Screening of antioxidant and antimicrobial activities of Caesalpinia bonducella Flem., leaves (Caesalpiniaceae)

  • The study was aimed at evaluating the antioxidant and antimicrobial activities of methanol extract of Caesalpinia bonducella leaves (MECB) (Family: Caesalpiniaceae). The effect of MECB on antioxidant activity, reducing power, free radical scavenging (DPPH radical, nitric oxide radical, superoxide anion radical, hydroxyl radical and hydrogen peroxide radical scavenging), total phenolic content and antimicrobial activities were studied. The antioxidant activity of MECB increased in a dose dependent manner. About 50, 100, 250 and 500 g of MECB showed 53.4, 61.2, 69.1 and 76.2 % inhibition respectively on peroxidation of linoleic acid emulsion. Like antioxidant activity, the effect of MECB on reducing power increased in a dose dependent manner. The free radical scavenging activity of MECB was determined by DPPH radical scavenging method. The potency of this activity was increased with increased amount of extract. MECB was found to inhibit the nitric oxide radicals generated from sodium nitroprusside $(IC_{50}\;=\;102.8\;g/ml)$ whereas the $IC_{50}$ value of curcumin was 20.4 g/ml. Moreover, the MECB was found to scavenge the superoxide generated by photoreduction of Riboflavin. MECB was also found to inhibit the hydroxyl radical generated by Fenton reaction, where the $IC_{50}$ value is 104.17 g/ml compared with catechin 5 g/ml, which indicates the antioxidant activity of MECB. The MECB capable of scavenging hydrogen peroxide in a concentration-dependent manner. The amounts of total phenolic compounds were also determined. Antimicrobial activities of MECB were carried out using disc diffusion methods with five Gram positive, four Gram negative and four fungal species. The results obtained in the present study indicate that MECB leaves are potential source of natural antioxidant and antimicrobial agents.

녹색 및 자색 콜라비 착즙액의 항산화 활성 (Antioxidant Activities of Green and Purple Kohlrabi Juices)

  • 본 연구에서는 녹색 및 자색 콜라비 착즙액의 이화학적 특성(pH 및 $^{\circ}Bx$), 총 페놀함량, 항산화 활성(ORAC 지수, DPPH radical 소거능, ABTS radical 소거능, FRAP 및 reducing power), 아질산염 소거능, 피부 섬유아세포에서의 세포 보호 효과 및 ROS 생성억제 효과를 측정하였다. 총 페놀 함량은 녹색 콜라비 착즙액(153.74 mg GAE/mL)에서 자색 콜라비 착즙액(122.55 mg GAE/mL)보다 더 높게 나타난 반면, ORAC 지수의 경우 자색 콜라비 착즙액($664.32{\mu}MTE/mL$ )에서 녹색 콜라비 착즙액($572.74{\mu}MTE/mL$)보다 더 높게 나타났다. DPPH radical 소거능, ABTS radical 소거능, FRAP 및 reducing power에서는 콜라비 착즙액 10, 50, 100% 농도에서 농도 의존적으로 항산화 활성이 증가하는 경향을 나타내었으며, 같은 농도로 비교하였을 때, 녹색 콜라비 착즙액에서 자색 콜라비 착즙액보다 높은 항산화 활성을 보였다. 아질산염 소거능에서도 녹색 콜라비 착즙액이 자색 콜라비 착즙액보다 높은 효능을 나타내었다. 피부 섬유아세포에서는 녹색 및 자색 콜라비 착즙액을 동결 건조하여 사용하였으며 10, 25, $50{\mu}g/mL$의 농도에서 독성을 나타내지 않았다. Hydrogen peroxide로 산화적 스트레스를 유도한 상태에서 세포 보호 효과를 측정한 결과 $10{\mu}g/mL$의 농도에서는 녹색 및 자색 콜라비 착즙액에서 hydrogen peroxide를 처리한 군과 차이를 나타내지 않았으나, 25, $50{\mu}g/mL$의 농도에서는 농도 유의적으로 세포보효효과가 증가하였다. 또한 녹색 콜라비 착즙액에서는 양성대조군으로 사용한 항산화 물질인 NAC 수준까지 세포생존율이 증가하였다. $H_2$-DCFDA 염색을 통하여 관찰한 ROS 생성 억제 효과는 세포 보호 효과와 유사한 경향으로 관찰되었다. 이상의 결과를 종합하여 볼 때 녹색 및 자색 콜라비 착즙액이 다양한 항산화 모델에서 효능을 나타내었으며, 피부 섬유아세포에서 세포 보호 효과 및 ROS 생성 억제효과가 관찰되어 기능성 식품원료로서의 활용도가 매우 넓을 것으로 판단된다.

