saururus chinensis

  • In order to find the new hepatoprotective agents from natural products, the isolation and identification of biological active components of the roots of Saururus chinensis has been carried out. A MeOH extract of this plant showed the significant hepatoprotective effect on tacrine-induced cytotoxicity in Hep G2 cells. Five lignans including sauchinone, manassantin A, manassantin B, saucerneol B, and di-O-methyltetrahydrofuriguaiacin B were isolated and identified by spectroscopic evaluation. (omitted)

  • In the course of hepatoprotective screening for domestic plants. the aerial parts of B. vulgaris var. cycla exhibited hepatoprotective activity which was determined by using the primary cultures of rat hepatocytes injured by H2O2. Bioactivity-guided separation for this plant gave a new flavonoid (1) and the known compounds (2-4), which structures were elucidated by 1H-NMR, HMQC, 1H-1H COSY and HMBC as compound 1, apigenin 8-C-, 7-O-di-$\beta$-D-glucopyranoside, compound 2, vitexin 2"-O-$\beta$-D-glucopyranoside, compound 3, (+)-dehydrovomifoliol, and compound 4, 3-hydroxy-5$\alpha$, 6$\alpha$-epoxy-$\beta$-ionone.

  • Objectives : Present study investigated hepatoprotective effect of Haegan-jeon extract (HE) and tried to elucidate molecular mechanism involved. According to molecular mechanism, present study optimized herbal composition of HE (op-HE) and compared in vitro and in vivo hepatoprotective effects of op-HE to HE. Methods : For in vitro experiments, HepG2 cells were exposed to arachidonic acid (AA, $10{\mu}M$) and iron ($5{\mu}M$) for inducing oxidative stress. Cell viability, GSH contents, $H_2O_2$ production, mitochondrial membrane potential, immunoblot and reporter gene assay were performed to investigate cytoprotective effects and responsible molecular mechanisms. For in vivo experiments, hepatoprotective effect of HE and op-HE were assessed on $CCl_4-induced$ liver injury mice model. Results : HE pretreatment prevented AA+iron-mediated hepatocytes apoptosis. In addition, AA+iron-induced mitochondrial dysfunction, $H_2O_2$ production, glutathione depletion were reduced by HE pretreatment. In addition, nuclear factor erythroid 2-related factor 2 (Nrf2) phosphorylation, antioxidant response element (ARE)-driven reporter gene activity, and antioxidant genes expression were increased by HE. Based on reporter gene and MTT assays, we found that op-HE consisting three medicinal herbs also significantly increased transactivation of Nrf2 and reduced the AA+iron-mediated cytotoxicity. Moreover, in $CCl_4-induced$ liver injury mice model, HE-op had an ability to ameliorate $CCl_4-mediated$ increases in serum alanine transferase and aspartate aminotransferase activity, hepatic degeneration, inflammatory cell infiltration, and collagen deposition. Hepatoprotective effects of op-HE were comparable to those of HE. Conclusions : Present study suggests that op-HE as well as HE exhibit hepatoprotective effect against oxidative stress-mediated liver injury via Nrf2 activation.

  • Dalwoneum is the one of Chinese traditional prescription used for the treatment of liver disease. This prescription consists of Arecae Semen (12 g), Magnoliae Cortex (9 g), Amomi tsao-ko Fructus (3 g), Anemarrhenae Rhizoma (12 g), Paeoniae Radix (9 g), Scutellariae Radix (12 g), and Glycyrrhizae Radix (3 g). Water extract of Dalwoneum showed the significant hepatoprotective effect on tacrine-induced cytotoxicity in Hep G2 cells. (omitted)

  • Through a modification of allicin structure a disagreeable odor and chemical instability of allicin can be improved. 3-Alkoxy-6-allylthiopyridazine derivatives exhibit a superior effect for prevention and treatment of hepatic diseases induced by carbon tetrachloride and aflatoxin B1 and for prevention of human tissues from radiation. These compounds inhibit also efficiently SK-Hep-1 cell proliferation through induction of apoptosis. So another 4,5-mono- or di-substituted 3-alkyloxy-6-allylthiopyridazine derivatives were synthesized on purpose to find out SAR of allylthiopyridazine in hepatoprotective and hepatotherapeutic acitivitis and to develop more effective drug candidate.

  • Methanolic extract of Cassia tora leaves was evaluated for its hepatoprotective activity in rats by inducing hepatotoxicity with paracetamol (acute model). The extract at a dose of 400 mg/kg orally exhibited significant protective effect by lowering the serum levels of transaminase (SGOT and SGPT), bilirubin, and alkaline phosphatase (ALP). The effects produced were comparable to that of a standard hepatoprotective agent.

  • Saururus Chinensis Baill (Saururaceae) has been used as folk medicine for analgesics, beriberi, edema, hepatitis, and icterus, etc. Hepatoprotective effects of Saururus chinensis Baill (SCB) administration on function of the biochemical parameters in liver of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) treated rats were investigated. After 7 days from TCDD(1$\mu\textrm{g}$/kg) injection, SCB(200mg/kg) was administered into rats intraperitoneally for 4 week.s We examined the antioxidative enzymatic activity by measuring the level of AST and ALT in serum and SOD, Catalase, GPx, GSH and GSSG in liver tissue of rats.   • Background : A traditional Oriental medicine, GongJjn-dan (GJD), is one of the most well-known tonic agents in Korea. Among 6 types of GJD components, antler, red ginseng, and Cornus fructus have shown antioxidant effects, while EtOH-induced tissue damage may be a consequence of oxidative stress. Objectives & Methods : The hepatoprotective effects of GJD were evaluated on EtOH-mediated experimental liver damaged rats at 50, 100, 250 and 500mg/kg comparing with 100mg/kg of silymarin as a reference drug in the present study. Test substances were dosed once a day for 60 days with oral administration of 20% ethanol 2.5ml/100g body weight twice a day (equivalent to 7.9g ethanol/kg/day). Each of 8 rats per group was selected using body weight at 10 days after acclimatization. Experimental animals were sacrificed after 60 days of continuous oral treatment of test substances with 20% ethanol treatment, and changes on the body weight, liver weight, and serum AST and ALT were observed. Results : There were dramatic decreases of body weight and increases of liver weight and serum AST and ALT. Similar inhibition effects on the EtOH-induced hepatic damages were detected between equal dosages of GJD and silymarin. Conclusion : Based on these results. it is concluded that GJD showed clear hepatoprotective effects on EtOH-induced hepatic damage.
