Antimutagenic Effect and Active Compound Analysis of Kale Juice in Salmonella Assay System (Salmonella 실험계에서 케일즙액의 항돌연변이의 효과와 활성획분)

  • Antimutagenic effects of kale juice on the mutagenicity induced by $B_{1}(AFB_{1})$ N-methyl-N'-N-nitrosoguanidine(MNNG) in Salmonella assay system were studied. The freeze dried kale juice significantly reduced the mutagenicity induced by $AFB_{1}$ in Salmonella typhimurium TA100 and TA98. However, the kale juice exhibited less inhigbitory effect on the mutagenicity induced by MNNG as the concentrations of the juice sample increased. Also, kale juice after dialysis (>12,000, Mw) appeared to have 42.3∼89.5% of inhibitory effects against $AFB_{1}$, however, the dialyzate did not show any inhibitory effect against MNNG. To separate and identify the antimutagenic compounds from the kale juice, the dialyzates were further fractioned by using Sepharose CL-6B-200 gel filtration. Fraction number 13 showed the strong antimutagenic activity against $AFB_{1}$, and the fraction exhibited positive results of a characterized colour reactions of protein, carbohydrate and phenolic compound. Therefore, one of the possible active compounds from the kale juice was supposed to a glycoprotein(Mw. 270,000) which seemed unstable with heating.

Inhibitory Effect of Kale Juice on the Growth and DNA Incorporation of Human Cancer Cells

  • The inhibitory effects of kale juice on the growh and DNA incorporation of human cancer cells, using HT-29 colon cancer cells, MG-63 osteosarcoma cells, AGS gastric adenocarcinoma cells and K-562 leukemia cells, were studied. The growth of human cancer cells were inhibited in the presence of kale juice (10, 20 nd 40$\mu$l/ml) and the effects were the juice concentration- and incubation time-dependent up to 6 days. When 20$\mu$l/ml of kale juice was added to the media of HT-29, MG-63, AGS and K-562 cancer cells, the cell growth after 6 or 4 days of incubation was retarded by 83~95% of control group. Morphological changes of HT-29 colon cancer cells wre studied under inverted microscope. As the concentration of kale juice increased up to 20$\mu$l/ml, degree of cell aggregation was decreased. Moreover, the DNA incorporation o AGS gastric adenocarcinoma cells and MG-63 osteosarcoma cells which were labeled with [$^3$H] thymidine was significantly reduced after 2 days of incubation at 37$^{\circ}C$ with kale juice. Therefore, we concluded that kale juice strongly decreased the growth of various human cancer cells.

Gamma-Irradiation Provides Microbiological Protection While Maintaining Sensory Quality Change of Fresh Kale Juice During Storage

  • The effect of gamma-irradiation on microbiological growth in kale juice during storage was studied. Fresh kale juice was prepared and irradiated at 0, 1, 3, 5, 7, 10, and 15 kGy. D values for total bacteria, yeast and mold, Salmonella, E. coli, and Pseudomonas were 3.6, 4.0, 3.2, 1.4, and 1.6 kGy, respectively. E. coli and Pseudomonas were eliminated completely at 5 and 7 kGy, respectively. Gamma-irradiation also reduced total viable bacteria during storage. Therefore, these results indicate gamma-irradiation can prevent microbial spoilage of fresh kale juice by inactivating pathogenic microorganisms.

Nitrite Scavenging Effect of Methaol Fraction Obtained from Green Yellow Vegetable Juices (녹즙추출물의 아질산염 소거능에 대한 연구)

  • The objectives of this study were to investigate biological activities such as nitrite scavenging effect of water and methanol fractions obtained from each green juice(Angelica keiskei, kale, carrot) under different pH conditions, and antioxidant vitamin contents in green juice made of green yellow vegetables. The result shows that nitrite scavenging effect of each green juice was 14.0~91.2% in model system. The methanol fraction provided higher effect than the water fraction. The nitrite degradation was the highest at pH 1.2. Nitrite scavenging effect of methanol fraction obtained from kale juice was 91.2% at pH 1.2 which was the highest among tested samples in this experiments. Consequently, extracts of kale juice were shown to provide the greatest functional properties among the vegetables tested. Contents of vitamine E and C in the extracts of kale were higher than the others.

Therefore the aim of this work was to investigate for prevention in cardiovascular disease, Total cholesterol concentration in liver were significantly lower kale juice and sodium dextrothyroxine(1.25mg/kg diet) fed groups than the other groups and trigly-ceride concentration was significantly lower Ganodrma lucidum fed group than in control group;. Total cholesterol concentration in testis were lower other groups than in control group and triglyceride concentration were significantly lower cholesterol Ganoderma lucidum and kale juice groups in control group. Phospholipid concentration in testis was higher sodium dextroth-yroxine fed group thanin control group.

