Factors Affecting the Inosine Monophosphate Content of Muscles in Taihe Silkies Chickens

  • In this paper, changes in the concentration of inosine monophosphate (IMP) of muscles in Taihe Silkies chickens from 2 to 28 weeks of age were studied. The results showed that: (1) with increasing age, IMP content of muscles decreased continuously. (2) the relationship between body weight and IMP content of musculus pectoralis major was significantly negative (p<0.01), so was body weight and IMP content of musculus peronaeus, and their coefficients of correlation were -0.45, -0.38 respectively; the relationship between IMP contents of musculus pectoralis major and that of musculus peronaeus was more significantly positive (p<0.01), and its coefficient of correlation was 0.59. (3) by multiple regression analysis, changes of IMP content of muscles depended on the weeks of age and body weight. (4) from 2 to 28 weeks of age, the heritabilities of IMP content of musculus pectoralis major were calculated between 0.33-0.48, and those of musculus peronaeus were calculated between 0.51-0.69.

  • 1. 분화과정에 따른 균체로부터 추출한 총 RNA량은 포자형성 직전에 증가함을 알았다. 2. 추출한 RNA를 가수분해하여 P.E.I. Cellulose TLC를 전개한 결과 AMP유도체와 IMP가 존재함을 확인하였다. 3. 포자형성을 전후하여 IMP와 AMP의 RNA에 대한 함량비율은 일정함을 알았다. 4. 포자형성 전기의 균체에 아데노신, 구아노신, 이노신, 크산토신을 처리한바 포자형성이 10배 이상 촉진되었다. 5. 포자형성 전기의 균체에 A. bisporus, F. velutipes, L. edodes의 열수추출액을 처리한바 포자형성이 $1.5{\sim}2$배 촉진되었다.

Association of single-nucleotide polymorphisms in dual specificity phosphatase 8 and insulin-like growth factor 2 genes with inosine-5'-monophosphate, inosine, and hypoxanthine contents in chickens

  • Objective: This study aimed to identify the single-nucleotide polymorphisms (SNPs) in the dual-specificity phosphatase 8 (DUSP8) and insulin-like growth factor 2 (IGF2) genes and to explore their effects on inosine-5'-monophosphate (IMP), inosine, and hypoxanthine contents in Korean native chicken -red-brown line (KNC-R Line). Methods: A total sample of 284 (males, n = 127; females n = 157) and 230 (males, n = 106; females, n = 124) aged of 10 weeks old KNC-R line was used for genotyping of DUSP8 and IGF2 genes, respectively. One SNP (rs313443014 C>T) in DUSP8 gene and two SNPs (rs315806609A/G and rs313810945T/C) in IGF2 gene were used for genotyping by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and KASP methods, respectively. The Two-way analysis of variance of the R program was used to associate DUSP8 and IGF2 genotypes with nucleotide contents in KNC-R chickens. Results: The DUSP8 (rs313443014 C>T) was polymorphic in KNC-R line and showed three genotypes: CC, CT, and TT. The IGF2 gene (rs315806609A/G and rs313810945T/C) was also polymorphic and had three genotypes per SNP, including GG, AG, and AA for the SNP rs315806609A/G and genotypes: CC, CT, and TT for the SNP rs313810945T/C. Association resulted into a strong significant association (p<0.01) with IMP, inosine, and hypoxanthine. Moreover, the significant effect of sex (p<0.05) on nucleotide content was also observed. Conclusion: The SNPs in the DUSP8 and IGF2 genes might be used as genetic markers in the selection and production of chickens with highly flavored meat.

  • Adenine-guanine 동시영양요구성인 Brevibacterium ammoniagenes D-21530을 사용하여 5'-IMP을 생산할 경우, 5'-IMP생성에는 탄소원으로 fructose가 glucose보다 효율적이었다. 반면에 균 생육에는 glucose를 탄소원으로 사용하는 것이 좋았다. Glucose와 fructose를 혼합하여 사용할 때 균 생육을 적절히 조절하여 5'-IMP축적을 증가시켜 줄 수 있었으며 fructose와 glucose의 혼합당에 있는 fructose의 함유율을 20∼40%범위로 하였을 때 5'-IMP생성이 최대가 되었고 fructose 단독으로 사용할 때보다 약 40% 증가되었다. 또한 영양요구성물질인 adenine과 guanine의 첨가농도가 증가함에 따라 균생육이 촉진되나 150㎎/l 이상에서는 정상상태였고, 5'-IMP생성은 50㎎/l 까지는 첨가 농도에 따라 급격히 증가하나 그 이상에서는 감소하였다.

