Hypoglycemic and antioxidant effects of jaceosidin in streptozotocin-induced diabetic mice (Jaceosidin이 streptozotocin으로 유도된 당뇨 쥐의 혈당강하 및 항산화능에 미치는 영향)

  • Purpose: In this study, we investigated the effects of jaceosidin on blood glucose regulation in type 1 diabetic mice. Methods: C57BL/6 mice were divided into four groups; normal control (Normal), diabetes control (D-Control), diabetes low-jaceosidin (D-0.005%), and diabetes high-jaceosidin (D-0.02%). Type 1 diabetes was induced by streptozotocin and mice were then fed a diet containing jaceosidin for eight weeks. Fasting blood glucose, oral glucose tolerance test, insulin tolerance test, lipid peroxidation, and antioxidant enzyme activities were assessed. Results: Jaceosidin supplementation for eight weeks had no effect on body weight, organ weight, and blood lipid profiles. However, jaceosidin supplementation significantly lowered fasting blood glucose level and reduced insulin resistance. We also found that jaceosidin supplementation increased antioxidant capacity by enhancement of catalase and GSH-px activities. Conclusion: These results suggest that jaceosidin could be a therapeutic candidate to ameliorate hyperglycemia through increase of antioxidant enzyme activity.

Quantitative Analysis of Eupatilin and Jaceosidin in Artemisia herba (약쑥 함유 Eupatilin과 Jaceosidin 성분의 분석)

  • Artemisia Herb is the dry aboveground part of Artemisia asiatica (Compositae). This is listed officially in the Korean Herbal Pharmacopoeia and used mainly in stopping all kinds of bleeding, regulating menses and curing menstrual disorders as well as in alleviating pain. Eupatilin and jaceosidin were isolated from Sazabalsuk and Sajuarisuk. and identified by means of spectral methods $(UV,\;^1H\;and\;^{13}C-NMR,\;MS) $ and quantitative analysis using HPLC. Regression equations for two flavonoid component standards obtained from Artemisia asiatica were $y=76069_x\;+\;107329\;(r=0.9999^{**})$ for eupatilin, $y=76651_x\;+\;77399\;(r=0.9996^{**})$ for jaceosidin. The eupatilin content of Sazabalsuk was 240mg, jaceosidin was 10mg in 100 g dry leaves. The Sajuarisuk were 330mg, 123mg in 100g dry leaves, respectively.

Variation of Eupatilin and Jaceosidin Content of Mugwort (약쑥의 Eupatilin과 Jaceosidin 함량변이)

  • This study was carried out to select the high eupatilin and jaceosidin containing species and suppose the crop value of Mugwort (Artemisia sp.) investigating plant characteristics. Among the twelve collected mugworts, only one's of Ganghwa and Baekryeongdo contained the eupatilin and jaceosidin and the highest eupatilin and jaceosidin contents weye obtained from the Ssajuarissuk collected in Ganghwa. The result of seasonal variation in eupatilin content indicated the highest contents from May 23 to June 10. Eupatilin content of leaves in upper was higher than in lower leaves and content of middle leaves was not higher than in upper leaves. And the highest eupatilin content found in the leaves of the upper part of the plant, almost 2 times higher than those of the lower of the plant.

  • 동의보감에 수재되어 있는 약쑥은 식용뿐만이 아니고 한방에서는 출혈 및 지혈약으로 자궁출혈, 임신중의 출혈, 코피지혈 등에 효과가 크다고 알려져 왔다. 또한 부인병과 건위, 설사치료와 항암활성, HeLa cell에 대한 억제작용도 보고되어 있다. MeOH엑스가 HCl-EtOH유발법, $1\%$ 암모니아 수용액 유발법, HCl-aspirin 유발법등에 의하여 유발된 위 손상에 대하여 강한 억제 작용을 나타내고 있는데 이는 flavonoid의 일종인 eupatilin으로 확인된 바 있다. 이 물질이 5-lipoxygenase inhibitor ($ID_{50}=14{\times}16^{-6}M$) 작용과 prosta glandin synthase의 활성을 약간 억제한다고 보고되었다. 약쑥의 eupatilin과 jaceosidin을 분리하여 그 화학구조를 구명하였다. 이를 지표물질로 하여 HPLC 분석조건과 HPLC용 검체의 조제, 표준검량선을 작성하였다. 강화사자발쑥 100 g의 잎에는 eupatilin은 240 mg, jaceosidin은 10 mg함유되어 있고 싸주아리쑥에는 eupatilin은 330 mg, jaceosidin은 123 mg 함유되어 있음이 확인되었다.

