Effects of histamine on the ruminal smooth muscle motility of cattle (소(牛)의 제1위 평활근 운동성에 대한 histamine의 효과)

  • Effects of histamine on the ruminal smooth muscle motility of cattle were investigated in the longitudinal and circular smooth muscle strips. In order to these experiments, specimens were obtained from 35 korean native cattles, 3-4 years old, in Kwang-ju area slaughterhouse. Smooth muscle strips of rumen were made from sample, and then measured the isometric contraction with physiograph in $37{^{\circ}C}$ organ bath. The results were as follows : 1. Histamine caused two different types of response(a contraction or a relaxation) on the smooth muscle of cattle rumen. These responses increased in dose dependant manner. 2. Pyrilamine($H_1$-receptor antagonist) completely blocked contraction in all the preparation and converted the response into relaxation. 3. Cimetidine($H_2$-receptor antagonist) completely blocked relaxation in all the preparation and converted the response into contraction. 4. The contraction induced by histamine($10^{-3}M$) was not Mocked by cholinergic, adrenergic blocker or hexamethonium. 5. The contraction induced by histamine($10^{-3}M$) was markedly inhibited in the $Ca^{2+}$ free(or EDTA 2Na) Kreb's solution and by verapamil.

Antipruritic effect of curcumin on histamine-induced itching in mice

  • Itching is a common clinical symptom of skin disease that significantly affects a patient's quality of life. Transient receptor potential vanilloid 1 (TRPV1) receptors of keratinocytes and peripheral nerve fibers in skin are involved in the regulation of itching as well as pain. In this study, we investigated whether curcumin, which acts on TRPV1 receptors, affects histamine-induced itching in mice, using behavioral tests and electrophysiological approaches. We found that histamine-induced itching was blocked by topical application of curcumin in a concentration-dependent manner. In ex-vivo recordings, histamine-induced discharges of peripheral nerves were reduced by the application of curcumin, indicating that curcumin acts directly on peripheral nerves. Additionally, curcumin blocked the histamine-induced inward current via activation of TRPV1 (curcumin $IC_{50}=523nM$). However, it did not alter chloroquine-induced itching behavior in mice, which is associated with transient receptor potential ankyrin 1 (TRPA1). Taken together, our results suggest that histamine-induced itching can be blocked by topical application of curcumin through the inhibitory action of curcumin on TRPV1 receptors in peripheral nerves.

  • This Experimental study was done to research effects of Fagopyrum esculentum Moench(Me-mil) Extract on the anti-allergic effects. The results were obtained as follows : 1. All concentrations of Fagopyrum esculentum Moench Extract(Flours and seeds) inhibit histamine release under the vascular permeability response to intracutanenous injection. The result is proportion to concentration. But, rutin can't get considerable result. 2. All concentrations of Fagopyrum esculentum Moench Extract inhibit histamine release under the vascular permeability response to intraperitoneal injection of SD Rat in comparison with Diphenhydramine which is typical anti-histamine drug. 3. All concentrations of Fagopyrum esculentum Moench Extract inhibit histamine release under the vascular permeability response to per-oral during the three or five days. 4. In the result of quantification of histamine induced Compound $\frac{48}{80}$, Flours and seeds of Fagopyrum esculentum Moench Extract inhibit histamine release. 5. Among the fracination of Fagopyrum esculentum Moench Extract, $CHCl_3$ fraction inhibit histamine release effectively. 6. In the result of genetic manifestative inhibition about the Human mast cell treated PMA and A23187, Fagopyrum esculentum Moench Extract effect in the IL-4 and TNF-${\alpha}$ except IL-5. According to the above results, it is suggested that Fagopyrum esculentum Moench(Me-mil) Extract has anti-allergic effect.

Exposure Assessment of Biogenic Amines as a Chemical Hazard in Commercial Anchovy Fish Sauce from South Korea (멸치액젓의 화학적 위해요소로서의 바이오제닉 아민 노출 평가)

  • This study was conducted to evaluate the chemical hazards of 74 kinds of commercial anchovy fish sauce in Korea by measuring their biogenic amine content. The biogenic amines detected in commercial anchovy fish sauce were tryptamine, putrescine, cadaverine, tyramine, spermidine, spermine, and histamine. Among them, histamine content was the highest with an average of 1,439.62±812.62 mg/kg. Excessive intake of histamine could result in chemical food poisoning characterized by high blood pressure, headache, and diarrhea. Accordingly, by simulating histamine exposure (repetition=10,000 times) based on the daily intake of 0.969 g of domestic anchovy fish sauce, we found that the estimated average histamine exposure was 24.49±55.48 ㎍/kg bw/day and that the margins of exposure (MOE) value were 139.18 at 1% and 66,141.07 at 95% of 10,000 simulations (9,500 times). As such, it was confirmed that 9,500 out of 10,000 simulations of MOE analysis significantly exceeded MOE 10. Therefore, the risk of exposure to histamine from ingestion of commercial anchovy fish sauce is approximately very low.