머위 분획물의 In Vitro 라디칼 소거능 및 신경세포의 산화적 스트레스 보호 효과 (In Vitro Radical Scavenging Effect and Neuroprotective Activity from Oxidative Stress of Petasites japonicus)

  • This study was focused on the evaluation of radical scavenging effect and the protective activity against oxidative stress of the extract and fractions from Petasites japonicus. P. japonicus was extracted with methanol and then fractionated into 4 fractions [n-butanol, ethyl acetate (EtOAc), methylene chloride, and n-hexane]. The extract and fractions showed strong 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity. Among all the fractions, particularly, the EtOAc fraction showed the strongest effect with the $IC_{50}$ value of $0.02{\mu}g/ml$. In addition, the fractions also showed strong hydroxyl radical scavenging activity and nitric oxide scavenging activity as well. Furthermore, cell viability generated by the P. japonicus extract and 4 fractions were examined under C6 glial cellular model. The C6 glial cells showed high generation of reactive oxygen species (ROS) and decrease in cell viability by the treatment generator of hydrogen peroxide. However, the production of ROS formation was decreased by the treatment of the fractions of P. japonicus and also founded that the EtOAc fraction led to significant increase in the cell viability at concentration $100{\mu}g/ml$. Results from this work indicated that P. japonicus showed protective effects against oxidative stress and its EtOAc fraction may be served as a useful natural antioxidant.

Paraquat 유도독성에 대한 Ginkgo biloba Extract의 독성경감효과(I) (Scavenging Effects of Ginkgo biloba Extract on Paraquat Induced Toxicity)

  • Reactive oxygen species (ROS) are highly reactive molecules due to their unpaired electron. They have been suspected as one of the major tissue damage inducers in biological metabolic systems. Antioxidant enzymes, such as catalase and superoxide dismutase, could not repair all the oxidative damages resulting from those excessive toxic ROS. It is, therefore, urgent to develop effective antioxidants to relieve from the oxidatire damages. In this study antioxidative effects were investigated by using two flavonoids such as quercetin and naringenin and a flavonoid-rich extract, Ginkgo biloba extract in combination with paraquat that is known as a strong generator of oxygen radicals. The results are summeringed as follows: 1. To assess radical scavenging ability reduction concentrations (IC$_{50}$) of 1,1-diphenyl-2-picrylhydrazine (DPPH) within 15 minutes were measured. The values of the IC$_{50}$ of quercetin and Ginkgo biloba extract were 15.4 $\mu$M and 13.2$\mu$g/ml, respectively. Their radical removing activities showed concentration-dependent manners. 2. In the hydrogen peroxide assay by using PMS-NADH system, quercetin, naringenin and Ginkgo biloba extract led to removing hydrogen peroxide in concentrationdependent manner whose removing abilities at 100$\mu$M or 100 $\mu$g/ml were 75.6, 25.8 and 26.0%, respectively. 3. In the hydrogen peroxide-induced rat blood hemolysis assay all three compounds led to similar effects whose hemolysis inhibition ratios at 100$\mu$M or 100$\mu$g/ml were 68.0, 5.14 and 55.8%, respectively. 4. In the xanthinee oxidase assay by measuring degree of NADH oxidation in the presence of hypoxanthine and xanthinee oxidase, both quercetin and Ginkgo biloba extract showed excellent activities showing 42.8 and 24.2% inhibiting xanthine oxidase activity at 100$\mu$M or 100$\mu$g/ml concentrations, respectively.