  • This study was investigated the effects of water soluble extract of Ganoderma lucidum kale juice and soidium dextrothyroxine on lipid components in serum of hypercholesterolmemic rats in vivo in order to prevent in cardiovascular disease. Total cholesterol concentrations in serum were significantly increased after feeding cholesterol diet group compared with control group and were significantly increased after feeding cholesterol diet group compared with control group and were lower in Ganoderma lucidum kale juice and sodium dextrothyrioxine diet groups than in control group. Ratio of high density lipoprotein cholesterol concentration to total cholesterol were significantly highest in sodium dextrothyroxine fed groups among diet groups and were higher in Ganodrma lucidum and kale juice fed group than in control group. Phospholipid concentrations in seurm were significantly lower in Ganoderma lucidum kale juice and sodium dextrothyroxine(1.25mg/kg diet) fed groups than in control group and triglyceride concentra-tions were lower in Ganderma lucidum sodium dextrothyroxine fed groups than in other groups. Triiodothyronine concentration in serum were lower in Ganoerma lucdum and kale juice fed groups than in the other groups while it was higher in sodium dextrothyroxine diet group than in other groups. Tetraiodothyronine concentrations is serum were remakably higher in sodium dextrothyroxine fed group than in other groups. Blood glucose concentration was lower in cholesterol diet group than in other groups but was higher in sodium dextrothyroxine diet group than in other groups.

  • BACKGROUND/OBJECTIVES: Glutathione s-transferase (GST) is involved in the formation of a multigene family comprising phase II detoxification enzymes, involved in the detoxification of reactive oxygen species. This study evaluated whether daily supplementation with kale juice could modulate levels of plasma antioxidant vitamins and oxidative stress-related parameters. We further examined whether this modulation was affected by combined GSTM1 and T1 polymorphisms. SUBJECTS/METHODS: Totally, 84 subclinical hypertensive patients having systolic blood pressure (BP) over 130 mmHg or diastolic BP over 85 mmHg, received 300 mL of kale juice daily for 6 weeks. Blood samples were drawn before start of study and after completion of 6 weeks. RESULTS: After supplementation, we observed significant decrease in DNA damage and increase in erythrocyte catalase activity in all genotypes. Plasma level of vitamin C was significantly increased in the wild/null and double null genotypes. The plasma levels of ${\beta}-carotene$, erythrocyte glutathione peroxidase activity, and nitric oxide were increased only in the wild/null genotype after kale juice supplementation. CONCLUSIONS: The effect of kale juice was significantly greater in the GSTM1 null genotype and wild/null genotype groups, suggesting possibility of personalized nutritional prescriptions based on personal genetics.

Volatile Sulfur Compounds, Proximate Components, Minerals, Vitamin C Content and Sensory Characteristics of the Juices of Kale and Broccoli Leaves (케일 및 브로콜리잎즙의 함황 향기성분, 일반성분, 무기질, Vitamin C 함량 및 관능적 특성)

  • To utilize the leaves of broccoli, the shape of which was similar with that of kale leaves, as a vegetable juice, the extracts from two types of vegetable leaves were subjected to the analyses of proximate com ponents, volatile sulfur compounds, vitamin C and minerals. The sensory evaluation of the juices, prepared from kale and broccoli leaves, were performed by duo trio test and scoring test. Among eight varieties of broccoli, four varieties('Pilgrim', 'Greenbelt', 'Salinas' and 'Shasta')were not significantly different from kale(p<0.05), based on the duo trio sensory test. Sulfur and nitrogen compounds present in the dichloromethane extracts of kale and broccoli leaves 'Pilgrim' were identified as 3 butenyl, allyl, butyl and 4 methylsulfinylbutyl isothiocyanate(sulforaphane), 4,5 epithiovalero, 4,5 epithio 3 hydroxyvalero, benzenepropane, 3 hydroxy 4 propene, 4 methylthio butane, 3 hydroxy 3 phenylpropane, 5 methy lsulfinylpentane, 4 methoxyphenyl 3 hydroxypropane nitrile and dimethyl trisulfide by GC/MSD analysis. Proximate components were observed to be relatively similar between kale and broccoli leaves. Vitamin C and sugar content were higher in broccoli leaves(125∼180mg% and 8∼12°Brix) than in kale(101mg% and 7oBrix). In mineral analysis by AA and ICP Mass, 'Pilgrim' showed a higher content of K, Fe, Mn, Zn and Se. In separate experiment, sensory scores of bitter taste and astringent taste were lower and scores of over all taste and over all acceptability, higher in broccoli('1243') juice than those in kale juice.