Effect of Sea Buckthorn Leaves on Inosine Monophosphate and Adenylosuccinatelyase Gene Expression in Broilers during Heat Stress

  • The trial was conducted to evaluate the effects of sea buckthorn leaves (SBL) on meat flavor in broilers during heat stress. A total 360 one-day-old Arbor Acre (AA) broilers (male) were randomly allotted to 4 treatments with 6 replicates pens pretreatment and 15 birds per pen. The control group was fed a basal diet, the experimental group I, II and III were fed the basal diet supplemented with 0.25%, 0.5%, 1% SBL, respectively. During the 4th week, broilers were exposed to heat stress conditions ($36{\pm}2^{\circ}C$), after which, muscle and liver samples were collected. High performance liquid chromatography (HPLC) was performed to measure the content of inosine monophosphate (IMP); Real-Time PCR was performed to determine the expression of the ADSL gene. The results showed that the content of breast muscle IMP of group I, II and III was significantly increased 68%, 102% and 103% (p<0.01) compared with the control, respectively; the content of thigh muscle IMP of group II and III was significantly increased 56% and 58% (p<0.01), respectively. Additionally, ADSL mRNA expression in group I, II and III was increased significantly 80%, 65% and 49% (p<0.01) compared with the control, respectively. The content of IMP and expression of ADSL mRNA were increased by basal diet supplemented with SBL, therefore, the decrease of meat flavor caused by heat stress was relieved.

Underlying mechanisms of phosphodiesterase 10A and glutamate-ammonia ligase genes that regulate inosine monophosphate deposition and thereby affect muscle tenderness in Jingyuan chickens

  • Objective: Inosine monophosphate (IMP) is a key factor that imparts of meat flavor. Differences in the IMP content in the muscles were evaluated to improve chicken meat quality. Methods: For this study, the IMP content was detected by high performance liquid chromatography. The gene expression profiles of Jingyuan chickens with different feeding patterns and different sexes were analyzed by RNA-sequencing (RNA-seq). Results: Breast muscle IMP content in free-range chickens was extremely significantly higher than that of caged chickens (p<0.01). Breast muscle IMP content in hens was also higher than that of cocks, but the difference was not significant. Correlation analysis showed that the breast muscle IMP content in caged hens and cocks was negatively correlated with the shear force, and the breast muscle IMP content in free-range hens was significantly negatively correlated with the shear force (p<0.05). The two key genes associated with IMP synthesis in chickens with different feeding patterns were glutamate-ammonia ligase (GLUL) and phosphodiesterase 10A (PDE10A). Bioinformatics analysis revealed that the GLUL and PDE10A genes are involved in glutamine biosynthesis and purine salvage pathways respectively. In addition, GLUL expression was positively correlated with the IMP content in caged and free-range chickens, and PDE10A expression was significantly positively correlated with the IMP content in caged and free-range chickens (p<0.05). Conclusion: These findings will facilitate the comprehension of the deposition of IMP in the muscles and thereby aid the process of selection and breeding of good quality local chickens.

전통 장류를 이용한 소스제품에 접종된 Bacillus cereus spore의 발아에 미치는 온도 및 inosine monophosphate 첨가 효과 (Effect of Temperature and Inosine Monophosphate on Spore Germination of Artificially Inoculated Bacillus cereus in Traditional Korean Fermented Sauces)

  • 장류를 이용한 소스 3종류(간장, 된장 및 고추장소스)에 대해 Bacillus cereus spore를 인위적으로 접종하고 $40^{\circ}C$ 혹은 $80^{\circ}C$ 처리구 및 $40^{\circ}C$ 혹은 $80^{\circ}C$에 10 mM IMP 첨가 처리구로 나누어 포자의 발아를 유도한 다음 이들을 다시 $80^{\circ}C$로 처리하는 방법을 통해 발아효과를 측정하였다. 간장소스의 경우 $40^{\circ}C$$80^{\circ}C$ 처리에서 모두 0.8 log CFU/g 수준의 발아효과를 보였고 여기에 10 mM IMP를 첨가 시 $40^{\circ}C$에서는 2.25 log CFU/g, $80^{\circ}C$ 처리에서는 1.21 log CFU/g의 발아수준을 보여 처리구 간에 유의적인 차이를 나타내었다. 한편 된장소스는 $40^{\circ}C$, 10 mM IMP 및 $80^{\circ}C$, 10 mM IMP 처리 시 대조구 대비 각각 0.46 log CFU/g 및 0.85 log CFU/g로서 간장소스의 경우보다 낮은 발아수준을 나타내었고 고추장 소스의 경우에는 발아효과가 거의 나타나지 않았다. 상기 결과를 통해 열과 IMP 처리는 간장소스 내 B. cereus spore의 발아 유도에 응용이 가능하며 된장소스 및 고추장 소스에서 발아효율을 높이기 위해서는 pH 등 다양한 환경인자를 고려한 연구가 필요할 것으로 생각된다.