Effect of antibacterial substances produced by probiotic lactic acid bacteria on histamine formation in rennet curd (렌넷 커드 내 히스타민 생성에 관한 프로바이오틱 유산균이 생산한 항균 물질의 영향)

  • Purpose of the present study was to investigate the factors affecting the production of antibacterial substances and histamine in rennet curd prepared by inoculation of histamine-producing lactic acid bacteria (LAB) and probiotic LAB. Probiotic Lactobacillus sakei PIL52 and Lactobacillus plantarum FIL20 produced strong antimicrobial agents against histamine-producing bacteria Lactobacillus brevis LAS129, Enterococcus faecium SBP12, and Enterococcus faecalis SBP58. The lactic acid and crude bacteriocin produced from the probiotic LAB inhibited histamine-producing bacteria in a concentration-dependent manner. As the number of probiotic LAB inoculated for the production of rennet curd increased, the antibacterial activity against histamine-producing bacteria was elevated due to the increased amount of lactic acid and crude bacteriocin in the sample. The growth of probiotic LAB as well as histamine-producing bacteria was inhibited by addition of 10% NaCl, thus the antibacterial substances and histamine contents in rennet curd were significantly lower than those of the control (P < 0.05). Meanwhile, the histamine content was not significantly increased when the rennet curd prepared by mixing probiotic LAB and histamine-producing bacteria was stored at $25^{\circ}C$ for 5 days. However, the amount of histamine detected in the rennet curd was significantly (P < 0.05) increased because the antibacterial activity of the bacteriocin produced by the probiotic LAB was decreased at $20^{\circ}C$ for 20 days.

Isolation and Identification of Histamine Degrading Bacteria from Kwamegi (과메기에서 histamine 분해능을 나타내는 세균의 분리 동정)

  • To isolate and identify histamine degrading bacteria from Kwamegi, bacteria were screened with restriction media containing histamine. Ten strains were selected through morphological and biochemical identification procedure followed by comparison with DNA sequence of 16 rRNA gene. And also, these strains were confirmed by the histamine degrading assay such as turbidity and enzymatic assay. The results of identification are as followings : Ewingella americana B791, Arthrobacter sp. R45S, Halomonas marisflava, Psychrobacter sp. 9B-7, Bacillus sp. LMC 21002, Psychrohacter cibarius BC-220, Bacillus megaterium KL-197 were identified showing homology of $99\%,\;95\%,\;98\%,\;99\%,\;99\%,\;99\%\;and\;98\%$, respectively. Three strains remain unidentified. Arthrobacter sp. R45S, H. marisflava, Bacillus sp. LMG 21002, B. megaterium KL-197 showed histamine degrading activity, whereas, Psychrobacter sp. 9B-7 only showed weak activity. Three unidentified strains also have histamine degrading activity. In contrast, E. american B791 and p. cibarius JG-220 did not show any significant activity of histamine degradation. The strains isolated from this study showed relatively fast growth rate and histamine degrading rate as compared to those from salted mackerel.

The Effect of Histamine on the Disappearance Rate of $D_2O$ from the Arterial Blood of Rabbits (토끼 동맥혈로 부터의 $D_2O$ 소실률에 대한 히스타민의 영향)

  • Disappearance rate of injected $D_2O$ from the arterial blood as well as the effect of histamine on the rate were studied in rabbits. The concentrations of $D_2O$ in the serial arterial samples obtained through a Polyethylene tubing inserted into the carotid artery were assayed by the freezing point elevation method of Reaser. At zero time 3 ml of isotonic $D_2O$ in normal saline was injected into the jugular vein and at the same time serial sampling of arterial blood started. The serial sampling interval was either 7.7 sec or 12.3 sec. In the histamine treated animals histamine diphosphate (0,5 mg of histamine base) was injected intravenously 30 minutes prior to the zero time. The following results were obtained. 1. $D_2O$ concentration in arterial plasma water, x, was empirically obtained by the sum of 2 exponential terms of time, $x=Ae^{-k1t}+Be^{-k2t},$ and its theoretical basis was sought. The first term of the right member of the equation was regarded to be attributable to the compartment P which possessed instantaneous exchange of water with plasma. The second term was postulated to represent the poorly exchangeable compartment. 2. The constant A of the equation was evaluated as 4,37% and 14.3% in the control and histamine treated groups, respectively. B was 1.19% in the control and 0.849% in histamine treated animals. 3. The disappearance rates determined were; $k_1=0.0519{\pm}0.0221\;sec^{-1}\;K_2=0.00454{\pm}0.00247\;sec^{-1}$ in the control group. $k_1=0.1137{\pm}0.0290\;sec^{-1}\;K_2=0.00499{\pm}0.00204\;sec^{-1}$ in the histamine group. 4. In the histamine treated animals the disappearance rate of the first term was larger than that of the control animals, suggesting an enlarged size of the rapidly exchangeable compartment with regard to the plasma water. On the other hand the constant B was decreased by histamine administration, suggesting a distribution of $D_2O$ in an enlarged volume. This view was also made clear by comparing the apparent asymptotes to which the concentration curves of $D_2O$ approached in respective groups. The asymptotes in the histamine treated group showed lower values.

