Gene Therapy for Mice Sarcoma with Oncolytic Herpes Simplex Virus-1 Lacking the Apoptosis-inhibiting Gene, icp34.5

  • A mutant herpes simplex virus 1, mtHSV, was constructed by inserting the E. coli beta-galactosidase gene into the loci of icp34.5, the apoptosis-inhibiting gene of HSV. The mtHSV replicated in and lysed U251 (human glioma cells), EJ (human bladder cells), and S-180 (mice sarcoma cells), but not Wish (human amnion cells) cells. With its intact tk (thymidine kinase) gene, mtHSV exhibited susceptibility to acyclovir (ACV), which provided an approach to control viral replication. An in vivo test with mtHSV was conducted in immune-competent mice bearing sarcoma S-180 tumors, which were treated with a single intratumoral injection of mtHSV or PBS. Tumor dimensions then were measured at serial time points, and the tumor volumes were calculated. Sarcoma growth was significantly inhibited with prolonged time and reduced tumor volume. There was microscopic evidence of necrosis of tumors in treated mice, whereas no damage was found in other organs. Immunohistochemical staining revealed that virus replication was exclusively confined to the treated tumor cells. HSV-1 DNA was detected in tumors, but not in the other organs by a polymerase chain reaction analysis. From these experiments, we concluded that mtHSV should be a safe and promising oncolytic agent for cancer treatment.

A study on the Determination of Trace Se and Bi in the Scalp Hair by Hydride Generation- Inductively Coupled Plasma Atomic Emission Spectrometry (수소화물발생 유도결합플라즈마 원자방출분광법에 의한 머리카락 시료 중 미량의 Se와 Bi의 분석에 관한 연구)

  • A method to determine the trace amount of Se and Bi in the scalp hair using the hydride generation inductively coupled plasma atomic emmission spectrometry was studied. The optimum operation conditions of ICP for hydride generation are 0.6~0.8L/min for the carrier gas flow rate, and 6mm above the induction coil for the observation height. Hydrochloric acid concentrations for the optimum hydride generation conditions were greater than 1.5M when 2.5% $NaBH_4$ and NaOH were used, and greater than 0.5M when 2.5% $NaBH_4$ and 0.1% NaOH were used. Severe interference effects are observed from transition metals such as Cu and Ni, and they could be circumvented by the coprecipitation with lanthanum hydroxide.

Method and proficiency test for analysis of toxic metals in livestock products by ICP/MS (ICP/MS를 이용한 축산물의 유해중금속 분석방법 및 숙련도 검증)

  • The analytical method of trace toxic metals in livestock products was confirmed and validated through certified reference material (CRM) and the international proficiency tests. There are some difficulties to determine low levels of toxic metals in livestock products because of interferences due to the matrix. The recoveries of CRM (NIST 1577c) ranged from 73.9 to 119% for lead and from 86.4 to 111% for cadmium in bovine liver. The international proficiency tests were carried out with the milk powder and cocoa powder samples including metals provided by Food Analysis Performance Assessment Scheme (FAPAS$^{(R)}$, UK). The test samples were prepared by microwave digestion using solution of $HNO_3:H_2O_2:H_2O$ (v/v/v = 5 : 2 : 4) and analyzed by ICP/MS. The analytical result of cadmium in milk powder was $121{\mu}g/kg$ with -0.3 of the z-score compared to the assigned value of $131{\mu}g/kg$ by FAPAS$^{(R)}$. The analytical results of lead and cadmium in cocoa powder were $29.2{\mu}g/kg$ and $97.6{\mu}g/kg$, respectively, which satisfied the assigned values of $34.2{\mu}g/kg$ for lead and $126{\mu}g/kg$ for cadmium by FAPAS$^{(R)}$. It is verified that the analytical method is accurate and reliable to determine trace lead and cadmium in livestock products by microwave digestion and ICP/MS.