영지(靈芝)의 모노뉴크레오티드 성분의 분포에 관한 연구 (Studies on Distribution of the Mononucleotides in Ganoderma lucidum)

  • Ganoderma lucidum (Fr.) Karsten을 실험재료로 하여 자실체형성 전후의 RNA를 추출 정량하였다. 버섯의 자실체 형성 전후에 걸쳐 총 RNA함량에 차이가 있었으며 자실체 형성 전에 총 RNA함량이 두드러지게 많았다. 버섯의 추출물인 RNA를 알카리가수분해한 것을 P.E.I. cellulose TLC로 전개한 결과 GMP와 XMP가 확인되었으며 미확인물질로서 한개의 spot가 나타났다. 이것은 위치로 보아 CMP가 아닌가 생각된다. 영지의 RNA추출물을 HPLC로 분석한 결과는 XMP와 GMP가 확인되었으며 그 함량은 각각, 자실체 형성 전의 GMP는 7.l9mg%, XMP는 8.43mg%, 형성후의 GMP는 4.02mg%, XMP는 3.67mg%의 함량분포를 나타내었다.

전통 장류로 제조한 양념육의 숙성 중 맛성분 변화 (Changes in Taste Compounds of Seasoned Pork with Korean Traditional Sauces during Aging)

  • 한국 전통 장류를 이용한 양념 돈육의 맛성분에 관한 연구로 돼지고기의 등심을 채취하여 $5{\time}15{\time}5$cm 크기로 자른 후 육을 동일한 비율의 양념액에 침지하여 $1{\pm}1^{\circ}C$에서 28일간 숙성하면서 양념육(간장소스 양념육, T1; 고추장소스 양념육, T2; 된장소스 양념육, T3)의 품질 특성을 조사하였다. Citric acid와 fumaric acid는 숙성 14일 높게 나타났으며, 고추장소스로 양념한 돈육이 다른 처리구에 비해 높은 함량을 나타내었다. Propionic acid와 lactic acid는 다른 처리구에 비해 된장소스로 양념한 돈육에서 높게 나타났다. 유리 아미노산은 간장소스로 양념한 돈육에 비해 된장과 고추장소스로 양념한 돈육에서 높게 나타났으며, 숙성기간에 따라 모든 처리구에서 약간 증가하였다. 숙성기간동안 IMP 는 감소한 반면, inosine은 증가하였으며, inosine은 간장소스로 양념한 돈육에 비해 된장과 고추장소스로 양념한 돈육에서 높게 나타났으며, 숙성기간에 따라 모든 처리구에서 증가하였다. 관능검사는 숙성기간의 증가에 따라 향과 맛은 물론 연도와 다즙성도 증가되어 기호성도 숙성 초기에 비해 증가되었다. 이상의 결과를 요약해 보면 기존의 양념육은 간장소스가 대부분이었으나 고추장소스나 된장소스를 활용한다면 맛과 품질이 우수한 다양한 양념육 생산이 가능할 것으로 사료된다.

New Retention Mechanism of Mononucleotides with Buffer Concentrations in Ion-suppressing RP-HPLC

  • HPLC separation of ionic samples tends to be more complicated and difficult to understand than that of non-ionic compounds. On the other hand, band spacing is much more easily manipulated for ionic than for neutral samples. Ion-suppression RP-HPLC method was used with organic modifier and aqueous buffer solution. In this work, five mononucleotides of cytidine-5-monophosphate (5-CMP) disodium salt, uridine-5-monophosphate disodium salt (5-UMP), guanosine-5-monophosphate disodium salt (5-GMP), inosine-5-monophosphate disodium salt (5-IMP), and adenosine-5-monophosphate disodium salt (5-AMP) were examined. Acetic acid and sodium phosphate were used as buffers, and methanol as an organic modifier. A new relationship between the retention factor and the buffer concentration at a fixed modifier content (5% of methanol) could be expressed by following: K = (k(sub)-1 + k(sub)0 (k(sub)B/k(sub)S)/(1 + (k(sub)B/k(sub)S) C(sub)B(sup)a), where C(sub)B was the concentration of buffer. Using this relationship, the calculated values closely matched the experimental data. The derived relationship showed that as the buffer concentration increased, the retention factor approached a certain value, and this was buffer dependent.