유기용매에서의 CiP [coprinus cinereus peroxidase]의 안정성 (In the presence of organic solvent stability of CiP [coprinus cinereus peroxidase])

  • CiP를 이용한 유기합성은 주로 유기용매와 완충용액을 섞어서 반응을 한다. 하지만 단순히 유기용매 상에서 효소의 활성도를 측정한 것이 아니라, 과산화수소가 있을 경우에 효소의 활성도를 측정한 결과는 예상과는 다르게 순수한 완충용액에서 보다 유기용매를 포함하고 있는 것이 더 높게 나타났다. 또한 유기용매의 비율을 증가가 될수록 효소의 활성도가 더 높게 나타났으며, 특히 2-propanol이 33%가 포함이 된 경우에는 순수한 완충용액보다 효소의 활성도가 크게 증가가 되는 것을 관찰을 할 수가 있었다. 이러한 효소의 활성도가 유기용매가 포함이 된 조건에서 높게 나는 이유는 유기용매가 CiP에 의해 생성된 반응성이 높은 라디칼을 포집을 하여서, CiP의 활성 부위인 heme의 파괴를 막는 것에 크게 기여하는 것으로 판단이 되었다.

남성생식세포 Sertoli cell에 미치는 복분자(覆盆子)의 항산화 효과 (Antioxidant Effect of Rubi Fructus on TM4 Sertoli Cells)

  • Objectives : The purpose of this study was to examine the antioxidant effects of the extract of Rubi Fructus on TM4 Sertoli cells. Methods : The extract was studied for diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity and cell viability assays on Sertoli cells. In addition, hydrogen peroxide-induced oxidative stress on Sertoli cells were examined by MTT assay. The antioxidant enzyme of Cu/Zn SOD, Mn SOD, catalase protein expression on Sertoli cells were also measured. Results : The results showed that the extract scavenged DPPH radical dose-dependent manner. The extract showed no cytotoxicity at concentration of 1, 5, 10, 50, $100{\mu}g/ml$. The hydrogen peroxide-induced cytotoxicity of Sertoli cells was protected to 88.3% by the extract at concentration of $100{\mu}g/ml$. Cu/Zn SOD and Mn SOD protein expression were significantly increased on Sertoli cells, but catalase protein expression was not significantly changed. Conclusions : In conclusion, the extract of Rubi Fructus has antioxidant effects on Sertoli cells and protect male reproductive system against oxidative stress.

호박분말 효소가수분해물의 항산화활성 (Antioxidant Activities in Enzymatic Hydrolysates of Pumpkin Powder (Cucurbita spp.))

  • 본 연구는 늙은호박, 단호박 및 애호박을 다양한 탄수화물 분해효소로 소화하여 조제한 효소가수분해물의 총 폴리페놀 함량과 항산화 활성을 측정하였다. 조회분, 조단백질 및 조섬유는 애호박이 높았으며, 탄수화물은 단호박이 높았고, 조지방은 서로 비슷하였다. 애호박 분말의 모든 효소처리군의 총 폴리페놀 함량은 늙은호박과 단호박의 가수분해물에 비하여 좀 더 높았다. 애호박 분말의 AMG와 Termamyl 소화로 조제된 효소가수분해물의 DPPH 유리기 소거활성 또한 늙은호박 및 단호박에 비하여 매우 높게 나타났다. 한편, Visozyme 처리 가수분해물을 제외한 늙은호박과 단호박의 대부분 가수분해물은 애호박보다 더 높은 superoxide anion 소거활성을 나타내었다. 애호박 가수분해물(Ultraflo 예외)의 hydrogen peroxide 소거활성은 다른 호박의 효소 분해물 보다 약간 높았으나, hydroxyl radical 소거활성은 모든 호박 품종에서 14% 미만으로 낮았다. Nitric oxide 소거활성은 단호박 및 애호박의 Viscozyme 분해물에서 매우 효과적이었으며, 다른 효소가수분해물의 경우에도 BHT보다는 낮았으나 $\alpha$-tocopherol보다 높은 활성을 나타내었다.

