  • This study determined plasma glucose, free fatty acid(ffa), epinephrine, norepinephrine, creatine the phosphokinase(CPK), calcium (Ca), magnesium(Mg) and hydroxyproline, Ca and Mg in the heart muscle. Thirty six male Sprague Dawley rats average weighing 110g divided into two diet group : normal Mg group(0.05% Mg), Marginal Mg group(0.005% Mg). After the rats were on experimental diets for 2 weeks, rats for each group were randomly assigned to stress and no stress group and 30 minutes IMMB stress was administered to stress group every day for two weeks. The following were found : 1) Rats fed normal magnesium diet showed decreased plasma glucose and non signficant change in norepinephrine and CPK with IMMB stress, while magnesium deficient rats showed elevated plasma glucose, norepinephrine and CPK with stress. 2) Myocardial hydroxyproline and plasma epinephrine were not significantly different among groups. 3) In the rats fed adequate magnesium, stress increased plasma magnesium while rats on marginal magnesium diet did not show significant change with stress. 4) Plasma and myocardial calcium were increased in rats fed marginal Mg diet, but did not show any significant difference with stress.

  • Collagen is a family of proteins which consists of several genetically distinct molecular species and is intimately involved in tissue organization, function, differentiation and development. The purpose of this study was to investigate the concentration of different hydroxyproline (Hyp) fractions viz., total, free, peptide-bound, protein-bound, soluble- and insoluble-collagen hydroxyproline (Hyp) in various bovine tissues. Results showed that liver had the highest concentration of free Hyp followed by kidney, brain, spleen, lungs, muscle and heart. Liver also had the highest concentration of peptide-bound collagen Hyp followed by kidney, heart, spleen, lungs, brain and muscle. The concentration of protein-bound collagen Hyp was highest in the liver, followed by kidney, spleen, lungs, muscle, brain and heart. Total Hyp was highest in the liver, followed by kidney, spleen, brain, heart, muscle and lungs. Liver also had significantly high concentration of collagen as compared to other tissues examined (P<0.001). Spleen had the significantly higher concentration of soluble-collagen Hyp when compared to other tissues (P<0.001). This was followed by heart, muscle, lungs, brain, kidney and liver. Heart had the highest concentration of insoluble-collagen Hyp followed by lungs, kidney, liver, muscle, spleen and brain. The variation among the insoluble-collagen Hyp concentration of heart and muscle, spleen and brain was significant (P<0.001). We speculate that these differences could be due to the variation in turn over of rate of collagen metabolism in this species.

  • To prevent the denaturation of ficin(EC 3.4.22.3) that is a proteolytic enzyme in fig(Ficus carica L.), fig conserve was heated to 55oC. The fig conserve was added as a tenderizer to native Korean cattle beef(KCB), dairy cattle beef(DCB), castrated dairy cattle beef(CDCB), and imported beef(IB). The composition of free amino acids, hydroxyproline content, shear force, cooking loss, morphological changes and sensory evaluation were then investigated to observe the effect of tenderizing beef with fig conserve. Free amino acids and cooking loss of treated beef were higher than those of control, whereas hydroxyproline and shear force were lower. Glutamine in treated beef decreased relatively but asparagine increased. Hydroxyproline was found, in increasing order of abundance, in DCB, CDCB, IB and KCB. By portion, loin was higher than tenderloin in free amino acids, hydroxyproline and shear force but was lower in cooking loss. Observation with a light microgram revealed a surprising loss of muscle fiber in treated beef. In sensory evaluation of uncooked beef, the control was redder than the treated beef(p<0.01~p<0.05). Treatment with fig conserve increased the juiciness of both cooked CDCB and IB(p<0.001) and decreased their hardness(p<0.01~p<0.001).

  • To study the effect of menopause and dietary protein level on Ca metabolism, ovariectomy (OVAX) and sham operations were performed in 16-week old female rats. Each treatment group was fed for 16 weeks either 5%(L) or 50%(H) casein diets, forming SH, SL, OH, OL groups. High protein groups(SH, OH) showed higher Ca and hydroxyproline excretion in urine. Urinay hydroxyproline was also higher in OVAX, which tells the possibility of increased bone resorption by OVAX and by high dietary protein. At 16th wee, however, urinary Ca and hydroxyproline of SH caught up with OH group, whereas those of OL remained higher than SL. Therefore it seems that high dietary protein overrides the effect of OVAX with time. Urnary protein measured at 8th week was higher in high protein groups, especially in OH. GFR was not differ significantly among groups at 8th week. At 16th week, however, high protein groups showed twice the GFR value of low protein groups. Therefore the increase of urinary Ca and hydroxyproline in SH and OH groups can be explained partly by the increased GFR. The tendency of increased GFR and urinary excretion of protein, Ca, and hydroxyproline was most obvious in OH group. It seems that the effect of high protein diet is likely to accelerated by ovariectomy. The effect of Ovax and dietary protein on the composition of femur, scapular, and lumbar bones, was not pronounced. However, when only the high protein groups were compared, OVAX resulted in the reduction of bone weight, ash and Ca contents, especially in femur. The reason that was no significant effect on bone might be due to the short experimental period to induce that was no significant effect on bone might be due to the short experimental period to induce the changes on bone composition and dietary Ca content used in this experiment may have been high enough to prevent bone loss.

  • The effect of insulin on the collagen metabolism in dental pulp and gingiva was studied using Sprague-Dawley rat and ³H-proline was injected intraperitoneally and then sacrificed according to predetermined schedule. Lower incisor pulp and gingiva were removed immediatedly and hydrolysed with acid. After digestion, proline and hydroxyproline were separated and analysed quantitatively employing thin-layer chromatography and radioactivity of each amino acid was counted by liquid scintillation counter. The label incorporated into lower incisor pulp and gingiva and radioactivity of ³H-hydroxyproline were increased in insulin administered group. And disappearance rate of ³H-hydroxyproline was decreased. Those results indicated that amino acid, I.e. proline transport and collagen biosynthesis were increased in dental pulp and gingiva by insulin administration.

  • Resolution of enantiomers of DNS-amino acids has been achieved by a reversed phase liquid chromatography with an addition of a copper(Ⅱ) complex of N-benzyl-L-hydroxyproline to the mobile phase. N-Benzyl-L-hydroxyproline was prepared and used as a chiral ligand of copper(Ⅱ) chelate for the optical resolution. The pH and the concentration of copper(Ⅱ) chelate, organic solvent, and buffer agent in the mobile phase all affect the optical resolutions of dansyl amino acids. The elution orders between D and L-DNS-amino acids were different depending on the structure of the side chain of the amino acids. The retention mechanism for the chiral separation of the dansyl amino acids can be illustrated by the equilibrium of ligand exchange and by hydrophobic interaction with $C_{18}$ stationary phase. The chiral separation can be illustrated with cis and trans effect of the ligand exchange reaction.

  • Hydroxyproline (HYP) is a post-translational product of proline hydroxylation catalyzed by an enzyme prolyl 4-hydroxylase which plays a crucial role in the synthesis of all collagens, because the 4-hydroxyproline residues are essential for the folding of the newly synthesized collagen polypeptide chains into triple-heical molecules. Considering the role of collagen and its significance in many clinically important diseases such as liver cirrhosis, a great deal of attention has been directed toward the development of an assay at cell-based system.