  • This study has been focused on the influence of Fe(III) incorporation into framework of SAPO-34(FeAPSO-34s;/su/Fe = 40,20 and 5) on the catalytic performance of methanol conversion. By rapid crystallization method, the XRD,SEM,ICP,TG-DTA, and BET sufrace areas. With an increase in the Fe content incorporated to the framework, the crystallinity identified from the intensity of XRD peaks slightly decreased and the particle size observed from SEM photographs decreased also. On the other hand, the acid sites in crystal decreased kin the Fe-incorporated samples, and the selectivity to ethylene for FeAPSO-34 catalyst on methanol conversion was enhanced compared with the nonmetal incorporated(SAPO-34)

Distribution Characteristics of Hazardous Heavy Metals in Ginseng and Wood-cultivated Ginseng (인삼 및 산양삼의 부위별 유해중금속 분포 특성)

  • The present study was carried out to identify the distribution characteristics of heavy metals in ginseng and wood-cultivated ginseng reduction of dietary exposure. Samples of ginseng and wood-cultivated ginseng were collected from 14 and 5 regions across Korea, respectively. Lead (Pb), cadmium (Cd) and arsenic (Ar) were detected by ICP-MS after microwave digestion, whereas aluminum (Al) was determined using ICP. Cultivated ginseng peels were 16.2% of whole root, while the peels and fine roots amounted to 21.8% and 16.8% of whole wood-cultivated ginseng, respectively. Taking into account the weight and concentration of the heavy metals by root part, their distribution ratios were calculated and compared. The cultivated ginseng peels contained 40.3% Pb, 25.9% Cd, 47.6% As, and 89.9% Al. Meanwhile, heavy metals consisting of 27.2% Pb, 28.2% Cd, 48.3% As, and 56.8% Al were distributed in the peels of the wood-cultivated ginseng. There was no significant difference between the peels and fine roots of the wood-cultivated ginseng with respect to the distribution proportion of heavy metals, except for Al. These results indicate that the level of dietary exposure to heavy metals could be reduced by peeling ginseng and wood-cultivated ginseng prior to consumption.

Investigation of Acids on the Germanium Analysis by HG-ICP-AES (HG-ICP-AES를 이용한 Germanium 분석에 있어서 보조산에 대한 연구)

  • The present work is aimed to evaluate the conditions of the hydride generation (HG) for germanium analysis by inductively coupled plasma (ICP)-atomic emission spectrometry (AES). Twelve different kinds of acids were used such as phosphoric, hydrochloric, nitric, sulfuric, perchloric, boric, tartaric, malic, oxalic, tannic, citric, and acetic acid. It was found that phosphoric acid yielded the maximum efficiency of hydride generation. Also, efficient hydride generation was obtined with the buffer solutions containing phosphate ions over a wide range of pH. In addition, in the presence of phosphoric acid the interference caused by metals was suppressed in the hydride generation of germanium. As the concentrations of a reducing agent and a stabilizing increased the hydride generation efficiency and the acid concentration proviaing the maximum intensity were increased. By using an analytical method developed in this study, the contents of germanium in water and rock samples were determined. The detection limit of germanium in the presence of phosphoric acid was $0.03{\mu}g/L$.

Relationship between Cerebral Arteriovenous Oxygen Difference and Development of Delayed Cerebral Infarction in Patients with Severe Head Injury (중증 뇌손상 환자에서 뇌동정맥 산소함유량차이와 지연성 뇌경색 발생과의 관계)

