Inhibition Effect of Zizania latifolia on Apoptosis Induced by $H_2O_2$ in Neuro2A Cell ($H_2O_2$로 유발된 Neuro2A 신경세포고사에 대한 줄풀의 억제 효과)

  • The purpose of this study was to examine the inhibition effect of Zizania latifolia that has been used heart disease, Diabetes Mellitus and Skin disease for a long time on apoptosis induced by $H_2O_2$ in Neuro2A cell. Neuro2A cells were cultivated in RPMI(GibcoBRL) with $5\%$, FBS and treated with $H_2O_2$, and Zizania latifolia. We measured the cell viability and analyzed DNA fragmentation. Activity of PARP, Cytochrome C, caspase-9, caspase-3, p53, p21, Bax and Bcl-2 in the cell was examined by using western blot. The cell viability in Zizania latifolia treatment (60ug/ml<) decreased significantly compared with that of none treatment. (p<0.001) Zizania latifolia increased cell viability about twice as much as that being injury by $H_2O_2$. (Zizania Latifolia 20ug/ml, $H_2O_2$ 200uM, P<0.001) DNA fragmentation developed by $H_2O_2$, but was not developed in Zizania latifolia treatment. PARP, Cytochrome C, caspase-9 and caspase-3 activated all by $H_2O_2$ but were not activated in Zizania latifolia treatment.. P53, P2l and Bu activated by $H_2O_2$, and Bcl-2 got into inactivation. But the opposite results appeared in Zizania latifolia treatment. In conclusion, these results suggest that Zizania latifolia inhibit the development of DNA fragmentation and apoptosis by $H_2O_2$ and the antioxidant action of Zizania latifolia is effective. More researches about effect of Zizania latifolia are considered to need.

  • To prevent human body injury from oxidative stress, antioxidants are very important and many research about antioxidants are generally being conducted. Hydrogen peroxide($H_2O_2$) that is one of vitality oxygen species has been seen that cause various diseases, DNA damage and gene change. Comparative Study on the Inhibition Effect on Apoptosis in Neuro2A Cell on the Region of Zizania Latifolia(Radix, Rhizoma, Herba) (고장초의 부위별(뿌리, 줄기, 전초) Neuro2A 신경세포고사에 대한 억제 효과 비교 연구)

And the purpose of this study was that we made a comparative study on the inhibition effect of apoptosis in Neuro2A cell on the region of Zizania latifolia(Radix, Rhizoma, Herba). Neuro2A cells were cultivated in RPMI(GibcoBRL) with 5% FBS and treated with $H_2O_2$ and Zizania latifolia(Radix, Rhizoma, Herba). Separately we measured the cell viability and analyzed DNA fragmentation. Activity of PARP, Cytochrome C, caspase-9, caspase-3, p53, p21, Bax and Bcl-2 in the cell was examined by using western blot. The results obtained were as Follows: The cell viability in all of Zizania latifolia (Radix, Rhizoma, Herba) treatment (60ug/m1<) decreased significantly compared with that of none treatment(p<0.001). Zizania latifolia Radix increased cell viability was most effective of three regions. But we had no significant difference among three regions. All of Zizania latifolia (Radix, Rhizoma, Herba) increased cell viability about twice as much as that being injury by $H_2O_2$,(Zizania Latifolia (Radix, nhizoma, Herba) 20ug/m1, $H_2O_2$ 200uM, p<0.001). DNA fragmentation developed by $H_2O_2$, but was not developed in all of Zizania latifolia (Radix, Rhizoma, Herba) treatment. Anti-nociceptive and Anti-inflammatory Properties of Ilex latifolia and its Active Component, 3,5-Di-caffeoyl Quinic Acid Methyl Ester