팔물탕의 항산화 효과와 자외선으로 유도된 각질형성세포 손상에 대한 보호효과 (Antioxidant and Protective Effects of Palmul-tang on Ultraviolet B (UVB)-induced Damage in Human Keratinocytes)

  • Objective : In this paper, we investigated the anti-oxidative capacities and protective effects of water extract of palmul-tang (PMT) against Ultraviolet B(UVB)-induced oxidative damage in human keratinocytes(HaCaT). Method : To evaluate the anti-oxidative activities of PMT, we measured scavenging activities on 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation and reducing power of PMT. To give an oxidative stress to HaCaT cells, UVB was irradiated with $40mJ/cm^2$ to HaCaT cells. To detect the protective effects of PMT against UVB, we measured cell viability, apoptotic bodies and reactive oxygen species(ROS) generation. Results : PMT showed the anti-oxidative activities by scavenging DPPH radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation. Also PMT showed high reducing values. The UVB-induced oxidative conditions led to the cell apoptosis. However, treatment with PMT reduced oxidative stress conditions, including inhibition of cell apoptosis and expression of ROS. Conclusion : PMT had anti-oxidative activities and exhibited protective effects against UVB on HaCaT cells. PMT would be useful for the development of cosmetics treating UVB-induced skin aging.

Quantitative Analysis of Compounds in Fermented Insampaedok-san and Their Neuroprotective Activity in HT22 Cells

  • Insampaedok-san is a traditional medicine used for the treatment of colds. We investigated several compounds in Insampeadok-san, and tested their neuroprotective and anti-oxidative activities after fermentation with Lactobacillus. The amounts of four marker compounds (ferulic acid, hesperidin, 6-gingerol and glycyrrhizin) and unidentified compounds in Insampaedok-san (IS) and fermented Insampaedok-san (FIS) were measured and compared by an established HPLC-DAD method. Neuroprotective activity of IS and FIS extracts was evaluated and compared after glutamate-induced neurotoxicity in HT22 cells. Anti-oxidative activity of IS and FIS was also compared in DPPH free radical, hydroxyl radical and hydrogen peroxide scavenging activity assays. Contents of two compounds, ferulic acid and glycyrrhizin were decreased while 6-gingerol was increased by fermentation. FIS showed more potent neuroprotective activity than IS. DPPH, hydroxyl radical and hydrogen peroxide scavenging was slightly increased by FIS when compared to IS. In conclusion, fermentation with Lactobacillus can vary the amounts of the marker compounds in IS and improve neuroprotective and anti-oxidative activities of IS.

  • The ethanol extract of peony root (Paeonia Lactiflora Pall, Paeoniaceae) and its major active components including gallic acid and methyl gallate were evaluated for their protective effects against free radical generation and lipid peroxidation. And protective effects against hydrogen peroxide-induced oxidative DNA damage in a mammalian cell line were performed. The ethanol extract of peony root (PRE), gallic acid and methyl gallate were shown to possess the significant free radical scavenging effect against 1,1-diphenyl-2-picryl hydrazine (DPPH) radical generation and were revealed the inhibitory effect of lipid peroxidation as expressed by malondialdehyde (MDA) formation. They were also found to strongly inhibit hydrogen peroxide-induced DNA damage from NIH/3T3 fibroblasts, assessed by single cell gel electrophoresis. Furthermore, oral administration of 50% PRE (50% ethanol extract), gallic acid and methyl gallate potently inhibited micronucleated reticulocyte (MNRET) formation of mouse peripheral blood induced by KBrO3 treatment in vivo. Therefore, PRE containing gallic acid and methyl gallate may be a useful natural antioxidant by scavenging free radicals, inhibition of lipid peroxidation and protecting oxidative DNA damage.