Quality characteristics of fermented vinegar prepared with Seomaeyaksuk (Artemisia argyi H.) extract (섬애약쑥 주정 추출물로 제조한 발효식초의 품질특성)

  • This study was carried out to investigate the quality characteristics of vinegars containing jaceosidin and eupatilin using Artemisia argyi H. ethanol extract (AEE). 10% malt extract (ME) and water extract of Artemisia argyi H. (AWE) were also prepared for vinegar production. Three kinds of materials were mixed in the same amount to prepare vinegar as follows; CO (ME, water, 18% edible ethanol), SE (ME, water, and AEE), SW (ME, AWE, and 18% edible ethanol) and SM (ME, AWE, AEE). All samples were fermented by Acetobacter pasteurianus A8 at $30^{\circ}C$ for 25 days and analyzed at 10, 15, 20 and 25 days. The pH decreased significantly during the fermentation. pH was lower in SE and SM than CO and SW. The acidity increased significantly during the fermentation, and was highest in SM (4.44%) at 25 days of fermentation. The concentration of acetic acid was higher than other organic acids for all vinegars. Jaceosidin and eupatilin were not detected in both CO and SW, but both were detected in the SE and SM. At 25 days of fermentation, jaceosidin and eupatilin concentrations in SE and SM were 6.49-6.88 mg/kg and 2.23-2.24 mg/kg, respectively. From these results, we confirmed that production of vinegar containing jaceosidin, eupatilin and phenolic compounds can be prepared by using Artemisia argyi H. edible ethanol extract.

Chemical Component of Mugwort (Artemisia princeps Pampanini) leaves Collected in Korea (수집약쑥의 유효성분 함량변이)

  • Mugwort, Artemisia princeps Pampanini, has been used as natural medicine traditionally in the far east countries including Korea. This study was performed with the object of provoding basic data an leaf quality improvement in mugwort. The 210 mugwort were collected in 48 locations from 2004 to 2006, leaves of 210 genotypes collected were analyzed for eupatilin, jaceosidin and sesamin content. The eupatilin content was averaged to 43.8mg (100 g leaves) and ranged from 0 to 228.0 mg. The average jaceosidin and sesamin contents were 12.8 mg and 3.58 mg and the ranges of those were 0 to 73.8 mg and 0 to 6.56 mg, respectively. The eupatilin content of mugwort harvested in the western region was higher compared with those harvested in the resemble altitude of the southern and eastern of Korea.

Anticoagulant and Antiplatelet Activities of Artemisia princeps Pampanini and Its Bioactive Components

  • Artemisia princeps Pampanini (AP) has been used as a traditional medicine in Korea, China and Japan and reported to exhibit various beneficial biological effects including anti-inflammatory, antioxidant, anti-atherogenic and lipid lowering activities; however, its antiplatelet and anticoagulant properties have not been studied. In the present study, we evaluated the effects of an ethanol extract of Artemisia princeps Pampanini (EAP) and its major flavonoids, eupatilin and jaceosidin, on platelet aggregation and coagulation. To determine the antiplatelet activity, arachidonic acid (AA)-, collagen- and ADP (adenosine diphosphate)-induced platelet aggregation were examined along with serotonin and thromboxane A2 ($TXA_2$) generation in vitro. The anticoagulant activity was determined by monitoring the activated partial thromboplastin time (aPTT) and prothrombin time (PT) in vitro. The data showed that EAP and its major flavonoids, eupatilin and jaceosidin, significantly reduced AA-induced platelet aggregation and the generation of serotonin and $TXA_2$, although no significant change in platelet aggregation induced by collagen and ADP was observed. Moreover, EAP significantly prolonged the PT and aPTT. The PT and/or aPTT were significantly increased in the presence of eupatilin and jaceosidin. Thus, these results suggest that EAP may have the potential to prevent or improve thrombosis by inhibiting platelet activation and blood coagulation.

  • In order to search for biologically active compounds from edible plant sources, the aerial parts of Sajabalssuk (Artemisia herba) were extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH and $H_2O$, successively. From the EtOAc fraction, four compounds were isolated through the repeated silica gel and ODS column chromatographies. From the results of physico-chemical data including NMR, MS and IR, the chemical structures of the compounds were determined as eupatilin (1), jaceosidin (2), apigenin (3) and eupafolin (4). Among them, compounds 3 and 4 were isolated for the first time from Sajabalssuk (Artemisia herba).