  • Objective : This study was performed to evaluate the relationships among intracranial pressure(ICP), cerebral perfusion pressure(CPP), and cerebral arteriovenous oxygen difference($AVDO_2$) which were used as parameters of adequacy of cerebral blood flow to support cerebral metabolism after severe head injury and also to examine the association between delayed cerebral infarction and outcome. Material and Method : The authors studied the ICP, CPP and $AVDO_2$ before and after treatment on 34 head-injured patients from June 1996 to December 1997 and examined the association with the change of an ICP, CPP and $AVDO_2$ following treatment and the development of delayed cerebral infarction. Sixteen patients underwent craniotomy for hematoma evacuation and eighteen patients received mannitol to decrease ICP. Results : The development of delayed cerebral infarction was demonstrated in 3(42.9%) out of 7 patients in no improvement group and 13(48.1%) out of 27 patients in improvement group with an increased ICP following treatment. Also, the development of delayed cerebral infarction was demonstrated in 8(50%) out of 16 patients in no improvement group and 8(44.4%) out of 18 patients in improvement group with a decreased CPP following treatment. The association with changes of ICP and CPP following treatment and development of delayed cerebral infarction was not statistically significant(p>0.01). However, 11(78.6%) out of 14 patients who demonstrated an increase in $AVDO_2$ and 5(25%) out of 20 patients who demonstrated a decrease in $AVDO_2$ following treatment developed delayed cerebral infarction. No improvement(reduction) in $AVDO_2$ following treatment was significantly associated with the development of delayed cerebral infarction(p<0.01). All of 16 patients with delayed cerebral infarction showed poor prognosis. Conlcusion : The change of $AVDO_2$ rather than those of ICP and CPP was considered more important factor for the development of the delayed cerebral infarction and poor outcome.

  • An ion-association chromatography was applied for the separation and determination of individual rare earth elements (REE) contained in mineral monazite. Prior to the determination, the group separation of REE was achieved by a cation exchange column of Dowex 5OW-X8 resin. The quantitative recovery of REE by the resin column, free from coexisting elements in monazite, was confirmed with radioactive tracers as well as with ICP-MS. Individual REE at ppm level was separated on reversed-phase column ($\mu$-Bondapak $C_{18}$) using gradient elution from 0.05 to 0.3 M $\alpha$-hydroxyisobutyric acid at pH 4.6. The individual REE was detected at 546 nm following post-column reaction with PAR (4-(2-pyridylazo)-resorcinol monosodium salt).

  • Mineral and volatile flavor compounds of Pimpinella brochycarpa N., a perennial Korean medicinal plant of the Umbelliferae family, were analyzed by inductively coupled plasma-atomic emission spectroscopy (ICP-AES) and simultaneous steam distillation extract (SDE)-gas chromatography mass spectrometry (GC/MS), head space solid phase micro-extraction (HS-SPME)-GC/MS. Mineral contents of the stalks and leaves were compared and the flavor patterns of the fresh and the shady air-dried samples were obtained by the electronic nose (EN) with 6 metal oxide sensors. Principal component analysis (PCA) was carried out using the data obtained from EN. The 1st principal values of the fresh samples have + values and the shady air-dried have - values. The essential oil extracted from the fresh and the shady air-dried by SDE method contain 58 and 31 flavor compounds. When HS-SPME method with CAR/PDMS fiber and PDMS fiber were used, 34 and 21 flavor compounds. The principal volatile components of Pimpinella brachycarpa N. were ${\alpha}$-selinene, germacrene D, and myrcene.

A Study on the Cation Extraction and Separation in Cement Industrial By-products for Applications to the Carbonation Process (탄산화 공정 적용을 위한 시멘트 산업부산물 내 양이온 추출 및 분리 연구)

  • A cement industrial by-product was used as a Ca source for the carbonation process. It was confirmed that the most of cement industrial by-products was composed of CaO and KCl through ICP and XRD analyses. The optimal extractant type and concentration was 1.5 M of hydrochloric acid, and the solid/liquid ratio was 0.1 g/mL. It was assumed that the cation extraction efficiency was dependent of the pair ions and their binding formation and also the solubility from extraction efficiency results by varing extractants. After extraction process, it was also confirmed that the cation could be selectively separated from the solution with respect to the kind of additives and the injection order. When NaOH was injected into the solution to control pH values ranging from 9.5 and 13, impurities and $Ca(OH)_2$ were precipitated, whereas the separated K ion was precipitated in the form of KCl under the injected $C_2H_5OH$.