  • It has been well known that histamine is presumably the causative material of an outbreak of allergy-like food Poisoning from eating of the meat of dark-fleshed fishes. The present paper was conducted to elucidate the changes in histamine content in the muscle of the dark-fleshed fishes, such as, common mackerel, Scomber japonicus, gizzard-shad, Konosirus punctatus and small sardine, Sardinops melanosticta, under different condition of storage and process. In general, the formation of histamine was markedly different depending upon the kind of fish and the storage temperature. The highest value of the amount of histamine was observed in common mackerel showing thirty folds of gizzard-shad, and gizzard-shad showed the lowest value. Referring to temperature, the formation of histamine at $10^{\circ}C$ was observed more rapid than that of at $25^{\circ}C$. According to the each products, the higher value observed in the sample of sun dried, and those of salted and hot-air dried were similar and boiled-dried of small sardine Was observed the lower value.

Inhibitory Activity of Pigmented Rice Bran Extract to the Allergic Inflammation in Basophilic Cell Line and Peritoneal Mast Cells (호염구세포주와 복강 비만세포에서 유색미 겨 추출물의 알레르기 염증 억제활성)

  • The effects of the extracts from the bran part of pigmented rices on inflammation was evaluated by determining their inhibitory action on the histamine and ${\beta}-hexosaminidase$ release, together with inflammatory cytokine productions ($IL-1{\beta},\;TNF-{\alpha}$ and IL-6). Examination of the inhibitory effects on the histamine and ${\beta}-hexosaminidase$ release from a basophilic cell line RBL-2H3 cells and rat peritoneal mast cells (RPMC) showed that the pigmented rice extract inhibited these inflammation-mediating substances (10.19% and 110.03% inhibition in histamine and ${\beta}-hexosaminidase$ release, respectively), while normal brown rice extract rather increased their release. For RPMC, the pigmented rice extract was found to have 8 or 3-fold stronger inhibitory activity than normal brown rice toward histamine or ${\beta}-hexosaminidase$ release, respectively. Expression of $IL-1{\beta},\;TNF-{\alpha}$ and IL-6 was measured as the representative inflammatory cytokine species showed that the pigmented rice extract had a higher inhibitory activity than the normal rice counterpart. ELISA analysis for determining cytokine release demonstrated a more effective blockading ability of the pigmented rice to the release of $IL-{\beta},\;TNF-{\alpha}$ and IL-6 compared to normal rice. These results showed us the superiority of the pigmented rice bran extract not only in suppressing the release of inflammation-mediating substances such as histamine and ${\beta}-hexosaminidase$, but also in repression of the inflammatory cytokine expression.

The Modulatory Role of Spinally Located Histamine Receptors in the Regulation of the Blood Glucose Level in D-Glucose-Fed Mice

  • The possible roles of spinal histamine receptors in the regulation of the blood glucose level were studied in ICR mice. Mice were intrathecally (i.t.) treated with histamine 1 (H1) receptor agonist (2-pyridylethylamine) or antagonist (cetirizine), histamine 2 (H2) receptor agonist (dimaprit) or antagonist (ranitidine), histamine 3 (H3) receptor agonist (${\alpha}$-methylhistamine) or antagonist (carcinine) and histamine 4 (H4) receptor agonist (VUF 8430) or antagonist (JNJ 7777120), and the blood glucose level was measured at 30, 60 and 120 min after i.t. administration. The i.t. injection with ${\alpha}$-methylhistamine, but not carcinine slightly caused an elevation of the blood glucose level. In addition, histamine H1, H2, and H4 receptor agonists and antagonists did not affect the blood glucose level. In D-glucose-fed model, i.t. pretreatment with cetirizine enhanced the blood glucose level, whereas 2-pyridylethylamine did not affect. The i.t. pretreatment with dimaprit, but not ranitidine, enhanced the blood glucose level in D-glucose-fed model. In addition, ${\alpha}$-methylhistamine, but not carcinine, slightly but significantly enhanced the blood glucose level D-glucose-fed model. Finally, i.t. pretreatment with JNJ 7777120, but not VUF 8430, slightly but significantly increased the blood glucose level. Although histamine receptors themselves located at the spinal cord do not exert any effect on the regulation of the blood glucose level, our results suggest that the activation of spinal histamine H2 receptors and the blockade of spinal histamine H1 or H3 receptors may play modulatory roles for up-regulation and down-regulation, respectively, of the blood glucose level in D-glucose fed model.