Analysis of Antioxidant Nutrients in Green Yellow Vegetable Juice (녹즙의 항산화 영양성분 분석)

  • The present study was attempted to investigate the contents of antioxidant nutrients in green juice of Angelica keiskei, kale, carrot, celery, cucumber. All 5 green juice were found to contain antioxidant nutrients such as ${\beta}-carotene$, vitamin E, vitamin C, Se, Cu, Mn, Zn and phenols. ${\beta}-carotene$ contents of carrot, kale, Angelica keiskei, celery and cucumber juice were 5909.0, 3285.3, 1398.4, 176.7 and 61.4 mg/l00 g, respectively. ${\alpha}-tocopherol$ contents of 5 green juices were $0.04{\sim}1.41$ mg/l00 g. Kale juice had highest ${\alpha}-tocopherol$ and celery juice contained lowest, kale juice also contained highest vitamin C (105.1 mg/l00 g) and Angelica keiskei, cucumber, celery and carrot had vitamin C contents of 31.4, 9.3, 7.5 and 5.2 mg/l00 g, respectively. Angelica keiskei had highest amount of antioxidant minerals among the sample such as Cu (0.40 mg/l00 g), Mn (0.5 mg/l00 g), Zn (0.24 mg/l00 g), Se $(0.61\;{\mu}g/kg)$. Kale was the second best source of antioxidant minerals which were Cu (0.45 mg/l00 g), Mn (0.22 mg/l00 g), Zn (0.24 mg/l00 g), Se $(0.33\;{\mu}g/kg)$. However, celery and cucumber were poor in antioxidant minerals. Total phenolic contents of 5 green juices were $0.01\;{\sim}0.05%$, kale juice also had highest total phenolics among the samples.

Study on the Content of ${NO_3}^-$ in Green Vegetable Juice by Different Sorts, Harvesting Time, Mixing Rate of Vegetable, Storage Condition and Manufacturers (채소 모재료의 종류, 수확시기별, 부위별 혼합비율, 저장조건 및 생산회사에 따른 녹즙의 ${NO_3}^-$ 함량차이)

  • After the consumption of green vegetable juice by Korean increase rapidly, the ${NO_3}^-$ intake through green vegetable juice have been ignored to consider for the calculation of daily ${NO_3}^-$ intake. It is necessary to collect the basic data on the ${NO_3}^-$ content in green vegetable juice by different sorts, harvesting time and mixing rate of vegetable, manufacturers and storage conditions for the next calculation of daily ${NO_3}^-$ intake for Korean. Followings are the research results from monitoring and laboratory experiment related with ${NO_3}^-$ and vitamine C in green vegetable juice. The ${NO_3}^-$ content of angelica plaant(tomorrow's leaf) and kale were higher in spring than those in summer and autumm. The highest value of ${NO_3}^-$ content in tomorrow's leaf and kale were 4.85 and 2.94 times higher compare to the lowest value. The average ${NO_3}^-$ content in the midribs of tomorrow's leaf and kale were 7.5 and 2.1 times higher than those in leafblades. It indicate the green vegetable juice made from leadblade of tomorrow's leaf and kale might be better compare to those from midrib in terms of ${NO_3}^-$ content. The content of ${NO_3}^-$ and vitamine C as affected by the timecourse after juice making were decreased rapidly compare to those by storage temperature in case of carrot, kale and cucumber juice. It show the positive comelation between the content of ${NO_3}^-$ and vitamine C in carrot, kale and cucumber juice regardless of room temperature(${NO_3}^-$) or cold temperature(${NO_3}^-$). The content of ${NO_3}^-$ and vitamine C of green vegetable juice by P company were the highest among the manufactuers. The lower content of ${NO_3}^-$ and vitamine C of green vegecable juice by TW company and GB company compare to P company is due to dilution with water to produce the juice. The content of ${NO_3}^-$ of green vegetable juice which were available in market showed 143ppm in carrot juice, 506ppm in tomorrow's leaf juice, 669ppm in wild water celery juice, 985ppm in kale juice, whereas the content of vitamine C were 43ppm in carrot juice, 289ppm in wild water celery juice, 353ppm in kale juice and 768ppm in tomorrow's leaf juice. It was calculated that people take 253mg by tomorrow's leaf juice, 335mg by wild water celery juice, 483mg by kale juice if they drink 500ml of green vegetable juice perday, and it suggest to excess 1.16, 1.53 and 2.21 times respectively only by green vegetable juice consumption.