  • The present study was conducted to investigate anti-nociceptive and anti-inflammatory effects of the leaves of Ilex latifolia Thunb (I. latifolia) in in vivo and in vitro. Writhing responses induced by acetic acid and formalin- and thermal stimuli (tail flick and hot plate tests)-induced pain responses for nociception were evaluated in mice. I. latifolia (50 - 200 mg/kg, p.o.) and ibuprofen (100 mg/kg, p.o.), a positive non-steroidal anti-inflammatory drug (NSAID), inhibited the acetic acid-induced writhing response and the second phase response (peripheral inflammatory response) in the formalin test, but did not protect against thermal nociception and the first phase response (central response) in the formalin test. These results show that I. latifolia has a significant anti-nociceptive effect that appears to be peripheral, but not central. Additionally, I. latifolia (50 and $100{\mu}g/mL$) and 3,5-di-caffeoyl quinic acid methyl ester ($5{\mu}M$) isolated from I. latifolia as an active compound significantly inhibited LPS-induced NO production and mRNA expression of the pro-inflammatory mediators, iNOS and COX-2, and the pro-inflammatory cytokines, IL-6 and $IL-1{\beta}$, in RAW 264.7 macrophages. These results suggest that I. latifolia can produce antinociceptive effects peripherally, but not centrally, via anti-inflammatory activity and supports a possible use of I. latifolia to treat pain and inflammation.

Comparative Analysis of Anti-oxidative, Anti-inflammatory, Anti-allergy, and Whitening Effects of Different Solvent Extracts from Zizania latifolia (고장초 추출 용매의 에탄올 함량에 따른 항산화, 항염증, 항알러지, 미백 활성 비교 분석)

  • This study was performed to evaluate the anti-oxidative, anti-inflammatory, anti-allergy, and whitening effects of Zizania latifolia ethanol extracts prepared from 5 different ethanol concentrations (10, 30, 50, 70, and 90%). As the ethanol concentration in the extraction solvent was increased, the radical scavenging activities also increased. The inhibitory activity of Z. latifolia ethanol extracts on nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells tended to increase as the content of ethanol increased. The highest inhibitory activity was obtained with 70% ethanol extract. The antiallergy effects of Z. latifolia ethanol extracts were tested by measuring the release of ${\beta}-hexosaminidase$ in IgE-sensitized RBL-2H3 cells. The suppressive effect of Z. latifolia ethanol extracts increased in a dose-dependent manner as the proportion of ethanol increased, except for the 10% ethanol extract. Furthermore, the inhibitory effects of Z. latifolia ethanol extracts against melanin production in ${\alpha}-melanocyte$ stimulated hormone (MSH)-stimulated B16F0 cells increased as the ethanol ratio increased, and 70 and 90% ethanol extracts showed similar inhibitory activities to arbutin, a positive control, at $250{\mu}m$. The present study confirmed the efficacy of Z. latifolia ethanol extracts in various areas, demonstrating antioxidative, anti-inflammation, antiallergy, skin protective, and skin whitening effects, with no cytotoxicity. It could be used as a raw material in functional foods, as well as in cosmetics.

Dyeing of Silk Fabrics Using Zizania latifolia Turcz. Extract (줄풀을 활용한 견직물의 염색)

  • The purpose of this study is to investigate the proper dyeing condition, color fastness and functionality of silk fabrics dyed with Zizania latifolia Turcz. FT-IR spectrum analysis suggests that colorants of Zizania latifolia Turcz. are tannins. The dyeing of the silk fabrics using Zizania latifolia Turcz. was very good even without pretreatment or mordanting treatment. Optimal dyeing conditions of silk fabrics were colorant concentration of 200%(o.w.f.), dyeing temperature of $100^{\circ}C$, dyeing time of 60 minutes and dyebath pH of 5.5. Color fastness of dyed silk fabrics to washing, rubbing, perspiration and light was 4~4-5, 5, 4~4-5(acidic), 4-5(alkaline) and 2 respectively. UV protection rate and deodorization rate of silk fabrics dyed with Zizania latifolia Turcz. were improved. Reduction rate(Staphyloccus aureus) of silk fabrics was excellent at 98.3%. As a result, it was confirmed that the Zizania latifolia Turcz. could be used as an eco-friendly functional natural dye.

  • Zizania latifola has used for folk medicine in Korea. It is effective in treating skin trouble and sensitivity skins. The purpose of this study was to develop high valuable types of bath preparation of Zizania latifolia. The natural materials were manufactured by the mixed ratio of Zizania latifolia. Product was manufactured by the mixed ratio of Zizania latifolia and 3 natural materials. Green tea was highest in sensory evaluation as similar with Zizania latifolia. optimum condition of mixed ratio was adaptive 50% Houttuynia cordata, 30% green tea with 20% Artemisia vularis. Product's sensory evaluation was showed best in mixing 40% and Zizania latifolia extract 60%. There were optimal condition for manufacturing of cleansing. Therefore, The development of a bath preparation used by Zizania latifolia is very useful for manufacturing cleansing. We suggest that it help for improve skin trouble and make healthy skin.

The Purification Capacity of Zizania latifolia on Wetlands of Munpyeong Stream

  • This study examined the changes of water quality in relation to distribution of hydrophytes, and the purification capacity of Zizania latifolia to improve the effluent from Munpyeong stream from March 1997 to December 1999. While the concentration of nitrogen and phosphorous in water were increased during the farming season, those decreased, during the streaming down to paddy and drainage areas. In investigated sites, the Z. latifolia was dominant community according to the development of the natural wetlands. Furthermore, it formed a large community owing to its high adaptability to environmental changes in the agriculture lands. In September, the leaves productivity of the Z. latifolia were 4,032g D.W/$m^2$and roots were 7,680gD.W/$m^2$. The purification capacity of the Z. latifolia for NH$_3$-N, $No_3$-N, and PO$_4$-P were 13.41, 17.07, and 4.58 respectively during 5 days. The results suggested that it needs to establish wetlands vegetated by hydrophytes to improve the water quality of the effluent from agricultural lands.

Change in Community Composition and Soil Carbon Stock Along Transitional Boundary in a Sub-Tropical Forest of Garhwal Himalaya

  • The aim of the present study was to assess the effect of transitional boundary on community composition and soil carbon stock. Five vegetation types were recognized horizontally along the transitional strip based on the dominance of tree species i.e., Pure Anogeissus latifolia forest (P.AL), mixed Pinus roxburghii and Lannea coromandelica forest (M.PR&LC), pure Pinus roxburghii forest (P.PR), mixed Pinus roxburghii and Lannea coromandelica (M.PR&LC) and pure Anogeissus latifolia forest (P.AL). The results revealed that Anogeissus latifolia was reported dominant tree in the outer transitional boundaries of the forest, which reduced dominance of trees towards middle where Pinus roxburghii was found dominant. The soil carbon stock was reported higher in the Anogeissus latifolia dominant forest and reduced with the dominance of Pinus roxburghii in the middle site. Both the species are growing close to one another and competing for survival, but the aggressive nature of Anogeissus latifolia particular in this region may change new growth of Pinus roxburghii and will enhance soil carbon stock. But high anthropogenic pressure on Anogeissus latifolia tree species could be limited chance to further its flourish.

Taxonomic position of Taxus cuspidata var. latifolia endemic to Ulleung Island (울릉도 회솔나무(Taxus cuspidata var. latifolia)의 분류학적 위치)

  • The purpose of this study is to review the taxonomic position of Taxus cuspidata var. latifolia endemic to Ulleung Island with related taxa T. cuspidata var. cuspidata, T. caespitosa, and T. cuspidata var. nana based on external morphological characters and DNA barcoding study. T. cuspidata var. latifolia was similar to T. cuspidata var. cuspidata in the arbor, straight trunk, and symmetric arrangement of leaf. But the unique differences between T. cuspidata var. latifolia and T. cuspidata var. cuspidata were leaf size and the exposure of seed from aril. Additionally, sequences of four chloroplast DNA regions including matK, rbcL, trnL intron and trnL-trnF spacer regions were analyzed. Korean Taxus species and their related taxon T. cuspidata var. nana were strongly supported as a monophyletic group in neighbor-joining analysis. Taxus cuspidata var. latifolia showed 100% sequence identity to related taxa. Korean endemic T. caespitosa is also distinguishable from related taxa by prostrate stems and spiral arrangement of leaf. The examinations of external morphology and DNA barcoding study suggest that the taxonomic position of T. cuspidata var. latifolia should be maintained as a variety of T. cuspidata.
